[The relationship between immunosuppression and oxygen free radicals in burned patients].

The serum immunosuppression effect and its relation to oxygen free radicals were studied in patients with thermal injury (n = 11, total body surface area 20%-83%). The serum MDA, sIL-2R increased significantly in burned patients (P < 0.01 all). The serum of burned patients depressed the IL-2 productin of normal human PBMC (P < 0.01). Antioxidants decreased the serum sIL-2R in burned patients and attenuated the suppressive effect IL-2 production of normal human PBMC (P < 0.05 all). The results suggest that oxygen free radicals participate in the serum immunosuppression effect in burned patients.

[Ex vivo study of ultraviolet-B inactivating immunocompetence and hematopoietic potential of cord blood].

OBJECTIVE: To determine the differences of the effect of ultraviolet-B(UVB) on lymphocyte and hematopoietic progenitor cell in cord blood. METHODS: The viability, proliferative response and stimulatory activity in mixed lymphocyte culture (MLC), colony forming unit-granulo-monocyte (CFU-GM) colony growth and percentage of CD34+ cell of cord blood in Hanks medium after 0, 5, 10, 20, 50 mJ/cm2 UVB-irradiation were compared. RESULTS: Like the proliferative response and stimulatory activity, CFU-GM colony also showed UVB-irradiation dose dependent decrease. However, much less colonies were destroyed by UVB irradition less than 10 mJ/cm2. More obvious differences were found when 20%(vol/vol) bovine serum was added to the MNC medium. The percentage of CD34+ cell remained unchanged after UVB exposure. CONCLUSION: A lower dose of UVB exposure can slectively inactivate cord blood lymphocyte and antigen presenting cell while sparing hematopoietic progenitor cells. The data indicates the possibility of utility of UVB irradiation for graft-versus-host disease prophylaxis in cord blood transplantation.

[Chiral HPLC determination of conversion and enantiomeric excess of enzyme catalyzed stereoselective esterification of racemic ibuprofen].

In the study of enzyme catalyzed kinetic resolution of racemates, it is imperative to assay how the optical yield varies with chemical conversion. In this paper, a method using one-time injection to determine enantiomeric excess and conversion of the stereoselective esterification of racemic ibuprofen with n-butanol catalyzed by lipase was developed with a commercially available HPLC CSP column Regis(S, S) Whelk-01. In the linear range of detector, all peak areas of products and substrates are proportional to their concentrations. Because the total mole concentration remains unchanged (equal to the initial value of ibuprofen) in the reaction process, the conversion could be calculated from the peak areas, provided the ratio of response factors was known. The calibration curves of two ibuprofen enantiomers with racemic ibuprofen as external standard were overlapped, indicating fiR = fiS. By investigating the variation of peak areas of products and substrates against conversion (determined by external standard), the ratio of peak area-concentration response factor of ibuprofen butyl ester to that of unreacted ibuprofen was determined to be 1 through linear regressions, from which the conversion could be directly determined by the self normalization of the peak areas. With a mobile phase of IPA/hexane/HAc/triethylamine (15/85/0.2/0.05, V/V, flow rate 0.4 mL/min), the resolution of ibuprofen enantiomers was sufficient for precise enantiomeric purity determination.

[Enantiomeric high performance liquid chromatographic separation of epinephrine and analogous using chiral mobile phase additives].

The enantiomeric resolution of epinephrine, isoproterenol, ephedrine was studied by reversed phase HPLC. beta-CD, DM-beta-CD, TM-beta-CD were used as chiral mobile phase additives. The effects of the concentration of DM-beta-CD and the concentration of methanol on resolution of D,L-epinephrine were investigated. The results showed: D,L-epinephrine and D,L-isoproterenol can be separated by beta-CD, DM-beta-CD, TM-beta-CD, but D,L-ephedrine can't. Hydrogen bonding plays an important role in the chiral recognition mechanism. In the separation of isoproterenol, the enantioselectivity of TM-beta-CD was better than those of beta-CD and DM-beta-CD. As the concentration of DM-beta-CD changed from 0.04 mmol/L to 1.00 mmol/L, the resolution varied slightly. The experiment of the effect of methanol concentration on Rs showed: when the ratio of methanol/water was 40/60, the Rs reached the maximum. The coefficients of variation of retention time of D-epinephrine and L-epinephrine were 1.3% and 1.4% respectively. It is demonstrated that the chromatographic systems with a dynamically-generated stationary phase with methylated-beta-cyclodextrin are versatile tools for enantiomer separation. This technique leads to column with excellent time stability and good reproducibility of the enantio-selectivity.

