[The relationship of C fiber to 5-hydroxytryptamine in spinal cord of rat in electroacupuncture analgesia].

By using spectrofluorophotometry, it was observed that electroacupuntrure at "Huantiao" points of rat could decease the content of 5-hydroxytryptamine (5-HT) and increase the content of 5-hydroxyindoleacetic acid (5-HIAA) in spinal cord. However, the above effects of electroacupunture were obviously reduced after the C fibers of rats had been destroyed by capsaicin, which suggesting that capsaicin-sensitive C fiber might take part in the roles of electroacupuncture's promoting the release and utilization of 5-HT in spinal cord.

[The effect of salmeterol on the activity of neutrophil chemotactic factor in patients with asthma].

OBJECTIVE: The activity of neutrophil chemotactic factor (NCF) in serum of 40 patients with asthma on their acute attacks was measured with membrane filter method. METHODS: Of the 26 patients, 13 were administed salmeterol 50 micrograms bid by inhaler and another 13 took procaterol 50 micrograms bid orally for four weeks. RESULTS: It was found that in patients with asthma NCF was 82 +/- 17 cell/10 HP, which was significantly higher than that in normal subjects (33 +/- 5 cell/10 HP, P < 0.01). After treatment with salmeterol the decrease in NCF activity was 26 cell/10 HP (P < 0.01), while the decrease in another group treated with procaterol was 8 cell/10 HP (P > 0.05). The difference between the two treated groups was significant (P < 0.01). In salmeterol treated group the decrease in NCF activity was more remarkable than the improvement in lung function. CONCLUSIONS: It was considered that the result may be related to the anti-inflammatory effect of salmeterol.

[Studies on the expression of desmin and myosin in experimental astrogliosis].

The dynamic changes of desmin and myosin in rat brain at different periods after stab wound were studied by immunohistochemistry and quantitation methods. The expression of desmin in astrocytes markedly increased and reached its peak on the 7th day and maintained this high level up to 30 days. The zone of reactivity was initially limited to the wound vicinity but spread with time to the entire ipsilateral cortex. The intensity weakened gradually with the increase of distance from the lesion. Myosin was positive mainly in the margin of the wound and partly in the molecular layer of the cortex. The results indicated that the enhancement of desmin expression was closely related to the reactive astrogliosis, whereas the expression of myosin may be a marker of astrocyte hyperplasia. The significance of these responses was discussed.

[Effects of iodine and thyroid hormone deficiency during brain development on activities of cholinergic neurone-related enzymes in central nervous system of rats].

To study the effects of iodine and thyroid hormone deficiency during brain development on activities of enzymes of central acetyl choline metabolism, activities of choline acetylcholinetransferase (CHAT) and acetylcholinesterase (AChE) in hippocampus and cerebral cortex, basal forebrain and cerebellum in 20-day-old rats were determined. Results showed that CHAT and AChE activities in hippocampus, cerebral cortex and basal forebrain in iodine deficiency and hypothyroid rats were significantly lower than those in controls. A ratio of membranebound AChE to soluble AChE, activities of inextractable AChE, and percentage of its activities in the four sections of the brain in the trial rats were significantly lower, and activities of soluble AChE and its percentage of activities were significantly higher than those in controls. It suggests iodine and thyroid hormone deficiency could affect maturation of CHAT and disturb transformation and maturation of various molecular types of AChE and delay distinctly the development of cholinergic neurone during the critical period of brain development in rats.

[Expression and clinical significance of PCNA in renal pelvic and ureteral cancer].

