[Association of peripheral and central blood pressure with the alpha-adducin Gly460Trp polymorphism in a Chinese population].

OBJECTIVE: To investigate the association of peripheral and central blood pressure with the alpha-adducin Gly460Trp polymorphism in Chinese. METHODS: We randomly selected 6 villages from JingNing County, ZheJiang Province. We invited nuclear families to take part in our study. We measured each participant's blood pressure at the non-dominant arm by means of a standard mercury sphygmomanometer at subjects' homes. Five consecutive readings were averaged for analysis. Central blood pressures were obtained by use of SphigmoCor pulse wave analysis system. The observers administered a standardized questionnaire to collect information on smoking habits, alcohol consumption and use of antihypertensive drugs. Venous blood was sampled and the adducin genotype was determined by restrictive fragment length polymorphism (RFLP). RESULTS: Four hundred and forty-two subjects included 230 (52.0%) women, and 116 (26.2%) hypertensive patients, of whom 49 (11.1%) took antihypertensive drugs. The frequencies of alpha -adducin GlyGly, GlyTrp and TrpTrp genotypes were 21.3%, 54.5% and 24.2%, respectively. There was no association between the alpha-adducin Gly460Trp polymorphism and peripheral systolic and diastolic blood pressure and pulse pressure. However, both before and after adjustment for sex, age, age(2), body-mass index, current smoking, alcohol intake, and antihypertensive treatment, the alpha-adducin polymorphism was significantly (P < 0.02) associated with central systolic blood pressure and central pulse pressure. After adjustment, central systolic blood pressure (+/- SE) averaged 122.5 +/- 3.5, 114.1 +/- 1.5 and 109.1 +/- 1.8 mm Hg (P = 0.01) in the GlyGly, GlyTrp and TrpTrp subjects, respectively. The corresponding values for central pulse pressure were 39.4 +/- 1.3, 36.4 +/- 1.0 and 32.9 +/- 0.9 mm Hg (P = 0.002), respectively. CONCLUSIONS: In the JingNing population, the adducin 460Trp allele was associated with lower levels of central systolic pressure and pulse pressure.

[Association of single nucleotide polymorphism in matrix metalloproteinases promoter with susceptibility to ovarian cancer].

OBJECTIVE: To investigate the association of single nucleotide polymorphism in matrix metalloproteinase (MMP)-1 and MMP-3 promoter with susceptibility to ovarian cancer. METHODS: The genotype of MMP-1 and MMP-3 gene promoter region was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 122 ovarian cancer patients (ovarian cancer group) and 151 unrelated healthy women (control group). RESULTS: The frequencies of 2G and 1G alleles in ovarian cancer group were 68.0%, 32.0% and in control group 66.9%, 33.1%, with no significant difference between the two groups (P > 0.05); the genotype frequencies of 1G/1G, 1G/2G, 2G/2G in ovarian cancer group (16.4%, 31.1% and 52.5%) was not significantly different from that in control group (16.6%, 33.1% and 50.3%) (P > 0.05). Compared to 1G/1G genotype, neither 2G/2G nor in combination with 1G/2G genotype significantly modified the risk of developing ovarian cancer. The adjusted odds ratio was 1.05 (95% CI = 0.53-2.07) and 1.00 (95% CI = 0.52-1.90), respectively. The frequencies of 5A and 6A alleles in MMP-3 in ovarian cancer group were 17.2%, 82.8% and in control group 20.2%, 79.8%, with no significant difference between the two groups (P > 0.05). No significant difference in genotype (5A/5A, 5A/6A and 6A/6A) distribution between ovarian cancer and control groups was observed, either. Compared to 6A/6A genotype, 5A/5A plus 5A/6A genotype did not significantly modify the risk of developing ovarian cancer, the adjusted odds ratio was 1.34 (95% CI = 0.81-2.23). 2G allele of MMP-1 and 6A allele of MMP-3 were in linkage disequilibrium (chi(2) = 56.53, P < 0.01). CONCLUSION: MMP-1 and MMP-3 promoter polymorphism is not associated with the susceptibility to ovarian cancer.

[Adsorption behaviors of chitosan and the analysis of FTIR spectra].

