Milk production, estimated phosphorus excretion, and bone characteristics of dairy cows fed different amounts of phosphorus for two or three years.

Diets containing 0.31, 0.39, or 0.47% P on a DM basis were fed to 10, 14, and 13 multiparous Holstein cows, respectively, for a full lactation. Most of the cows (33) were fed similar amounts of P in the previous one or two lactations. The objective was to obtain information on bone integrity after prolonged feeding of different amounts of P. At the end of the experiment, a section (-20 cm) of the 12th rib was surgically removed. The bone was tested for strength and analyzed for ash and P content. The shear strength and the energy required to deform the bone to the point of fracture did not differ among treatments. Bone specific gravities were 1.50, 1.57, and 1.55 for the three treatments. Ash and P content of the bone, measured in wet weight, dry weight, and wet bone volume, were similar for the 0.39 and 0.47% P treatments, but tended to be lower with the 0.31% P treatment. Milk production of cows in all groups was high, averaging >11,900 kg for the 308-d lactation. Feeding P at 0.31% of dietary DM over two to three lactations appeared to decrease P concentration of bone, but the decrease was not severe enough to affect bone strength. Dietary P at 0.39% did not affect bone P content or strength. Dietary P content of 0.31% appears to be borderline deficient for cows producing >11,900 kg/308 d.

Cracked dry or finely ground high moisture shelled corn as a supplement for grazing cows.

Feeding shelled corn as the primary supplement in cracked dry form or ground high moisture form to grazing cows was studied in an experiment using 32 cows in late lactation. Each of the two forms of corn accounted for 74.7% of the concentrate supplement, which was fed at 9 kg/d (DM). The experiment was 13 wk, during which cows grazed permanent native pasture containing mostly grasses and some clover. Milk yield was 2.4 kg/d higher for cows fed ground high moisture shelled corn than for those fed dry cracked shelled corn. Milk fat content was lower (3.28 vs. 3.67%), but protein content was higher (3.26 vs. 3.15%) for the high moisture corn supplement. Results suggest that high moisture corn can serve as a better supplement for grazing cows than dry corn, likely by providing more metabolizable energy and microbial protein due to more fermentable and digestible starch.

Investigation on the rheological properties of hepatocellular carcinoma cells and their relevance to cytoskeleton structure.

OBJECTIVE: To investigate the relevance of the rheological properties, i.e., viscoelasticities and adhesion to basement membrane components coated surface, of both hepatocytes and hepatocellular carcinoma (HCC) cells to the cytoskeleton structure. METHODS: Micropipette aspiration technique was adopted to measure viscoelastic coefficients and adhesion forces to 2 microg/ml collagen IV/1.25 microg/ml laminin coated surface of the cells. Two kinds of cytoskeleton perturbing agents, colchicine and cytochalasin D, were used to treat both HCC cells and hepatocytes and the effects of these treatments on cell viscoelastic coefficients and cell adhesion forces were investigated. RESULTS: Upon treatment of cells with colchicine in a concentration range of 1 to 60 mg/L, the elastic coefficients, especially the first elastic coefficient K1, and adhesion forces of hepatocytes generally tended to increase or increased significantly while, in contrast, viscoelastic coefficients and adhesion forces of HCC cells decreased obviously. Upon treatment of cells with cytochalasin D in a concentration range of 0.25-5.00 mg/L, viscoelastic coefficients of both hepatocytes and HCC cells decreased uniformly, with a larger magnitude for the decrease in elastic coefficients and adhesion forces of HCC cells than for those of hepatocytes. Adhesion forces of hepatocytes and HCC cells onto collagen IV/laminin coated surface varied similarly as viscoelastic coefficients under the action of the cytoskeleton perturbing agents. A significant positive correlation existed between changes of HCC cell adhesion forces on collagen IV/laminin coated surfaces and those of cell elastic coefficients (P<0.01). CONCLUSIONS: The effects of cochicine and cytochalasin D on rheological properties of HCC cells differed significantly either in ways or extents from those on rheological properties of hepatocytes. These results might reflect the difference in the state of cytoskeleton structure and function among these two kinds of cells.

[Application of time-frequency analysis on medical signal processing].

The frequency content of many biomedical signals can change rapidly with time. Conventional Fourier spectral analysis techniques are insufficient for analyzing the time-varying spectral content of these signals. By mapping a one-dimensional function of time(or frequency), the time-frequency representation can localize the signal energy in both the time and frequency directions. It has been shown that many biomedical signal problems may benefit from time-frequency analysis. The basic method of time-frequency analysis and its application in biomedical signals processing are introduced.

[Reduction of FHIT gene expression in primary lung cancer: relationship with the proliferation and apoptosis of tumor cell].