To determine the relationship between PCNA expression and pathological stage, cellular grade, recurrence and prognosis of renal pelvic and ureteral cancer, research on PCNA proliferating index in 47 cases of renal pelvic and ureteral cancer we studied by immunohistochemistry S-P method. PCNA proliferating index of renal pelvic and ureteral cancer increased with pathologic stage and cellular grade. PCNA proliferating index of T3 + T4 was significantly higher than that of T1 + T2 (P < 0.01). PCNA proliferating index of G3 was significantly higher than that of G1 + G2 (P < 0.001). Those with higher PCNA proliferating index (III + IV) had higher bladder cancer recurrent rate after operation (60%, 12/20), those with lower PCNA proliferating index (I + II) had lower bladder cancer recurrent rate (22.2%, 6/27). P < 0.01. 35% (7/20) of those with III + IV PCNA proliferating index had simultaneous polyorganic urothalial cancer, 7.4% (2/27) of those with I + II PCNA proliferating index had simultaneous polyorganic urothelial cancer, P < 0.05. 7.4% (2/27) of those with I + II PCNA proliferating index had local recurrence and metastasis after operation, 40% (8/20) of those with III + IV had local recurrence and metastasis, P < 0.05. The 5-year survival rate of I + II was 86.7%, the 5-year survival rated of III + IV was 35.7% (5/14), P < 0.05. The findings suggest that PCNA proliferating index connects with pathological stage and cellular grade of renal pelvic and ureteral cancer. It is an important characteristic of low differentiation, invasiveness and simultaneous or sequentially polyorganic urothelial cancer. It may be an important prognostic indicator of renal pelvic and ureteral cancer.

[Hashimoto's disease with thyroid cancer: report of six cases].

Hashimoto's disease associated with thyroid cancer is not common. From January 1976 to December 1994, a total of 95 patients with Hashimoto's disease were treated surgically. Six (5 Papillary 1 folicular) of 95 patients had thyroid cancer which was concomitantly revealed pathologically. They were all women with mean age of 30.3 years. The size of tumor was 0.3-2.0 cm. Follow-up for 1-17 years showed that they were all alive and healthy. Two patients were misdiagnosed as having anaplastic carcinoma but the definitive diagnosis by immunohistochemical analysis was thyroid malignant lymphoma (B-cell type). Hashimoto's disease and thyroid cancer relationship, diagnosis, and treatment are discussed.

[Metastatic potential correlates with cell surface carbohydrate profiles of nasopharyngeal carcinoma].

In order to investigate the changes in glycoprotein structure between non-metastatic and metastatic cells of nasopharyngerl carcinoma (NPC), peanut agglutinin (PNA), ulex europeaus (UEA-I) and concanavalin ensifomis agglutinin (ConA) staining were used to examine 102 nasopharyngeal carcinoma tissues (84 primary tumors and 18 metastatic lymph nodes) with the avidin-biotinperoxidase complex method. PNA reaction was positive in most cells of nasopharyngeal carcinomas regardles of in histopathologic type, the degree of differentiation and metastasis. The levels of UEA-I receptors apparently increased during nasopharyngeal carcinoma progression from non-metastatic to metastatic tumors. The binding to ConA clearly decreased in the cells of metastatic cases and metastatic lymph node. So, the increased expression of UEA-I receptors and decrease of ConA receptors on tumor cells might have been implicated in the expression of metastatic potential.

[Dynamic changes in soluble interleukin-2 receptor levels after rat corneal allograft].

PURPOSE: To evaluate the diagnostic utility of serum IL-2R level test in corneal allograft rejection. METHODS: Serum soluble IL-2R was detected by radioimmunoassay in rat corneal allograft model. RESULTS: Levels of soluble IL-2R were markedly elevated prior to the clinical diagnosis of rejection. CONCLUSIONS: Marked elevations of soluble IL-2R are associated with rejection, measurement of soluble IL-2R may have a useful value in the early diagnosis of corneal allograft rejection.

[Pharmacological comparative study on baiqian and baiwei].

The decoction of Cynanchum glaucescens and the ethanol extract of C. stauntonii show significant antitussive and expectorant actions. The decoctions of C. stauntonii or C. glaucescens have antiasthmatic and anti-inflammatory effects. The decoction of C. atratum has some expectorant effect but no antitussive or antiasthmatic actions. On the other hand, the decoction of C. versicolor cannot relieve cough nor sputum. The results indicate that both C. atratum and C. versicolor cannot be used as C. glaucescens or C. stauntonii.

[Changes of inorganic elements in senescent scales of Fritillaria thumbergii Miq. at the late stage].

By means of inductively coupled plasma-atomic emission spectroscopy (ICP-AES), the changes of inorganic elements in the senescent scales of Fritillaria thunbergii at the late stage were determined. The results showed that the main trends of the changes of P, S and K were continuously removing, while those of most elements such as Al were less removing. The inorganic elements for the growth of new organs were absorbed mainly from soil by the root.

