[Toxic studies on various processed products of Fructus Psoraleae].

Toxic studies on Fructus Psoraleae (FP) and its processed products are carried on. LD50 is 37.7 +/- 0.54 g crude drug/kg in oral administration of raw FP in comparison with 43.25 +/- 6.1/kg in that of Salt-bake one in mice. When 5.0 g/kg is given by oral for 14 days, the wet weight of testicles, preputial galnds, prostate and semianl vesicle is decreased in raw FP and its stir-bake tested groups. The wet weight of preputial glends, prostate, seminal vesicle in Lei Gong's Procession group, or prostate, and seminal vesicle in salt-steam group is also decreased to a certain extent. But no significant change on the weight of those sex organs is found in liquor-bake or salt-bake procession group, The glomerulal cell hyperplasia and cloudy swelling of renal tubules are induced by administration of raw FP in pathological examination.

[Comparision on the effects of ALI aeute liver injury of three species of concha haliotidis in mice].

The liver injury mice model with Haliotis ruber (Leach), Haliotis disus hannai Ino and Haliotis lacvigata (Donovan) was studied. The results indicated three species Haliotidis could versus carbon tetrachloride-induced ALI aeute liver injury in mice. Comparision of reduced serum glutamic-pyruvic transaminass (SGPT) activity showed Halitis disacus hannai Ino > Haliotis Lacvigata (Donovan) > Haliotis ruber (Leach). The Liver starch was determined. It is suggested protection liver effect of Haliotis discus hannai Ino and Haliotis lacvigata (Donovan) are most powerful.

[Experimental studies on the expansion of cord blood CD34+ cells and the improvement of their grafting efficiency].

OBJECTIVE: To elucidate the expanding response of cord blood CD34+ cells to human hematopoietic growth factors (HGFs) and evaluate their grafting efficiency. METHODS: CD34+ cells were isolated from umbilical cord blood by using a high-gradient magnetic cell sorting system (MACS), expanded with HGFs in a liquid culture system and evaluated of grafting efficiency with a panning assay on an adult bone marrow stroma. RESULTS: The percentage of CD34+ CD38- subset in cord blood was much higher than that in bone marrow or peripheral blood. Treatment with a combination of HGFs, including SCF, IL-3, IL-6, G-CSF,and Epo increased hematopoietic progenitor cells by nearly 100 fold and nucleated cells by more than 500 fold. 68.0%-89.6% of the CD34+ cells treated with HGFs (GM-CSF or IL-3, alone or in combination) seeded in the pre-irradiated marrow stroma, whereas only 36% of the untreated ones did. CONCLUSION: It is possible to expand cord blood CD34+ cells ex vivo and to improve their grafting efficiency with HGFs.

Characteristics of risk-taking behaviors, HIV and AIDS knowledge, and risk perception among young males in southwest China.

A cross-sectional survey was undertaken to describe risk-taking behaviors and to assess the knowledge and risk perception of HIV and AIDS among young males aged 18 to 29 years in 82 villages in Longchuan, Yunnan, China, in 1994. Information on demographic, behavioral, and drug-using factors, and knowledge of HIV transmission and prevention, and risk perception was collected using an interviewer-administered anonymous questionnaire. A total of 1,548 individuals were interviewed and 433 drug users, including 52 nonsharing injectors and 140 sharing injectors, were identified. Over half the individuals scored 0 on HIV knowledge, but knowledge was greater among nonsharing drug injectors. Most drug injectors had initiated drug injection after 1990. The reported incidence continues to increase in all three major ethnic groups. Sharing of equipment was common (73%) among injectors. Drug users were four times more likely to have had premarital or extramarital sex, but condoms were used by only 2.5%. Thus, factors promoting spreading of HIV are common in this area. We recommend that a community-based intervention program, targeting both young men and women, be implemented and evaluated in Longchuan as soon as possible.

Use of polymerase chain reaction in the diagnosis of toxocariasis: an experimental study.

In this paper we report the usefulness of polymerase chain reaction technique in the diagnosis of visceral larva migrans in a mouse model. Liver samples obtained from two set of experimentally infected mice (10, 100, 1,000 and 10,000 embryonated Toxocara canis eggs per mouse) along with the eggs of T. canis, T. cati and Ascaris suum were included in this study. Polymerase chain reaction (PCR) was performed using Toxocara primers (SB12). The first PCR product electrophoresis revealed very thin positive bands or no bands in liver samples. However, on second PCR a clear-cut bands were observed. No positive band was shown by A. suum eggs. Our findings thus indicate the usefulness of PCR technic in the diagnosis of visceral larva migrans (VLM) in liver biopsy materials specifically by means of double PCR using the primer SB12.

Quantitative analysis of light sensitivities in central 30-degree visual field of normal Chinese and glaucoma patients.