OBJECTIVE: To investigate the role of FHIT(fragile histidine triad) gene in oncogenesis and progression of human lung cancer and explore the relationship of FHIT gene expression with the proliferation and apoptosis of tumor cells. METHODS: The expression of FHIT gene and that of Ki-67 were detected in 166 lung cancer samples and 37 lung benign lesion tissues as control by immunohistochemistry, and the apoptotic level of tumor cell was detected by terminal deoxynucl neotidyl transferase mediated dUTP nick end labelling(TUNEL). RESULTS: (1)The expression level of FHIT gene was found to be significantly lower in lung cancer tissues than in benign lesion tissues (P<0.01); (2)The expression level of FHIT gene was closely related to histological classification, cancer cell differentiation, P-TNM stages and lymph node involvement in lung cancer patients (P<0.05); (3)Lasting and heavy smoking might be one of the important reasons of reduction of FHIT gene expression in lung cancer patients; (4)The postoperative survival time of patients in high FHIT expression group was significantly longer than those in low expression group (P<0.05); (5)The reduction of FHIT gene expression might be related with the over-proliferation and suppression of apoptosis of lung cancer cell(P<0.01). CONCLUSION: FHIT gene may play a role in the regulation of proliferation and apoptosis of lung cancer cell. And reduction of FHIT gene expression may be related to the pathogenesis and development of lung cancer.

[Trend of changes in mortality of cardiovascular diseases in some areas of Beijing during 1984 to 1998].

OBJECTIVE: To understand the main causes of deaths affecting population health and the trend of mortality of cardiovascular disease (CVD) in Beijing area during 1984 to 1998. METHODS: Monitoring data for cardiovascular diseases in some areas of Beijing during 1984 to 1998 were collected from MONICA project and used to analyze the distribution of disease, mortality caused by cardiovascular diseases and its age-adjusted mortality. Yearly average change in cardiovascular disease-specific mortality was calculated with regression analysis. RESULTS: Cardiovascular disease-specific mortality ranked the first place of the causes of deaths in Beijing population during 1984 to 1998, accounting for 46.0% of the total deaths. Cancer ranked the second, accounting for 26.5% of the total. Overall mortality, cardiovascular-specific mortality and cerebro-specific mortality all lowered significantly during the past 15 years, with yearly average reductions of 2.9%, 2.7% and 3.5%, respectively. Mortality of coronary heart disease significantly rose. CONCLUSIONS: Cardiovascular disease was the top cause of death in Beijing population during 1984 to 1998. Mortality of coronary heart disease significantly rose in the past 15 years. It is a pressing matter of the moment to prevent cardiovascular diseases in the population.

[The cytotoxic effects of the adriamycin magnetic albumin microspheres combined with external magnetic fields on the malignant tumor cells].

This study sought to assess the inhibitory effects of the adriamycin magnetic albumin microspheres (ADM-MAMs) on Walker-256 malignant tumor cells in vitro induced by the permanent magnetic fields. The cultured Walker-256 cells were divided into three groups; the group of ADM-MAMs combined with magnetic fields, the group of AMD-MAMs without magnetic fields, and ADM group. The growth states of the cells were observed and photographed under the inverted microscope. The inhibitory rates(IR) were assayed by the modified MTT colorimetric method. The results showed that the IR of the ADM-MAMs group were similar to those of the ADM group (P > 0.05), but the group of ADM-MAMs combined with magnetic fields had obviously higher IR and significant changes of the cells' shapes. These findings indicate that the anticancer effect of ADM-MAMs on malignant tumor cells is similar to that of ADM, and such effect can be increased by the combined use of ADM-MAMs and external magnetic fields.

[RGD peptides and endothelialization of biomaterials].

Endothelialization can remarkably improve blood compatibility of biomaterials and promote their clinical application, especially the uses of cardiovascular grafts. The attachment and growth of endothelial cells on artificial surfaces is the critical factor affecting the process of endothelialization. Many adhesive proteins contain the Arg-Gly-Asp(RGD) sequence which plays an important role in cell adhesion, migration and growth. In this review, the interaction of RGD peptides sequence and integrins, the influential factors of RGD peptides bioactivity and its application in endothelialization of biomaterials were discussed.

[Investigation on the mechanics of adhesion to the selective extracellular matrix coated surfaces of lung cancer cells].

