Structural motifs encoded by individual exons of the human neurokinin-1 receptor gene interact differentially with selective agonists and antagonists.

Three chimeric receptors were constructed by exchanging exons between human neurokinin NK1 and NK3 receptor genes. The N-terminal sequences of these chimeric receptors are encoded by exon 1, exon 1-2, or exon 1-3 of the NK1 receptor gene, whereas the remaining C-terminal sequences of these chimeric receptors are encoded by corresponding exons of the human NK3 receptor gene. Substance P bound with high affinities to all three chimeric receptors, suggesting that in addition to the common structures composed of conserved amino acid residues among neurokinin receptors, structural elements encoded by the first exon of the human NK1 receptor gene may also play an important role for substance P binding. On the contrary, potent NK1 antagonists L703,606 and SR140,333 did not show any detectable binding to these chimeric receptors. In accordance, sequences encoded by exon 4, and possibly exon 5, are likely to contain important structural motifs that may directly or indirectly influence the binding of these antagonists. Further comparison of the binding affinities of highly selective NK1 agonists, [Sar9, Met(O2)11] substance P, substance P methyl ester, and septide, revealed that each agonist may interact differently with the human NK1 receptor. These results show that the exon-exchanging technique can be a useful tool for studying structure-function relationships of receptors in which exon-intron junctions are fully conserved among receptor subtypes.

Properties of AT1a and AT1b angiotensin receptors expressed in adrenocortical Y-1 cells.

Adrenocortical Y-1 cells were stably transfected with the AT1a and AT1b subtypes of the rat angiotensin (ANG)IIAT1 receptor cDNA to study the pharmacological and functional properties of the two receptors. Selected clones of transfected cells expressing the AT1a or AT1b receptor subtypes bound the native ligand ANG II and the peptide antagonist [Sar1,Ile8]ANG II with similar affinities, but they differed in their relative affinities for the nonpeptide antagonist losartan (half-maximal inhibitory concentration 9.7 and 4.7 nM), ANG III (126 and 33 nM), and the peptide antagonist [Sar1,Gly8]ANG II (6.2 and 1.2 nM). Photoaffinity labeling of the expressed receptors revealed a single component of 65 kDa for both receptor subtypes, suggesting that both receptors were glycosylated in a similar manner. The sensitivity of 125I-ANG II binding to AT1a and AT1b receptors to guanine nucleotides was unaffected by pertussis toxin treatment. ANG II stimulated the formation of inositol phosphates and increased the level of cytoplasmic Ca2+ in both At1a- and AT1b-transfected Y-1 cells. However, ANG II had little effect on forskolin-induced adenosine 3',5'-cyclic monophosphate accumulation, causing only minor inhibition in AT1a-transfected cells and slight enhancement in AT1b-transfected cells. These data indicate that AT1a and AT1b receptors show small but significant differences in their binding pharmacology and, upon activation, are coupled through Gq/G11 to the phosphoinositide-Ca2+ signaling pathway. However, neither AT1a nor AT1b receptors exhibit coupling to Gi and inhibition of adenylate cyclase when expressed in murine adrenal tumor cells.

[Construction of shuttle vector containing delta-endotoxin gene of Bacillus thuringiensis].

The Bacillus thuringiensis toxin gene CryIA(c) was inserted into the shuttle vector pBE-2 to construct pAMY for expressing the B.t. gene in both Gram-negative and -positive bacterial systems. pAMY was introduced into wild type Bacillus cereus, B.brevis and B.subtilis by electroporation. Transformants containing delta-endotoxin gene produced proteins reacted with B.t. crystal protein antibody. Upon biological toxicity tests, the transformants gave a mortality of 100% against Ostrinia furnacilis, 58.8% against Heliothis armigera and 100% against Heliothis assulta. The ability of promoting plant growth of the original strains is retained.

[Morphological changes in intracranial and extracranial arteries of autopsy cases of cerebrovascular diseases].

OBJECTIVE: To determine the distribution and severity of arterial lesions of intracranial and extracranial arteries in stroke patients in Chinese. METHODS: We studied 120 consecutive autopsy cases including 100 cases of stroke (50-80 years) and 20 cases of cancer as controls (50-80 years). Medium intracranial arteries and their primary branches, small intracerebral arteries, arterioles and extracranial carotid arteries (from 16 cases only) were examined with light microscopy. RESULTS: The stroke patients had significantly lower mean LA-VA ratios of the medium sized intracranial arteries and their primary branches, small intracerebral arteries and arterioles than the controls (P < 0.001). The stroke patients had higher mean VW-VR ratios of these arteries than the controls (P < 0.001). Atherosclerotic narrowing of the medium sized intracranial arteries and their primary branch were more severe than those of the extracranial caotid arteries. Moreover, the proximal portion of the primary branch arteries was much more severe than that of extracranial caotid arteries (P < 0.01, P < 0.05) in the stroke patients. CONCLUSION: Stroke patients had atherosclerotic lesions of the intracranial medium sized arteries and their primary branches and sclerotic lesions of the small intracerebral arteries and arterioles. These pathological changes resulted in thickening arterial walls and narrowing arterial lumens.

