Glycophorin variants and contents of sialic acid and total sulfhydryl groups on erythrocyte membranes of residents in a malaria hyperendemic area.

OBJECTIVE: To conduct a screening survey of glycophorin (GP) variants and observe the content changes of sialic acid (SA) and total sulfhydryl (SH) groups on the erythrocyte membranes among residents in a tertian malaria hyperendemic area of Guizhou Province. METHODS: GP variants were detected in the erythrocyte hemolysates of 173 local residents at two villages of Libo County by SDS-PAGE on 10% to 15% gradients gel and Western immunoblotting. Their SA and total SH group contents were estimated in erythrocyte membranes by spectrophotometric methods. 114 healthy subjects in Changsha and 49 individuals at a neighbouring village of the above area showing low morbidity of malaria served as normal and endemic controls respectively. RESULTS: Three distinct types of GP variants were found among 19 propositi in this hyperendemic area. The incidence of GP variants was 7.9% (8/101) at Yaolu Village whose population was mainly composed of Yao ethnic group; while that of Buyi ethnic group at Maolan Village was higher (15.3%; 11/72). The erythrocyte membrane contents of SA in residents at both villages exhibited a very significant tendency of decline (P < 0.01), whereas those of total SH groups increased prominently in residents of Yaolu Village only (P < 0.05). CONCLUSIONS: The frequency of GP variants in this hyperendemic area does not depend upon the severity of malarial prevalence. The evident reduction of SA contents in the residents may be related to the breaking down of the SA residues on membrane GPs by the invasion of Plasmodium vivax.

[Changes of thromboxane A2 receptors and their mRNA expression and effects of their antagonist phenol red in the lung of acute lung injury of rats].

OBJECTIVE: To investigate the changes of thromboxane A2 receptors and the influence of their antagonist phenol red, and to study in molecular level the mechanism of action of thromboxane A2 in acute lung injury. METHODS: Acute lung injury model of rats was produced by oleic acid, and phenol red (5 mg/kg, 50 mg/kg) was injected via vein 5 minutes before oleic acid injection. Changes of thromboxane A2 receptors and their mRNA expression in the lung were determined at 6 hours after oleic acid injection. RESULTS: Down-regulation of thromboxane A2 receptors were observed, but their mRNA transcription increased significantly in the lung after oleic acid injection. The injection of phenol red before oleic acid could induce down-regulation of thromboxane A2 receptors and abolish the increase of their mRNA transcription after oleic acid injection. Phenol red could reduce LBI, RLW and improve PaO2. Histological examination of lung tissues demonstrated that the degree of lung injury had been reduced significantly. CONCLUSION: Thromboxane A2 plays an important role plays in pathological processes of acute lung injury. Phenol red in vivo could effectively reduce severity of acute lung injury by blocking thromboxane A2 receptors and regulating the expression of thromboxane A2 receptors.

[E2-induced prolactin-secreting tumor of heteroplasty pituitary in renal capsule of the rats].

OBJECTIVE: A prolactin-producing tumor induced by 17-beta-estradiol (E2) in isotransplanted pituitaries under renal capsules of SD rats were studied. METHODS: Male SD rats (30 days old) were transplanted with an isologaus pituitary under renal capsule and treated with subcutaneous implantation of an empty or E2-laden silastic capsule. RESULTS: After treated with E2 for 60 days and 120 days, both the eutopic and ectopic pituitaries were more than three times heavier than those from control rats, accompanied by hyperprolactinemia, and the body weight of rats decreased significantly. The effects of E2 on the weight of eutopic pituitary and the concentration of plasma PRL were time-dependent. In situ hybridization was employed to measure the levels of PRL mRNA expression in cells from the eutopic and ectopic pituitaries 120 days after treatment of E2. The PRL mRNA contents in both the eutopic and ectopic pituitary cells were much greater than those in untreated pituitary cells. But PRL mRNA levels showed no significant difference between the eutopic and ectopic pituitary cells. CONCLUSIONS: Our previous data has shown that prolactinomas can be induced by chronic treatment of E2 in eutopic pituitaries of SD rats. In present study it appeared that E2 exerted similar effect on the ectopic pituitaries which were distant from the hypothalamus. Our results also demonstrated that E2 could promote the PRL gene expression in both the eutopic and ectopic pituitary cells, and there was no significant difference between them. Our data suggested the possibility of PRL-producing pituitary tumors could originate from anterior pituitary.