The adhesion properties of tumor cells with extracellular matrix(ECM) are closely associated with their invasion and metastasis. Our work reported here was intended reveal the relevant biomechanical and biorheological manifestations of human lung cancer. Using micropipette aspiration technique, we investigated quantitatively the adhesive mechanics properties of high metastatic human giant cell carcinoma(PG) cells as well as low metastatic adenocarcinoma(PAa) cells of lung based on cell culture in vitro. The results showed that the adhesion forces of PAa and PG cells to collagen IV were significantly higher than those to glass surfaces, but at the lower concentrations(1.00 microgram/ml and 2.00 micrograms/ml) of collagen IV, the amplitude for the increase of adhesion forces of PG cells were less than the amplitude for that of PAa cells, and most of the adhesion force values of PAa cells to the coated surfaces of incorporation of laminin along with 2 micrograms/ml collagen IV were significantly greater than those of PG cells. At the lower concentrations(0.625 microgram/ml for PAa cells, and 0.625 microgram/ml, 1.25 micrograms/ml for PG cells) of laminin tested, the adhesion force values of PAa and PG cells all decreased, but the amplitude and level for the decreased values of adhesion forces of PG cells were greater than those for the PAa cells. In conclusion, the adhesive and proteolytic behaviour of cancer cells to extracellular matrix might be mediated mainly by tumor cell membrane receptors such as integrin receptors and laminin receptors, it might affect the biological characteristics and the metastasis of the tumor cells. The results may benefit to explain some questions in biomechanical views about how the highly metastatic PG cells are prone to migration and invasion.

[Wet FGD process on rotating-stream-tray tower with magnesium-enhanced lime].

Wet flue gas desulfurization(FGD) process with magnesium-enhanced lime slurry was conducted on a rotating-stream-tray tower. Changes of SO2 removal efficiency and slurry pH with time were experimentally determined. Effects of magnesium sulfate concentration were investigated and the lime utilization was calculated according to experimental data. In comparison with lime FGD process, adding magnesium sulfate could effectively enhance SO2 removal efficiency; when the adequate magnesium sulfate concentration was 0.2 mol/L, the removal efficiency was more than 60% with two rotating-stream-trays, while the removal efficiency of lime process was only 45%. To achieve higher SO2 removal efficiency, the slurry pH should be controlled between 6.0 and 7.5. Magnesium sulfate could also promote lime dissolution, slow down descent of slurry pH and increase lime utilization; when magnesium sulfate concentration was 0.2 mol/L, lime utilization could be increased by more than 5 percentage point under adequate conditions.

[Degradation of trichloroethylene by discharge plasma process].

Two types of discharge and two kinds of discharge reactors was employed to decompose trichloroethylene (TCE) in air. The results showed that both pulsed discharge and AC discharge were efficient to the degradation of TCE. Under the pulsed discharge with peak voltage of 42 kV, gas resident time of 15 s and TCE initial concentration of 1350 mg/m3, the degradation efficiency of TCE was close to 100% in the hallow reactor and the higher voltage results in a higher degradation efficiency. For the AC discharge, the degradation efficiency of TCE reached 90% with applied voltage of 30 kV. Meanwhile, increasing the applied frequency of AC voltage can enhance the degradation of TCE. The energy consumption for TCE degradation by AC discharge was higher than that by pulsed discharge. The energy consumption for TCE degradation efficiency of 80% by pulsed and AC discharge were 4.9 W.h/m3 and 116 W.h/m3, respectively. In addition, the energy consumption in the packed reactor was higher comparing with that in the hallow one.

Molecular cloning and characterization of a 21 kDa protein secreted from Trichinella pseudospiralis.

Recombinant protein was produced from the cDNA library of Trichinella pseudospiralis, which seemed to form part of the excretory-secretory (ES) products. The library was constructed from cDNA of muscle larvae at 1 month post-infection, and immunoscreened with antibody against T. pseudospiralis ES products. A clone, designated Tp21-3, contained a cDNA transcript of 657 bp in length with a single open reading frame, which encoded 172 amino acids (19617 Da in the estimated molecular mass). The predicted amino acid sequence of clone Tp21-3 had a similarity of 76% to that of clone ORF 17.20 (GenBank under accession number U88239) from T. spiralis. The recombinant fusion proteins encoded by clone Tp21-3 were produced in an Escherichia coli expression system and affinity purified. On Western blotting analysis, Tp21-3 recombinant proteins migrated at 40 kDa and reacted to antibody against T. pseudospiralis ES products and T. pseudospiralis-infected sera. Sera were developed against Tp 21-3 recombinant proteins, which reacted to a single band migrating at 21 kDa in crude worm extract and ES products from T. pseudospiralis on Western blotting analysis, and reacted with stichocytes of T. pseudospiralis on immunohistochemical staining.

[Mutations of mitochondrial 12S rRNA gene in type 2 diabetes].