[Study of p53 protein expression in cancers of small intestine and its relationship to clinical course and prognosis].

In order to study the relationship of p53 gene mutation with the occurrence and prognosis of cancer of small intestine, expression of p53 protein was examined. The results showed that p53 protein expression was high in 75% of small intestine cancer, and 21.1% positive in tissues close to cancer. In 7 cases of small intestinal adenoma only one was positive. Sixteen cases of normal tissue of intestine showed negative expression of p53 protein. The study also found that the degree of p53 protein expression was significantly correlated with the degree of tumor cell differention, invasion, metastasis and prognosis.

[Clinical diagnostic problems of the female coronary heart disease evaluated by coronary artery angiography].

The clinical data of 100 female cases with suspected coronary heart disease (CHD) undergoing coronary artery angiography (CAA) were analysed. CHD was defined as an estimated diameter stenosis of 75% and more in the main coronary arteries or coronary stenosis of 50%-75% with evidence of cardiac ischemia. Forty-eight case were confirmed as having CHD by CAA. The remaining 52 cases were confirmed as non-CHD and served as the control group, including 49 cases with normal coronary arteries and 3 cases with stenosis of less than 50% in the right coronary artery or left anterior descending artery. The risk factors of CHD and clinical characteristics of chest pain were compared for the two groups. The data demonstrated that menopause was a special risk factor for females and the incidence of CHD was increasing with aging after menopause. However, the clinical characteristics of chest pain in female CHD were usually atypical. When chest pain, whether clinically typical or atypical, were present after menopause and accompanied by evident risk factors, the probability of CHD would be high. If there was only chest pain without any risk factor after menopause, the diagnosis of CHD would be less likely.

Effect of radix Salviae miltiorrhizae on nitric oxide in cerebral ischemic-reperfusion injury.

It has been postulated that nitric oxide (NO) is produced in significant amount on reperfusion of ischemic brain and could cause brain edema and cell death. This study was undertaken in an attempt to determine whether Radix Salviae Miltiorrhizae (RSM) could reduce cerebral NO contents in 4-vessel occlusion rat model after 30 min of global ischemia and 15 min of reperfusion. There was a significant increase in cerebral NO level in untreated animals as compared with sham-operated ones. By pretreatment with RSM the cerebral NO concentration was reduced to normal level. The possibilities of neuroprotective role of RSM in cerebral ischemic-reperfusion injury were discussed.

Radix Salviae miltiorrhizae treatment results in decreased lipid peroxidation in reperfusion injury.

In our pervious investigations we have found that Radix Salviae Miltiorrhizae (RSM) can reduce neurological deficits and ameliorate cellular damages, and its protective actions are associated with its effects on cerebral edema, monoamines, neuropeptides, neurotransmitter aminoacids, thromboxane A2, peroxidation and down-regulation of c-fos gene expression in ischemic animals. The purpose of present study was to investigate the protective effect of RSM against lipid peroxidation in 4-vessel occlusion and reperfusion rat model. We found that in RSM-treated rats the cerebral SOD activity is significantly increased, while the cerebral MDA level is significantly decreased when compared with untreated animals. The results suggested that RSM can reduce the lipid peroxidation and afford cerebroprotection against reperfusion injury.

[DNA quantitative analysis of small bowel carcinoma and its clinical significance].

By using computer cytophotometric analysis, aneuploid as well as other 12 parameters of tissue DNA content were measured in 28 cases of small bowel cancer, 5 cases of intestinal adenoma, 6 cases of normal intestinal tissues. The number of aneuploid tumors in intestinal cancer was much higher than in intestinal adenoma. 7 DNA parameters (Entropy of DNA, 2CDI, DNA- grade of neoplasia, 5C Exceeding Rate, 9C Exceeding Rate, Diploid Deviation Quotient and Mean Ploidy) were the valuable indexes that could determine the quality of intestinal lesions, benign or malignant. DNA content didn't correlate with the clinically differentiated degree of pathological tissues, but aneuploid intestinal cancers had a higher malignant degree than non-aneuploid ones, and were more liable to invade and metastasize with low postoperative survival-rate and poor prognosis. 5 DNA parameters (DNA-Index, Medal Value, DNA-grade of neoplasia, Stemline Ploid and 9C Exceeding Rate) had close relation to the prognosis of intestinal cancer, possessing important value to the prognostic analysis of intestinal cancer.

[Modified duodenum-preserving resection of the head of the pancreas].

