[Determination of the selenomethionine content in grain and human blood].

The authors used the method of cyanogen bromide-fulorimetry to determine the trace amounts of bound selenomethionine (SeMet) in corn, rice, wheat, soybean and human blood. The contents of SeMet in corn samples were found to be 9.2-19014.1 ng/g, and 45.5% to 82% of the total Se in corn samples were in the form of SeMet. Like corn, the proportion of Se in the form of SeMet in total Se in rice, wheat and soybean were 54.9%-86.5%, 50.4%-81.4% and 62.9%-71.8% respectively. The results showed that SeMet is the major chemical form of Se in grain samples determined. The contents of SeMet in two human blood samples (Se content 56.4 ng/g and 71.8 ng/g) were determined as 28.3 ng/g and 53.4 ng/g respectively.

[The Chinese total diet study in 1992--macro elements and trace elements].

Ten macro elements and trace elements were directly determined in the 1992 total diet study conducted in four districts of China from north to south. The intake of these elements was evaluated. The levels of intake for seasonal variation and for different age groups were reported. The results showed that the intakes of Ca, P, K, Mg, Zn, Se and Cu were low in children. The intake of Ca, P and Zn was insufficient for adults. The intakes of other seven elements reached or approached their RDA levels, but there were big differences among the 4 districts. The relationships between intakes of Ca/P and Na/K as well as Fe and Cu and iron difficiency anemia were discussed.

[Drilling decompression of the proximal tibia for osteoarthritis of the knee].

OBJECTIVE: To investigate the effect of drilling decompression of the proximal tibia for the Osteoarthritis of the Knee. METHOD: The drilling decompression of the proximal tibia were performed in 21 patients (38 knees) of osteoarthritis with 99mTc-MDP scintingraphy and the intraosseous phlebography made pre-postoperatively. Knee function scores and scales were evaluated. RESULT: During 6-12 months follow-up, the preoperative score of the knee function was 18-48 (average 30.78) and the postoperative score was 61-96 (average 87.95); the rates of fair and good of knee scales rose from 5% preoperatively to 86.84% postoperatively. CONCLUSION: Drilling decompression of the proximal tibia is a simple, safe, less traumatic and effective method to relieve the pain and improve the function for the early osteroarthritis of knee, For late cases it is a method to relieve pain. 99mTc-MDP scintingraphy can be used as a method to diagnose the early stage of the osteoarthritis.

[The expression of proliferating cell nuclear antigen and p53 protein in salivary gland tumours].

The expressions and distributions of PCNA and p53 protein were studied with anti-PCNA and anti-p53 protein MoAbs, using immunohistochemical staining method in salivary gland tumour. The relationship between PCNA and p53 protein was also studied. Results indicated: Malignant mixed tumour (MmT) had a higher proliferating index (PI) of PCNA. The PI could be used as one of adjuvant diagnostic criteria; and as an important parameter for grading mucoepidermoid carcinoma (MEC); The positive expression of p53 might be a good tumour marker for MmT; Tumour tissues with positive expression of p53 had been shown higher PI of PCNA, however there was no significance difference in our group; It remained to be studied later.

[Immunohistochemical study of c-erbB-2 and p53 protein in breast papillomatosis and its malignant transformation].

OBJECTIVE: To evaluate the levels of c-erbB-2 and p53 protein in different stages of papillomatosis of breast and its malignant transformation and value for early diagnosis. METHODS: Levels of c-erbB-2 and p53 protein were detected by immunohistochemistry in 184 paraffin-embeded blocks. RESULTS: c-erbB-2 protein expression was seen in all stages of papillomatosis with different degrees of proliferation and in tissues undergone malignant transformation. The rate of overexpression increased with the degree of proliferation. The p53 protein expression was not found in papillomatosis with mild to severe proliferation. It was only overexpressed in 3.5% of carcinoma arising from papillomatosis, but increased to 25% when invasion occurred. CONCLUSION: c-erbB-2 assay may be helpful for detection of early malignant change of the highly proliferative papillomatosis, especially when histopathological evidence of malignancy is still lacking. p53 protein is of no significant diagnostic value in these lesions.

[Pathological study of en bloc breast sections in papillomatosis and its malignant transformation].

