[A study on the diagnostic criteria for adult acute myeloid leukemia with morphology, immunology and cytogenetics].

This study was undertaken to establish the more reasonable criteria for diagnosis of acute myeloid leukemia (AML). 82 cases diagnosed initially or finally as AML were analyzed with morphology, immunology and cytogenetics (MIC). The results revealed that 89.0% of the pretherapy morphology conformed to MIC and 93.9% of the immunology conformed to it. 4 cases with hybrid acute leukemia (HAL), one case with acute undifferentiated leukemia (AUL) and one case with acute B-Acute lymphocytic leukemia were confirmed with MIC. The positive expression of myeloid markers on samples from 76 cases of AML was followed by CD33 > CD13 > CD65, SI6 > CD15 > CD11b > CD14, but not specific for AML subtypes. The lymphoid antigens CD2, CD7, CD10 and CD19 were positive in minority of the cases of AML, but CD2+ and CD7+ were easily found in M3 and M1 speerately. 55.4% of the patients with AML in this group showed abnormality in cytogenetics. Typical t(8;21) or its variants was found in 14/24 cases of M2 and one case of M1; t(7;11) (P15;P15) in one case of M2; t(15;17) in 4/7 cases of M3; and inv(16) in one case of M4E0. It is shown that MIC classificassion is more helpful than any signgle one of the three in diagnosis of AML, especially of HAL, AUL, Mo.

[Effects of acupuncture on lipid, TXB2, 6-keto-PGF, alpha in simple obese patients complicated with hyperlipidemia].

The authors observed the changes pf symptoms and signs, obesity index, lipid index (TC, TG, LDL-C, HDL-C), atherosclerosis in dex (AL), ratio of waist centimetre to hip centimetre (W. C/H. C). TXB2 and 6-Keto-PGF1 alpha in 34 simple obesity patients complicated with hyperlipidemia before and after acupuncture so as to make clear the influence of acupuncture on pathogenic factors led up to circular diseases. The results showed that the marked weight loss effect was achieved in the cases by acupuncture, while the level of TC, TG, LDL-C, HDL-C, AI, W.C./H.C, TXB2, 6-Keto-PGF1 alpha in the patients were finely regulated. It suggests that the acupuncture treatment not only treated obesity and hyperlipidemia, but also resisted the pathogenic factors led up to circular diseases.

[Study of electrophysiological properties of large conductance K(+)-channels in outer hair cells from the guinea pig with patch-clamp technique].

The basic electrophysiological characteristics of large conductance K(+)-channels in the basolateral cell membrane of outer hair cells (OHC) from the guinea pig cochlea were studied with patch-clamp technique. Acutely isolated OHC preparations were obtained by enzymatic digestion and excised patches were made in cell-attached configuration to record single-channel currents led by patch-clamp micropipette and fed to patch-clamp amplifier. Data were sampled and analyzed on-line with a computer. The result showed that the typical properties of single channels of large conductance K(+)-channels were: 1. The channel was activated by depolarization of membrane and it showed a marked voltage-dependence. The channel had a unitary conductance of about 133 pS and the channel current reversed at about -30mV under asymmetrical K+ concentration gradient. 2. Tetraethylammonium (TEA, > 10 mmol/L), a potassium channel blocker, dose-dependently abolished the outward current. 3. The open probability (Po) and mean open time of this channel was markedly increased with elevation of cytoplasmic Ca2+ concentration. These results suggest that a population of voltage-dependent and Ca(2+)-activated K+ channel exists in the basolateral membrane of OHC of the guinea pig; it is characterized by a very high unitary conductance and high sensitivity to Ca2+ and is blocked by K+ channel blocker. This work provided valuable basic materials for further study on the regulation and drug influence of the type of single channels.

[Interleukin-6 and immune state in children with nephrotic syndrome].

