[Changes in various blood platelet function tests in rabbits exposed to whole-body irradiation].

With a selected spectrum of coagulation tests the functioning capacity of thrombocytes was investigated in rabbits exposed to a whole body irradiation by means of 60Co radiation with a LD 5/30. A reduced retraction could be proved for times of irradiation (the 5th, 8th, 11th, 21st, 35th, and 56th day). A reduced formation of malondialdehyde could be identified in thrombocytes on the 8th and 21st day after irradiation. No changes could be found in determining adhesiveness, platelet aggregation caused by ADP, and PF3A and PF3F tests. In the course of additional investigations (coagulation time in unprepared and siliconized glass tubes, thromboelastogramme, activated partial chromboplastine time), significant changes of coagulation time could be observed in siliconized glass tubes on the 8th, 11th, 21st, and 56th day following irradiation.

[Flavellagic acid influence on blood coagulation and hemostasis disturbed by whole body irradiation].

By experimentations in vitro with human blood, we found out that an addition of flavellagic acid influences the initial stage of the coagulation system by activating the XIIth coagulation factor. So there is another substance besides the ellagic acid that activates in solution the XIIth coagulation factor. Our experimentations on sound rats showed that an intravenous administration of flavellagic acid in a dose of 0.7 mg/kg body weight has a short-term hemostatic action and that the r and k values and the m epsilon value in the thrombelastogram are increased. There was a similar effect in rats receiving an intravenous injection of flavellagic acid on the twelfth day after a whole-body irradiation with 8.0 Gy.

Ceruloplasmin in Hodgkin's disease.

In 56 patients with Hodgkin's disease different clinical and histological stages the activity of polyphenoloxidase was repeatedly examined in the course of the disease as an indicator of serum ceruloplasmin concentration. In untreated patients the ceruloplasmin concentration was on average twice as high as in healthy controls. Repeated examinations made in the course of the disease demonstrated a good correlation of the values with the clinical condition of the patient, with the exception of some terminal stages and conditions following repeated chemotherapy, when liver damage was expected. The decrease to control levels in the course of therapy indicated the onset of remission. In contrast, increased levels indicated progression of the disease and necessity of treatment prolongation. Excluding intercurrent infections, higher ceruloplasmin levels above those found at remission were a clear sign of the progression. The correlation of ceruloplasmin values and erythrocyte sedimentation rate (1 h) was highly significant. The diagnostic and prognostic value of ceruloplasmin and/or serum copper determination is discussed.

Functional imaging of uptake of radiopharmaceuticals as a method for tumor visualisation in rats.

Dynamic studies of kinetics of 99mTcO4, 57Co-Bleomycin, 67Ga, A2 fraction of 57Co-Bleomycin in Yoshida sarcoma bearing rats were undertaken. Then the regional variations of rate of the initial uptake of a radiopharmaceutical were examined using functional representation of the rate constants as a function of position. It was concluded that--for radiopharmaceuticals with a high initial uptake rate--the method resulted in visualization of tumor and in suppression of the blood background, within five to eight minutes after administration. This approach based on an evaluation of the parameters of the radiopharmaceutical kinetics in organs and tumor makes possible the use of short-lived radionuclides for tumor visualization and could offer some possibilities of distinguishing the tumor from other accumulating organs.
