Identification of ventral lateral geniculate nucleus cells projecting to the pretectum and superior colliculus in the cat.

Among 235 histologically identified cells of the ventral lateral geniculate nucleus (LGV) in the cat, 66 responded antidromically to electrical stimulation of the pretectum (PT) and/or superior colliculus (SC): 22 projected to PT, 22 to SC and 22 to both sites. The LGV cells were innervated by optic tract fibers corresponding to axons of X- as well as W-type retinal ganglion cells.

Selective localization of calpain I (the low-Ca2+-requiring form of Ca2+-dependent cysteine proteinase) in B-cells of human pancreatic islets.

An immunohistochemical study was performed to localize two distinct Ca2+-proteases (low-Ca2+-requiring calpain I and high-Ca2+-requiring calpain II) and their specific inhibitor (calpastatin) in human pancreas using the respective monospecific antibodies. Strongly positive staining by anti-calpain I antibody was found in pancreatic islets, specifically in B-cells, whereas the exocrine pancreatic tissue showed essentially no positive immunostaining. No such specific staining was found with anti-calpain II antibodies or anti-calpastatin antibodies. The results suggest that the Ca2+-dependent proteolysis in B-cells can be triggered by a small rise of the intracellular Ca2+ concentration without serious interference by the endogenous inhibitor.

Dentoskeletal and tongue soft-tissue correlates: a cephalometric analysis of rest position.

The relationship between dentoskeletal and tongue soft-tissue variables has been quantified in a sample of 60 adult females with normal and anterior open-bite malocclusions. Three lateral rest-position head films were obtained for each subject. A principal component analysis reduced the data base and six significant canonical correlations were identified. The first canonical correlation (r1 = 0.962) represented a size-related correlation factor between the two groups of variables. Subjects with characteristics of a short face syndrome and some evidence of overbite had tongue tips positioned below the lower occlusal plane (r2 = 0.929). In contrast, skeletal open-bite subjects (r4 = 0.759) revealed tongue tips ahead of and above the lower incisor teeth with the mandible in the rest position. Undererupted mandibular teeth (r5 = 0.666) were associated with a reduced tongue height and an inferior epiglottis; short tongue length (r6 = 0.563) correlated with a linear combination of upright central incisors, a small overjet, a low ANB angle, unerupted maxillary and mandibular teeth, and a steep occlusal plane. The multivariate statistical analysis extracted clinically significant associations between tongue soft-tissue and dentoskeletal variables. Tongue posture at rest in skeletal open-bite subjects appeared to be related to incisor position.

The stacking interactions in 7-methylguanine-tryptophan systems, a model study for the interaction between the 'cap' structure of mRNA and its binding protein.

Stacking interactions were shown by spectroscopic and X-ray crystallographic studies to be formed between the tryptophan and the protonated 7-methylguanine derivatives. These interactions would be in part responsible for the specific interaction between the 5'-terminal capped structure of mRNA and its binding protein.

A novel guanine-guanine base pairing: crystal structure of a complex between 7-methylguanosine and its iodide.

7-Methylguanosine, one of the biologically important minor nucleosides, could be crystallized as a complex of its zwitterionic form and its iodide, and the crystal structure was determined by the X-ray diffraction method. The crystals belong to the triclinic space group P1 with the unit cell dimensions: a = 7.678(1), b = 18.094(3), and c = 5.711(1) A, alpha = 79.32(1), beta = 80.14(1) and gamma = 76.90(1) degrees. The structure was solved by the heavy atom method and refined by the least-squares method to give a final R index of 0.075. The novel reverse Watson-Crick type base pairing observed between a positively charged molecule and a deprotonated one indicates that the deprotonation at the N(1) position promoted by the alkylation at the N(7) position may interrupt the formation of the normal Watson-Crick type GC base pair. The conformations about the glycosidic bond and the sugar puckering are quite different between the two molecules: the former has anti and C(4')-exo,C(3')-endo and the latter syn and C(1')-exo-C(2')-endo.

N-Sulfomethylation of guanine, adenine and cytosine with formaldehyde-bisulfite. A selective modification of guanine in DNA.

When guanine-, adenine- and cytosine-nucleosides and nucleotides were treated with formaldehyde and then with bisulfite, stable N-sulfomethyl compounds were formed. N2-Sulfomethylguanine, N6-sulfomethyladenine, N4-sulfomthylcytosine and N6-sulfomethyl-9-beta-D-arabinofuranosyladenine were isolated as crystals and characterized. A guanine-specific sulfomethylation was brought about by treatment and denatured single-stranded DNA with formaldehyde and then with bisulfite at pH 7 and 4 degrees C. Since native double-stranded DNA was not modified by this treatment, this new method of modification is expected to be useful as a conformational probe for polynucleotides.

The crystal structure of a major metabolite of thyroid hormone: 3,3',5'-triiodo-L-thyronine.

