[Effect of acetylcholine receptor antibody derived from myasthenia gravis on the rat central neuronal nicotinic acetylcholine receptor].

Immunoreaction between acetylcholine receptor antibody (AChRab) derived from myasthenia gravis (MG) and neuronal nicotinic acetylcholine receptor (AChR) and the action of AChRab on rat brain stem auditory transmission were investigated by immunocytochemistry and brain stem auditory evoked potential (BAEP) in a search for abnormalities indicating central nervous system (CNS) involvement in this disorder. 25 microliters of AChRab were injected into the ventricular system of rats once every 48 hours for periods of up to a week BAEP examination was taken before and after injection of AChRab. Immunocytochemical investigation of the rat brain sections provided evidence that the neuronal nicotinic AChR like immunoreactivity is widely distributed throughout the rat CNS such as cerebral cortex, hippocampus, motor neurons in brain stem, auditory nucleus cerebellar cortex, etc. Analysis of the immunocytochemical results showed that the distribution property of AChRab in rat CNS correlated well with known neuronal nicotinic AChR subtypes e.g., alpha 7 and alpha 4. BAEP inter peak latency was significantly prolonged in rats after injection of AChRab as compared with the control rats and the rats prior to injection of AChRab. The results indicated that cerebroventricular administration of AChRab not only cause dysfunction of brain stem auditory transmission but also leads to development of experimental autoimmune MG symptoms. It is concluded that AChRab influences the rat brain stem auditory transmission and it is suggested that the pathogenic effect of AChRab is not only limited at neuromuscular junction, but also involves the central neuronal nicotinic AChR.

Adenoviral vectors efficiently target cell lines derived from selected lymphocytic malignancies, including anaplastic large cell lymphoma and Hodgkin's disease.

We have hypothesized that adenoviral vectors might mediate gene transfer into cell lines derived from human lymphocytic malignancies, such as lymphoma, lymphocytic leukemia, and myeloma. A panel of 33 cell lines was studied for their ability to be transduced by an adenoviral (AD) vector encoding the Escherichia coli beta-galactosidase gene (AD-betagal). A cytochemical assay and a flow cytometry assay both demonstrated that a subset of lymphocytic cell lines can be efficiently transduced by adenoviral vectors. In particular, three of three anaplastic large cell lymphoma lines, two of two Hodgkin's disease cell lines, two of seven Burkitt's lymphoma cell lines, and three of five myeloma cell lines exhibited efficient gene transfer. The ability of an AD vector expressing the thymidine kinase (tk) gene from herpes simplex virus-1 (AD-tk) followed by ganciclovir (GCV) to kill 11 of these lymphocytic cell lines was studied. In eight of the cell lines tested, more than 68% of the cells were killed by AD-tk/GCV. Similar results were obtained using an adenoviral vector expressing the wild-type p53 tumor suppressor gene (AD-p53). Thus, AD-tk/GCV and AD-p53 both demonstrated efficient killing of these cell lines. These data document that adenoviral vectors are valuable reagents for the introduction of genes into selected lymphocytic cell lines. These data also suggest that adenoviral vectors might be useful for gene therapy of subsets of lymphocytic malignancy.

[Comparison of mechanical properties between human aortic valve and pulmonary arterial valve].

We have investigated the mechanical properties of Chinese aortic and pulmonary valves by means of uniaxial tension. The parameters include the stress-strain curve, the maximum tangent modulus E (the slope of linear segment) in the curve, the tensile strength delta b, and the ultimate strain epsilon b. The results have shown that E, delta b, and epsilon b values between aortic valve and pulmonary valve exist significant differences (significant level = 0.01), but the mechanical property differences between circumferential direction and radial direction in the pulmonary arterial valve are not significant, that means, the valve appears small anisotropic behaviour.

[Three-dimensional reconstruction of sinusoids from serial sections].

The three-dimensional image of sinusoids has been reconstructed by computer image processing using a new method of registration. By observing the reconstructed image of sinusoids, we can easily understand the anatomy of sinusoids. Theoretical analysis and experimental results suggest that the method is feasible and practicable.