OBJECTIVE: To investigate the characteristics of breast papillomatosis, its malignant transformation and relation with main ductal papilloma. METHODS: A histopathological study by en bloc breast subserial sections was performed on 14 cases of highly proliferative papillomatosis and 22 cases of carcinoma arising from the highly proliferative papillomatosis. RESULTS: The two types of lesions were widely scattered over the sections, covering more than two quadrants in 71.4% (10/14) and 63.3% (14/22) of the breast, respectively. Crisscross and histological transition of the two lesions were observed frequently in the sections of carcinoma arising from the highly proliferative papillomatosis. These two kinds of lesion often coexisted with main ductal papilloma. CONCLUSION: Papillomatosis is a breast precancerous lesion and it may have similar pathogenetic mechanism as main ductal papilloma.

[Cloning and restriction analysis of saw1--a key gene for differentiation of Streptomyces ansochromogenes].

saw1--a essential gene for sporulation of Streptomyces ansochromogenes was cloned, 7 kb of DNA fragment in Streptomyces plasmid pIJ702 could recover the sporulation of saw1-deficient strain W51 which could not form spore with white phenotype of aerial hyphae. The recombinant plasmid was deleted by some restriction enzymes, the result showed that 2.5 kb DNA fragment in pIJ702 still has function to recover sporulation of W51. The sequencing and expression of saw1 gene is carrying out at present.

Changes on receptor expression and production of interleukin-2 in circulating lymphocyte population after open heart surgery.

To evaluate the change of perioperative cell mediated immunity after cardiac operation with cardiopulmonary bypass (CPB), so as to provide some information for timely prevention and treatment against postoperative immunological disorder, 40 patients were studied. By searching for the effects of CPB and anesthesia, interleukin-2 receptor (IL-2R) expression upon the surface of peripheral blood mononuclear cells (PBMC), as well as interleukin-2 (IL-2) production in vitro was traced 55 min after anesthesia, at end of CPB, on postoperative 1, 7, and 14 day versus preanesthesia control. Our data demonstrated that expression of IL-2R on PBMC was significantly suppressed in all comparing with the baseline value, meanwhile, IL-2 production in vitro also statistically dropped. However, no statistical difference was found on perioperative IL-2R expression and IL-2 synthesis in the cholecystectomy group. We conclude that postoperative immunological disorder seems to be the main factor, which could be denoted as reduced IL-2R expression on PBMC and IL-2 synthesis in vitro for sepsis, even multiple system organ failure developed after cardiac surgery.

[Establishment of a coupling method of high performance liquid chromatography and capillary gas chromatography].

An on-line coupling system of normal phase microbore HPLC and capillary GC has been established, in which retention gap technique with on-column injection and partially concurrent solvent evaporation with early vapor exit were used. The conditions suitable for the coupling system of HPLC and CGC must be determined experimentally since the factors have to be adjusted with respect to solvent evaporation temperature and column inlet pressure etc. The suitable solvent evaporation temprature for the mobile phase used for this system, n-hexane, was about 70 degrees C. A temperature much, lower or higher than 70 degrees C will influence the GC analysis. In addition, solvent evaporation temperature and column inlet pressure are interacting, that is, higher solvent evaporation temperature needs higher inlet pressure. At the above evaporation temperature, the suitable inlet pressure for this system was about 0.2MPa. In order to minimize the extra-column effect on the LC peaks, thin transfer-line and connectors of zero-dead volume between LC column and GC column must be used. The separation and analysis of aromatics in a light diesel fuel was performed by using this system with 15cm x 2mm i.d. YGW (5microm) silica LC column and 60m x 0.25mm i.d. OV-101 GC column under the above conditions. The analytical results show that the system of on-line coupling of HPLC with CGC is a reliable analytical tool for microcomponents in complex mixtures, especially for those requiring sample preparation prior to GC.

[Method of cyanogen bromide-fluorimetry determination of trace amount of selenomethionine in grain and blood].

Selenomethionine (SeMet) reacted with cyanogen bromide (BrCN) quantitatively forms CH3SeCN. After extracted with CHCl3, the Se of CH3SeCN is acid-digested to Se(IV). Then 2,3-diaminonaphthalene is used to determine the fluorescent Se value of 4,5-benzopiaselenol. The determination limit of this method was 3 ng/g SeMet. The accuracy of 10-500 ng Se in SeMet standard was 91.8%-97.6%. RSD was 1.9%-6.3%. Recoveries for grain and blood were 92.3%-96.7%. RSD was 2.7%-5.1%. The RSD for samples was 2.7%-9.0%. Selenocystine, selenocystiene selenite and methionine did not interfere with the determination.

A comparative study on morphology and immunohistochemistry of rhabdomyosarcoma and embryonal skeletal muscles.