This paper studied the adsorption behavior of lab-made chitosan for acid dyeing waste water and basic dyeing waste waters, based on the effects of adsorbing time, deacetylation degree of chitosan, and pH of waste water. The results showed that the adsorption of basic fuchsin and saffron by the chitosan almost reached equilibrium state in 20 min. The adsorption capacities of the chitosan for dyes was comparable with activated carbon. Moreover, its dosages were only 2/3 of the latter. The adsorption process for Acid Red presented a dynamics character of first order reaction. FTIR spectroscopy was used to analyze the adsorption mechanisms of chitosan for different kinds of dyes. It was found that a great deal of hydroxide radicals in the chitosan molecule participated in the adsorption of basic fuchsin and saffron.

Isolation and characterization of heterotrophic nitrifying bacteria in MBR.

The study presented the method for isolating the heterotrophic nitrifiers and the characterization of heterotrophic nitrification. Continuous tests via a membrane bioreactor (MBR) were operated under the controlled conditions to proliferate the nitrifiers. Heterotrophic nitrifying bacteria were isolated from the system in which the efficiency of total nitrogen (TN) removal was up to 80%. Since no autotrophic ammonium and nitrite oxidizers could be detected by fluorescence in situ hybridization (FISH), oxidized-N production was unlikely to be catalyzed by autotrophic nitrifiers during the heterotrophic nitrifiers' isolation in this study. The batch test results indicate that the isolated heterotrophic bacteria were able to nitrify. After 3 weeks incubation, the efficiencies of the COD removal by the three isolated bacterial strains B1, B2, and B3 were 52.6%, 71.7%, and 77.7%, respectively. The efficiencies of the TN removal by B1, B2, and B3 were 35.6%, 61.2% and 68.7%, respectively.

[Expression of KAI1, MRP-1, and FAK proteins in lung cancer detected by high-density tissue microarray].

BACKGROUND & OBJECTIVE: The genesis, development, invasion, and metastasis of tumor are closely correlate with alterations of multi-genes. At present, the roles of Kang-ai-1 (KAI1), motility-related protein-1 (MRP-1), and focal adhesion kinase (FAK) in lung cancer have seldom been reported. This study was designed to investigate the roles of KAI1, MRP-1, and FAK in tumorigenesis and development of lung cancer, and their values in diagnosis and predicting the prognosis of lung cancer. METHODS: The expression of KAI1, MRP-1, and FAK proteins in a high-density tissue microarray containing 240 spots were detected by SP immunohistochemistry. RESULTS: The positive rates of KAI1 and MRP-1 were significantly lower in primary lung cancer than in normal lung tissue (25.9% vs. 100%, 42.6% vs. 100%, P<0.05). The positive rate of FAK was significantly higher in primary lung cancer than in normal lung tissue (44.4% vs. 10.0%, P<0.05). The expression of KAI1, FAK, and MRP-1 in primary lung cancer had no correlation with age and gender of the patients, and macroscopic and histological type of tumor, but had correlations with tumor differentiation, clinical stage, and lymph node metastasis. In addition, the expression of MRP-1 had correlation with histological type of tumor; the positive rate of MRP-1 was significantly lower in small cell lung cancer than in non-small cell lung cancer (0 vs. 50.0%, P<0.05). KAI1 expression was negatively correlated to FAK expression (rs=-0.458, P<0.05); MRP-1 expression was positively correlated with KAI1 expression (rs=0.535, P<0.05), and negatively correlated with FAK expression (rs=-0.438, P<0.05). CONCLUSIONS: The abnormal expression of KAI1, MRP-1, and FAK proteins are related to invasion and metastasis of lung cancer. Combined detection of the 3 proteins may be useful in predicting the development and prognosis of lung cancer.

[Molecular cloning and expression of anti-tumor adhesion peptide (beta3)].

To block tumor cell adhesion, inhibit tumor metastasis and recurrence, the anti-adhesion peptide-trimeric beta peptide (DLYYLMDLSYSMKGGDLYYLMDLSYSMKGGDLYYLMDLSYSMK, beta3) was designed. The DNA fragment of beta3 was cloned into expression vector pET-His and the fusion protein His-beta3 was expressed in E. coli. BL21(DE3)plysS. After 1.5 hours' induction with IPTG, His-beta3 peptide was expressed significantly amounting to 10% of the insoluble proteins and 4% of the total proteins. 20mg of beta3 peptide was obtained from one litter culture medium after purification by using metal-chelating sepharose 6B FF. The purity of beta3 is 92.2% according to Gel-Pro analysis. The anti-adhesion effects of beta3 peptide, beta1 peptide (DLYYLMDLSYSMK) and GRGDS on the hepatocellular carcinoma cell line SMMC-7721 and the high metastasis hepatocellular carcinoma cell line HCCLM6 were studied. The result showed the beta3 blocked the adhesion of HCCLM6 cells and SMMC-7721 cells to fibronectin (FN) specifically. The inhibition effect was dose-dependent and time-dependent and the inhibition rate of beta3 was higher than three times concentration of beta1 and GRGDS. This suggested that pET-His-beta3/BL21(DE3)plysS was a suitable expression system for beta3, and the expressed beta3 specially inhibited the adhesion of cancer cells.