OBJECTIVE: To examine the difference in the light sensitivity in the central visual field between the normal subjects and patients with or without the early visual field damages and propose the criteria to identify the possible early disturbances in the visual field. METHODS: Light sensitivities at the 76 locations in the central field were measured in 108 normal eyes of 75 normal volunteers aged from 20 to 78 years, and 82 eyes of 53 patients with early open-angle or closure-angle glaucoma on the Humphrey Field Analyzer (Model 610), using the Central-30-2 program. RESULTS: No sex and eye differences were found. The significant differences in the light sensitivities existed between the age of 20-45 years and the age of 46-78 years in the normal subjects. The lower limit of 80% normal range for each location in the central field of the two age groups was calculated. The light sensitivity decreased by 2-3 dB at every 10 degree from the eccentricity. The light sensitivity in the inferior field was 1-2 dB higher than that in the superior field. There was no statistical difference between the nasal and the temporal field. For the patients with the field defects, the light sensitivity of many locations decreased by 4-8 dB, and by 1-2 dB in the patients without the field defects as compared with the normal controls. CONCLUSIONS: Based on comparisons with the normal controls and the lower limit of 80% normal range, we proposed the following criteria for classification of the damages in the glaucoma visual field. Grade I: light sensitivity reduces by 1-3 dB at a cluster of locations in the field; Grade II: light sensitivity reduces by 4-6 dB at a cluster of locations in the field; Grade II: light sensitivity reduces by more than 6 dB at a cluster of locations in the field.

[The role of NADPH in the development of neonatal jaundice with G6PD deficiency].

OBJECTIVE: To investigate the role of NADPH in the development of neonatal jaundice with G6PD deficiency. METHODS: The enzyme activities of G6PD, catalse (Cat) and glutathione peroxidase (GSH-px) were measured by quantitative determination of enzyme activity. The level of MDA was analyzed with alpha-thiobarbituric acid and the level of NADPH was determined with modified Nisselbaum JS's. Comparisons of these markers between G6PD normal and deficient erythrocytes were made before and during the incubation of the erythrocytes with H2O2. RESULTS: The level of MDA, which was 36 +/- 8n-mol.L-1.gHb-1, was increased and that of NADPH, which was 1792 +/- 106mumol.L-1.gHb-1, was decreased in jaundiced neonates with G6PD deficiency compared with those with normal G6PD activity. When the cells were incubated with H2O2, the level of NADPH and the activities of Cat and GSHpx in erythrocytes with normal G6PD activity increased at first, and then turned to decrease as the incubation lasted longer than 30 minutes. But in G6PD-deficient erythrocytes all these markers decreased continuously as the cells were incubated with H2O2. CONCLUSIONS: The diminished capability of generation of NADPH in G6PD-deficient erythrocytes may contribute directly to the more extensive peroxidation of the cells. The defect capacity of generation of NADPH, which resulted in the weakened capability of antiperoxidation and finally the lysis of erythrocytes, was one of the important mechanisms in the development of jaundice in G6PD-deficient neonates.

[Carotid body tumor (report of 4 cases)].

Four cases of carotid body tumor are reported, and the diagnosis and treatment are also discussed. We consider that color Doppler flow image and carotid angiography, especially digital subtraction angiography, are of great value to the diagnosis of carotid body tumor. In addition, the collateral cerebral cross-flow was promoted preoperatively with the use of compressing exercises of the carotid artery, and controlled by transcranial Doppler. This means to provide the basis for choosing the operative opportunity and for managing the carotid artery. The operative method was dependent on preoperative color Doppler flow image, carotid angiography, and relationship between the tumor and the carotid artery during operation.

[Studies on relationship between passive smoking and lung cancer in non-smoking women].

A population-based case-control study was conducted in Beijing, as part of Sino-Monica Project, to explore etiology of lung cancer in non-smoking women. Results showed that there was certain association between passive smoking and lung cancer in non-smoking women, with an OR of 2.52, P < 0.05, and its strength also increased with cigarette-years of smoking. A statistically significant risk of lung cancer was observed in non-smoking women, as their passive cigarette-years accumulated to more than 200. Adenocarcinoma of the lung was more possibly induced by environmental tobacco smoke (ETS) (OR = 2.32, P < 0.05), and it seemed to have little relation between ETS and squamous lung cancer (OR = 1.04, P < 0.05).

[Investigation on the viscoelastic properties of hepatocytes and their relavances to cytoskeleton structure].

Using micropipette aspiration technique, the authors investigated the viscoelastic properties of human fetal hepatocytes (HFH) and hepatocellular carcinoma (HCC) cells. And further, the effects of two cytoskeleton interferents, cytochalasin D and colchicine on the viscoelastic properties of HFHs were investigated. The results showed: the elastic coefficients K1 and K2 of HCC cells were significantly higher than those of HFHs; the action of cytochalasin D led to an obvious decrease in elastic coefficients K1, K2 as well as the viscous coefficient mu of HFHs; under the action of colchicine, the K1 value of HFHs increased while the K2 and mu values decreased obviously or tended to decrease. A brief discussion was presented about these results.

[Multiple factors analysis of formation of corneal haze after photorefractive keratectomy].

VISX 20/20 excimer laser was used to correct myopia from 1.00 to 16.00 diopters on 529 myopia eyes in 306 patients. All the patients were followed up for 3 to 12 months (8.1 +/- 3.1 months). At the 12th postoperative month, 0.5-3 grade corneal haze occurred in 62.1% of eyes in high myopia eyes, and only 8.8% in low and moderate myopia eyes (P < 0.01). The factors of formation of the corneal haze were the central corneal thickness, methods of removing epithelium and the number of laser pulses. The results suggest that the photorefractive keratectomy is regarded safe in correction of low and moderate myopia, but not in high myopia.