Relationship between apoptosis of brain cells and free radical scavenging ability in rats during acute intermittent hypoxia.

It has been found that plateau hypoxia could induce apoptosis in rats' brain, but the mechanism was not clear. In order to study it further, we utilized acute intermittent hypoxia model to investigate reduced gliutathione (GSH), oxidized gliutathione(GSSG), super-oxide dismutase (SOD) and programmed cell death (PCD) or apoptosis (AP) in rats' brain. The results showed that with the increase of hypoxic exposure duration, apoptotic cells increased, the ratio of GSH/GSSG and the activity of SOD dropped. Regression analysis showed that there was a negative power relationship between the number of apoptotic cells and the ratio of GSH/GSSG or between the number of apoptotic cells and the activity of SOD in a limited range. It was concluded that there would be an intrinsic relationship between apoptosis and free radical scavenging ability in the organ.

Detection of human papillomavirus DNA and oncoprotein overexpression are associated with distinct morphological patterns of tonsillar squamous cell carcinoma.

Human papillomavirus (HPV) DNA has been detected in approximately 15% of squamous cell carcinomas (SCCs) of the head and neck. Recent studies have shown a predilection of HPV for certain anatomical sites, especially the tonsillar region, with viral DNA identified in approximately 60% of SCCs of the Waldeyer's tonsillar ring. This study was undertaken to determine whether there are differences in morphology or in oncogene expression in SCC of the tonsil with and without detectable HPV DNA. Twenty-two SCCs of the tonsil were analyzed for the presence of HPV DNA by polymerase chain reaction (PCR) using both a consensus primer set (My09/My11) and type-specific primers. Viral transcription was established in both primary and metastatic tumors by RNA in situ hybridization. The morphology of invasive SCC was classified into three subtypes: well keratinized (K-SCC), intermediate keratinized (I-SCC), and poorly keratinized (P-SCC). Expression of p53, pRB, and cyclin D1 (bcl-1) were studied by immunohistochemistry. In these cases (6 K-SCCs, 2 I-SCCs, and 14 P-SCCs), HPV DNA was detected in 14 (64%), with 11 containing HPV-16 (10 P-SCCs, 1 I-SCCs, and 0 K-SCCs) and 1 each containing HPV-33, HPV-59, and an unclassified HPV type (all P-SCCs). Viral oncoprotein E6/E7 transcription was demonstrated in 7 of 7 HPV-16-positive tumors. Cyclin D1 protein overexpression was detected in the majority of HPV-negative tumors (7 of 8 cases), whereas it was minimal or absent in 13 HPV-positive tumors. Overexpression of p53 protein was detected in 3 HPV-negative K-SCCs. In the HPV-positive tumors, fewer malignant cells expressed pRB and the staining was less intense than in the HPV-negative cancers. HPV DNA and E6/E7 expression, especially HPV-16, is detected in the majority of tonsillar SCCs and is almost exclusively associated with a poorly keratinized tumor histology. Decreased expression of cyclin D1, pRB, and p53 in tumors with HPV DNA is consistent with the known effects of the viral oncoproteins on the cellular protein. The morphology of the HPV-positive tumors suggests that HPV may have a predilection for a population of nonkeratinizing squamous cells or that the virally transformed cells inhibit keratinization of the tumor cells. Well keratinized tonsillar SCCs lack HPV DNA and are associated with overexpression of cyclin D1 protein and/or p53, suggesting that mechanisms that alter the cell cycle regulatory proteins, either by interaction with viral oncoproteins or by changes in the cellular proteins themselves, is critical for tumorigenesis of tonsillar SCC.

Osteogenesis of electrically stimulated bone cells mediated in part by calcium ions.