[Repair of pulp defect of thumb by free palmaris brevis musculocutaneous flap].

It is difficult to repair the pulp defect of finger with good function. Here reported two cases of pulp defect of the thumb which were repaired with free palmris brevis musculo-cutaneous flap. The flap was designed as the following: taken the line crossing the pisiform and metacarpo-phalangeal joint of the little finger as the longitudinal axis of the flap. The proximal end of the flap was at the level of pisiform and the distal end was the distal transverse palmar crease. The radial border was the radial side of the palmris brevis and ulnar border was the ulnar edge of the palm. The flap should not be larger than 6.5 cm x 2.5 cm. The flap was dissected with proper ulnar vessels and the accompanying arterial branch and superficial branch of the ulnar nerve. The flap was transferred to repair the pulp defect of the thumb. The vessels were anastomosed with the radial artery and cephalic vein at the snuffbox. The nerves were sutured to the digital nerve. Three months after operation, the defect was healed and the sensation of pulp was recovered. The donor area was painless and without sensation disturbance. The motion of the little finger was normal. The function of the thumb was restored. It was concluded that the palmaris brevis musculocutaneous flap is one of the best donor region to repair pulp defect of thumb.

[Choice of treatment for fracture of distal tibia with ankle joint involvement].

It is difficult to treat the intra-articular fracture of distal tibia or Pilion's fracture. From 1987 to 1995, 43 cases of Pilon's fracture were admitted and treated with different methods. After treatment, they had been followed up for 1 to 8 years. According to Riiedi's Classification, there were type I 12 cases, type II 21 cases and type III 10 cases. The methods used on these patients included manual reduction and plaster of paris immobilization, calcaneous tubercle traction and plaster of paris immobilization, open reduction and internal fixation with Kirschner wires, and open reduction and internal fixation with AO plate. The outcome was evaluated according to Ovadia's criteria, for type I fracture, 8 cases were treated with conservative method with a satisfactory rate of 79.17% and 4 cases were treated with operation with a satisfactory rate of 91.67%; for type II, conservative method for 12 cases and the satisfactory rate was 33.33%, and operation for 9 cases with a satisfactory rate of 70.37%; for type III, conservative method for 2 cases, with poor result, and operation for 8 cases with satisfactory rate of 79.17%. The indication for conservative treatment was type I fracture. It showed that for Pilon's fracture, the outcome of open reduction and internal fixation was superior to that of the conservative treatment, especially in those having internal fixation with AO plate.

[Repair and functional reconstruction of war wounds of limbs].

From 1979 to 1994, reparative and reconstructive surgery were used to repair the war injuries of skins, bones, blood vessels and nerves of the limbs in 800 cases. A systematic clinical study was carried out. Many new operative methods were used and the results of treatment were good. Innovations and modifications were made in technique. In 120 cases of war injuries having soft tissues defects including skin and muscles, various tissue transplantations were used with the hope to accomplish one-staged repair of the defect and reconstruction of motor function of muscle. To those infections of bone and joint in war injuries, following early eradication of infected focus, transplantation of musculo-cutaneous flap or omental graft was immediately carried out with the aim to obtain primary healing of the wound. In the treatment of bone defects from war wounds with loss of skin and muscles, the vascularized skeleto-cutaneous graft was used. In the treatment of 150 cases of injury of peripheral nerve from forearms, the result of good to fair rated 68.8 percent for upper extremity and that for lower extremity, it was 62.2 percent. Following the early repair of 500 cases of injury of peripheral blood vessels, the patency rate of the blood vessel was 90 percent. The result following by pass vascular graft in the treatment of forearms injury of blood vessels even with very poor local condition was still very successful.

[A comparative study of laryngeal reinnervation from the ansa cervicalis].

A comparative study of four methods of laryngeal muscle reinnervation in dogs is presented. Twenty-eight cases were divided into four groups to undergo main branch and branch of ansa cervicalis nerve anastomosis, and nerves implantation an neuromuscular pedicles transfer respectively for restoration of vocal cord adduction on left sides. The results showed that the four procedures seemed to induce effective reinnervation of adductor muscles. But the main branch of ansa cervicalis nerve suture was superior to the other methods among which little difference was noted in the functional recovery, electrophysiological activity and muscle strength. It demonstrated that main branch of ansa nerve suture was the best procedure for treatment of unilateral vocal cord paralysis among the four methods.

