Effect of angiotensin converting enzyme inhibitors on tolerance of nitroglycerin.

The aim of this study was to assess whether the angiotensin-converting enzyme inhibitors captopril and enalaprilat could change the tolerance of nitroglycerin in isolated rat aortae. In aortic rings precontracted with potassium chloride, captopril (1 microM) but not enalaprilat (0.1 microM) incubation potentiated the responses to nitroglycerin. It is suggested that captopril can reduce the tolerance of nitroglycerin because it is a sulfhydryl group donor.

[Association of plasma insulin with blood pressure and sodium sensitivity in children].

To study the relationship between plasma insulin level, sodium sensitivity and blood pressure, 150 children aged from 10 to 15 years with high blood pressure and 150 children matched for age and sex but with normal blood pressure were measured for plasma insulin with radioimmunoassay and sodium sensitivity with the tests of oral saline load and furosemide sodium-volume depletion and followed for 5 years. The results showed that (1) In children with HBP and positive family history of hypertension (FH+), the plasma insulin level was significantly higher than that in the respective control group (P < 0.01); (2) In children with high plasma insulin level, the percentiles of systolic blood pressure kept high and had a rising tendency during the follow-up period; (3) In children with sodium sensitivity, the plasma insulin level was higher than that in children with sodium resistance (P < 0.01) and the plasma insulin level correlated positively with blood pressure increasing rate of post-load (P < 0.05). It is suggested that insulin resistance was associated with sodium sensitivity, and they might play a role in the development of hypertension in childhood.

[Synovial fluid on non-synovial tendon: experimental study].

The non-synovial tendon was placed in the synovial cavity of rabbit knee joint to carry out tissue culture in vivo. The surface of the cultured tendon was covered with a smooth membrane-like tissue which possessed the morphologic characteristics of the synovial membrane identified by histological and electron microscopic examinations. The tendon segments were kept free in the cavity of knee joint and adhesions were not observed. It is suggested that non-synovial tendon could transform into a synovial tendon in the environment filled with synovial fluid.

[t(14:18) chromosomal translocation and over expression of bcl-2 protein in follicular type non-Hodgkin's lymphoma].

Polymerase chain reaction (PCR) and immunohistochemical staining methods were used to detect the bcl-2/IgH fusion gene [t(14:18)] and the overexpression of bcl-2 protein in 28 cases of follicular lymphomas and 18 cases of reactive follicular hyperplasia. The results showed that the bcl-2/IgH fusion gene was present in 57% of follicular lymphomas and that this translocation was also present in 33% of the reactive follicular hyperplasia cases. The over expression of bcl-2 protein was found in 78% of follicular lymphomas, but none in follicular hyperplasia, therefore bcl-2 protein staining may be a useful immunohistochemical method to differentiate follicular lymphoma and follicular hyperplasia.

[The differences in corneal shape between myopic and normal eyes].

The corneal shapes of 35 eyes with moderate and mild degrees of myopia compared with those of 35 normal eyes were studied by a computerized video keratoscope (TMS-1). The results show that the surface regularity index (SRI), the refractive power at central optical zone, the simulated keratoscopic reading (Simk), the average corneal power of zones 3mm, 5mm, 7mm in diameter centered at the corneal center of the eyes with moderate and mild degrees of myopia are greater than those of the normal eyes (P < 0.05). However, the surface asymmetry index (SAI), the differences of corneal powers between the first and the 15th ring and between the first and the 25th ring in both groups are of no statistical significant differences (P > 0.05). It is suggested that the corneal refractive power of myopic eye be much higher than that of the normal eye. The authors propose the first time that the pathogenesis of some eyes with moderate and mild myopia may be the increase of corneal refractive power which results from traction of sclera near the limbus due to accommodation and discuss it in detail.

Identification of 2-[125I] iodomelatonin binding sites in the thymus of mice and its significance.

The melatonin binding sites in membrane preparations of the mouse thymus were demonstrated using 2-[125I] iodomelatonin as a radioligand. The binding sites were stable, saturable, reversible and of high affinity. Studies on specificity of 2-[125I] iodomelatonin binding suggested that the 2-[125I] iodomelatonin binding sites are highly specific for melatonin. These binding sites fulfilled the standard criteria for receptors. Our work suggested that melatonin should have direct regulatory action on immune system mediated through the melatonin binding sites. Studies on the circadian rhythm showed that there existed the circadian rhythm in the binding capacity for 2-[125I] iodomelatonin in the mouse thymus with the peak values at 12:00-16:00 and the trough values between 00:00 and 4:00. The subcellular distribution of 2-[125I] iodomelatonin binding sites in the mouse thymus was in the following descending order: nuclear > mitochondrial > microsomal > cytosolic fraction. There was also an age-related decrease in 2-[125I] iodomelatonin binding in the mouse thymus. This is correlated with the involution of the thymus.