OBJECTIVE: To investigate mutations at points 1310, 1438 and 1442 of mitochondrial 12S rRNA gene in Chinese type 2 diabetes and screen for new mutations associated with type 2 diabetes in this area. METHODS: The mitochondrial DNAs (mtDNAs) of 86 patients with type 2 diabetes and those of 70 normal controls were analyzed using PCR-SSCP and PCR product direct sequencing technique. RESULTS: One patient was found with the mitochondrial DNA G --> A mutation at point 1438, and one with the G two head right arrow A mutation at point 1442, whereas none of the normal controls was found to have mutation at these two points. No C --> T mutation at nucleotide pair 1310 of the mitochondrial 12S rRNA gene was identified. CONCLUSION: The G --> A mutation at points 1438 and 1442 is likely to be associated with the development of type 2 diabetes; the occurrence of the C -->T mutation at point 1310 is probably rare in the blood cells, and the development of type 2 diabetes may be related to heterogeneity in mitochondrial genetic changes.

[Adenovirus-mediated transfer of human wild-type p53, GM-CSF, and B7-1 genes efficiently produces growth suppression and apoptosis of hepatocellular cells in vitro].

OBJECTIVE: To investigate the tumor suppressor activity of recombinant adenovirus vector expressing the human wild-type p53, GM-CSF, and B7-1 proteins (designated as BB-102) in human hepatocellular carcinoma cells (HCC) in vitro. METHODS: The wild-type p53 BEL-7402, mutant p53 HLE, and HuH-7 HCC cell lines were infected with BB-102 at MOI of 50 in vitro. Immunohistochemical assay was used to determine p53 expressed by BB-102. Tumor suppressor activity of the expressed p53 was identified by terminal deoxynucleotidy I transferase (TdT) assay in BB-102-infected HCC cell lines. RESULTS: p53 protein was found to express in a dose-dependent manner in BB-102-infected HCC cell lines. The proliferation of HCC cell lines were suppressed significantly at the average rates of 58.5%, 81.5%, and 71.1% for BEL-7402, HLE, and HuH-7 respectively from 4 to 10 days, accompanying inducing apoptosis in BB-102-infected HCC cell lines. CONCLUSIONS: Besides the expression of B7-1 and GM-CSF, BB-102 is able to express p53 protein in independent manner and exerts its anti-tumor activity, which suggests that BB-102 may be useful for gene therapy against HCC in vivo.

[Evaluation on comprehensive benefits of different regulation models of agroforestry ecosystem in snail river beach].

The economic, ecological, social and comprehensive benefits of different regulation models of agroforestry ecosystem in Nangeng river beach were evaluated by AHP. The results showed that agroforestry ecosystem was significantly superior to reed(Phragmites communis) system. Different regulation models of agroforestry ecosystem had different benefits. The sheet-thinning model had the highest economic, social, and comprehensive benefits, and it was the optimal regulation model. For all thinning models, the key approach to further raise benefits was to choose some intercrop variations, which had a high economic value, especially a high ratio of output to input. For no-thinning model, keeping on intercropping was the effective measure to raise the social and comprehensive benefits.

[Application of laparoscopic technique in splenectomy].

OBJECTIVE: To investigate the application and effect of laparoscopic splenectomy (LS) in the treatment of spleen diseases. METHODS: Under CO2 pneumoperitoneum, laparoscopic splenectomy was performed on 43 patients. General conditions before and after operation, platelet counting, complications and recovery of patients were studied. RESULTS: Laparoscopic splenectomy was performed without complications in all the patients. In the patients with ITP, the effective rate of LS was 89.3%. The difficulty of operation was identified when the diameter of the spleen was over 15 cm assessed by ultrasonic examination, and the operative time was usually prolonged. CONCLUSIONS: LS is one of the best choices when proper indications exist and good techniques under laparoscopy are provided.

[New advances in research of combined pollution].

Combined pollution has become one of the important directions of environmental science at present. In this paper, new advances of studies on combined pollution were summarized. The studies of inorganic combined pollution, inorganic-organic combined pollution and organic combined pollution were reviewed. The mechanisms of combined pollution were approached from chemistry, physiology, enzymology, cytology and molecular biology. Then some problems and developing aspects on the research of combined pollution were put forward.

[The influence of hydraulic characteristics on wastewater purifying efficiency in vertical flow constructed wetlands].

The hydraulic characteristics of vertical flow constructed wetlands was investigated, and the influence of these characteristics on wastewater purifying efficiency was studied as an emphasis. The results of the experiments in small scale plots and medium scale plots showed the plant roots, with the physical and biological effects, play a significant role in hydraulics and the wastewater purifying efficiency. The other characteristics such as retention time, effluent velocity and influent load also directly affect the wastewater purifying efficiency of constructed wetland. To get a better understanding of the hydraulics will be crucial to the improvement of purifying efficiency and facilitate the practical use of constructed wetland.