[Study on the cytotoxic and DNA damaging effects in oral mucosal fibroblasts by areca nut extract].

An aqueous areca nut extract (ANE) was tested for its cytotoxic effects on cultured human embryo oral mucosal fibroblasts (HE-OMF) in vitro by using the trypan blue and thiazolyl blue (MTT) assay. The ANE decreases the cell survival rate in a dose-dependent manner (P < 0.05). So was the extract in inducing damage on the cellular DNA of HE-OMF in vitro examined by the nick translation assay. The increase in counts per minute (CPM) values was significant (P < 0.05) for comparing all four concentration groups tested, The results suggests that aqueous ANE is highly cytotoxic and capable to induce DNA damage on cultured HE-OMF. It may have potential carcinogenic effect on the oral mucosal membrane of whom habitually chewing the areca nut frequently for quite a long time. Futher study is required to illustrate the detail process and study the mechanism of these effects.

Elimination of the hydrolytic water molecule in a class A beta-lactamase mutant: crystal structure and kinetics.

Two site-directed mutant enzymes of the class A beta-lactamase from Staphylococcus aureus PC1 were produced with the goal of blocking the site that in the native enzyme is occupied by the proposed hydrolytic water molecule. The crystal structures of these two mutant enzymes, N170Q and N170M, have been determined and refined at 2.2 and 2.0 A, respectively. They reveal that the side chain of Gln 170 displaces the water molecule, whereas that of Met170 does not. In both cases, the catalytic rates with benzylpenicillin are reduced by 10(4) compared with the native enzyme. With nitrocefin, the N170Q mutant enzyme exhibits an approximately 800-fold reduced rate compared with the native enzyme and in addition, a fast initial burst with stoichiometry of 1 mol of degraded nitrocefin/mol of enzyme. Stopped-flow kinetic experiments establish that the rate constant of the burst is 250 s-1, a value comparable with the rate of acylation of the native enzyme. Two structurally based mechanisms that explain the kinetic properties of the N170Q beta-lactamase are proposed, both invoking a deacylation-impaired enzyme due to the elimination of the hydrolytic water molecule. The catalytic rate of the N170M mutant enzyme with nitrocefin is reduced by approximately 50-fold compared with the native enzyme, and the slow progressive inhibition that is revealed indicates that the hydrolysis proceeds via a branched pathway mechanism. This is consistent with the structural data that show that the water site is preserved and that Met170 occupies part of the space that is required for substrate binding. The short contacts between the substrate and the enzyme may lead to structure perturbation and inactivation.

The primary structure of p38 gamma: a new member of p38 group of MAP kinases.

We have identified a third member of the p38 group of MAP kinase termed p38 gamma. The cDNA for this MAP kinase encodes an 367 amino acid polypeptide that is slightly greater than 60% identical to p38 and p38 beta. Expression of its mRNA is primarily in muscle with no detectable expression in Northern blots using RNA from other tissues. In contrast to p38 and p38 beta, p38 gamma, fails to phosphorylate ATF-2 or MAPKAP kinase 2 but does like other MAP kinases phosphorylate MBP. In vivo kinase assays using protein extracts from skeletal muscle reveal that a 7-kDa protein is phosphorylated by p38 gamma but not by other members of this group of kinases. We suggest p38 gamma may have unique functions when compared with other members of the p38 group due to its restricted tissue expression and apparent substrate preferences.

Single-unit responses to whole-field visual stimulation in the pretectum of Rana pipiens.

