An orphan seven-transmembrane domain receptor expressed widely in the brain functions as a coreceptor for human immunodeficiency virus type 1 and simian immunodeficiency virus.

Both CD4 and an appropriate coreceptor are necessary for infection of cells by human immunodeficiency virus type 1 (HIV-1) and most strains of HIV-2. The chemokine receptors CCR5 and CXCR4 are the major HIV-1 coreceptors, although some virus strains can also utilize alternative coreceptors such as CCR3 to infect cells. In contrast, most if not all simian immunodeficiency virus (SIV) strains use CCR5 as a coreceptor, and many SIV strains can use CCR5 independently of CD4. In addition, several orphan seven-transmembrane receptors which can serve as HIV-1 and SIV coreceptors have been identified. Here we report that APJ, an orphan seven-transmembrane domain receptor with homology to the angiotensin receptor family, functions as a coreceptor for a number of HIV-1 and SIV strains. APJ was expressed widely in the human brain and in NT2N neurons. APJ transcripts were also detected by reverse transcription-PCR in the CD4-positive T-cell line C8166, but not in peripheral blood leukocytes, microglia, phytohemagglutinin (PHA)- or PHA/interleukin-2-stimulated peripheral blood mononuclear cells, monocytes, or monocyte-derived macrophages. The widespread distribution of APJ in the central nervous system coupled with its use as a coreceptor by some HIV-1 strains indicates that it may play a role in neuropathogenesis.

An apparent case of nonsymmetrical and sustained strand-specific hemimethylation in the Dc8 gene of carrot.

The Dc8 gene of carrot (Daucus carota L.) shows differential expression during embryo development. Changes in methylation patterns of a segment of about 500 bp (from base +120 to base -446) of Dc8 allele 6 were investigated by treating genomic DNA, extracted from embryogenic callus at different stages of development, with sodium bisulfite to modify nonmethylated cytosines. Following asymmetric (strand-specific) amplification, base sequences for samples from each developmental stage were determined for each strand directly from the PCR products or from cloned PCR products. Different methylation patterns were detected in the two strands. The 5' to 3' sense (coding) strand was almost completely nonmethylated, whereas almost all the cytosines in the 3' to 5' (template) strand were methylated. By 71 days after transfer to embryo-inducing medium, few methylcytosines remained; those that were present were generally near the TATA box or in a region beyond -300. The cytosines that were methylated were not limited to CG or CNG sequences. The difference in the extent of methylation between the two complementary strands implies either that there is a mechanism for strand-specific methylation, or that complementary sequences can differ greatly in sensitivity to bisulfite treatment or PCR amplification.

Localization of nicotinamide adenine dinucleotide phosphate-diaphorase reactivity and nitric oxide synthase immunoreactivity in the lumbosacral dorsal root ganglia in guinea pigs.

This study examined the distribution of reduced nicotinamide adenine dinucleotide phosphate diaphorase (NADPH-d) reactivity and nitric oxide synthase (NOS) immunoreactivity in the lumbosacral dorsal root ganglia (DRG) in male guinea pigs. A differential distribution of NADPH-d reactivity and NOS immunoreactivity was detected in neurons of DRG at different segmental levels. There were numerically more intensely stained NADPH-d and NOS reactive cells in the rostral (L1-L3) DRG compared with those at the caudal (L6-S4) levels. In the corresponding DRG, NADPH-d reactivity was not paralleled by NOS immunoreactivity. This was evidenced by the wide distribution of afferent neurons in the lumbosacral DRG stained for NADPH-d, yet only a small number of them exhibited NOS immunoreactivity. Double labelling study has shown that some of the NADPH-d positive neurons were NOS negative. Ultrastructurally, NADPH-d reaction product was associated with the membranes of various subcellular organelles, including the rough endoplasmic reticulum (rER), Golgi saccules, mitochondria and some segments of the nuclear envelop, whereas NOS immune-precipitate was patchily distributed throughout the cytoplasm. Present results suggest that nitric oxide (NO) may function as a neurotransmitter in the afferent pathways at lumbosacral segments. On the other hand, in view of their marked disparity in numbers and the lack of total one-to-one correspondence, it seems likely that the NOS positive neurons represent only a subpopulation of the NADPH-d positive cells in the lumbosacral DRG.

