Three-dimensional model of the human craniofacial skeleton: method and preliminary results using finite element analysis.

The purpose of this study was to develop a three-dimensional finite element model of the craniofacial skeleton using a dry human skull. The model consisted of 2918 nodes and 1776 solid elements, and was used to investigate the biomechanical effect of a distally directed orthopaedic force on the craniofacial complex. The force was applied at the level of the maxillary first molar. The results indicated that in response to the force system applied: the nasomaxillary complex displaces in a backward and downward direction and rotates in clockwise sense; the nasomaxillary complex, including the zygomatic bone, experiences high stress levels in comparison with those at the remaining bones; the stress distribution in the maxillary basal bone area is relatively uniform; and the stress distribution across the opposing surface of the bony margins of the sutures is non-uniform.

Multiple forms of growth inhibitors secreted from cultured rat liver cells: purification and characterization.

It was found that a non-tumorigenic epithelial cell line from the liver of a Buffalo-strain rat (BRL) secreted into the culture medium various inhibitors of the growth of BRL and RSV-BRL (tumorigenic BRL transformed by infection of Rous sarcoma virus). The secreted inhibitors were classified into two types: one inhibited the growth of BRL to a greater extent than that of RSV-BRL (non-tumorigenic BRL growth inhibitor, NGI), and the other, vice versa (tumorigenic BRL growth inhibitor, TGI). Two NGI (NGI-I and NGI-II) and two TGI (TGI-I and TGI-II) were highly purified from the serum-free conditioned medium. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis without 2-mercaptoethanol, NGI-I and II gave protein bands with molecular weights (Mr) of 56,000 and 21,000, respectively. TGI-I and II gave a band that migrated faster than bromophenol blue marker dye, but they did not pass through an ultrafiltration membrane with an Mr cutoff of 5,000. In the presence of a reducing reagent, only NGI-II showed a decrease of Mr, from 21,000 to 11,000. NGI and TGI showed 50% growth inhibition with BRL and RSV-BRL, respectively, at 5-15 ng/ml in the medium containing 10% fetal calf serum. NGI and TGI all were stable to 1 M acetic acid (pH 2.3) and 6 M urea, but labile to 5 mM dithiothreitol or trypsin. Of the eight cell lines tested, NGI-I was most effective on BRL, NGI-II on BRL and HSC-3 (human tongue squamous carcinoma), and both TGI-I and II on RSV-BRL.

Distribution of calpains and calpastatin in human blood cells.

The occurrence and molecular sizes of calpains and calpastatin in the lysates of human erythrocytes, platelets, lymphocytes/monocytes, and polymorphonuclear cells were studied by immunoelectrophoretic blot analysis. The basic uniformity among these cells of the 85-kDa and 83-kDa heavy subunits of low- and high-Ca2+-requiring calpains I and II, respectively, and of the 29-kDa light subunit was confirmed. Molecular diversity of calpastatin species, ranging from 70 kDa to 107 kDa, among different blood cells was also shown. The obtained data are consistent with those known for other animal tissues, thus settling hitherto uncertain or rather controversial issues on the distribution of calpains and calpastatin in human blood cells.

Platelet factor XIII is activated by calpain.

The action of calpain (EC 3.4.22.17; Ca2+-dependent cysteine proteinase) on platelet factor XIII has been studied. Calpain I activated platelet factor XIII up to 76% of the maximum level observed with thrombin. Activation was accompanied by the limited proteolysis of the a subunit of platelet factor XIII to produce a 76 kDa fragment which was comparable to the proteolytic product by thrombin. Activation of platelet factor XIII by calpain was inhibited by EDTA, leupeptin, and endogenous calpain-specific inhibitor calpastatin. These findings suggest that calpain is responsible for the intracellular activation of platelet factor XIII.

High-performance liquid chromatographic assay of transglutaminase and its application to the purification of human erythrocyte transglutaminase and platelet factor XIII.

A high-performance liquid chromatographic method was developed for the assay of transglutaminase [EC 2.3.2.13] activity. Casein and dansylcadaverine were used as substrates and the reaction was stopped by adding an excess amount of EGTA. Casein-bound dansylcadaverine was separated from free dansylcadaverine by high-performance liquid chromatography on a TSK SW gel column on the basis of the differences in the molecular weight and hydrophobicity. The sensitivity was approximately 0.04 nmol of casein-bound dansylcadaverine in the assay mixture. With this assay method, human erythrocyte transglutaminase and platelet factor XIII were purified by successive chromatographies on DEAE-cellulose and Sephacryl S-300, which were common for both enzymes, followed by Blue Sepharose CL-6B and DEAE Bio-Gel A for erythrocyte transglutaminase or Phenyl-Sepharose CL-4B for platelet factor XIII. The purification factors and activity yields were 15,300-fold and 22% for erythrocyte transglutaminase and 43.8-fold and 33% for platelet factor XIII.

Specific localization of tissue-type transglutaminase in adrenocorticotropin-producing cells of the human pituitary gland as demonstrated by immunohistochemistry.