Culture selected and expanded osteoblastic cells may be able to be reintroduced into massive skeletal defects to accelerate cell mediated regeneration of skeletal tissues, especially in bone ingrowth in total joint replacement, fracture healing, and osteoporosis. In vitro osteogenic cell culture is a useful model in studying the mechanism of bone metabolism under direct current stimulation. In this study, an osteoblastlike cell line was isolated from newborn rat calvaria. The osteogenic processes of the in vitro cultured cell line were studied by cytochemical, electron microscopic, and energy dispersive x-ray analysis techniques that resembled those observed in membrane bone ossification centers in vivo. Direct current stimulation of 100 microA/cm2 accelerated greatly the proliferation and calcification of the in vitro cultured cells. Intracellular free calcium ion metabolism was measured with an Adherent Cell Analysis and Sorting Machine. Under direct current stimulation, intracellular free calcium ion concentration increased an average of 2.3 times of the original level, which may play a key role in regulating osteogenesis and bone metabolism.

[The types and distributive features of gene mutation on beta-thalassemia in three districts in northwest China].

OBJECTIVE: To sum up the results of gene analyses on beta-Thalassemia in three districts in northwest China and discuss the types and distributive features of the mutation. METHODS: Polymerase china reaction in combination with dot-blot hybridization of allele-specific oligonucleutide probes(PCR-ASO). RESULTS: In the gene analysis carried out in 85 probands with beta-Thalassemia, twelve types of gene mutation were identified from five nationalities. CD8(-AA), CDs8-9(+G), CDs27-28(+C) heterozygote and [-28(A-->uG). CD17(A-->uT)/N] double heterozygote were firstly assayed in these districts besides the common types in Chinese, of which, [-28(A-->uG). CD17(A-->uT)/N] as double gene mutation in the same chromosome is rare. CONCLUSION: There are significant differences in the distribution and nationality features of gene mutation types for beta-Thalassemia in the three districts, so the three districts as a whole is an area with special distributive features of the disease.

Targeted induction of collagenase and stromelysin by relaxin in unprimed and beta-estradiol-primed diarthrodial joint fibrocartilaginous cells but not in synoviocytes.

Temporomandibular joint (TMJ)-related diseases have a high female-to-male predilection and, unlike similar diseases of other joints, occur primarily during reproductive years. Although a role of female reproductive hormones has been proposed in the etiopathogenesis of these diseases, no direct evidence exists to link female reproductive hormones to TMJ disorders or to define the mechanisms by which these hormones may predispose to TMJ disease. Because relaxin, a 6-kd polypeptide hormone, alters the matrix composition of pubic symphyseal fibrocartilage and has been implicated in systemic joint hypermobility, synovial joints--particularly those with a large component of fibrocartilaginous tissues such as the TMJ--are potential but unproved target sites for its matrix-remodeling activity. The purpose of these studies was to determine the effects of relaxin on the expression of tissue-degrading enzymes, matrix metalloproteinases (MMPs), and their inhibitors, tissue inhibitors of metalloproteinases (TIMPs), in unprimed and beta-estradiol-primed TMJ disc fibrocartilaginous cells. Early-passage disc fibrocartilaginous cells were exposed to increasing concentrations of relaxin, and the cell-conditioned medium or mRNA was assayed for MMPs and TIMPs. Relaxin produced a dose-dependent induction of collagenase-1 (MMP-1) and stromelysin-1 (MMP-3), but minimal modulation of TIMP-1 and TIMP-2 expression in the fibrocartilaginous cells. Priming of these cells with beta-estradiol potentiated their MMP-inductive response to relaxin such that the maximal expression of collagenase-1 and stromelysin-1 occurred at 10- to 100-fold lower concentrations of relaxin in estrogen-primed than in unprimed cells. By contrast, beta-estradiol alone caused a dose-dependent decrease in these MMPs. Finally, relaxin's induction of collagenase-1 and stromelysin-1 was specific to the fibrocartilaginous cells, because in both unprimed and estrogen-primed synoviocytes, relaxin produced a dose-dependent decrease in these MMPs. These findings implicate relaxin alone, or in combination with beta-estradiol, in the degradative remodeling of the fibrocartilaginous disc and suggest a mechanism by which relaxin may predispose women to TMJ disease.

Ethnic stratification in northwest China: occupational differences between Han Chinese and national minorities in Xinjiang, 1982-1990.

The debate on market reforms and social stratification in China has paid very little attention to China's ethnic minorities. We explored rising occupational stratification by ethnicity in the Xinjiang Uygur Autonomous Region. Analyses of census data from 1982 and 1990 pointed to educational disadvantages faced by ethinic minorities as the most plausible explanation for the change. Multivariate analysis revealed a significant increase in the effect of education on high-status occupational attainment but no change in the effect of ethnicity. Net of education, ethnic differences in high-status occupational attainment were negligible. In contrast, large ethnic differences in manufacturing and agricultural occupations persisted after education and geography were statistically controlled.

