Characterization of the epitope recognized by a monoclonal antibody directed against the largest subunit of Drosophila RNA polymerase II.

The epitope recognized by monoclonal antibody MAb215 generated previously against Drosophila melanogaster RNA polymerase II was mapped to amino acid residues 806-820 of the largest, 215 kDa, subunit located in a region conserved within the largest subunits of pro- and eukaryotic RNA polymerases. The affinities of MAb215 and of a recombinant single-chain Fv fragment (scFv215) were determined for binding to the enzyme as well as the fusion protein and synthetic peptides used for epitope mapping. In addition, amino acid residues of the epitope important for binding to MAb215 were identified using peptides carrying single amino acid substitutions. The epitope is not involved in the polymerization reaction or the DNA unwinding process since no inhibitory effects of the monoclonal antibody were observed in nonspecific in vitro transcription using denatured calf thymus DNA or double stranded oligo dC-tailed T7 DNA as template. In contrast, MAb215 inhibits accurate in vitro transcription from the Krüppel gene promoter and from the adenovirus-2 major late promoter. Preincubation of template DNA with the nuclear extract had no effects on inhibition supporting the notion that the epitope does not participate directly in the formation of preinitiation complexes. The same inhibitory effects were observed using scFv215. The results provide further evidence that recombinant antibody fragments produced in Escherichia coli possess the same specificity and similar affinity as their parental antibodies and demonstrate that scFv fragments are useful tools for analysis of transcriptional processes.

Study of behavior of rats with gluten-induced enteropathy.

Association of different psychological and neurological disturbances with gluten intake in coeliac patients was repeatedly described. In the present study gluten-induced enteropathy was elicited in rats by prolonged intragastric administration of gliadin from birth to 10 weeks. Various neurological (contact and visual placing reactions, equilibrium on horizontal bar) and behavioral tests (open field and Morris water maze task) were used to assess the possible deficits. No substantial differences were found in the behavior of rats fed with gliadin compared with those fed with bovine serum albumin (control group). The only difference found between control and experimental rats was that gliadin-fed rats showed slightly higher emotionality in the open field test. It is concluded that prolonged application of gliadin to young rats at enteropathy-inducing dosages does not modify their behavior.

Suppression of the indirect pathway of T cell reactivity by high doses of allopeptide.

T helper cells, which recognize allopeptides processed and presented by self APC, contribute to the generation of both cellular and humoral immune responses against allogeneic transplants. We have explored the hypothesis that the indirect T cell recognition pathway is initiated by soluble MHC antigens and that it can be suppressed by high doses of synthetic peptides corresponding to the dominant alloepitope. T cells from a DR11/7 responder were immunized in vitro with recombinant HLA-DR4 (rDR4). Experiments using partially overlapping synthetic peptides showed that the resulting T cell line (TCL) recognized a single dominant epitope mapping within residues 69-88 of the first domain of the DR4 molecule. In vitro immunization with synthetic allopeptides corresponding to other polymorphic regions, were unable to elicit T cell reactivity against rDR4, although at least one of these peptides (corresponding to residues 13-27) was immunogenic, behaving like a cryptic epitope. The rDR4-specific TCL expressed a limited TCR repertoire and provided help to autologous B cells for the production of specific antibodies. The T cell blastogenic response as well as the transcription and secretion of IL-4 (but not IL-2) was efficiently suppressed by high doses of the dominant allopeptide. These findings support the concept that selective immunointervention of indirect allorecognition can be achieved by use of high doses of antigen or TCR vaccination, as proposed for autoimmune diseases.

Computerized interactive morphometry in the differential diagnosis of irradiated prostates.

Radiation therapy is becoming a treatment of choice for many patients with prostatic carcinoma. Distinguishing radiation change in prostate glands from carcinoma may be difficult. In this study we objectively assessed, by morphometric methods, the nuclear characteristics of benign and malignant prostates with a history of radiation treatment (125I implant with or without prior external beam radiation). This is part of our continuing efforts to achieve difficult differential diagnoses by analyzing perimeter, diameter and nuclear profile area of cells or interest and applying methods of statistical classification. Biopsies were performed 18-36 months following implant therapy. Eleven cases with residual prostate tumor constituted the malignant group. These were compared to 20 benign cases (benign glands in the 11 carcinoma cases plus 9 other cases with no residual carcinoma). Immunohistochemical staining with keratin 903 was performed on all cases. Differences in the nuclear parameters were most evident in the average nuclear profile areas (32.5 microns 2 for the malignant groups vs. 39.6 for the benign) and in the mean maximal cord length (diameter) (7.4 microns for the malignant group vs. 9.0 for the benign). Classification, however, is based on the size distribution plots of nuclear profile areas, which, in the malignant cases, had a sharper peak at lower value, while the benign cases had higher value and a broader peak with a trailing off into the larger values. This study emphasized the marked nuclear alterations that occur in irradiated prostates.(ABSTRACT TRUNCATED AT 250 WORDS)

Multiple subpial transection for treatment of intractable epilepsy.

On the basis of experimental study, we applied multiple subpial transection (MST) to treat 50 patients with intractable epilepsy in which epileptigenic lesion involved functional areas such as pericentral gyrus, postcentral gyrus, Broca's area, Wernicke's area, visual cortex, etc. They were followed up for 6 to 40 months. Complete control of seizures was obtained in 32 patients, significant reduction of seizure (more than 50%) in 13, reduction (less than 50%) in 3, and no effect in 2. The total effective rate was 96%. No functional defect was found in all patients. The mechanism of the disease and surgical technique were discussed in detail. We consider that MST could replace some standard excisional therapy for local epilepsy.