OBJECTIVE: To investigate the histologic and histogenetic relationship between various types of rhabdomyosarcomas (RMS) and embryonal skeletal muscle (ESM) for further understanding of the histogenesis, classification and pattern of differentiation of RMS. METHODS: Fifty cases of variant types of RMS and 20 cases of ESM at different gestational ages were available. All specimens were stained with HE, PAS, Van Gieson, Masson, phosphotungstic acid hematoxilin and with antibodies for the demonstration of vimentin, desmin, HHF-35 and myoglobin by ABC method. RESULTS: The results showed that the order of positive expression and the intensity of positive reaction of the different immunohistochemical staining were consistent with the degree of differentiation of the tumor and the development of the ESM. It is obvious that each type of RMS is composed of tumor cells in different degree of differentiation and is derived from primitive mesodermal cells which are capable of potential differentiation towards mature skeletal muscles. CONCLUSIONS: Based on the results of this study, an ideal histologic classification of RMS should reflect not only the cell morphology and histologic structures but also the degree of differentiation of the tumor cells.

[Glycosylated high density lipoprotein in diabetes related to pregnancy induced hypertension].

OBJECTIVE: To investigate the glycosylation of high density lipoprotein (glc-HDL) and its specific binding to human lung fibroblasts (HLF) receptors in gestational diabetes woman and to study its relationship with pregnancy induced hypertension (PIH). METHODS: The glc-HDL was measured by fluorimetry and the specific binding of HDL to HLF receptors was measured with enzyme linked immunoreceptor assay in 32 gestational diabetic woman group 1, 34 group II, 36 normal pregnant (NP) and 32 normal non-pregnant (NNP) women. RESULTS: The levels of glc-HDL and the intracellular total cholesterol in diabetic pregnant woman group I and II were significantly higher than those in the NP and the NNP (P < 0.01). The cell binding, internalization and degradation of glc-HDL were higher than that in the NP and the NNP. CONCLUSION: The change of HDL level and lipid deposition during the process may be one of the causes for arteriole spasm in PIH.

Effects of protein calorie malnutrition on tuberculosis in mice.

Infectious diseases and malnutrition represent major burdens afflicting millions of people in developing countries. Both conditions affect individuals in industrialized nations, particularly the aged, the HIV-infected, and people with chronic diseases. While malnutrition is known to induce a state of immunodeficiency, the mechanisms responsible for compromised antimicrobial resistance in malnourished hosts remain obscure. In the present study, mice fed a 2% protein diet and developing protein calorie malnutrition, in contrast to well-nourished controls receiving a 20% protein diet, rapidly succumbed to infection with Mycobacterium tuberculosis. Malnourished mice exhibited a tissue-specific diminution in the expression of interferon gamma, tumor necrosis factor alpha, and the inducible form of nitric oxide synthase in the lungs, but not the liver. The expression of these molecules critical to the production of mycobactericidal nitrogen oxides was depressed in malnourished animals in the lungs specifically at early times (< 14 days) after infection. At later times, levels of expression became comparable to those in well-nourished controls, although the bacillary burden in the malnourished animals continued to rise. Nevertheless, urinary and serum nitrate contents, an index of total nitric oxide (NO) production in vivo, were not detectably diminished in malnourished, mycobacteria-infected mice. In contrast to the selective and early reduction of lymphokines and the inducible form of nitric oxide synthase in the lung, a marked diminution of the granulomatous reaction was observed in malnourished mice throughout the entire course of infection in all tissues examined (lungs, liver, and spleen). Remarkably, the progressively fatal course of tuberculosis observed in the malnourished mice could be reversed by restoring a full protein (20%) diet. The results indicate that protein calorie malnutrition selectively compromises several components of the cellular immune response that are important for containing and restricting tuberculous infection, and suggest that malnutrition-induced susceptibility to some infectious diseases can be reversed or ameliorated by nutritional intervention.

The unpredicted high affinities of a large number of naturally occurring tachykinins for chimeric NK1/NK3 receptors suggest a role for an inhibitory domain in determining receptor specificity.

