[Computer-assisted identification of basic substances].

Abstract

The potential applications of a personal computer with integrated database software in toxicologic analysis are discussed. A toxicologic database including 370 basic compounds, is presented that was built up without knowledge of a higher programming language and was used as a base for drug identification in normal analysis by PC, DC and GC.

Comparison of conventional and fast gas chromatography in human plasma fatty acid determination.

A fast gas chromatographic technique for the determination of fatty acids in human plasma was developed. Its validation and comparison to a conventional method are here reported. The fast method significantly reduced the time required for analysis by a factor of 5 (total time of 3.2 min) while maintaining a similar resolution. Reproducibility of qualitative and quantitative data was measured in both applications. The results demonstrated that the applied fast gas chromatography (GC) conditions do not affect the analytical quality of the assays, allowing a short analysis time.

Isolation and identification of 2-methoxy-3,5-dimethylpyrazine, a potent musty compound from wine corks.

The compound responsible for a "fungal must" taint evident in industry assessments of wine corks was identified as 2-methoxy-3,5-dimethylpyrazine. The identification was made on the basis of gas chromatography/odor analyses, collection of material using micropreparative techniques, determination of chemical properties of collected material, and comparison by gas chromatography/mass spectrometry with an authentic sample, synthesized from 2-hydroxy-3,5-dimethylpyrazine. 2-Methoxy-3,5-dimethylpyrazine is an extremely potent compound with an unpleasant, musty, moldy aroma and an aroma threshold in a white wine of 2.1 ng/L. While its contribution to the frequency and intensity of cork taint in bottled wine is yet to be established, it has been assessed by some wine industry personnel as second only to 2,4,6-trichloroanisole as a cause of cork taint in Australian wine.

Quantification of solid cell material by detection of membrane-associated proteins and peptidoglycan.

Quantification of solid cell material (cell debris) is necessary for the optimisation of the efficiency of bioseparations. Cell debris can be quantified by detection of a component present in the cell wall that can act as a marker for cell debris. Membrane-associated proteins have previously been used as a marker for cell debris. This marker was quantified by SDS-PAGE with densiometry. In this paper cell debris quantification methods are presented that are faster and more accurate, i.e. membrane-associated protein quantification with the Protein 50 Labchip of Agilent Technologies, or that make use of peptidoglycan as marker for cell debris, i.e. a spectrophotometric muramic acid assay.
