[Study on observation indexes of rational dosage of atropine in treatment of acute organophosphorus insecticides poisoning].

In order to obtain nursing observation indexes in treatment of acute organophosphorus insecticides poisoning (AOIP) with rational atropine dosage, three nursing observation indexes: abnormal secretion, body temperature and pulse were summarized with the theory obtained from a earlier study in our hospital. The rational atropine dosage for treatment of AOIP should be enough to antagonize the accumulation of acetylcholine but should not cause too much body feedback regulation. Regulate atropine dose by changes of these three indexes, 171 patients with AOIP (Group A) were treated and observed, and compared with 184 patients (Group B) treated with traditional observating method (atropinization index). The results showed that the atropine dose used in Group A was markedly less than that in Group B (P < 0.001), the medication duration was shortened (P < 0.001), the complication and fatality rate were decreased (both P < 0.05). It indicated that these observation indexes are scientific and reliable, practical and of spreading value.

[Primary and secondary structure of 3'--end of the large subunit ribosomal RNA of silkworm Attacus ricini and evolution implications as inferred from the gene sequence].

The DNA sequence of the 3'-end of LSu-rRNA coding region of silkworm Attacus ricini was determined. By comparisons with the corresponding sequence of H. sapiens, X. leavis, H. momus, A. albopictus, D. melanogaster, C. elegants, S. pombe, M. musculus and M. racemosus, we found that the coding sequence we obtained is very conserved. Phylogenetic tree inferred by Neighbor-joining Method showed that the rate of evolution of insects was much faster than vertebrate. In addition, the secondary structure has also worked out it is highly conserved.

Responsiveness of cytosolic free calcium concentration in cultured rat pulmonary arterial smooth muscle cells: confocal microscopic measurement.

Rat pulmonary arterial smooth muscle cells were primarily cultured. alpha-actins in pulmonary arterial smooth muscle cells at day 7 of culture were observed by an immunofluorescence staining method using anti-alpha-actin antibody. Microfluorimetry of Ca2+ signals in fluo-3-loaded single smooth cell at day 7 of culture was performed by a laser-scanned confocal imaging system. The effects of several kinds of Ca(2+)-mobilizing drugs on cytoplasmic Ca2+ concentration ([Ca2+]i) were examined. KCl, a depolarizing agent, and norepinephrine, an alpha-adrenergic agonist, equally increased [Ca2+]i. Angiotensin II, a receptor agonist, and caffeine, a Ca(2+)-induced Ca2+ releaser, elevated [Ca2+]i in the same manner but was more potent than KCl and norepinephrine. Br-A23187, a Ca2+ ionophore, most potently increased [Ca2+]i. The present results suggest that drug receptors on plasma membrane, Ca2+ entry pathways and Ca(2+)-releasing mechanisms act normally, and that our cultured pulmonary arterial smooth muscle cells may be a good model for the study on the essential role of Ca2+ in vasoconstriction.

Sensitization of cis-platinum by a recombinant adenovirus vector expressing wild-type p53 gene in human ovarian carcinomas.

Mutations of the tumor suppressor wild-type p53 gene have been implicated in the development of resistance to anticancer drugs. We have examined the role of wild-type p53 in resistance to cis-diamminedichloroplatinum (II) (CDDP) in human ovarian cancer cells using a recombinant adenovirus containing human wild-type p53 cDNA (Adwtp53). In this study we used the human ovarian A2780 tumor cells (wtp53), which are sensitive to CDDP and A2780/CP tumor cells (nonfunctional/mutant p53) and are resistant to CDDP. Studies show that introduction of wtp53 protein via adenovirus gene transfer into A2780/CP cells significantly sensitized these cells to CDDP cytotoxicity, indicating wtp53 was involved in resistance to CDDP. We found that introduction of wtp53 protein also resulted in growth arrest of A2780/CP tumor cells whereas the parent A2780 cells were significantly less sensitive to Adwtp53. This synthesis of wtp53 protein induced by Adwtp53 in A2780/CP cells resulted in a significant increase in the expression of Bax protein without significantly effecting the expression of bcl2 protein, and induced a dose-dependent increase in the nucleosomal DNA fragmentation. The presence of CDDP further enhanced this apoptosis, causing a 30-fold sensitization of A2780/CP cells to CDDP. These results indicate that mutation of p53 protein in A2780/CP ovarian tumor cells resulted in the resistance to CDDP and that combination of wtp53 gene and CDDP may result in sensitization of mutant p53-containing tumors to chemogenetherapy.