[The role of endothelin-1 and tumor necrosis factor-alpha in the development of asthma sarcoidosis].

In order to investigate the interaction of endothelin-1 (ET-1) and tumor necrosis factor-alpha (TNF-alpha) and their activity in the pathogenesis of asthma, ET-1 plasma and BALF levels were measured, and the amount of TNF-alpha released from alveolar macrophage (AM) was also tested. As in vitro the airway smooth muscle cells were stimulated with TNF-Alpha, the release of ET-1 was also observed. The results showed that the amount of ET-1, TNF-alpha were both increased than in the control group, at the same time, TNF-alpha could stimulate smooth muscle cells to secrete ET-1. We concluded that the release of ET-1 induced by TNF-alpha might play a certain role in development of airway hyperresponsiveness and airway remodeling.

[Determination of red-cell immunology in patients with laryngeal carcinoma].

In order to study the changes of the red-cell immunology, the rosette rates of the red cell C3b receptor (RBC-C3bRR) and red cell immune complex (RBC-ICR) were measured in 30 patients with laryngeal carcinoma. The results showed that: (1) the RBC-C3bRR and RBC-ICR were lower in patients with laryngeal carcinoma than those with polypus of the cord and the normal controls (P < 0.01); (2) the RBC-C3bRR and RBC-ICR of the patients with laryngeal carcinoma did not change in about twenty days after operation (P > 0.05). The results suggest that the red-cell immunological function is lowered in the patients with laryngeal carcinoma.

[Adherent mechanism of the mutans of Actinomyces viscosus T14V].

Three mutans of Actionmyces viscous T14V lacking type1 (5951), type 2 (5519) and both types of fimbriae (147) were selected in this study. We compared the ability of their adsorption to the test-tube wall, the result of the cellular coaggregation between these mutant strains and Streptococcus sanguis 34, Streptococcus sanguis black bova, Streptococcus mutans ingbritte, Streptococcus sobrinus 6715 and analysed the chemical characterization of cell walls of the three mutans. We found that the specific fimbriae and cell hydrophobicities had great effect on the bacteria adsorption or coaggregation.

[Functional rehabilitation of the urinary bladder after transection of cauda equina].

12 dogs were divided into three groups of L4 root crossover anastomosis S2 root, sacral root free anastomosis and control. All dogs received urodynamic test before and after operation and before sacrifice. The results of cystometry demonstrated the detrusor reflex had returned 7 months after operation in two groups of nerve renovation. The characteristics of axonal regeneration were revealed by immunohisto-cytochemistry and the count of nerve fibres numbers was compared with that by morphometric analysis in proximal and distal end of anastometric stoma. The comparison displayed a significant difference between nerve anastomosis groups and control group. Neural tracing study with horseradish peroxidase (HRP) in two dogs with the operation of L4 crossover S2 showed that lumbar somatic motor axons had regenerated successfully into the bladder and replaced previous sacral parasym-pathetic. Urethral sphincter function was still poor and urethral pressure was not changed post-operatively, though the electromyograme showed some improvement. The method of lumbar root or free sacral root anastomosis with sacral root is practicable for restoration of bladder function.

[Characterization of human coagulation factor IX cDNA expressed in cultured cells and in transgenic mice].

Two recombinant genes containing human coagulation factor IX cDNA driven by SV40 early promoter or by mouse MT promoter were introduced into the cultured mouse fabroblasts, respectively. It was found that both of them could be expressed in the cultured cells. Then, the two recombinant genes were microinjected into the male pronuclei of fertillized mouse eggs for generating transgenic mice harbouring the introduced genes, respectively. However, we did not observed that the two genes could be expressed in transgenic mice. These results suggest that the in vivo expression of human coagulation factor IX cDNA be controlled by some cis regulatory element (s).

[Comparison of MIVQUE and REML with Monte Carlo simulation].

A comparison of two methods for estimation of variance component, MIVQUE and REML, was carried out on four data sets with Monte Carlo simulation. The model used was the sire model containing herd-year-season(HYS) effect (fixed), sire group effect(fixed) and sire effect(random), which is widely used in dairy cattle breeding. The largest data set consisted of 12847 records with 47 sires and 778 HYSs, which is corresponding to the milk yield data available currently in Beijing area. The smallest data set comprised 200 records with 148 HYSs and 20 sires. The criterion for the comparison were the bias and variance, either theoretical or empirical based on 1000 repeated simulations, of the estimates. The results show that for the larger data sets the two methods are little different from each other: Bias < 1% of the true values, correlation approximately 1, and variance (MIVQUE) approximately variance (REML). For the smaller data sets the MIVQUE was significantly better than REML. It was also shown that for REML the sample size like the first data set can satisfy its large sample properties of asymptotical unbiasedness and efficiency.