[A study on the relationship of primary hypertension in elderly and their quality of life in Changsha City].

To study the relationship between hypertension and their quality of life in Changsha City, in the elderly primary questionnaire concerning quality of life developed by the Chinese Geriatric Association was used to investigate 555 aged workers. A community-based control programme on hypertension and it's impact was analysed. The results of stepwise regression suggested that physical status and function of daily activities were both subjective and objective determinants of importance. Morbidity of hypertension was the highest among all chronic diseases influecing aged worker's health status. The total score of quality of life and activities in aged people with hypertension was obviously low. There were many problems in the community-based control program on hypertension in urban districts. The treatment on systolic hypertension has positive effect. Strenthening community-based control of hypertension has important impact on improving the quality of life among aged people.

[A study on the risk factors of none-insulin-dependent type diabetes mellitus].

A 1:1 matched case-control study was conducted to investigate the risk factors for NIDDM in 1995. A total of 100 pairs of case and controls matched on sex, age and hopital of admission. Bivariate analysis revealed 7 factors significantly associated with NIDDM. Conditional logistic regression analysis showed that 3 of the 7 fectors: obesity, greasy and family history of diabetes were risk factors for NIDDM. That might play important roles in the etiology of NIDDM.

[Molecular epidemiology of TT virus infection in some parts of China].

OBJECTIVE: To study the molecular epidemiology of new hepatitis TT virus (TTV) infection, its distribution in population and its role in pathogenesis of hepatitis in some parts of our country. METHODS: TTV DNA in serum samples was detected by nested-polymerase chain reaction, partial gene of various geographic strains of TTV was cloned and sequenced, and their genetic variation was analyzed. RESULTS: Forty-four of 112 cases with non-A to G hepatitis from Shenzhen, Guangdong province, Nanjing, Jiangsu province, Beijing and Shengyang, Liaoning province were positive for TTV DNA with a positive rate of 42.9%, but only three of 102 cases of hepatitis A-G was positive, with a positive rate of 2.9% (chi 2 = 42.8, P < 0.01). Positive rate of TTV DNA was significantly higher in blood donors with abnormal ALT but without infection markers of hepatitis A-G (34.6%) than in those with normal ALT (16.8%) (chi 2 = 4.5, P < 0.05). Sequencing analysis showed that more than 98% of their nucleotides were analogous between strains of TTVCHN002 from Shenzhen and Nanjing, and TTVSHB015 from Beijing, and more than 97% analogous between the above-mentioned strains and Japanese ones. CONCLUSION: There existed TTV infection in both north and south China. TTV infection correlated closely with abnormal ALT, which might be an important pathogen for non-A, non-B, hepatitis G. There were somebodies infected with TTV in the normal healthy population, similar to that of "chronic carrier status" in hepatitis B.

Negative correlation between cyclophilin mRNA level in leukocytes of renal allograft recipients and ciclosporin concentration in whole blood.

AIM: To study mRNA level of cyclophilin in the white blood cells (WBC) of the renal allograft recipients (RAR) and its correlation with ciclosporin concentration in whole blood. METHODS: The cyclophilin mRNA levels and beta-actin as controls in the WBC of 47 RAR were measured by quantitative reverse transcription polymerase chain reaction. The blood ciclosporin assay utilized monoclonal antibody fluorescence polarization immunoassay. RESULTS: With the increase of ciclosporin concentration in whole blood (from 62 micrograms.L-1 to 678 micrograms.L-1), relative cyclophilin mRNA level in the WBC of RAR decreased nonlinearly (from 1.1 to 0.03, r = 0.8195). CONCLUSION: There was a negative correlation between the mRNA level of cyclophilin in the WBC of RAR and the ciclosporin concentration in whole blood.

[Histological and electrophysiologic changes of injury of dorsal root ganglia].