The distribution of transglutaminases, the Ca2+-dependent protein cross-linking enzymes, in the human pituitary gland was investigated by immunohistological methods using specific antibodies. Tissue-type transglutaminase was specifically localized in ACTH-producing cells, and the cells producing GH, PRL, TSH, FSH, and LH contained no appreciable amount of the enzyme. No detectable plasma-type transglutaminase (coagulation factor XIII) was found in pituitary tissue. In a previous study we demonstrated that ACTH-producing cells contain very little Ca2+-dependent proteinases (calpain), but a remarkable amount of their inhibitor, calpastatin. Pituitary gland cells producing hormones other than ACTH contained calpains, but no detectable calpastatin. These results collectively suggest that intracellular substrate proteins in ACTH-producing cells are protected from Ca2+-dependent degradation and are substrates for Ca2+-dependent cross-linking catalyzed by the tissue-type transglutaminase. In other pituitary gland cells, conversely, the intracellular substrate proteins are more likely to undergo Ca2+-dependent degradation than cross-linking.

Chemical control of Simulium ochraceum Walker (Diptera: Simuliidae) larvae in an onchocerciasis endemic area of Guatemala.

Chemical control against larvae of Simulium ochraceum, the principal vector of onchocerciasis, was carried out from 1979 to 1984 in a 91.3 km2 area of Guatemala where onchocerciasis is endemic. The control operation was divided chronologically into three phases according to the different tactics employed. Phase 1 (1979-1981), using briquettes of fat and detergent containing 10% temephos, was effective only in perennial streams. Phase 2 (1981-1982), which limited the application target to small streams with discharges of 0.1-1 litre/sec, was not effective. Successful control was achieved by Phase 3 (1982-1984), which consisted of fort-nightly applications of 5% temephos water dispersable powder in fixed doses of 24 g (1.2 g active ingredient) to every 50-100 m stretch of all streams with discharge rates of 0.1-50 l/sec. Vector biting rates were reduced by 97.8% in 1982 to 1983 and 97.6% in 1983-1984. The biting density of S. ochraceum at all five stations was reduced to less than 1.9/man/hour, the proposed critical level for long term transmission of onchocerciasis. The biting density of the S. metallicum/horacioi complex was not apparently affected by this operation. Through analysis of the density of S. ochraceum at various distances from the untreated areas, infiltration of the flies was found to be rare, if the distance was beyond 2 km. Fly-round surveys proved to be practical as a surveillance method for detecting larval breeding in untreated or improperly treated streams. The cost for a nation-wide vector control operation was presented.

Visual response properties of ventral lateral geniculate nucleus cells projecting to the pretectum and superior colliculus in the cat.

The visual properties of cells in the cat ventral lateral geniculate nucleus (LGV) identified antidromically from the pretectum and/or superior colliculus (projection cells) were studied in comparison with those of LGV neurons which could not be activated antidromically (non-projection cells). ON-phasic receptive fields (RFs) were relatively predominant in 27 projection cells, whereas ON-tonic RFs were found more commonly in the non-projection group. The distribution of the RF centers revealed a centroperipheral gradient of the visual field representation within the LGV that the central visual field was more densely organized.

Reversed distribution of calpains and calpastatin in human pituitary gland and selective localization of calpastatin in adrenocorticotropin-producing cells as demonstrated by immunohistochemistry.

The immunohistochemical distribution of Ca2+-dependent cysteine proteinases (calpains I and II) and their endogenous inhibitor calpastatin in normal and adenomatous human pituitary tissue was studied using specific antibodies. The distributions of calpain and calpastatin were dissimilar in human pituitary gland, i.e. ACTH-immunoreactive cells were strongly positive for calpastatin and negative for calpains. PRL-, GH-, FSH-, and TSH-producing cells were negative for calpastatin, but moderately positive for calpains, especially for calpain II, the high Ca2+-requiring form of the enzyme. Similar results were found in pituitary adenoma tissue. These findings indicate that each type of cells producing a specific hormone is equipped with a different balance of the enzyme-inhibitor system involved in the Ca2+-dependent degradation of intracellular proteins.

Geographical distribution of the prevalence of nodules of Onchocerca volvulus in Guatemala over the last four decades.

In Guatemala a national nodulectomy campaign has been carried out since 1935 in three operational areas, namely the Eastern, Central and Western endemic zones. The prevalence of nodules was calculated in each 1 km grid square in these zones in each decade and plotted on mesh maps. A total of nine foci of high endemicity, three in each operational area, was found in the decade 1940-49. Nodule prevalence was concentric in each endemic focus; high in the centre and gradually falling towards periphery. The foci were found mainly on Tertiary or Quaternary-Tertiary volcanic rocks in the Eastern and Central zones, or on non-carbonate sedimentary rocks in the Western zone. Nodule carriers were found at altitudes from 220 to 2200 m above sea level. The altitude of the foci of high endemicity varied, being low in the Eastern zone, medium in the Central zone and high in the Western zone. The nodule prevalence declined from the decade 1940-49 to the decade 1970-79 in most of the grid squares. The Santa Rosa focus in the Eastern zone virtually disappeared before 1970. The extent of endemic area having a nodule prevalence of 1% or higher decreased from 2384 km2 in the decade 1940-49 to 1424 km2 in the decade 1970-79. The area recommended for vector control operation against Simulium ochraceum was estimated at 803 km2.

Intestinal absorption enhancement of coenzyme Q10 with a lipid microsphere.

Coenzyme Q10 (ubidecarenone; CoQ10) was administered at a dose of 60 mg/d either using two conventional formulations or in a lipid microsphere to dogs. Plasma concentrations of CoQ10 were determined by HPLC. The bioavailability of CoQ10 from the lipid microsphere formulation was superior to conventional formulations. These results suggest that three daily administrations of the new formulation should be suitable for therapeutic use.