Characterization of satellite cells derived from chickens with the low score normal (LSN) muscle weakness.

Myogenic satellite cell clones were established from the pectoralis major muscle of chickens with the low score normal (LSN) muscle weakness and controls. The percentage of cells which attached to substrata and began to proliferate was higher for the control line than the LSN line (55% vs 30%). Furthermore, of those clones which initiated growth, 63% of the control cells and only 32% of the LSN cells proliferated to confluence in 25 cm2 tissue culture flasks. Proliferation rates were significantly lower with LSN satellite cells than with controls (p < 0.05). LSN satellite cells were less responsive to the mitogenic effects of chicken serum (p < 0.05) and differentiation rates were lower compared with controls (p < 0.05). There was a greater (p < 0.05) number of insulin-like growth factor receptors on LSN satellite cells compared with controls. The IGF receptor binding affinities (Kds) between the two cell lines were similar (p > 0.05). The results suggest that a defect in satellite cell physiology may contribute to the skeletal muscle weakness seen in the LSN line.

Changes in intracellular and extracellular calcium concentration of the myocardial cells during heart failure in children.

OBJECTIVE: To explore the relationship between heart failure and changes in intracellular calcium concentration of the myocardium. METHODS: The intra-and extracellular concentration of ionized calcium and total calcium of myocardium in 11 cases of heart failure was measured using calcium fluorescence indicator Fura-2 and atom absorption spectrophotometry. The activity of the erythrocyte membrane pump was determined with hemolysate chemical method. RESULTS: The concentration of ionized calcium in myocardial cells and the erythrocyte was significantly higher in the patients with heart failure (280.85 +/- 47.8 nmol/L, 1.76 +/- 0.04 F335/F385) than in those without heart failure (121.88 +/- 13.15 nmol/L, 1.47 +/- 0.08 F335/F385). Total calcium in the erythrocyte was also increased markedly in the patients with heart failure, but the activity of the erythrocyte membrane pump was lower than in those without heart failure. The intracellular calcium of the peripheral erythrocyte and the activity of membrane pump returned to normal after the heart failure was cured. CONCLUSION: There is excessive calcium accumulation in the myocardium and erythrocyte and the latter may be cause of the disturbance of myocardial diastolic function during heart failure.

Expression of proliferating cell nuclear antigen in Wilms' tumor.

OBJECTIVE: To investigate the expression of proliferating cell nuclear antigen (PCNA) and its clinical significance in Wilms' tumor. METHODS: PCNA immunoreactivity on paraffin-embedded tissues was assessed retrospectively in 35 cases using labelled streptaridin biotin (LSAB) procedure, and its association with clinical and other biological features was studied. RESULTS: Twelve tumors (34.29%) expressed PCNA. Expression rate increased with the advance of stage (P < 0.05). The S-phase fraction and proliferation index (PI) in children with PCNA positive expression were higher than those in children with negative PCNA expression (P < 0.05). The survival rate (16.67%) in patients with positive PCNA expression was significantly lower than that (73.91%) in no-expression ones (P < 0.01). CONCLUSION: PCNA is a sensitive indicator for cell proliferation and is clinically useful in identifying a poor prognostic group of patients with Wilms' tumor.

Percutaneous transluminal coronary angioplasty for acute myocardial infarction: initial experience in 43 cases.

OBJECTIVE: To evaluate the initial experience with the use of percutaneous transluminal coronary angioplasty (PTCA) in the set of acute myocardial infarction (AMI). METHODS: Between October 1993 and April 1996, PTCA was performed in 43 patients (34 male, 9 female) with initial onset of AMI. These patients ranged in age from 40 to 74 years (mean +/- SD, 60.3 3 +/- 9.2). Primary PTCA was performed in 16, rescue PTCA in 8, and semi-elective PTCA in 19 patients. RESULTS: The initial success was achieved in 95.3% (41/ 43). Failed angioplasty occurred in 2 patients, with acute reocclusion after rescue PTCA (this patient died of cerebral stroke 3 days later) in one patient, and small local intimal dissection during semi-elective PTCA in another patient. CONCLUSION: PTCA was a safe and effective approach in the treatment of AMI in the present study patients, with a high primary success and low acute complication rate.

[The effects of vibratory acoustic stimulation on fetal heart rate and body movement of normal fetuses at different gestational ages].