Injury of dorsal root ganglia (DRG) may cause sensory and motor dysfunction. In order to investigate the changes of somato-sensory evoked potential (SEP) and histological characteristics of DRG in different causes and different periods of injury, fifty-two rabbits were chosed to build the models. The rabbits were divided into 4 groups: Control group (n = 4); mechanical compressing group (n = 16); inflammatory injury group (n = 16); and treatment group (2% lidocaine with hydroprednisone was administered locally, n = 16). After one to eight weeks, SEP was determined and samples of DRG were obtained to observe the histological and ultrastructural changes every week. The result showed that the gap junction of microvascular endothelium in DRG had been destroyed by the mechanical compression was the major cause of the vessel permeability increasing. The increasing of endothelial pinocytic vesicles transportation and widening of endothelial gap junction were the main causes of inflammatory irritation of DRG. The local infiltration with 2% lidocaine and hydroprednisone could obviously ameliorate inflammatory injury in DRG.

[The effect of growth hormone on wound healing rate in adult burns].

OBJECTIVE: To determine the wound healing effect of rHGH in adult burn patients. METHODS: 16 patients with burn wounds covering over 60% of total body surface (TBSA) were enrolled in this placebo controlled prospective study. They were comparable in nutrient intake, TBSA, and full thickness burn area. rHGH group patients were given rHGH subcutaneously in the dose of 0.3u.Kg-1 at 8 am each morning for 10 days beginning from POD 1. The control group patients were given normal saline as placebo. All the patients received scar excision within 4 days postburn, and the excision wounds were covered with autologous skin pulp grafting. Serum amino acid profile was analyzed at day 1 and day 20 post burn. Healing time of burn wound area and donor site was recorded. Wound healing rate was assessed at day 30 after day. RESULTS: 1. The healing time of autologous skin pulp grafting and donor site, and the length of hospital stay were significantly shorter in GH group patients than control group. 2. The amino acid profile showed no difference between two groups at day 1 and was significantly better in GH group at day 20. CONCLUSION: rHGH could enhance the wound healing rate, improve amino acid profile, and reduce the length of hospital stay of severe burn patients.

[Protein status and antioxidant capability of residents in endemic and nonendemic areas of Keshan disease(KD)].

From a survey of residents in endemic and nonendemic areas of Keshan disease(KD), it was found that the plasma cystine, tryptophan and lysine as well as blood selenium content were lower, while the plasma lipid peroxide was higher in endemic areas than those in non-endemic areas. These results suggests that the selenium deficiency coexisting with cystine and tryptophan insufficiency decreases the antioxidant defense of the organism and may be one of the important factors for the development of KD. The role of sulfur-containing amino acid in antioxidant defense is discussed emphatically.

[Clinical evaluation of several tumor markers in the diagnosis of primary hepatic cancer].

OBJECTIVE: To evaluate the practical significance of alphafetoprotein(AFP), alpha-L-fucosidase(AFu), gamma-glutamyltransferase(GGT), sialic acid(SA) and carcinoembryonic antigen(CEA) in the diagnosis of primary hepatic cancer(PHC). METHODS: AFP, AFu, GGT, SA and CEA in the serum were studied in 67 patients with PHC, 8 with hepatic metastasis of gastrointestinal cancer, 89 with benign liver diseases and in 131 controls without liver diseases. RESULTS: According to statistics, the sensitivity of AFP, AFu, GGT, SA and CEA in the diagnosis of PHC was 65.4%, 70.8%, 34.8%, 60.2% and 36.4% repectively; their specificity was 92.6%, 98.1%, 87.1%, 94.5% and 77.8% respectively. CEA in hepatic metastasis of gastrointestinal cancer was higher than that in the other groups of patients. Simultaneous detection of AFP, AFu GGT and CEA gave a diagnostic sensitivity and specificity in PHC of 81.4% and 91.6% respectively. CONCLUSION: AFP and AFu are superior to GGT, SA and CEA in the diagnosis of PHC; simultaneous detection of AFP, AFu, GGT and SA might increase the diagnositic sensitivity and specificity of PHC. CEA was less helpful in the diagnosis of PHC, but might be used to differentiate PHC from hepatic metastasis of gastrointestinal cancer.