Two independent conformations of the thyroinactive thyroid hormone metabolite, 3,3',5'-triiodo-L-thyronine (rT3) were determined by X-ray diffraction methods. The conformations show significant difference in the lettering geometry when compared with those of the thyroactive thyroxine (T4) and 3,5,3'-triiodo-L-thyronine (T3). The diphenyl ether conformation of the two conformers of rT3 is an anti-skewed one, in which the torsion angles, phi (C5-C4-O4-Cl') are 8 degrees and -6 degrees, and phi' (C4-O4-Cl'-O6') are 86 degrees and 87 degrees. This conformation is in contrast to a twist-skewed one of T4 and T3. The difference in the binding abilities between T4, T3 and rT3 to thyroxine binding carrier proteins in serum or to a nuclear receptor protein may be explained by the characteristic solid-state conformations of these metabolites.

Analysis of body functions using a clinical liquid chromatograph.

A clinical liquid chromatograph, which consists of a completely automated liquid chromatograph combined with a microcomputer for diagnosis, and its application to body function analyses are described. The analytical rate for urinary ultraviolet-absorbing constituents using anion-exchange chromatography was 12 samples per day, and the reproducibilities for retention time and peak area were less than 3% and 4%, respectively. Diagnostic methods for kidney functions using the chromatograms are discussed.

Formation of cyanate and carbamyl phosphate by electric discharges of model primitive gas.

A mixed gas of nitrogen, carbon dioxide, and hydrogen was discharged over 100 ml of 0.2 M NaHCO3 solution in a 5 liter discharge apparatus which simulates the primitive Earth. The formation of cyanate, which is one of the possible primitive condensing agents, was demonstrated by the detection of [Cu(Py)2] (NCO)2 that was formed by the addition of copper sulfate-pyridine reagent to the solution. In a series of experiments the partial pressures of nitrogen and carbon dioxide in the starting gas were fixed at 10 cm Hg and 20 cm Hg, respectively, whereas that of hydrogen was varied between 5, 10, 15, 20, 25, and 30 cm Hg. The discharges were continued for one week. The rate of appearance of cyanate was strongly dependent upon the partial pressure of hydrogen. The maximum rate of the production of cyanate at the initial stage of the discharge was in the case of 10 cm Hg of hydrogen, in which condition the starting gas is in a predominantly oxidized state. In this case the concentration of cyanate reached about 0.012 M after one day. Another discharge experiment was carried out with 0.2 M phosphate solution, and the production of carbamyl phosphate was demonstrated through the formation of ATP by the incubation of the discharged solution with ADP and carbamyl phosphokinase.

Ureteric pain in patients with chronic renal failure on hemodialysis. Diagnostic approach with ultrasonography and computer tomography.

Chronically hemodialyzed patients who experienced ureteric pain were studied using ultrasonography and computer tomography. 6 of 45 dialysis patients complained of flank, lumbar or lower abdominal pain and 4 of these sonography or computer tomography. Ultrasonography was diagnostically superior to computer tomography. Renal stones were observed in 23 out of 45 cases by computer tomography. Ultrasonographic images of small renal stones were sometimes equivocal as compared with computer tomography, but low density stones, not shown by computer tomography, were shown by ultrasonography in a few cases. Therefore, both methods, in combination, are of great value in the diagnosis of asymptomatic renal stones as well as ureteral obstruction in the contracted kidney.

Synthesis and molecular conformation of 2',3'-O-isopropylidene-5'-deoxy-6(R),5'-cyclo-5,6-dihydrouridine.

The title compound (hereafter abbreviated as 6(R),5'-cyclo-hUrd) is synthesized from 2',3'-O-isopropylidene-5'-deoxy-5'-iodouridine and its molecular structure has been determined by X-ray analysis. 6(R),5'-Cyclo-hUrd crystallizes in space group C2 with Z = 4, and unit-cell dimensions a = 11.220 (2), b = 6.393 (1), c = 18,963 (3) A and beta = 107.98 (1) degrees. The structure was solved by direct interpretation of the three-dimensional Patterson function and refined to a final R index of 0.063. In the crystal the glycosyl torsion angle chi CN is 60.7 degrees (anti conformation) and the dihydrouracil ring adopts a half-chair conformation. The puckering of the ribose ring is an unusual O(1')-exo (P = 267 degrees, tau m = 47 degrees). The coupling constants of the 1H-NMR spectrum measured in C2HCl3 solution indicate that the overall conformation of 6(R),5'-cyclo-hUrd found in crystal is also maintained in solution. The theoretical calculations of coupling constants by the finite perturbation theory (FPT) intermediate neglect of differential overlap, self-consistent field molecular orbital (INDO SCF-MO) method indicate that the deviation of the observed coupling constants of sugar ring protons from those predicted by applying modified Karplus-type formula to the X-ray structure could be due to the strains around the sugar ring carbon atoms attached by protons.