A ribozyme-mediated, gene "knockdown" strategy for the identification of gene function in zebrafish.

The zebrafish system offers many unique opportunities for the study of molecular biology. To date, only random mutagenesis, and not directed gene knockouts, have been demonstrated in this system. To more fully develop the potential of the zebrafish system, an approach to effectively inhibit the expression of any targeted gene in the developing zebrafish embryo has been developed. This approach uses a transient, cytoplasmic, T7 expression system, injected into the fertilized zebrafish egg to rapidly produce high levels of a ribozyme directed against the mRNA encoded by the targeted gene to inhibit its expression. In a demonstration of this strategy, expression of the recessive dominant zebrafish no tail gene was effectively inhibited by using this strategy to yield a phenotype identical to that resulting from a known defective mutation in this same gene. This, ribozyme-mediated, message deletion strategy may have use in determining the function of genetic coding sequences of unknown function.

Inverse relationship between membrane lipid fluidity and activity of Na+/H+ exchangers, NHE1 and NHE3, in transfected fibroblasts.

This report presents a study of the effects of the membrane fluidizer, benzyl alcohol, on NHE isoforms 1 and 3. Using transfectants of an NHE-deficient fibroblast, we analyzed each isoform separately. An increase in membrane fluidity resulted in a decrease of approximately 50% in the specific activities of both NHE1 and NHE3. Only Vmax was affected; KNa was unchanged. This effect was specific, as Na+, K+, ATPase activity was slightly stimulated. Inhibition of NHE1 and NHE3 was reversible and de novo protein synthesis was not required to restore NHE activity after washout of fluidizer. Inhibition kinetics of NHE1 by amiloride, 5-(N,N-dimethyl)amiloride (DMA), 5-(N-hexamethyl)amiloride (HMA) and 5-(N-ethyl-N-isopropyl)amiloride (EIPA) were largely unchanged. Half-maximal inhibition of NHE3 was also reached at approximately the same concentrations of amiloride and analogues in control and benzyl alcohol treated, suggesting that the amiloride binding site was unaffected. Inhibition of vesicular transport by incubation at 4 degrees C augmented the benzyl alcohol inhibition of NHE activity, suggesting that the fluidizer effect does not solely involve vesicle trafficking. In summary, our data demonstrate that the physical state of membrane lipids (fluidity) influences Na+/H+ exchange and may represent a physiological regulatory mechanism of NHE1 and NHE3 activity.

Involvement of cdc2-mediated phosphorylation in the cell cycle-dependent regulation of p185neu.

We previously reported cell cycle-dependent negative regulation of p185neu (decreased tyrosine phosphorylation and kinase activity, with electrophoretic mobility retarded by serine/threonine phosphorylation) in M phase and the escape of mutation-activated p185neu* from this regulation. Our present results showed that retardation of electrophoretic mobility occurs independently of the cells' transformed status. We found that normal p185neu lost its ability to dimerize in the M phase. We demonstrated a physical association between cdc2 (a serine/threonine kinase, active in M phase) and p185neu. We showed that the carboxy terminal portion of p185neu is phosphorylated in vitro by cdc2. Many phosphopeptides (at least three phosphoserine residues) unique to the M phase were identified, and the in vivo and in vitro phosphopeptide patterns were superimposable. In contrast, mutation-activated p185neu* dimerized in the M phase with no changes in electrophoretic mobility, failed to associate with cdc2 and no unique phosphoserine residues could be identified in the M phase (data not shown), consistent with the escape of p185neu* from cell cycle-dependent regulation. Our results suggest that this escape is an intrinsic property of the mutation-activated p185neu* independent of its ability to transform cells. Our results also suggest the involvement of serine/threonine kinases such as cdc2 in the cell cycle-dependent negative regulation of p185neu.

Heat shock factor 1 represses Ras-induced transcriptional activation of the c-fos gene.

