Quantitative analysis of the development of genital organs from the urogenital sinus of the fetal male mouse treated prenatally with a 5 alpha-reductase inhibitor.

The role of 5 alpha-dihydrotestosterone (DHT) in the development of the genital organs and in the differentiation of the genital tract into prostate, coagulating gland (CG), bulbo-urethral gland (BUG) and seminal vesicle (SV) in male mice exposed prenatally to the 5 alpha-reductase inhibitor 6-methylene-4-pregnene-3,20-dione (6-MP) has been examined quantitatively. Female ICR mice were given 7 daily s.c. injections of the inhibitor (400 mg/day) starting on day 12 of gestation and the experiment was terminated on day 19 when the fetuses were removed by Caesarean section. In the prenatally 6-MP-exposed male mice the anogenital distance was significantly shorter than in the controls. Feminization of the nipples and hypospadias of the phallic urethra were noted. Development of prostate, CG and BUG was significantly suppressed. SV and testis development were not affected. These results lend further support to the conclusion that DHT is necessary for the development of the urogenital sinus (prostate, CG and BUG) and penis, and for the regression of the nipples in male mice. Reproductive abnormalities were not found in 90-day-old mice of both sexes exposed to 6-MP in utero. The 6-MP-exposed male and female mice had a normal reproductive capacity when mated with normal mice. These results show that 6-MP-induced growth retardation of reproductive organs is evident on day 19 of gestation, but that such retardation is no longer apparent in the adult.

Cell separation system studied by mixed culture of single wild strain with tetrads-forming mutant strain of Micrococcus luteus.

Mixed culture study of singly occurring wild strain IFO 3333 of Micrococcus luteus and a tetrads-forming mutant strain MT, in the absence or presence of trypsin, supported our previous assumption that at least two kinds of separation systems were involved in cell separation of M. luteus, the one having a physiological role in cutting off the outermost layer of the cell wall (separation system-Om) and the other in cutting off the inner layer of the "proper" cell wall or the septum (separation system-In). The separation system-Om of IFO 3333 insensitive to trypsin substituted, freely from the cells, for that of MT sensitive to trypsin.

Polyovular follicles in mouse ovaries exposed neonatally to diethylstilbestrol in vivo and in vitro.

In 35-day-old C57BL/Tw female mice given daily injections of 1 microgram diethylstilbestrol (DES) for 5 days from the day of birth, a significantly higher incidence of polyovular follicles (PF) were found in the ovaries than in those of age-matched control mice. Ovaries of prepubertal mice treated neonatally with oil or DES (DES mice) showed an enhancement of ovulation and luteinization following a combined treatment with eCG and hCG. Tubal ova in DES mice treated with eCG plus hCG were surrounded by many granulosa cells. Incidence of PF in control mice was not changed by eCG plus hCG treatment. In contrast, PF incidence in DES mice was reduced by prepubertal injections of eCG plus hCG. A high incidence of PF was also found in newborn mouse ovaries transplanted for 30 days into ovariectomized adult hosts given DES injections, but not in ovaries transplanted into intact or ovariectomized DES-untreated hosts. When neonatal ovaries were cultivated in a serum-free medium containing DES for 5 days and then transplanted into ovariectomized hosts, PF were formed in the grafts, but not in DES-unexposed grafts. Oocytes from PF in DES mice were found to have a smaller capacity for fertilization when examined in vitro. The present study also demonstrated that neonatal ovaries exposed to estrogen in vivo or in vitro (which produces PF in prepubertal hosts) are capable of responding to gonadotropins given later, resulting in a reduction of PF incidence, and that exogenous estrogen acts directly on neonatal ovaries to induce PF.

Abnormal development of the os penis in male mice treated neonatally with tamoxifen.

The os penis of male C57BL/Tw mice given 5 daily injections of 100 micrograms tamoxifen (Tx) starting on the day of birth (day 0) was examined at ages of 5-60 days; the bones of males given Tx injections for 5 days starting at 0-10 days and of those given neonatal injections of 100 micrograms clomiphene or nafoxidine were examined at 60 days. In the control males given the vehicle alone, the proximal segment of the os penis, composed of a compact cell mass found at day 0, developed at 5 days into the membrane bone with bone marrow and hyaline cartilage; the distal segment, composed of mesenchymatous cells until 10 days, developed at 30 days into fibrocartilage characterized by a distribution of type I collagen. By contrast, in Tx mice, fibrocartilage in the distal segment, and hyaline cartilage characterized by a distribution of type II collagen, and bone marrow in the proximal segment disappeared by 30 days. The maximum area of the proximal and distal segments gradually increased with age in control mice, whereas the proximal segment area remained unchanged in Tx mice. In clomiphene and nafoxidine mice at 60 days, the proximal segment was composed of hyaline cartilage; however, the distal segment lacked fibrocartilage. Hyaline cartilage in the proximal segment and fibrocartilage in the distal segment disappeared in all 60-day-old mice given Tx starting within 5 days. Neonatal castration did not suppress the formation of bone marrow and fibrocartilage in the os penis, though the bone size was smaller than in the intact controls. Formation of spines on the glans penis skin was suppressed by Tx given within 5 days.(ABSTRACT TRUNCATED AT 250 WORDS)

Chemical treatments of the cell packets induced from a Micrococcus luteus mutant and teichuronic acids on the packet surface.

