Influence of oestrogen and androgen on modelling of the mandibular condylar bone in ovariectomized and orchiectomized growing mice.

Oestrogen and androgen exert a substantial influence on bone metabolism, but any differences in their influence on modelling of the condyle, a mandibular growth site, have not been fully clarified. The purpose here was to examine histological and histochemical differences in the condyle of ovariectomized (OVX) or orchiectomized (ORX) mice given injections of oestrogen (E(2), 17 beta-oestradiol) or non-aromatizable androgen (DHT, 5 alpha-dihydrotestosterone). Eight-week-old C57BL/6J mice (n=170) were used: they were divided equally into six experimental groups (OVX, ORX, OVX+E(2), ORX+E(2), OVX+DHT, ORX+DHT), and non-treatment male and female control groups. In each experimental group, five mice were killed 2,4,8 and 12 weeks after OVX and ORX. Oestrogen or androgen were given daily after the surgery by subcutaneous injection of E(2) or DHT. Increases in the number of tartrate-resistant acid phosphatase-positive cells induced in the OVX and ORX mice from 4 to 12 weeks after surgery were obviously suppressed by E(2) and DHT. The trabecular bone volume in the OVX and ORX mice treated with DHT had only increased at 12 weeks after surgery, whereas the E(2) injected mice exhibited a substantial increase from 4 to 12 weeks after surgery. E(2) injected into the OVX and ORX mice increased the trabecular bone volume earlier than did DHT, and both E(2) and DHT suppressed osteoclast differentiation similarly during the same period. These results suggest that metabolic responses of osteoclasts and osteoblasts to E(2) and DHT may be different, producing somewhat different patterns of bone modelling in males and females.

A histopathological study of the role of periodontal ligament tissue in root resorption in the rat.

Whether periodontal ligament (PDL) tissue is capable of inducing root resorption was examined. The distal root of the rat molar was sectioned at the furcation and the PDL tissue removed from the root (non-PDL group, n=40). The distal root with the PDL intact was also prepared (PDL-intact group, n=40). The roots were transplanted into the dorsal skin of the rat. On the 1st, 3rd, 5th, 7th, 10th, 14th, 21st or 28th day after transplantation, the roots were removed together with surrounding dorsal subcutaneous tissue and were fixed, demineralized and embedded in paraffin. Serial sections from each block were stained with haematoxylin and eosin or by the tartrate-resistant acid phosphatase (TRAP) method to observe root-resorbing cell formation. Cyclo-oxygenase-2 (COX2) was also detected immunohistologically to examine prostaglandin E(2) production. On the 7th day after transplantation, multinucleated root-resorbing cells with TRAP were observed in the PDL-intact group. The number of TRAP-positive cells peaked on the 10th day after transplantation. COX2-positive cells were observed in PDL during the early experimental stages. No root resorption was seen in the non-PDL group. These results suggest that PDL tissue is involved in the formation of root-resorbing cells and root resorption.

Evaluation of rapid methods for the determination of okadaic acid in mussels.

AIMS: Two different screening methods, a Buffalo Green Monkey cytotoxicity test and a biosensor test, have been considered to replace the official mouse bioassay in monitoring for okadaic acid (OA) levels in mussels. METHODS AND RESULTS: Diarrhoetic shellfish poison-contaminated mussels from the Adriatic Sea were assayed in parallel by means of the mouse bioassay and both alternative methods. Both the cytotoxicity test and the biosensor test showed high sensitivity (OA 0.01 mg g-1 hepatopancreas and 0.002 mg g-1 hepatopancreas, respectively) and a high correlation with the mouse bioassay (r=0.932, P < 0.001 and r=- 0.850, P < 0.001, respectively). CONCLUSION: Both methods are efficacious, quick, inexpensive and provide data on the amount of toxin present in mussels. SIGNIFICANCE AND IMPACT OF THE STUDY: Both methods, besides allowing the simultaneous assay of a great number of samples, comply with the ethical need to reduce the use of animals in the laboratory.