[Antisense N-ras1 gene inhibits the proliferation of VSMC induced by bFGF after arterial injury].

OBJECTIVE: To develop a new way to prevent the process of restenosis after arterial injury, we transfer the recombinant antisense N-ras1 gene to the cultured SMCs and studied the influence of gene therapy with antisense N-ras1 on the SMCs proliferation induced by bFGF. METHODS: With the recombinant antisense N-ras1 plasmids (fpGv1-MT-N-ras1) transfered to the cultured SMC, we studied the effect on the SMCs proliferation induced by bFGF. RESULTS: The results of the experiment indicated that the count of cells transfered antisense N-ras1 was 16.8 +/- 1.3 x 10(4), the control cells transfered fpGv1-MT retrovirus vector were 30.1 +/- 1.2 x 10(4) (cell/ml) (P < 0.001). cell [3H] thymedine incorporation was 7,643 +/- 672 cpm in gene therapy cells, and 15,131 +/- 138 cpm in vector control cells (P < 0.001). Gene therapy cells ras mRNA and p21 were significantly decreased by Northern blotting and Western blotting technique. CONCLUSION: These results suggest that antisense N-ras1 gene may inhibit the proliferation of cultured VSMC induced by bFGF.

Effect of gynostemma pentaphyllum mak on carcinomatous conversions of golden hamster cheek pouches induced by dimethylbenzanthracene: a histological study.

OBJECTIVE: To evaluate the effect of gynostemma pentaphyllum mak (GP) on carcinomatous conversions of leukoplasia of the golden hamster cheek pouch induced by dimethylbenzanthracene (DMBA). METHODS: 284 golden hamsters, ranging from 4 to 8 weeks of age, 80 to 100 grams in weight were randomly divided into three groups: control, model and experimental groups. Control group (8 hamsters) were sacrificed at week 0 and week 12 for control. Model group (123 hamsters) were pasted in bilateral cheek pouches with 0.5% DMBA, 3 times a week for establishment of experimental leukoplasia, and sacrificed at week 2 to week 9 (12-22 hamsters were sacrificed a week). Experimental group (153 hamsters) were divided into three subgroups. Subgroup A underwent simultaneous DMBA pasting and oral taking of GP, and were sacrificed at week 6 and week 9. Subgroup B underwent oral taking of GP for 12 weeks after completion of DMBA pasting. Subgroup C underwent DMBA pasting after completion of oral taking of GP for 12 weeks. RESULTS: The earliest epithelial dysplasia and carcinoma in situ occurred at week 3 and week 6 after DMBA pasting. There were significant differences between subgroup B and model group for 3 weeks' administration of DMBA (P < 0.05), and between subgroup C and model group (P < 0.01), while there was no difference between subgroup B and model group for 6 week's administration of DMBA (P > 0.05). CONCLUSIONS: GP could inhibit and reverse the carcinomatous conversions of leukoplasia of golden hamster cheek pouches, indicating its positive anticarcinogenic effect.

[The expression of nm23-H1 and p53 protein in nasopharyngeal carcinoma].

UNLABELLED: To evaluate the role of nm23-H1 and p53 protein expression in Nasopharyngeal Carcinoma (NPC), the S-P immunohistochemical method was used to detect the expression of nm23-H1 and p53 protein in 125 cases of NPC. RESULTS: The positive expression for nm23-H1 and p53 were 50.4%(63/125), 90.4%(113/125), respectively nm23-H1 expression in the NPC with cervical lymph node metastasis and without cervical lymph node metastasis were 27.9%(17/61) and 71.9%(46/64), respectively (P < 0.001). CONCLUSION: 1. Mutations of p53 may play a role in carcinogenesis of NPC. 2. The difference between the NPC with cervical lymph node metastasis and without cervical lymph node metastasis for nm23-H1 expression was significant. The nm23-H1 gene may play a role in antimetastasis of NPC.

Clinical features of the femoral head necrosis caused by gross teres ligament after reduction for the developmental dislocation of the hip.

