Comparative study on serum iron determination by different methods.

Abstract

Iron is an important trace element in humans. Depletion or overload iron status can result in a number of aberrant physiological functions. Determination of serum iron is one of the valuable test panels for iron status assessment. A comparative study was performed on 3 different methods of serum iron determination, nonprecipitating ferrozine colorimetric method, precipitating ferrozine colorimetric method, and iron liquicolor cleaning factor colorimetric agar-based (CAB) method (Human, Taunusstein, Germany). The correlation coefficients between nonprecipitating ferrozine colorimetric method and precipitating ferrozine colorimetric method, between nonprecipitating ferrozine colorimetric method and iron liquicolor cleaning factor CAB method, and between precipitating ferrozine colorimetric method and iron liquicolor cleaning factor CAB method were found to be 0.998 (P = .067), 0.999 (P = .067), and 0.951 (P = .934), respectively. For a setting with limited resources such as Thailand, use of the first analytical technique, nonprecipitating ferrozine colorimetric method, is recommended because of its low cost and ease of performance.

Selective and sensitive colorimetric sensing of carbosulfan based on BiO2-x/Bi2O2.75 nanosheets with excellent haloperoxidase-like activity.

The development of colorimetric methods based on directly inhibiting nanozyme activity for pesticide detection has attracted considerable attention. In this study, we report a novel colorimetric sensing strategy utilizing BiO2-x/Bi2O2.75 nanosheets (BiO2-x/Bi2O2.75 NSs) with haloperoxidase (HPO)-like activity for the rapid and sensitive detection of carbosulfan (CBS) in foods. Oxygen-vacancy-rich BiO2-x/Bi2O2.75 NSs with HPO-like activity were rationally constructed. Kinetic studies revealed a remarkable Michaelis-Menten constant (Km) of 0.014 mM for I-, indicating a higher affinity for iodide ions than other reported HPO-like nanozymes, as evidenced by its lower Km. Under acidic conditions, CBS tends to be hydrolyzed to produce reductive sulfide species, which directly inhibit the iodoperoxidase-like activity of BiO2-x/Bi2O2.75 NSs, enabling selective detection with a limit of detection (LOD) of 0.18 μg/mL and a linear range of 0.20-100 μg/mL. When the concentration of interfering pesticides and substances was 5 times that of CBS, the sensor remained unaffected, exhibiting excellent stability and specificity. This work contributes to the detection of CBS in complex food matrices, bridging the application gap of HPO-like nanozymes in pesticide detection and providing a promising method for food safety detection.

Modification of the Stewart biphasic colorimetric assay for stable and accurate quantitative determination of Pluronic and Tetronic block copolymers for application in biological systems.

Block copolymers are increasingly being applied in areas such as transfection, membrane sealing, site-specific targeting, and bionanoengineering yet there is a relative paucity of assays available for simple, stable and reproducible colorimetric determination of copolymer concentration in solution. We have focused on improving the accuracy and reproducibility of a modified version of the Stewart biphasic colorimetric assay for quantitative determination of Pluronic (poloxamer) and Tetronic (poloxamine) macromolecules. The optimized assay achieved linear response ranges in chloroform for commonly used copolymers such as poloxamine 904 (20-300 microg/ml), poloxamine 908 (10-400 microg/ml), poloxamer 402 (20-400 microg/ml), and poloxamer 407 (10-400 microg/ml). Variation in the type of chlorinated solvent used significantly increased assay sensitivity, presumably through macromolecular reorientation, affording increased access for copolymer-ferrothiocyanate complexation. This was found to be optimally favored by the planar geometry of the solvent cis 1,2-dichloroethylene. For application to biological systems copolymer-protein interactions were for the first time determined and were found to be dependent on the fraction of hydrophobic constituents of the block copolymers and protein type. For instance serum albumin was found to interact with copolymers of low hydrophilic-lipophilic balance values and poly(propylene oxide) contaminants, whereas this interaction was not significant with the relatively hydrophilic IgG. In such systems the colorimetric assay directly determines the fraction of unbound (free) copolymer in the presence of proteins.