Visually responsive single units were recorded from the pretectal region that includes the large-celled nucleus lentiformis mesencephali (nLM) in the leopard frog, Rana pipiens. During monocular stimulation of the contralateral eye, 60 single units responding to movement of a large-field, random-dot pattern were quantitatively analyzed using horizontal and vertical directions at each of four pattern velocities (0.4-40 degrees/s). All units were spontaneously active, motion sensitive, and the majority showed 'on'-'off' responses. Several different response profiles were observed, including velocity-sensitive units with peak response at 10 degrees/s, most of which showed directional selectivity, and speed-sensitive units that showed increasing spike frequencies as pattern velocity increased, but little or no directional selectivity. About one-third of all unit analyzed were direction-selective, and 55% of those responded optimally to the temporal-to-nasal (T-N) direction of motion. T-N units were recorded primarily from an area that lies dorsolaterally between nLM and the optic tectum, in the 'peri-nLM' region. The pronounced monocular optokinetic nystagmus (OKN) response asymmetry that occurs in anurans appears to be reflected in the response profiles of the T-N direction selective units.

PEA3 transactivates vimentin promoter in mammary epithelial and tumor cells.

We have used differential display RT-PCR method to detect the genes specifically activated or repressed between mammary tumor and normal mammary epithelial cells. One of the genes identified is vimentin. The vimentin gene is abundantly expressed in both human and mouse mammary tumor cells and its expression decreased dramatically in normal mammary epithelial cells. The expression of vimentin gene correlates with the expression of transcription factor PEA3. Since the promoters of human and mouse vimentin genes contain one PEA3 binding site we investigated the ability of PEA3 to transactivate the vimentin promoter in mouse mammary epithelial cell CLS1, mouse mammary tumor MMT and human mammary tumor cell lines MCF7 and MDA231. Our results suggest that PEA3 specifically transactivates vimentin promoter through PEA3 site. Among members of the ETS transcription factor family only Erg showed ability to transactivate vimentin promoter besides PEA3. Our results also suggest that NFkB site on the vimentin promoter may act as a positive regulatory element for the transcription of vimentin. In metastatic mammary tumors derived from mice carrying the polyoma middle T or neu transgene, PEA3 is overexpressed and vimentin has been shown to play a key role in the motility of cells. Our results suggest that one of the roles of PEA3 in mammary tumor is to participate the activation of vimentin gene whose gene product in turn contributes to the metastatic potential of mammary tumors.

Oscillatory bursting of phasically firing rat supraoptic neurones in low-Ca2+ medium: Na+ influx, cytosolic Ca2+ and gap junctions.

1. Whole-cell patch recordings were obtained from supraoptic nucleus (SON) neurones in horizontal brain slices of adult male rats. Low-Ca2+ or Ca(2+)-free perifusion medium induced oscillatory bursting activity in all sixty-nine cells displaying both phasic firing and depolarizing after-potentials (DAPs). In fifteen non-phasic cells without DAPs, Ca(2+)-free medium produced little or no oscillatory bursting. 2. Typical bursts started with rapid membrane depolarization, resulting in a plateau with superimposed action potentials, and ended several hundred milliseconds later in swift repolarization. Prominent bursting was observed at membrane potentials from -50 to -70 mV, with maximum amplitudes of 12.2 +/- 0.7 mV (mean +/- S.E.M.) around -70 mV. Development of oscillatory bursting was dependent on reduction of [Ca2+]o, with a threshold for the bursting < or = 1.2 mM Ca2+. 3. Bursting was abolished by addition of TTX, Co2+, Ni2+ or Mg2+ into the Ca(2+)-free medium, or by replacement of external Na+ with choline or Li+. Low concentrations of TEA or increased [K+]o prolonged burst durations and enlarged oscillation amplitudes. 4. Voltage-clamp techniques were used to examine the persistent Na+ current (INaP), and revealed that low [Ca2+]o shifted the threshold for INaP activation in a negative direction and enhanced the amplitude of this current. These changes in INaP were abolished by adding Co2+ or Mg2+ to Ca(2+)-free medium. 5. Direct diffusion of BAPTA or heparin into neurones or bath application of ryanodine suppressed bursting. Oscillations were also eliminated by the uncoupling agents heptanol, halothane or acidification. 6. CNQX, APV, bicuculline, CGP35348 (GABAB receptor antagonist), promethazine, atropine, d-tubocurarine and suramin had no obvious effects on oscillatory bursting. Blockers of transient Ca2+, or hyperpolarization-activating cation currents also did not alter bursting activity. 7. These results suggest that intrinsic burst activity in SON neurons perifused with low-Ca2+ or Ca(2+)-free medium involves enhanced Na+ influx through persistent Na+ channels, and requires the presence of rapid intracellular Ca2+ mobilization that might also explain the selective existence of oscillatory bursting in phasically firing cells. Intercellular communication through gap junctions appears to be important in determining neuronal activity of the neuroendocrine cells in low-Ca2+ medium.