OBJECTIVE: To observe the clinical features of the femoral head necrosis caused by a gross teres ligament after reduction of developmental dislocation of the hip and study its pathological causes. METHODS: Ten hips with necrosis of the femoral head after reduction of developmental dislocation of the hip were observed. X-ray images of the hip were retrospectively reviewed with regard to the acetabular index, the acetabular-head index, the shapes and the development of the femoral heads. The operative findings were analyzed. RESULTS: No growth or delayed growth of the femoral head and the acetabular index was found after reduction. The recovery of acetabulum was very slow. All of the cases showed the same features: femoral head necrosis, subluxation, and acetabular dysplasia. A gross teres ligament was proved during surgical operation. The femoral head recovered gradually after the operation. CONCLUSIONS: The gross tissue interposed (a gross teres ligament) is another important pathogenesis of femoral head necrosis after the reduction of developmental dislocation of the hip. It is necessary to recognize it before reduction by imaging examinations.

Lidocaine improving the cerebral protection by retrograde cerebral perfusion.

OBJECTIVE: To determine whether lidocaine can improve the cerebral protection provided by retrograde cerebral perfusion. METHODS: Sixteen mongrel dogs were placed on cardiopulmonary bypass and cooled to 20 degrees C. Retrograde cerebral perfusion was then carried out for 120 minutes, with the external jugular venous pressure kept at 3.33 kPa. Cardiopulmonary bypass was resumed, and the animals were rewarmed to 36 degrees C. The animals were divided into two groups. In the lidocaine group (n = 8), lidocaine was administrated continuously throughout the experiment. In the control group (n = 8), normal saline was given at the same rate. RESULTS: In both groups, cerebral tissue creatine phosphate and adenosine triphosphate concentrations and energy charge increased by the end of hypothermic cardiopulmonary bypass, decreased continuously during retrograde cerebral perfusion, and recovered gradually after resuming cardiopulmonary bypass. Nevertheless, they recovered to significantly higher levels in the lidocaine group than in the control group (creatine phosphate: 2.44 +/- 0.53 versus 1.61 +/- 0.49 mumol/g wet weight, P = 0.006; adenosine triphosphate: 0.71 +/- 0.18 versus 0.50 +/- 0.17 mumol/g wet weight, P = 0.029; energy charge: 0.59 +/- 0.10 versus 0.48 +/- 0.09, P = 0.044) by the end of the experiment. There was no significant difference between the two groups in the cerebral tissue water content (control group: 77.6% +/- 1.9%; lidocaine group: 77.6% +/- 1.3%). CONCLUSION: Continuous lidocaine infusion accelerates the recovery of cerebral tissue high energy phosphate contents after resuming cardiopulmonary bypass, but it has no effect on the formation of cerebral edema after retrograde cerebral perfusion.

[The role of apolipoprotein A-I in the very early stage of experimental hyperlipidemia].

OBJECTIVE: To find out the change of serum total cholesterol, HDL cholesterol, LCAT activity, and the expression of apoA-I mRNA in liver and intestine in the very early stage of experimental hyperlipidemia of rabbits. METHODS: Experimental hyperlipidemia models were established by cholesterol feeding in NZW rabbits. Animals of the experimental groups were sacrificed by the end of the 1st, 2nd, 4th and 7th week respectively. Serum total cholesterol (TC), free cholesterol (FC), LCAT activity, HDL were assayed. Slot blot were used to analyze the expression of apoA-I mRNA in liver and intestine. RESULTS: TC and FC were noticed to be elevated at the end of the 1st week after cholesterol feeding. The increase of LCAT activity was proportional to the levels of serum lipid. Serum HDL-C level did not drop down by the 7th week of cholesterol feeding. Analysis of lipid infiltration area in aorta of the experimental group rabbits showed apparent lipid disposition in intima since the 4th week of cholesterol feeding. Slot blot results showed that apoA-I mRNA level increased at the end of the 1st week in the small intestine. The expression in liver was much weaker than in intestine. CONCLUSION: In the very early stage of experimental hyperlipidemia, serum TC, FC, LCAT activity increase remarkably and apoA-I, mainly produced in the small intestine, plays an important role in activating LCAT.