IL-6 activities were determined in 37 children with idiopathic nephrotic syndrome (INS) by using an IL-6 dependent murine hybridoma-clone (MH60). The results showed that the levels of IL-6 activities in children with INS at the stage of onset were significantly higher than those of normal control group (P < 0.01), and the levels of IL-6 activities had positive correlation with the levels of urinary protein in 24 hours. The levels of IL-6 activities decreased remarkably at the stage of remission. CD+3 cells and the levels of serum IgG decreased significantly in those with INS; CD+8 cell increased and CD+4/CD-8 cell ratio decreased obviously in those with the simple nephrotic syndrome (SNS) at the stage of onset as compared with normal control group. These results indicate that there are immune disorders characterized by increased level of IL-6 activity in INS and suggest that IL-6 plays an important role in the development of INS.

[Improvement in model of mesangial proliferative glomerulonephritis of rat].

To establish a model of mesangial proliferative glomerulonephritis (MsPGN), we gave Sprague-Dawley rats staphylococcal enterotoxin B (SEB) intravenously twice in different times, bovine serum albumine (BSA) through digestive tract every other day, and Freud's adjuvant subcutaneously twice in different times, respectively. The results showed that measangial cells and matrix in glomeruli of rats began to proliferate at the end of 6th week and this change became obvious at the end of 8th week. The duration of experiment was shorter than that using only SEB, and the change of pathomorphology was stable. The experiment confirms the thesis that mucosal immunization of gastroenteric tract plays an important role in the development of MsPGN.

[Corneal endothelial decompensation of iridocorneal endothelial syndrome treated by penetrating keratoplasty].

OBJECTIVE: To evaluate the effect of penetrating keratoplasty in treatment of iridocorneal endothelial syndrome. METHODS: The clinical and pathologic evaluation records were retrospectively analyzed for 8 cases with iridocorneal endothelial syndrome treated by penetrating keratoplasty. Postoperatively, they were followed for 3 months to 6 years. RESULTS: Seven cases had visual improvement, five buttons kept being transparent and three buttons became opaque. CONCLUSIONS: The penetrating keratoplasty is an effective measure for treatment of corneal endothelial decompensation of iridocorneal endothelial syndrome. However, the successful operative rate of this syndrome is lower than that in cases with corneal leucoma without vascularization.

[Impact of platelet derived growth factor in the glomeruli of active lupus nephritis].

Platelet derived growth factor (PDGF) possesses diverse biological activities and plays an important role in the pathogenesis of glomeruli nephritis. In order to elucidate the relationship between PDGF and the disease activation in lupus nephritis, PDGF was observed in renal biopsy specimen from 9 cases of lupus nephritis (7 cases with active and 2 cases with inactive lesions) by immunohistochemistry using 4-layer PAP method. The mRNA expression of PDGF-A and -B chain, and the receptor of PDGF-B were analyzed by RT-PCR. It was found that the levels of mRNA expression of PDGF and its receptor were much higher in patients with active lesions in glomeruli as compared with those without active lesions, there changes were paralleled with the degree of hematuria in these patients. Noted that there was no correlation between the expression of PDGF and the cellular proliferation in glomeruli of lupus nephritis. These results indicate that PDGF is a critical mediator for inducing active lesion in glomeruli of lupus nephritis. The effect of PDGF antagonist in the regulation of glomerular damage in lupus nephritis need to further elucidate.

[Computer image analysis in the study of endolymphatic hydrops].

In search for an objective method to be used to judge the extent of hydrops, computer image analysis system was used in the study on guinea pig endolymphatic hydrops. The image information of the whole cochlea was collected, then the length of Reissener's membrane and section area of scala media were calculated based upon parameters such as ratio factors and point values. The results were as follows: 1) Hydrops observed with light microscope had no statistical difference. 2) Within 55 days postoperatively, the hydrops grew more obvious with time. 3) The hydrops was more extensive in the basal turn than in the apical turn, though more conspicuous cytopathologic change was found in the apical turn. The conclusions: 1) The current technique is objective, accurate and reliable. 2) Both the length of Reissener's membrane and section area of scala media can be used to measure the extent of hydrops.

Corneal topography research of myopia in Chinese.