Multiple forms of tRNA(Lys3) in HIV-1.

tRNALys3 is the primer for HIV-1 reverse transcriptase (RI) and is selectively incorporated into HIV-1 during viral assembly. While whole cell extracts of uninfected or infected cells contain only one detectable form of tRNALys3, multiple forms of tRNALys3 are detected in the virus released into the cell culture media. These tRNALys3 isoacceptors are found in HIV-1 produced from newly infected cord blood lymphocytes and from cells chronically infected with HIV-1, such as the lymphocytic cell line H9 and the monocytic cell lines U937 and PLB. They can be detected through the use of either RPC-5 column chromatography of tRNA aminoacylated with radioactive lysine or northern blot analysis using a tRNALys3-specific DNA hybridization probe. Both RPC-5 chromatography and northern blot analysis show the cytoplasmic form of tRNALys3 to be the major abundance form of tRNALys3 in the virus. Starting with the viral RNA isolated from HIV (PLB), the tRNALys3 species resolved by RPC-5 into peaks 2, 3, and 4 were deacylated and 3' end-labeled by heat-annealing the RNA in each peak to synthetic HIV genomic RNA, and extending the hybridized species one base using HIV-1 RT and radioactive dCTP. An electrophoretic comparison of the partial T1 digest pattern of purified human placental tRNALys3 with those of the RPC-5 resolved species showed that the labeled RNA species in each peak was tRNALys3. These radioactive tRNALys3 species retained their relative mobilities when rechromatographed on RPC-5. When total HIV (PLB) RNA was used as the source of primer/template, and similarly extended with RT in the presence of radioactive dCTP, the major priming tRNA resolved by RPC-5 had a chromatographic mobility identical to peak 3. This tRNA primer has a T1 digest pattern identifying it as tRNALys3. These results indicate that the major tRNALys3 species present in the virus is also the major tRNALys3 isoacceptor used as the primer for reverse transcription.

Whole cell patch recordings from forebrain slices demonstrate angiotensin II inhibits potassium currents in subfornical organ neurons.

Whole cell patch clamp recordings have been obtained from SFO neurons in a forebrain slice preparation. Basic electrophysiological characteristics recorded from these cells in current clamp mode showed a mean resting membrane potential of -57.0 +/- 2.5 mV (+/- SEM, n = 7), mean input resistance of 900 +/- 110 M omega (n = 7), and a mean spike amplitude of 68.6 +/- 4.1 mV (n = 7), accompanied by either irregular or no spontaneous activity. All cells also demonstrated a delayed return to baseline membrane potential following large hyperpolarizing current pulses indicative of the presence of a rapidly activated transient potassium current in these neurons. Voltage clamp recordings identified both rapid transient, and a sustained outward currents which demonstrated the characteristics of IA and IK respectively. While bath administration of angiotensin II (Ang) (10(-7) M) was without effect on IK in 4 of 4 neurons tested, IA was reduced by between 20 and 100% in the same 4 neurons. These data provide the first description of the basic electrophysiological characteristics of SFO neurons recorded in forebrain slice preparations. They also provide the first direct evidence suggesting that Ang may exert its control over the excitability of SFO neurons through modulation of IA in these cells.

RAG mutations in human B cell-negative SCID.

Patients with human severe combined immunodeficiency (SCID) can be divided into those with B lymphocytes (B+ SCID) and those without (B- SCID). Although several genetic causes are known for B+ SCID, the etiology of B- SCID has not been defined. Six of 14 B- SCID patients tested were found to carry a mutation of the recombinase activating gene 1 (RAG-1), RAG-2, or both. This mutation resulted in a functional inability to form antigen receptors through genetic recombination and links a defect in one of the site-specific recombination systems to a human disease.
