Replication of Zika Virus in Human Prostate Cells: A Potential Source of Sexually Transmitted Virus.

BACKGROUND: While Zika virus (ZIKV) is mainly transmitted by mosquitoes, numerous cases of sexual transmission have been reported during recent outbreaks. Little is known about which host cell types or entry factors aid in mediating this sexual transmission. METHODS: In this study, we investigated ZIKV cell tropism by infecting 2 types of human prostate cells with 3 contemporary ZIKV isolates from persons infected in the Americas. We used real-time quantitative polymerase chain reaction and immunofluorescence analyses to measure infection and flow cytometry to detect entry factor expression. RESULTS: Here we show that ZIKV infects, replicates, and produces infectious virus in prostate stromal mesenchymal stem cells, epithelial cells, and organoids made with a combination of these cells. We also show that prostate cells express several well-characterized flavivirus attachment factors. In contrast, dengue virus does not infect or does not replicate in these prostate cells, although it is known to use similar receptors. CONCLUSIONS: Our results indicate that ZIKV favors infection of stromal cells more so than epithelial cells in organoids, possibly indicating a preference for stem cells in general. Overall, these results suggest that ZIKV replication occurs in the human prostate and can account for ZIKV secretion in semen, thus leading to sexual transmission.

Cadmium concentration in tobacco (Nicotiana tabacum L.) from different countries and its relationship with other elements.

Cadmium accumulation in crop plants, such as tobacco (Nicotiana tabacum L.), can lead to human exposure to this carcinogenic metal. To better define actual Cd distribution in cured or processed tobacco leaves from chief tobacco-producing regions, we analyzed 755 leaf samples of three major types (Flue-cured, Burley, and Oriental) obtained from 13 countries during 2001-2003. This survey may help identify regions with low- or high-Cd concentrations in tobacco to obtain insight into the cause of the concentration extremes and to assist in defining strategies to reduce Cd in tobacco. Cadmium concentrations in the samples ranged from 0 to 6.78 microg g(-1), as determined by ICP-MS. Significant differences were found among types and among countries, but significant interactions between type and country were found. Variations in Cd concentrations were also found in all countries. Our results suggest an important contribution of the field (e.g., bioavailable Cd in soil, other soil characteristics) to the Cd concentration in tobacco. Finally, the correlation between the concentration of Cd and that of other elements differed among the types, which could be effectively discriminated based on the concentrations of 20 elements.

The Spanish royal philanthropic expedition to bring smallpox vaccination to the New World and Asia in the 19th century.

The New World was ravaged by smallpox for several centuries after the Spanish conquest. Jenner's discovery of the smallpox vaccine made possible the prevention and control of smallpox epidemics. In response to a large outbreak of smallpox in the Spanish colonies, King Charles IV appointed Francisco Xavier de Balmis to lead an expedition that would introduce Jenner's vaccine to these colonies. During the journey, the vaccine was kept viable by passing it from arm to arm in orphaned children, who were brought along expressly for that purpose and remained under the care of the orphanage's director. This expedition was the first large scale mass vaccination of its kind. The historic legacy of this pioneering event in international health should be revisited in the current era of persistent inequalities in global health.