OBJECTIVE: To study the effects of vibratory acoustic stimulation (VAS) on fetal heart rate (FHR) and fetal movement (FM) of normal fetu at different gestational ages. METHOD: 435 normal second and third-trimester pregnancies were studied using real time ultrasonography. FHR baseline was recorded. Then a five-second VAS was given and FM and (or) FHR was observed. The results of vibroacoustic stimulation testing (VAS-T), sound-provoked fetal movement (SPFM) and the amplitude of FHR acceleration were evaluated. RESULTS: (1) The fetus started to response to VAS from the 24th gestational week, but most could not reach the diagnostic criteria of VAS-T. The responses of FM were normal. (2) From 28 week on, 90% of the fetu showed marked FM and acceleration of FHR after VAS. Over 98% of the term babies were reactive. (3) The detective rates of SPFM appeared significantly earlier than that of VAS-T. CONCLUSION: It suggested that the development of fetal nervous system was matured at 28 weeks, and the development of motor nerves was earlier than that of the autonomic nerves.

[The role of TNF alpha in the allograft rejection of orthotopic rat liver transplantation].

OBJECTIVES: To measure tumor necrosis factors-alpha (TNF alpha) in rats on the 3rd, 64d days after orthotopic rat liver transplantation, and detect the expression of TNF alpha mRNA in the grafts. METHOD: Reverse transcription polymerase chain reaction (RT-PCR) was used. RESULTS: The TNF alpha serum levels on postoperative day 3, 6 were significantly higher in allograft group than in isogeneic graft group. RT-PCR analysis revealed that TNF alpha mRNA was present in allografts, while no TNF alpha mRNA expression occurred in isogeneic grafts. Our findings supported that TNF alpha plays an important role in the pathogenesis of acute allograft rejection, and suggested that sequential determinations of TNF alpha after operation may be of value in the prediction and diagnosis of acute rejection.

[The development of a microcomputerized fetal electrocardiograph].

A new microcomputerized Fetal Electrocardiograph (FECG) is presented in this paper. A two-grade configuration (front-end & back-end) is designed for the instrument. The front-end processor is based on a Digital Signal Processor (DSP) TMS32010 and the Adaptive Noise Cancelling (ANC) algorithm to extract FECG with non-invasive method. The processed results are sent to the back-end personal computer for real time display, analysis and record. Clinical tests show the instrument is designed smartly and operated easily, and can be applied widely to many medical situations.

[Ultrastructural study of tumor infiltrating lymphocyte in tissues of laryngeal squamous cell carcinoma].

By transmission electron microscopy tumor infiltrating lymphocyte (TIL) in laryngeal squamous cell carcinoma tissues of 14 cases was studied. The results showed that the infiltrating lymphocyte in varying degrees eristed in laryngeal squamous cell carcinoma tissues. Most of them were present in a motionless state, and the others showed the feature of activated metrocyte and closely contracted with cancer cells and resulted in distinct morphological changes both the cancer cells and themselves. Therefore, this study provided an important morphological evidence for TIL as killer and inhibiter to the growth of cancer cells.

[Effects of CD11/CD18 antibody on adhesive force between polymorphonuclear neutrophil and endothelial cell in response to burn serum in vitro].

UNLABELLED: Our previous studies have proved that a great number of polymorphonuclear neutrophils (PMN) adhered to endothelial lining and induced endothelial cell (EC) injury. This study was attempted to investigate the change in adhesive force between PMN and EC in response to burn patients' serum within 24 hours and effects of antibody against CD11/CD18 on PMN-EC adhesive force. Burn serum was isolated from 4 burn patients (III degrees 20%-50% TBSA) within 48 hours after burn injury. PMNs were isolated from 8 volunteers. Adhesive force between a PMN and a HUVEC was calculated by means of micropipette technique at 1 h, 3 h, 6 h, 12 h, 24 h after PMN or HUVEC incubated with burn serum or normal serum. Five individual PMN-HUVEC pairs were measured for each sample. All data were analyzed statistically using the t test. RESULTS: 1. The adhesive force between a single PMN-HUVEC pair increased sharply after PMN incubated with burn serum, which reached the maximun level at 1 h after incubation and remained at this level in the following 23 hours. However, the adhesive force was reduced by 78.5%, 72.5%, 75.2% at 1 h, 6 h, 24 h, respectively after the PMNs were pretreated with CD11/CD18 mAb; 2. The adhesive force between a single burn serum stimulated-HUVEC and PMN pair increased gradually, reached peak at 12 h and remained at this level in the following 12 hours. CONCLUSIONS: Burn serum could induce PMN-EC adhesion, with related to PMN aggregation in internal organs and EC damage after burn injury. Antibodies against CD11/CD18 could partially block PMN-EC adhesion and might reduce EC injury induced by activated PMNs.