Interaction of interleukin-6, tumour necrosis factor and interleukin-1 during Listeria infection.

Injected recombinant interleukin-6 (IL-6), tumour necrosis factor (TNF) and IL-1 all protect mice against experimental infection with Listeria monocytogenes. We have therefore investigated the interaction of these cytokines during infection. Treatment with recombinant (r)IL-6 enhanced TNF production by spleen cells during the first 2 days of infection. Anti-TNF antibody could totally abolish the protective effect of rIL-6, while the optimal protective function of TNF could not be achieved when IL-6 was neutralized by anti-IL-6 antibody. IL-1 induced a high level of IL-6 in the serum a short time after its administration, and neutralization of IL-6 totally abolished the protective function of rIL-1. The results thus provide further evidence for the complexity of cytokine interaction.

[Determinate titer of tuberculosis antibody with cytomembrane antigen].

Using tuberculous cytomembrane antibody and titer determining methodology, serum specimens from 165 pulmonary tuberculosis patients were analysed. The results showed: 75.7% of specimen is in high level, titer > or = 1:400.20% in low level, titer 1:100-1:200.4. 42% negative. This method is more sensitive than usually used PPD and polymeric OT antigen. At the same time, sputum specimens from 406 TB patients were examined for TB bacilli, the result demonstrated that the positive rate of sputum smear in the patients with titer > or = 800 is up to 95.71%-100%.1:400 is 69.86%, 1:100-1:200 is 14%-22.09%, < 1:100 is 10%.

Regulation of human clotting factor IX cDNA expression in transgenic mice.

To study the expression of human clotting factor IX cDNA in transgenic mice, which is an essential work on gene therapy for hemophilia B, 3 recombinant constructions containing different lengths of human clotting factor IX cDNA have been introduced into the cultured cells. All of the recombinant constructions were found to be expressed well in vitro. They were then microinjected into the male pronuclei of the fertilized mouse eggs respectively for generating transgenic mice. Unfortunately, none of them was expressed in any transgenic mice. These results show that the expression of the human clotting factor IX cDNA in the transgenic mice can be determined by cis regulatory element(s). As compared with the results from other related works, it is suggested that the cis regulatory element(s) is resided in the 5'-end non-coding region.

[Prognostic factors in patients with osteosarcoma].

The prognostic factors in 54 patients with osteosarcoma were studied by analyses of simple factor, multiple linear regression and stepwise regression. The results showed that the significant factors affecting prognosis were tumor cells' DNA content, radiological grade and surgical stage, while tumors' site, maximal diameter proliferating activity indicated by S-phase cell percentage, proliferating index and AgNOR counts had limited influence on the patients' outcome. Besides, patients' sex, age, duration of the symptom, tumors' radiological classification and pathological type bore no relation to survival rate.

[The effect of qing guang kang on the metabolism of retinal ganglionic cells in rats after artificial acute high intraocular pressure].

A modified cytochrome oxidase (CO) histochemical and computer image techniques were used to examine the effect of Qing Guang Kang (QGK) injection on the metabolism of rat retinas after artificial acute high intraocular pressure. The results show that 75% of the average arterial blood pressure as the perfusing pressure of the anterior chamber of rats can lead to the decrease of numbers of CO active retinal ganglionic cells and their grey values. QGK possess the protective and/or improving effects on the CO active retinal ganglionic cells of rat under artificial acute high intraocular pressure. In the study, the data of density and the grey value of CO active ganglionic cells in the experimental models with QGK injection are significantly different from those of the control group (P < 0.05).

[Changes in rat glomerular polyanion and ultrastructures induced by supernatants of cultured mononuclear cells from patients with steroid-response nephrotic syndrome].

The supernatant of cultured conA stimulated mononuclear cells from patients with steroid-response nephrotic syndrome was infused into rat kidneys. Renal tissue sections stained with colloidal iron were analyzed quantitatively by image analyzer. The mean width of foot processes and slits the volume fraction of foot processes and glomerular capilaries of electron photomicrograph were determined by using stereology in combination with the observation of glomerular ultrastructure. The studies showed that there was a significant decrease in glomerular polyanion in rat kidneys perfused with supernatant from patients in the active stage. The glomerular alterations included swelling and fusion of foot processes and narrowing of filtration slits. There were no above-mentioned alterations in both the patients in remission and the normal controls. The results demonstrated that supernatant of cultured mononuclear cells from patients with steroid-response nephrotic syndrome resulted in pathologic alterations in rat glomeruli similar to those observed in minimal change nephrotic syndrome, and suggested that the glomerular alterations might result from a factor produced by a kind of abnormal T lymphocytes.

Labeling and quantitative analysis of six lectin receptors of intracranial gliomas.

The distribution of six lectin receptors, WGA, RCA-1, LCA, PSA, PNA and SBA in 111 human gliomas, 8 human normal brain tissues and 11 reactive hyperplasia of astrocytes was observed by means of avidin-biotin-peroxidase technique. Their grays were also quantified with the image analysis instrument. The results showed that WGA and RCA-1 might be used as markers for distinguishing well-differentiated astrocytomas from the reactive hyperplasia of astrocytes especially the reactivity of astrocytes had a specific feature with RCA-1. The difference in quantities of WGA, RCA-1, LCA, PSA receptors between astrocytomas, ependymomas and oligodendrogliomas, medulloblastomas might conduce to the diagnosis and classification. For astrocytomas, it was also showed that a quantity of LCA and PSA receptors was correlated with the degree of cell differentiation. Therefore, they can be used as valuable markers of the differentiation of astrocytomas.