Three chimeric receptors were constructed by exchanging exon sequences between human NK1 and NK3 receptor genes. The resulting chimeric receptors not only retained high affinities for their natural ligands substance P and neurokinin B but also exhibited surprisingly high affinities for other naturally occurring tachykinins including neurokinin A, neuropeptide K, neuropeptide gamma, eledoisin, kassinin, physalaemin, and phyllomedusin. In contrast, these chimeric receptors displayed a wide range of variability in their affinities for non-naturally occurring ligands including selective agonists and antagonists of NK1, NK2, and NK3 receptors. Since the only common feature among these naturally occurring neurokinin peptides is the conserved C-terminal sequences, our data suggest that these conserved sequences must play the major role in conferring high affinity binding to the chimeric receptors. To explain the apparently "improved" affinities of these naturally occurring ligands for the chimeric receptors as compared with their affinities for the parent NK1 and NK3 receptors, we are proposing that certain inhibitory domains that are present in the NK1 and/or NK3 receptors are compromised in these chimeric receptors. Upon disruption of these inhibitory domains during the formation of chimeras, the naturally occurring ligands can interact more favorably with chimeric receptors through their conserved C-terminal sequences. Based on this hypothesis, the binding affinities of natural tachykinin ligands may be largely determined by their conserved C-terminal sequences, whereas receptor selectivities of these ligands are influenced more by the presence or absence of inhibitory domains rather than specific binding domains on their target receptors.

5-Fluorouracil suppresses nitric oxide biosynthesis in colon carcinoma cells.

Nitric oxide is an important cellular mediator that plays a role in regulating cellular proliferation of both normal and tumor cells. In the present study, we characterized nitric oxide production by the human colon adenocarcinoma cell line DLD-1 and examined the effects of 5-fluorouracil (5-FUra), an antimetabolite effective against colon tumors, on nitric oxide production. IFN-gamma was found to be a potent inducer of nitric oxide production in DLD-1 cells. This effect was dependent on L-arginine and blocked by the nitric oxide synthase inhibitors NG-monomethyl-L-arginine, nitroarginine, and aminoguanidine. Production of nitric oxide by DLD-1 cells was due to the expression of the inducible (type II) form of nitric oxide synthase. mRNA for the nitric oxide synthase was present in both untreated and IFN-gamma-stimulated cells, as determined by RT-PCR, suggesting that expression of enzyme is regulated posttranscriptionally. Treatment of DLD-1 cells with concentrations of 5-FUra that are not growth inhibitory or cytotoxic strongly inhibited their ability to express nitric oxide synthase and produce nitric oxide in response to IFN-gamma. This effect was not reversed with thymidine, indicating that inhibition of nitric oxide production was due to incorporation of 5-FUra into RNA. However, pretreatment of DLD-1 cells with 5-FUra before stimulation with IFN-gamma also suppressed nitric oxide production. Thus, inhibition of nitric oxide production was not due directly to incorporation of 5-FUra into the mRNA for nitric oxide synthase. Taken together, these data suggest that inhibition of nitric oxide biosynthesis in colon tumor cells by 5-FUra may underlie, at least in part, the efficacy of this antitumor agent.

Differential effects of costimulator signals and interleukin-2 on T cell receptor-mediated cell death of resting and activated CD4+ murine splenic T cells.

Postthymic T cell receptor (TCR)-mediated cell death offers the potential for creating antigen-specific transplant tolerance analogous to thymic clonal deletion. Murine specific CD4+ cell were rigorously purified by: (a) adherent cell depletion and (b) magnetic bead/monoclonal antibody (mAb) depletion of macrophages, Ia+ cells, mu-chain+ cells, NK cells, and CD8+ T cells. CD4+s were typically > 95% pure by flow cytometry. Resting CD4+s stimulated by plastic-immobilized anti-TCR/CD3 mAb wee shown to die in the absence of exogenous interleukin (IL)-2. Blasting CD4+s showed dose-dependent cell death upon religation of TCR/CD3 in the presence of IL-2; however, withdrawal of IL-2 from blasting CD4+s also resulted in cell death. Cell death was shown to be apoptotic by flow cytometry DNA content analysis. Anti-CD28 mAb, co-immobilized with anti-TCR/CD3 mAb, inhibited cell death of resting CD4+s in the absence of exogenous IL-2; however, anti-CD28 mAb showed minimal cell death inhibition of CD4+ blasts when TCR/CD3 was religated. In contrast, splenic adherent cells effectively inhibited cell death of blasting CD4+s induced by TCR/CD3 mAb religation. We conclude that TCR-mediated programmed cell death of highly purified splenic CD4+s is dependent upon activation state, availability of IL-2, and accessory cell or CD28 costimulator signals. Furthermore, IL-2 acts to protect against cell death in both resting and activated CD4+ T cells. IL-2 protection could be overcome by high concentrations of anti-TCR/CD3 mAb, which results in cell death of CD4+ blasts. In the effort to understand potential mechanisms of peripheral tolerance induction, these findings assist to distinguish and define conditions for antigen receptor-mediated programmed cell death of mature CD4+ T cells.