PURPOSE: To determine corneal topography parameters of myopia in Chinese and help for studies of causes of myopia, distingiush other corneal diseases (such as early keratoconus), further explore the accurateness and safeness of the clinical effects of radil keratotomy (RK) or photorefractive keratectomy (PRK). METHODS: Fifty-eight eyes with mild or moderate myopia were examined by computer-assisted photokeratography. (TMS-1 Computed Anatomy Inc. New York. NY) RESULTS: The corneal topography of most (68%) of the examined eyes showed symmetric bowtie pattern: the corneal surface of most (75%) of the examined eyes showed positive aspheric shape (steeper centrally, flatter peripherally); the refractive power of central cornea is at mean 44.165 +/- 1.429 D; the refractive difference between 1 to 25 ring in keratoscpe was at mean 1.655 +/- 0.785D; the mean SRI was 0.558 +/- 0.768, the mean SAI was 0.296 +/- 0.149; the mean Smik was 44.209 +/- 1.515D. The apex and visual axis were dislocation in all the cornea examined. CONCLUSIONS: The corneal surface refractive characterization of myopia might be relative to the clinical effects of RK or PRK.

[Inhibitory effects of co-cinobufotalin oral liquor on hepatitis B in vitro].

Co-Cinobufotalin Oral Liquor (CCOL) was studied for its ability to inhibit hepatitis B virus DNA replication, HBsAg and HBeAg expression in a HBV-transfected cell line (2.2.15 cell). The result showed that ID50 (the drug concentration that inhibits HBsAg or HBeAg secretion by 50%) was 0.08 mg/ml and 0.07 mg/ml on HBsAg and HBeAg respectively. CD50 (the drug concentration that reduces cell growth by 50%) was 2.5 mg/ml. TI (therapeutic index) was 31.3 and 35.7 respectively. The present data suggest that CCOL could exert a potent antiviral activity against HBV in vitro. Southern blot showed that CCOL inhibited HBV-DNA repication in a dose-dependent manner.

Molecular mapping of a pathogenically relevant BP180 epitope associated with experimentally induced murine bullous pemphigoid.

Bullous pemphigoid (BP) and herpes gestationis (HG) are subepidermal blistering diseases associated with an autoimmune response directed against BP180, an epidermal hemidesmosomal glycoprotein. The pathogenic relevance of this Ag/Ab system was established by the recent demonstration that IgG Abs reactive with the murine form of BP180 (mBP180) are capable of triggering a subepidermal blistering disease after passive transfer into neonatal BALB/c mice. The aim of the present study was to determine the fine specificity of the pathogenically relevant Abs in this experimental model of BP. Four high titer rabbit-anti-mBP180 antisera were included in this analysis--only two of which exhibited pathogenic activity in the passive transfer model. Immunoblot analysis using a panel of mBP180 deletion mutants revealed that each of the four rabbit sera reacted with at least three distinct sites on the mBP180 ectodomain; however, this technique failed to distinguish between the reactivity patterns of the pathogenic and nonpathogenic sera. An alternative technique, liquid phase immunoadsorption analysis, was used to identify one mBP180 antigenic site, comprising 9 to 12 amino acids and designated mBP1, that was specifically recognized by the two pathogenic sera. Pre-adsorption of pathogenically active IgG preparations with fusion proteins containing the mBP1 antigenic site resulted in complete blocking of immunofluorescence reactivity with the murine basement membrane zone (BMZ) and in complete neutralization of pathogenic activity. Anti-BMZ reactivity displayed by nonpathogenic Abs was not altered or diminished by pre-adsorption with this same mBP180 recombinant protein. These findings should help to elucidate the immunopathologic mechanisms responsible for human BP and HG and may have significant implications in the diagnosis and treatment of these autoimmune diseases.

Effect of kainic acid-induced status epilepticus on inositol-trisphosphate and seizure-induced brain damage in mature and immature animals.

We investigated the role of excitatory amino acids in the activation of the phosphoinositide pathway during kainic acid-induced seizures in mature and immature animals. Kainic acid caused more severe seizures in the immature animals, but no hippocampal damage or induction of phosphoinositide hydrolysis. In mature animals, seizures were mild but severe hippocampal damage was seen and was associated with a marked and sustained release of inositol-trisphosphate, suggesting a role of this pathway and intracellular calcium stores in seizure-induced brain damage.