[SHP2 and MKP5 in P2Y purinergic receptor-mediated prostate cancer invasion].

OBJECTIVE: To investigate the effects of protein tyrosine phosphatase-SHP2 and dual-specificity MAPK phosphatase-MKP5 on the activation of MAPKs and cell invasion induced by P2Y purinergic receptor in human prostate cancer cell lines with different metastatic potentials. METHODS: The wide type (-wt) SHP2, mutant type (-cs) SHP2 and wide type (-wt) MKP5 cDNA expression vectors were constructed and stably transfected into 1E8 cells (highly metastatic) and/or 2B4 cells (non-metastatic). The tyrosine phosphorylation of SHP2 was examined by immunoprecipitation. The activation of ERK1/2 and p38 induced by P2Y receptor agonist ATP was analyzed by Western blot with phospho-specific antibodies against the dually phosphorylated, active forms of ERK1/2 and p38. The in-vitro invasive ability through Matrigel was measured by boyden-chamber assay. RESULTS: ATP induced significant SHP2 phosphorylation, which was stronger and lasted longer in 1E8 than in 2B4. SHP2-wt enhanced the ERK1/2 activation induced by ATP in 2B4 cells, while SHP2-cs delayed and decreased this effect in 1E8 cells. Both SHP2-wt and SHP2-cs had no obvious influence on p38 activation. ATP stimulated cell invasion of both 1E8 and 2B4, while transfection of SHP2-wt into 2B4 cells further increased the invasive-stimulating ability of ATP (18.7% increase compared with ATP treatment alone). Transfection of SHP2-cs into 1E8 cells, however, antagonized the invasive-stimulating ability of ATP (40.9% decrease compared with ATP treated group). Up-regulation of MKP5-wt inhibited phosphorylation of p38 by ATP and reduced cell invasion stimulated by ATP (22.4% and 28.7% decrease compared with ATP treated group of 1E8 and 2B4, respectively). CONCLUSIONS: Both SHP2 and MKP5 play some roles in P2Y receptor-mediated activation of MEK/ERK, p38 signaling pathways and prostate cancer invasion. SHP2 positively regulates ERK activation and prostate cancer invasion, whereas MKP5 inhibits the invasion by suppressing p38 activation.

[Association of thymidylate synthase polymorphism with esophageal squamous-cell carcinoma].

OBJECTIVE: To explore the influence of 5'UTR tandem repeat and 3'UTR 6 bp deletion polymorphism of thymidylate synthase (TS) gene on the development and lymphatic metastases of esophageal squamous-cell carcinoma (ESCC). METHODS: Peripheral leucocyte DNA was extracted from 232 ESCC patients and 348 age- and sex-matched healthy controls. TS 5'UTR and 3'UTR genotyping in all study subjects was performed by PCR fragment analysis and PCR-RFLP analysis, respectively. RESULTS: The distribution of TS 5'UTR and 3'UTR variants in ESCC patients was not significantly different from that in healthy controls. However, individuals with 6 bp+/6 bp+ and 3R/3R genotypes significantly reduced the risk to ESCC development compared to those with other genotype combinations (adjusted OR = 0.32, 95% CI = 0.08-0.92). In addition, the frequency of 2R/3R genotype in ESCC patients with lymphatic metastases (40%) was significantly higher than that in lymph node negative cases (14.7%) (chi(2) = 10.11, P = 0.001). Compared to 3R/3R genotype, the 2R/3R genotype significantly increased the risk of lymphatic metastases in ESCC (adjusted OR = 3.68, 95% CI = 1.54-8.93). CONCLUSION: The genotyping of TS 5'UTR and 3'UTR polymorphisms might be used as a stratification maker for predicting susceptibility to ESCC. The TS 5'UTR 2R/3R genotype might be a candidate molecular marker to predict the potential of lymphatic metastases in ESCC.