[Experimental study of vascular smooth muscle cell apoptosis induced by Huotong Capsule].

OBJECTIVE: To explore the anti-atherogenic mechanism of Huotong Capsule (HTC) on vascular smooth muscle cell (VSMC) apoptosis. METHODS: VSMC proliferation model of cultured human embryonal thoracic aorta was established by stimulating VSMC with interleukin-1 (IL-1) and HTC was applied to observe the apoptosis inducing effect by transmission electron microscopy, DNA gel electrophoresis and flow cytometry. RESULTS: Typical apoptotic cellular changes were found after HTC treatment, the VSMC shrinked, the nuclei disappeared after chromatin agglutination, cellular membrane projected and wrapped the organelle to form apoptotic body. DNA gel electrophoresis showed "ladder" strand of DNA, a special phenomenon of cellular apoptosis. And an Ap peak of degraded DNA could be seen by flow cytometry, which widened and heightened when the concentration of HTC increased. The above-mentioned changes were not found in the blank control and untreated model group. CONCLUSION: HTC could induce apoptosis of human proliferated VSMC.

[Effect of different processing conditions on content of myrisiticin, volatile oil and fatty lipid in semen Myristicae].

The effect of different processing conditions on contents of myrisiticin, volatile oil and fatty lipid in Semen Myristicae was studied by orthogonal design. The result shows that these processing conditions could not influence the contents of myrisiticin, volatile oil and fatty lipid in the processed products of Semen Myristicae, suggesting that processing does not necessarily lower toxicity.

[Water-soluble ginsenosides in American ginseng].

OBJECTIVE: To have further comprehension of American Ginseng on the basis of its water-soluble components. METHOD: The water suspension of 80% methanol extract of American Ginseng was sequentially extracted with ethyl ether and n-butyl alcohol. The saccharides in the water-soluble portion were removed by macro-reticular resin column, and three compounds were obtained by silica-gel column chromatography. Their structures were confirmed on the basis of spectral analysis(IR, NMR, MALDI-MS) melting points and optical degrees. RESULT: Three compounds were identified as malonyl ginsenosides Rb1, ginsenoside Rb1 and Re respectively. Among them Rb1 and Re are known, while malonyl ginsenoside Rb1 was isolated from American Ginseng for the first time. Its structure was elucidated as (3 beta, 12 beta)-20-[(-6-O-beta-D- glucopyranosyl-beta-D-glucopyranosyl)oxy]-12-hydroxylammar-24-en-3-yl-O- [6-O-(carboxyacetyl)-beta-D-glucopyranosyl)]-beta-D-glucopyranoside. CONCLUSION: Water-soluble ginsenosides were isolated from the water-soluble extract of American Ginseng suggesting that malonyl ginsenosides exist both in American Ginseng and Ginseng.

[Heat shock response in experimental acute middle ear bacterial infection].

OBJECTIVE: To investigate the association between bacteria antigen and inner ear antigen and effects of stress on the expression spectrum and level of the antigen bearing hsp70 related epitope. METHODS: Use monoclonal antibody against mammiferous hsp70 to recognize the related antigens of Bacillus pyocyaneus, Bacillus proteus, Bacillus coli, Staphylococcus aureus and Klebsiella pneumoniae with modified western blotting. The expression level of hsp70 in unstressed guinea pig inner ear was tested. An animal model of middle ear acute infection by Klebsiella pneumoniae was established. The dynamic changes of mammiferous hsp70 epitope related proteins were tested with modified western blotting. RESULTS: All the bacteria antigens recognized by anti-mammiferous hsp70 antibody were not of the hsp70 family and the expression level of hsp70 in normal guinea pig was extraordinarily low. Klebsiella pneumoniae expressed 70,000 dalton and 17,000 dalton hsp after its invasion into guinea pig middle ear. 70,000 dalton antigen expressed most intensely at 3rd day of invasion and then fell. 17,000 dalton antigen was expressed most intensely at 5th day of invasion and then fell. The expression of 70,000 dalton hsp was much weaker than 31,000 dalton hsp at acute periods of middle ear infection. CONCLUSIONS: The elevated expression of hsp both in bacteria and inner ear during middle ear acute infection provides material base for immune impairment on inner ear induced by bacterial infection. The low molecular weight hsp may be involved in the inner ear autoimmune impairment.

