The possible role of squalene and its peroxide of the sebum in the occurrence of sunburn and protection from the damage caused by U.V. irradiation.

To clarify the role of squalene peroxide in the occurrence of skin damage from sunburn, the optimum condition of squalene peroxidation and the effect of squalene peroxide on cutaneous tissue were examined. Peroxidation of squalene was more easily induced than palmitoleic acid and oleic acid in the unsaturated lipid occurred in sebum. The peroxidation of squalene gradually occurred by U.V. irradation, and it is parallel to increases in the malonyldialdehyde production (production of lipoperoxide). This peroxidation easily carries out in the case of high temperature (40 degrees C than 30 degrees C), and in the case of low pH. Good correspondence was recognized among the spectrum of natural daylight, U.V. absorption spectrum of squalene and erythema curve. Squalene and its peroxide have an important role in the occurrence of sunburn, and/or protection from damage caused by U.V. irradiation.

[Amenorrhea due to weight loss].

The cessation of the normal menstrual cycle caused by rapid loss of weight was defined as amenorrhea due to weight loss. All patients developed secondary amenorrhea and 80% of 243 patients were young, unmarried women. More than half of the patients had consciously restricted food intake for cosmetic reasons. LH-RH test revealed that, at onset, the basal levels of LH were low and the LH responses were impaired. The basal and the stimulated levels of FSH were comparable to normal. As the amenorrhea improved, the basal and the stimulated levels of LH rose to the normal range while the FSH responses became greater than normal. There was no significant difference between the rate of weight loss and the responsiveness to LH-RH. The rate of induction of ovulation with clomiphene was 95% of 40 patients with 1st grade amenorrhea, while only 3.9% of 54 patients with 2nd grade amenorrhea ovulated following treatment clomiphene. 80 percent of 19 patients with 2nd grade amenorrhea ovulated following gonadotropin therapy. Within a year after LH-RH stimulation therapy, a resumption of the normal ovulatory cycle or a return to 1st grade amenorrhea was observed in 60%.

Clinical effect of intravenous immunoglobulin on chronic idiopathic thrombocytopenic purpura.

High dose immunoglobulin infusions showed a marked effect on platelet counts in eight out of nine chronic ITP patients and in one SLE patient. In the comparison of different IgG-preparations, the pepsin treated IgG F (ab')2 showed no platelet elevation while the sulfonated did. The elevated platelet count could not be maintained after discontinuation of IgG infusions, but in six out of ten patients the platelet level remained above the pretreatment values. This new treatment seems to be safe and effective in adulthood ITP.

Slow reacting substance in the exudate of allergic air pouch inflammation in rats.

The activity of slow reacting substance was examined in the exudate of the allergic inflammation in rats after partial purification by alkaline decomposition and XAD-8 anion-exchange column chromatography. The guinea-pig ileum contractile substance from the exudate collected at 1 hr after the challenge injection was attributed to leukotriene (LT) C4 and/or D4 since the activity was selectively antagonized by FPL 55712. Participations of histamine, acetylcholine and prostaglandin E2 were ruled out. However, the contraction pattern due to the partially purified substance collected at 24 hr after the challenge injection was different from that at 1 hr. The guinea-pig ileum was contracted much more slowly than by synthetic LT C4 or D4 and this contraction was not antagonized by FPL 55712. It was demonstrated that some unidentified materials exist in the exudate of the allergic air pouch inflammation which contract isolated guinea-pig ileum in a different manner from synthetic LT C4 or D4.

The relation of tooth extraction to anginal attack.

This clinical study retrospectively compared the relationship of anginal attack during dental extractions with factors such as blood pressure, heart rate, rate-pressure product, and electrocardiographic findings. The anesthetic procedure produced the greatest change in the various parameters. No direct relationship could be observed between anginal attack and doses of lidocaine, epinephrine, and premedicative drug or time required for treatment.

Studies on the contractility of follicular wall with special reference to the mechanism of ovulation in hens.

To investigate the mechanism of ovulation in the hen, histological and physiological investigations on the contractility of the follicular wall were carried out. Smooth muscle bundles in the loose connective tissue coat from the stalk to the equator showed spontaneous rhythmic contractions with an alternation of contraction and pausing periods of about 4 min. The administration of prostaglandins F2 alpha (PGF2 alpha), E2 (PGE2), ovarian oviposition inducing factor (OOIF), oxytocin and acetylcholine increased the frequency and amplitude of the contractions. The theca layer showed spontaneous contractions at a frequency of about 30 per 10 min. PGF2 alpha, OOIF, acetylcholine, adrenaline and noradrenaline induced tonic contractions, whereas PGE2, oxytocin and histamine induced relaxation. It is suggested that the contraction of the follicular wall promotes the rupture of the stigma at ovulation and the neurotransmitters as well as prostaglandins and OOIF may be involved in this event.

Levels of inhibin-F activity and steroids in human follicular fluid from normal women and women with polycystic ovarian disease.

To examine inhibin-F activity (FSH-suppressing activity) in human follicular fluid of polycystic ovary (PCO) patients, 13 follicles from 5 documented PCO patients and an additional 31 follicles from normal women in the follicular phase of the menstrual cycle were sampled, and inhibin-F activity was measured in rat anterior pituitary cell cultures. Inhibin-F activity was measured in follicular fluid after stripping steroids from the fluids using treatment with dextran and activated charcoal. Estrogen, progesterone, and delta 4-androstenedione in the follicular fluid were also determined by RIA. Estrogen and progesterone levels in follicular fluid from PCO follicles 3.9 +/- 0.34 mm in diameter were comparable with those in follicular fluid obtained from viable follicles (which had a delta 4-androstenedione to estrogen ratio of 10 or less) from normal women. delta 4-Androstenedione levels in PCO follicles were higher (P less than 0.01) than those in viable and atretic follicles of normal women. Inhibin-F levels in PCO follicles were comparable to those in viable follicles, but significantly greater (P less than 0.01) than levels in atretic follicles of normal women. If inhibin-F levels in both atretic and viable follicles of normal women were pooled, the levels were less (P less than 0.05) compared to the level in PCO follicular fluid. As an additional control, follicular fluid was collected from 90 follicles of normal women throughout the menstrual cycle, and follicular size was determined as well as inhibin-F and steroid content. Small follicles less than 8 mm; (comparable in size to the PCO follicles examined) obtained at each stage represented 79%, 24%, 0%, and 94% of the total follicles obtained in the early to midfollicular, late follicular, preovulatory, and luteal phases of the cycle, respectively. Inhibin-F activity in the fluids of these follicles was less than that in PCO follicular fluid. The average inhibin content of all of the small normal follicles was 197 +/- 34 U/10 microliter, which was significantly less (P less than 0.05) than the level in PCO follicles (332 +/- 13 U/10 microliters). These data represent the first observation of inhibin-F activity in PCO follicular fluid and suggest the possibility of involvement of inhibin-F in bringing about low or normal basal levels of FSH in the presence of elevated basal LH levels often observed in PCO patients.

Glycosylation of 3, 5, 3'-L-triiodothyronine and its biological activity.

The formation of glycosylated Hb by the nonenzymatic reaction between Hb and glucose suggests that glycosylation is a general reaction in the living body, we demonstrated by means of TLC and HPLC that T3 was glycosylated when reacted with glucose in absolute ethanol for a week. We demonstrated in a metamorphic study in tadpoles that the biological activity of thus glycosylated T3 was less than about 1/10 of that of T3. These findings suggest that glycosylation of substances with amino group other than serum proteins including Hb may occur and that glycosylated substances may differ in biological activities from the original substances.