[Detection of herpes viral genome in corneal buttons of quiescet herpes simplex keratitis with polymerase chain reaction].

PURPOSE: Herpetic keratitis is one of the major cause of blindness. The recent reports have proposed that the herpetic viruses may persist in cornea but provided noconclusive evidence. This study is to determine whether the cornea is another latent site of herpes simplex virus. METHODS: 18 corneal buttons of patients with quiescent herpes simplex keratitis which had quited for at least three months (mean 3.7 years) and were obtained by corneal transplantation were detected using polymerase chain reaction (PCR). The sequences of the primers are: P15'CATCACCGACCCGGAGAGGGAC, P25'GGGCCAGGCGCTTGTTGGTGTA. RESULTS: The positive HSV DNA were found in 17 corneal buttons out of 18 cases. CONCLUSION: The HSV genomes are present in the cornea of quiescent herpetic keratitis, which suggest that the cornea may be another latent site of herpetic virus. The PCR is a simple, specific and sensitive way to detect HSV genomes in cornea.

Benzimidazoles, potent anti-mitotic drugs: substrates for the P-glycoprotein transporter in multidrug-resistant cells.

P-glycoprotein is though to mediate the energy-dependent efflux of many structurally and functionally unrelated lipophilic compounds. Presently, the molecular mechanism underlying the binding and efflux of drugs by P-glycoprotein is not well understood. However, it has been suggested that two planar benzene ring structures and a cationic charge are commonly found in many drugs that interact with P-glycoprotein. The benzimidazoles (BZs) are potent anti-tumour, anti-fungal and anti-parasitic agents, whose mode of action is thought to result from their inhibition of microtubule functions. Although other classes of microtubule inhibitors, such as colchicine and vinblastine, have been studied extensively with respect to their interaction and efflux by P-glycoprotein, the BZ group of drugs has not been characterized. In this study, we have characterized the interaction of BZ with multidrug-resistant cells and found that resistant cells accumulated substantially less BZ compared with drug-sensitive cells. Furthermore, BZ was more toxic to sensitive than to drug-resistant cells, suggesting that BZ is likely to be a substrate for the P-glycoprotein drug efflux pump. In addition, we used a photoactive analogue of BZ ([125I]ASA-BZ) to demonstrate a direct binding between BZ and P-glycoprotein. Results showing that a molar excess of vinblastine, unmodified BZ, verapamil and rhodamine 123, but not colchicine, inhibited the photoaffinity labelling of P-glycoprotein by [125I]ASA-BZ confirmed the binding specificity of BZ to P-glycoprotein. Protease digestion of [125I]ASA-BZ photoaffinity labelled P-glycoprotein yielded two peptides that were similar to those obtained with other P-glycoprotein-associated drugs, e.g. azidopine and iodoaryl azidoprazosin. Taken together, these results demonstrate a direct and specific interaction between P-glycoprotein and BZ in a manner that is probably similar to other previously characterized P-glycoprotein-associated drugs.

Genomic organization and DNA sequence of the human catecholamine-sulfating phenol sulfotransferase gene (STM).

The human monoamine neurotransmitter-preferring phenol sulfotransferase (M-PST) plays an essential role in the sulfation of catecholamines, such as dopamine. The cDNA encoding M-PST has been reported, and we have recently identified cosmid clones from human chromosome 16p11.2 for this gene, STM. Plasmid subclones derived from the STM cosmid clones were subjected to dideoxynucleotide chain termination sequencing to determine the genomic organization and DNA sequence of STM. The gene encoding full-length STM is approximately 6.4 kb and contains 8 exons and 7 introns.

Loss of the imprinted IGF2/cation-independent mannose 6-phosphate receptor results in fetal overgrowth and perinatal lethality.

Murine embryos that inherit a nonfunctional insulin-like growth factor-II/cation-independent mannose 6-phosphate receptor (Igf2r) gene from their fathers are viable and develop normally into adults. However, the majority of mice inheriting the same mutated allele from their mothers die around birth, as a consequence of major cardiac abnormalities. These mice do not express IGF2R in their tissues, are 25-30% larger than their normal siblings, have elevated levels of circulating IGF2 and IGF-binding proteins, and exhibit a slight kink in their tails. These results show that Igf2r is paternally imprinted and reveal that the receptor is crucial for regulating normal fetal growth, circulating levels of IGF2, and heart development.