The XRCC4 gene encodes a novel protein involved in DNA double-strand break repair and V(D)J recombination.

The XR-1 Chinese hamster ovary cell line is impaired in DNA double-strand break repair (DSBR) and in ability to support V(D)J recombination of transiently introduced substrates. We now show that XR-1 cells support recombination-activating gene 1- and 2-mediated initiation of V(D)J recombination within a chromosomally integrated substrate, but are highly impaired in ability to complete the process by forming coding and recognition sequence joins. On this basis, we isolated a human cDNA sequence, termed XRCC4, whose expression confers normal V(D)J recombination ability and significant restoration of DSBR activity to XR-1, clearly demonstrating that this gene product is involved in both processes. The XRCC4 gene maps to the previously identified locus on human chromosome 5, is deleted in XR-1 cells, and encodes a ubiquitously expressed product unrelated to any described protein.

Effects of non-nucleoside inhibitors of human immunodeficiency virus type 1 in cell-free recombinant reverse transcriptase assays.

We have employed a cell-free human immunodeficiency virus type 1 (HIV-1) reverse transcriptase (RT) assay to study the effects of non-nucleoside inhibitors of RT (NNRTI) by directly monitoring specific HIV DNA products using a HIV-1 genome-derived template and an oligodeoxynucleotide primer. As previously shown by ourselves and others, nucleoside analog triphosphates, e.g. 3'-azido-3'-deoxythymidine triphosphate and 2',3'-dideoxyadenosine triphosphate, could directly inhibit HIV RT RNA-dependent DNA polymerase activity by causing chain termination, as visualized in a RT reaction that yields specific DNA products. In contrast, each of two NNRTIs, nevirapine and delavirdine, directly inhibited RT activity without causing chain termination effects. We also analyzed interactions between nucleoside analogs and NNRTIs or among NNRTIs by chain elongation/dNTP incorporation and/or steady-state kinetic assays. Combinations of nevirapine with the triphosphates of either the (-)-strand of 2',3'-dideoxy-3'-thiacytidine or 2',3'-dideoxyadenosine yielded additive/synergistic effects on RT activity. However, only an additive effect was observed when combinations of nevirapine and 3'-azido-3'-deoxythymidine triphosphate were employed. Combinations of nevirapine and delavirdine had an antagonistic effect on the inhibition of HIV-1 RT activity.

Tryptophan 250 on the alpha subunit plays an important role in flavin and aldehyde binding to bacterial luciferase. Effects of W-->Y mutations on catalytic function.

Bacterial luciferase is a heterodimer (alpha beta) that catalyzes the oxidation of FMNH2 and a fatty aldehyde, resulting in light emission. To explore the nature of the flavin binding site with respect to the role of tryptophan residues, the catalytic and binding properties of single-point mutants of Xenorhabdus luminescens luciferase with one of the eight tryptophans converted to a tyrosine residue were investigated by luminescence and fluorescence measurements. Conversion of tryptophans 194 and 250 on the alpha subunit to tyrosine had relatively large effects on the properties of luciferase with only minor changes in the properties on mutation of the other four tryptophans on alpha and the two on the beta subunit. Mutation of alpha W250 decreased the binding to FMNH2, FMN, aldehyde, and fatty acid, causing major changes in luminescence emission and decay. The results are consistent with alpha W250 interacting with flavin which in turn affects aldehyde binding. Mutation of alpha W194 did not affect the interaction with flavin or aldehyde but did change the relative rate of decay of light emission with aldehydes of different chain lengths as well as the activation energy for this process. Moreover, these results provide evidence for alpha W250, and to a lesser extent alpha W194, being in contact with the isoalloxazine ring of flavin, a proposal that has been recently made based on a model with flavin bound to the alpha subunit and anchored at a binding site for the phosphate moiety of FMN(H2) identified in the crystal structure of Vibrio harveyi luciferase [Fisher, A. J. Raushel, F. M., Baldwin, T. O., & Rayment, I. (1995) Biochemistry 34, 6581-6586].