The insulin-dependent biosynthesis of GLUT1 and GLUT3 glucose transporters in L6 muscle cells is mediated by distinct pathways. Roles of p21ras and pp70 S6 kinase.

Insulin binding results in rapid phosphorylation of insulin receptor substrate-1 to activate p21ras and mitogen-activated protein kinase. Insulin also activates the ribosomal protein S6 kinase (pp70 S6 kinase) independently of the Ras pathway. Chronic (18 h) treatment of L6 muscle cells with insulin increases glucose transport activity severalfold due to biosynthetic elevation of the GLUT1 and GLUT3 but not the GLUT4 glucose transporters. Here we investigate the roles of p21ras and pp70 S6 kinase in the insulin-mediated increases in GLUT1 and GLUT3 expression. L6 cells were transfected with the dominant negative Ras(S17N) under the control of a dexamethasone-inducible promoter. Induction of Ras(S17N) failed to block the insulin-mediated increase in GLUT1 glucose transporter protein and mRNA; however, it abrogated the insulin-mediated increase in GLUT3 glucose transporter protein and mRNA. Inhibition of pp70 S6 kinase by rapamycin, on the other hand, eliminated the insulin-mediated increase in GLUT1 but had no effect on that of GLUT3 in both parental and Ras(S17N) transfected L6 cells. These results suggest that the biosynthetic regulation of glucose transporters is differentially determined, with pp70 S6 kinase and p21ras playing active roles in the insulin-stimulated increases in GLUT1 and GLUT3, respectively.

Two CACGTG motifs with proper spacing dictate the carbohydrate regulation of hepatic gene transcription.

Regulatory sequences involved in the transcriptional induction of the rat S14 gene in response to increased glucose metabolism in the hepatocyte were investigated and compared with those of the liver-type pyruvate kinase (L-PK) gene. The carbohydrate response element (ChoRE) of the S14 gene was found to consist of two motifs related to the consensus binding site for the c-myc family of transcription factors, CACGTG. These two motifs are separated by five base pairs, a similar arrangement to that found in the L-PK ChoRE. In its natural context, the S14 ChoRE requires a novel accessory factor to support the full response glucose. This factor, as well as the factor hepatic nuclear factor-4, are both capable of binding to the L-PK gene to enhance its carbohydrate regulation. The need for an accessory factor for supporting the glucose response can be overcome in two ways. First, multimers of the ChoREs of either the L-PK or S14 genes can function independently to support the glucose response. Second, mutations in the S14 ChoRE that create a perfect match to the consensus CACGTG motif at each locus no longer require an accessory factor site. The spacing of the two CACGTG motifs, but not the nature of the bases within the spacer, are critical for control. These observations suggest that a carbohydrate responsive factor binds to both motifs in a highly specific spatial orientation to confer the response to increased carbohydrate metabolism.

Expression of syntaxin 4 in rat skeletal muscle and rat skeletal muscle cells in culture.

Syntaxins are a family of membrane proteins believed to participate in docking/fusing of arriving vesicles during membrane sorting and secretion. Of the six mammalian syntaxins known, only brain syntaxin 1A/1B has been biochemically characterized in its endogenous form. Syntaxin 4 mRNA is expressed in selective tissues including rat skeletal muscle, although it has not been studied at the protein level in any cell type. Therefore, we generated an affinity-purified antibody against syntaxin 4 to demonstrate that this 36 kDa protein is expressed in rat skeletal muscle and L6 muscle cells in culture. The content of the syntaxin 4 protein increased by 1.9-fold during differentiation of L6 myoblasts into myotubes. By subcellular fractionation the protein was mainly recovered in plasma membrane-enriched fractions of both red and white skeletal muscles and L6 myotubes. Coupled to the recent detection of vesicle associated membrane protein-2 and cellubrevin in skeletal muscle, syntaxin 4 may play a role in membrane traffic in this tissue.