Heat shock factor 1, the critical molecular regulator of the stress response is conserved throughout eukaryotic organisms and activates the transcription of heat shock genes. We now show that heat shock factor 1 inhibits the expression of c-fos, an immediate early gene that controls responses to extracellular stimuli for growth and differentiation. Heat shock factor 1 inhibits the transcription of the c-fos gene and antagonizes the activating effects of the signal transducing protein Ras on the c-fos promoter and on the promoter of another Ras responsive gene uPA. This property was specific for heat shock factor 1; c-fos repression was not seen with the structurally related protein heat shock factor 2. Repression involved different molecular mechanisms compared with those involved in transcriptional activation by heat shock factor 1 and specifically did not require binding to the c-fos promoter. Thus, in addition to its known role as a transcriptional activator of the cellular heat shock response, heat shock factor 1 also antagonizes the expression of Fos, a key component of the ubiquitous AP-1 transcription factor complex and as such could influence multiple aspects of cell regulation.

Palliation of AIDS-related primary lymphoma of the brain: observations from a multi-institutional database.

PURPOSE: To catalogue the presenting symptoms of patients with AIDS who are presumed to have primary central nervous system lymphoma (PCNSL). To document the palliative efficacy of cranial irradiation (RT) relative to the endpoints of complete and overall response for the respective symptoms. METHODS: An analysis of 163 patients with AIDS-related PCNSL who were evaluated at nine urban hospitals was performed. These patients were treated for PCNSL after the establishment of a tissue diagnosis or on a presumptive basis after failing empiric treatment for toxoplasmosis. All patients were treated between 1983 and 1995 with radiotherapy (median dose-fractionation scheme = 3 Gy x 10) and steroids (>90% dexamethasone). Because multiple fractionation schemes were used, prescriptions were converted to biologically effective doses according to the formula, Gy10 = Total Dose x (1 + fractional dose/alpha-beta); using an alpha-beta value of 10. RESULTS: The overall palliative response rate for the entire group was 53%. In univariate analysis, trends were present associating complete response rates with higher performance status (KPS > or = 70 vs. KPS < or = 60 = 17% vs. 5%), female gender (women vs. men = 29% vs. 8%), and the delivery of higher biologically effective doses (BED) of RT (Gy10 > 39 vs. < or = 39 = 20% vs. 5%). In multivariate analysis of factors predicting complete response, both higher KPS and higher BED retained independent significance. A separate univariate analysis identified high performance status (KPS > or = 70 vs. KPS < or = 60 = 71% vs. 47%), and young age (< or = 35 vs. > 35 = 61% vs. 40%) as factors significantly correlating with the endpoint of the overall response. In multivariate analysis, high performance status and the delivery of higher biologically effective doses of irradiation correlated significantly with higher overall response rates. CONCLUSION: Most AIDS patients who develop symptoms from primary lymphoma of the brain can achieve some palliation from a management program that includes cranial irradiation. Young patients with excellent performance status are most likely to respond to treatment. The delivery of higher biologically effective doses of irradiation also may increase the probability of achieving a palliative response.

Nitric oxide synthase activity in renal ischemia-reperfusion injury in the rat: implications for renal transplantation.

Cadaveric kidney transplants with delayed graft function have poorer graft survival by an unknown mechanism. Nitric oxide, produced by nitric oxide synthase (NOS), has a proven role in both recovery of ischemia and promotion of rejection. We therefore wished to study the patterns of NOS activity in a model of renal ischemia. The left renal pedicle of Fisher rats was occluded for 1 hr. Both kidneys were removed at various times and frozen. Renal NOS activity was measured by conversion of [3H]arginine to [3H]citrulline and the content of endothelial NOS isoenzyme (eNOS) was compared by Western blot. NOS activity increased significantly in the left ischemic kidney over the first 24 hr, from a control of 33.8 pmol/min/mg to 79.8 at 2 hr and 56.8 at 24 hr. NOS activity then dropped below baseline, returning to near normal levels at day 21. eNOS content was stimulated over the entire time course, consistent with the presence of an eNOS inhibitor. Oral treatment with the NOS substrate L-arginine at 5 g/L significantly hastened the return of serum creatinine to baseline, if simultaneous contralateral nephrectomy was performed. The lazaroid U74389G given perioperatively also improved renal function and hastened recovery of NOS activity. Because nitric oxide plays an important role in maintaining blood flow during recovery from renal ischemia, the observed decrease in NOS activity may be prevented by perioperative treatment with oral L-arginine and corticosteroids. In addition, U74389G may provide a clinically useful method of minimizing and/or shortening DGF, thereby improving graft function and survival.