[The study on the attrition of molars with occlusal hypersensitivity molars in elderly people].

OBJECTIVE: To investigate the attrition of molars in old people who suffered from occlusal hypersensitivity. METHODS: 113 maxillary and mandibular molars of old people (47-74 years old) who had dentine hypersensitivity caused by attrition were studied. The Lavelle's criteria were used. RESULTS: When the attrition reached to the e type, the rate of lesion rose up quickly (19.47%), the highest incidence was seen in g type (41.59%). The hypersensitivity occurred as 48.67% in the first molar, 51.33% in the second molar, 53.10% in enamel-dentin junction, and 38.05% in dentine concave site. CONCLUSION: The rate of hypersensitivity rises with the severity of attrition. There is no significant difference between the first molar and the second molar, but the incidence of hypersensitivity in enamel-dentin junction is significantly higher than in dentine concave site (P < 0.05).

[The influence of side-to-side portacaval shunt plus enhancement of perfusion of hepatic artery on cirrhotic liver: an experimental study].

OBJECTIVE: To for improving the perfusion of liver, we designed an operation of portacaval shunt (PCS) plus enhancement of perfusion of hepatic artery (EPHA). METHOD: 48 wistar rats were divided into 4 groups randomly. group I (normal control), group II (liver cirrhotic control), group III (PCS on liver cirrhosis), and group IV (PCS + EPHA on liver cirrhosis). The EPHA was performed by the ligation of the left gastric and splenic artery. Liver function tests, liver biopsy, and nuclide hepatobiliary dynamic imaging (NHDI) were performed on the 2nd week and the 6th month after operation. RESULT: In NHDI, the peak time was short (P < 0.05) and the excretive rate was high (P < 0.01). The liver function and the proliferative degree of the fibrotic tissue of the liver were perfected significantly (P < 0.05) in the group IV compared with the group III after operation. CONCLUSION: PCS + EPHA can increase the perfusion of the hepatic artery, maintain the liver function and delay liver cirrhosis, and overcome the side effects of PCS. It is a new surgical technique in treating portal hypertension.

[Preoperative staging of esophageal carcinoma: comparison of endoesophageal ultrasonography, computerized tomography and conventional clinical staging].

OBJECTIVE: To evaluate the value of different methods in preoperative staging of esophageal carcinoma. METHOD: Thirty-six patients undergoing resection of esophageal carcinoma were examined with endoesophageal ultrasonography (EU) before operation. Eight of 36 patients could not be evaluated because of severe esophageal stenosis. Computerized tomography (CT) was performed in 21 patients. Comparison was made between stagings based on EU, CT as well as the conventional clinical staging system, respectively with surgical-pathological staging after resection. RESULT: The accuracy of conventional clinical staging for esophageal cancer was only 36%. The accuracy of CT in T, N and pTNM staging was 38.1%, 57.1% and 47.6% respectively. For EU, the figures were 77.8%, 72.2% and 72.2%, and were improved to be 85.7%, 78.6% and 83.1% when untraversable patients were excluded. CONCLUSION: While conventional clinical staging system lacks accuracy in evaluating esophageal carcinoma, CT makes only moderate improvement. Based on direct morphological examination of lesions, EU is highly accurate in preoperative staging of esophageal carcinoma, especially in evaluating the extent of luminal wall invasion and regional lymph node involvement. Therefore, EU is of critical value in the therapeutic selection for esophageal carcinoma.

[Comparative molecular field analysis(CoMFA) of allylamine and benzylamine antimycotics].

"Active analog approach" has been employed to search the pharmacophoric conformation of the allylamine and benzylamine antimycotics. A local minimum energy conformation, which is very similar to the crystallographically determined coordinate of naftifine or terbinafine, has been applied to build all compounds. Comparative molecular field analysis (CoMFA) has been used to examine the correlations between the activities against 6 common human pathogenic fungi and the physicochemical properties of 62 allylamine and benzylamine compounds which had been aligned by rms fit rule. The predictive abilities of the resulting 3D-QSAR models have been tested by 8 new synthetic compounds and 5 reported compounds. Finally, we propose an interaction pattern between allylamine, benzylamine antimycotics and the active site of pseudoreceptor based on the results of 2D-QSAR and 3D-QSAR.