[Single nucleotide polymorphism in the matrix metalloproteinases promoter is associated with susceptibility to endometriosis and adenomyosis].

OBJECTIVE: To investigate the association of the matrix metalloproteinases (MMP) 1, 3 promoter single nucleotide polymorphism (SNP) with the susceptibility to endometriosis (EM) and adenomyosis. METHODS: The SNP of the MMP-1 and MMP-3 gene promoter region was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) among 100 endometriosis patients, 80 adenomyosis patients and 150 unrelated healthy women. RESULTS: (1) The frequency of 2G allelotype of MMP-1 in the EM and adenomyosis patients (79.0% and 79.4%, respectively) was significantly different from the control (67.0%) (P < 0.01). The frequencies of 1G/1G, 1G/2G and 2G/2G genotypes of EM and adenomyosis patients were significantly different from that in healthy controls (P < 0.05). Compared with 1G/1G genotype, both 2G/2G alone and in combination with 1G/2G genotype significantly increased the risk of developing EM (adjusted odds ratio was 3.65 and 3.25, 95% CI = 1.41-9.43 and 1.29-8.23, respectively), but only 2G/2G genotype significantly enhanced the risk of developing adenomyosis. (2) The frequencies of the 5A and 6A allelotype of MMP-3 among EM (14.0% and 86.0%, respectively) and adenomyosis patients (15.6% and 83.4%, respectively) were not significantly different from healthy controls (20.3% and 79.7%, respectively) (P > 0.05). The genotype distribution of 5A/5A, 5A/6A and 6A/6A in patients was not significantly different from controls (P > 0.05). Compared with the 6A/6A genotype, neither the 5A/5A alone nor in combination with the 5A/6A genotype significantly modified the risk of developing EM and adenomyosis. (3) The distribution of haplotype (1G/6A, 2G/6A, 1G/5A and 2G/5A) frequency of MMP-1 and MMP-3 SNP was significantly different between cases and controls. Compared with the 1G/6A haplotype, 2G/6A haplotype significantly enhanced the risk of developing EM, but not significantly enhanced the risk of developing adenomyosis. CONCLUSION: Individuals with the MMP-1 2G allelotype have significantly increased risk of developing EM and adenomyosis; MMP-3 promoter SNP is not associated with susceptibility to EM and adenomyosis, 2G/6A haplotype could be used as a stratified marker for EM.

Autologous cell based therapy for treating chronic infarct myocardium.

Recent experimental and clinical studies have shown that autologous cell based therapy using skeletal myoblasts or bone marrow-derived stem cells might have beneficial effects in chronic ischemic heart disease. The underlying concept is based on the repopulation of necrotic tissue by either readily contractile myoblasts or by bone marrow-derived stem cells. However, there is a need to resolve a number of issues for determining the better way to perform these treatments and, moreover, for assessing the real beneficial functional effect of each of these cell therapies. In this mini-review, we will discuss (i) the issues of the selection of chronic infarct animal to truly determine the impact of cell therapy on cardiac function recovery, and (ii) the evaluation of the bio-availability and the bio-distribution of transplanted cells. Some new investigational methodologies based on clinical end-points are also proposed.

[Relationship of interleukin-18 gene promoter polymorphisms with chronic hepatitis B in Chinese Han population].