Cell packets (MT packets) induced from a tetrads-forming mutant (strain MT) of Micrococcus luteus, both treated with chemical reagents and non-treated, were observed with a scanning electron microscope (SEM). The agglutinability of MT packets with antiserum containing anti-teichuronic acid antibody was examined. The binding of protein A-gold particles to the MT packets, mediated with the antiserum, was also observed with SEM. Gold particles were observed uniformly on the whole packet surface and also on the bridging structure formed by the outermost layer of the cell wall. Mild acid treatment, NaIO4-NaBH4 treatment and mild Smith degradation of the MT packets extremely decreased the agglutinability and binding of protein A-gold particles. The treatments gave a little influence on the surface feature and appreciably destroyed the regular packet structure. It was supposed that teichuronic acids distributed uniformly on the whole packet surface, naturally on the surface of the bridging structure too, and appreciably participated in the maintenance of the regular packet structure.

Morphometric analysis of the development of sexual dimorphism of the mouse pelvis.

Sex differences in the innominate bone of C57BL/Tw mice were studied morphometrically from the day of birth to 120 days of age. In neonatal male and female mice, a small cartilaginous spine was found on the basal part of ischium. This process disappeared in males within 24 hours after birth, whereas in females it remained until at least 30 days. Other sexual differences in the pubis and the ischium appeared at 30 and 120 days, respectively. The pubis in female mice was longer and thinner than that in the males, and the ischium in male mice was shorter and thicker than that in the females. Thirty-day-old female mice treated neonatally with testosterone or 5 alpha-dihydrotestosterone possessed pubic bones shorter and thicker than those of the age-matched untreated females. Pubes in male mice castrated at the day of birth were thinner than those in intact males. These findings suggest that the shape of the innominate bone is transformed to the male type under the influence of early postnatal androgen.

Immunological mechanisms of antitumor activity of some kinds of crude drugs on tumor necrosis factor production.

Previously, we described the antitumor activity of four crude drugs, A. capillaris, S. doederleinii, A. macrocephala and S. subprostrata. Tumor necrosis factor (TNF) can be produced in animals by injecting Bacillus Calmette-Guerin (BCG) or Corynebacterium parvum (CP) as priming agents and endotoxin (LPS) as an eliciting agent. However, because of their severe toxic action, it is inappropriate to use these agents to induce intrinsic TNF in cancer patients. In the present paper, we began with the investigation of a fluorometric method using ethidium bromide (EB) to assay TNF activity in serum in vitro, and then examined whether the four crude drugs displayed the priming and eliciting activities for TNF production. The TNF activity in a serum sample could be titrated using the fluorometric method in addition to a method using 3H-thymidine. The fluorometric method was superior to the radio-method as regards simplicity, safety and reproducibility. A. capillaris, S. doederleinii and S. subprostrata showed priming activity for TNF production in mice without the liver and spleen hyperplasia seen in the case of CP, S. subprostrata showed an eliciting activity in CP-primed mice. These results suggest that the crude drugs might be useful as inducers of intrinsic TNF in cancer patients.

Mechanisms of antitumor activity of aqueous extracts from Chinese herbs: their immunopharmacological properties.

Previously we reported the antitumor activity of 4 kinds of Chinese herbs: A. capillaris, S. doederleinii, A. macrocephala and S. subprostrata. Evidence has been also presented that A. capillaris shows the activity mainly through a direct tumoricidal action and the others display the activity through the enhancement of tumor-immune response. The present results are as follows: A. capillaris did not affect Meth A tumor-neutralizing activity (Winn's assay) in spleen cells of BALB/c mice bearing the primary tumor. The other herbs enhanced the activity on days 10 and 15 and prevented the decay of the activity on day 20 after the Meth A tumor transplantation. However, none of them enhanced the local graft versus host reaction (GvHR) induced in CBF1 mice by spleen cells of BALB/c mice. A. capillaris hardly affected the humoral antibody formation against sheep red blood cells in mice. On the contrary, the other 3 herbs enhanced the formation. Blast transformation of murine lymphocytes with PHA-P or LPS was suppressed by 10(-5) to 10(-4) g/ml of A. capillaris. A. macrocephala enhanced the response by LPS. It should be noted that A. capillaris showed no immunosuppressive action in spite of having a direct tumoricidal action in vitro. Further investigations will be required on the latter 3 herbs to clarify the relationship between their humoral immunity-stimulating activity and their tumor immunity-enhancing activity.

Immunological mechanisms of antitumor activity of some kinds of Chinese herbs: Meth A-induced delayed type hypersensitivity.

In the present paper, we confirmed that a delayed type hypersensitivity response can be elicited against Meth A tumor (Meth A-DTH) in BALB/c mice bearing the primary tumor. This response was augmented by lipopolysaccharide. We examined the effects of 4 kinds of Chinese herbs including A. capillaris, S. doederleinii, A. macrocephala and S. subprostrata on the Meth A-DTH, and the results were compared with that of the herbs on picryl chloride-induced delayed type hypersensitivity (PC-DTH). All of the herbs examined augmented the Meth A-DTH 10 days after the primary tumor transplantation, and S. doederleinii, A. macrocephala and S. subprostrata prevented the decay of the response on the 20th day, but A. capillaris did not. On the other hand, none of the herbs affected the PC-DTH. When both DTH responses were caused simultaneously in the same mouse, Meth A-DTH decayed 20 days after the transplantation but PC-DTH did not. In this case, the effects of these 4 herbs on Meth A-DTH and PC-DTH were essentially the same as those seen in the case of separate experiments. The previous and present results suggest that A. capillaris shows antitumor activity mainly through a direct cytotoxicity, although this herb might have certain components to enhance Meth A-DTH, and the other herbs display the activity through the enhancement of T cell-mediated tumor immunity, particularly tumor specific DTH.