OBJECTIVE: To investigate the polymorphisms of interleukin-18(IL-18) gene promoters, and to disclose whether such polymorphisms are associated with susceptibility to chronic hepatitis B in Chinese Han population. METHODS: Using polymerase chain reaction with sequence specific primers method, the authors detected the single nucleotide polymorphisms of the promoter region of IL-18 gene at position -607C/A and -137G/C in 231 patients with chronic hepatitis B and 300 normal controls. RESULTS: The frequency of CC genotype in IL-18 gene promoter region at position -607 was 0.22(66/300) in normal controls and 0.27(62/231) in patients. The frequency of CA genotype was 0.53(160/300) in normal controls and 0.50(116/231) in patients. The frequency of AA genotype was 0.25(74/300) in normal controls and 0.23(53/231) in patients. The frequencies of -137GG, GC and CC genotype were 0.67, 0.30 and 0.03 in normal controls respectively; whereas in chronic hepatitis B patients the frequencies were 0.79, 0.19 and 0.02. The genotype frequency of -137GG in chronic hepatitis B groups was significantly higher than that in normal controls(chi2: 8.55, P=0.003), but the frequencies of -607C/-137C and -607A/-137C haplotypes in chronic hepatitis B groups were significantly lower than those in normal controls. The association between genotype of IL-18 promoter region polymorphisms and hepatitis B virus(HBV) copies showed that the frequency of -607AA genotype in high HBV-DNA copies groups was lower than that in low HBV-DNA copies groups(chi2: 6.03, P=0.014). CONCLUSION: The polymorphisms of the promoter region of IL-18 gene at position -607C/A and -137G/C are correlated with chronic hepatitis B in Chinese Han population. The people with -137C allele in the promoter region of IL-18 gene may be protected against HBV infection, and the IL-18 -607AA genotype may be linked to HBV-DNA copy.

[Expression of urokiase-type plasminogen activator and matrix metalloproteinases in human tissue of abdominal aortic aneurysm].

OBJECTIVE: To explore the expression of urokinase-type plasminogen activator (u-PA) and matrix metalloproteinases in human abdominal aortic aneurysm (AAA) tissue. METHODS: Immunohistochemistry and in situ hybridization were employed to detect the protein and mRNA expression of u-PA, gelatinase A (MMP-2) and gelatinase B (MMP-9) in the tissue sections of 30 AAA specimens and 30 normal human abdominal aorta specimens. RESULTS: The expressions of u-PA and MMP-9 were mainly found in the macrophages in AAA tissue, but not in normal human abdominal aorta tissues. The expression of MMP-2 was found mainly in the smooth muscle cells in AAA tissues, but not in normal human abdominal aorta tissues. CONCLUSION: u-PA produced by macrophages may immediately activate proMMP-2 and proMMP-9, which play a pivotal role in the formation and development of AAA.

[Studies on the determination of protein with spectroscopic probe of o-chlorophenylfluorone-molybdenum (VI) complex in microemulsion medium].

The interaction and absorption spectral behavior of o-chlorophenylfluorone (o-CPF)-Mo(VI) and protein are studied in OP microemulsion medium. Under the optimum condition, the molar absorptivity of o-CPF-Mo(VI)-BSA system at 532 nm is 6.12 x 10(6) L x mol(-1) x cm(-1), and Beer's law is obeyed in the range of 0-14 microg x mL(-1) of BSA. Moreover, the binding number between the complex and BSA determined by molar ratio and slope ratio methods is 91. The mechanism of interaction of BSA and o-CPF-Mo (VI) is also discussed. OP microemulsion media introduced into the protein determination increased markedly the sensitivity of the system. The result of experiment indicates that the method possesses a high sensitivity as well as a high selectivity. It can be used to determine protein in human urine with satisfactory results.

Expression and significance of Survivin mRNA in lung cancer tissue microarray detected by FISH.

OBJECTIVE: To investigate the expression of Survivin mRNA in lung cancer tissue microarray (TMA) by fluorescence in situ hybridization (FISH) method, and determine the role and significance of it in lung cancer genesis and progress. METHODS: The expression of Survivin mRNA was detected by FISH method and TMA technology. Fifty-four cases of lung cancer and 10 cases of normal lung tissue were examined. RESULTS: Survivin mRNA was expressed in 66.7% (36/54) of lung cancer; the positive ratio of lung cancer was significantly higher than that of normal lung tissue (0/10; chi2 = 15.238, P < 0.05). The positive ratio of Survivin mRNA was significantly higher in poor differentiated cancer (20/24, 83.3%) than moderate and well differentiated cancer (16/30, 53.3%; chi2 = 5.40, P < 0.05). The positive ratio of Survivin mRNA was significantly higher in group with lymph node metastasis (27/32, 84.4%) than without lymph node metastasis (9/22, 40.9%; chi2 = 11.084, P < 0.05). The positive ratio of Survivin mRNA was significantly higher in stage III-IV(12/13, 92.3%) than stage I - II (24/41, 58.5%; chi2 = 5.066, P < 0.05). CONCLUSION: Survivin mRNA highly expresses in lung cancer, which is related to the progress and malignant behavior. Survivin may play a promoting role in lung cancer genesis and progress and provide a basis for estimating prognosis and treatment.