A modified procedure of duodenum-preserving resection of the head of the pancreas was used in treating three patients with benign lesions in the head of the pancreas. Compared with Beger's procedure, the modified procedure was simpler, and pancreaticojejunostomy on duodenal side was not necessary because of less remaining pancreas attached to the conjunction of the pancreatobiliary duct. The patients, recovered and ate food from 4 to 6 days after operation without symptoms of duodenal obstruction. The patient's digestive function was normal, and the pre- and postoperative pancreatic endocrine function was almost identical. This procedure can be used as the first-choice surgical procedure for benign lesions in the head of the pancreas. Problems in how to protect blood supply of the duodenum were also discussed.

A molecular pathologic study on apoptosis in retinoblastoma and the mechanism of spontaneous regression in retinoblastoma.

OBJECTIVE: The present study was designed to prove the existence of apoptosis in retinoblastoma (Rb) and to determine the pathogenic mechanism of spontaneous regression of Rb as well as the relationship between them. METHODS: Qualitative morphological study on Rb was performed by means of light microscope, electron microscope and TdT mediated biotin-dUTP nick-end labeling (TUNEL). Quantitative study was performed by automatic image analysis technology (AIAT) stained with Feulgen reaction. RESULTS: The characteristic regressed area occurred in all 47 cases of Rb. Morphological changes observed within Rb closely resembled the apoptotic cell described by Kerr et al in 1972. Under electron microscope, details of apoptosis were observed in 7 cases of Rb: the morphological sequence of events occurred in and around the cell nucleus. The morphology of the TUNEL labeling Rb cell was various. TUNEL labeling showed more positive cells in regressed area, while fewer in advanced area. AIAT revealed that apoptosis index (AI) in regressed areas was higher than that in other areas of Rb, DNA average ploidy (DP) consisted with the histology grade of Rb, the degree of hyperdiploid (DH) in metastatic area was distinctly higher than that in other areas (P < 0.05). CONCLUSIONS 1. Morphologic evidence proved the existence of apoptosis, especially more in regressed area of Rb. 2. Apoptosis may participate in the spontaneous regression of Rb. Apoptosis contributed to the spontaneous regression of Rb. 3. Tumor growth parameters of Rb (AI, DP, DH) obtained by AIAT may be used as quantitative index for pathologic classification, the selection of clinical treatment and the prognostic evaluation.

[A comparative study of quantification of morphological changes in intracranial and extracranial arteries between cerebral hemorrhage and infarction].

In order to compare the difference and similarity of the pathological changes in intracranial and extracranial arteries between cerebral hemorrhage (CH) and cerebral Infarction (CI), 120 autopsy cases were studied, including 42 cases of the CH, 58 cases of the CI and 20 cases of cancer as control. Medium sized intracranial arteries and their main branches, small sized intracerebral arteries, arterioles and extracranial carotid arteries (from 14 cases only) were examined with light microscopy. The ratio of lumen area to vessel area (LA-VA ratio) and the degree of narrowing of the vessels indicating the severity of arteriosclerosis were determined. Quantitative analysis was carried out with computer. The results were as follows: (1) The CH and CI patients had significantly lower mean LA-VA ratios of the medium intracranial arteries and their main branches, small intracerebral arteries and arterioles than the controls. (2) The CH patients had apparently lower LA-VA ratio of the small intracerebral arteries and arterioles than the CI patients. (3) The CI patients had significantly lower LA-VA ratio of the medium sized intracranial arteries and their main branches than the CH patients. Moreover, atherosclerotic narrowing of these vessels was also much more severe in the CI than in the CH patients. (4) Atherosclerotic narrowing of the extracranial carotid artery in the CI patients was slightly more severe than that in the CH patients. These data suggest that patients with cerebral stroke have frequently atherosclerotic lesions in the medium sized intracranial arteries and their main branches and sclerotic lesions in the small intracerebral arteries and arterioles. The principal lesion of the CH are in the small intracerebral arteries and arterioles, while those of the CI in the medium sized intracranial arteries and their main branches and sometimes in the extracranial carotid arteries.

[Protective effect of active components extracted from radix Astragali on human erythrocyte membrane damages caused by reactive oxygen species].

A study has been made on the protective activities of active components extracted from Radix Astragali by using reactive oxygen species initiated lipid peroxidation of purified human erythrocyte membrane. The results show that the total flavonoids of Astragalus and total saponins of Astragalus can significantly inhibit the membrane lipid peroxidation generated by O2.-, H2O2 and UV rays, while the total polysaccharide of Astragalus possesses weaker protective activity.

Neurokinin-3 receptors modulate dopamine cell function and alter the effects of 6-hydroxydopamine.

Neurokinin-3 receptor expression within rat midbrain dopamine neurons was demonstrated using a combination of in situ hybridization and receptor autoradiographic techniques. Continuous intranigral infusion of the neurokinin-3 receptor agonist senktide selectively increased striatal dopamine metabolism over a period of several days, followed by apparent development of tolerance. In contrast, in animals with moderate unilateral 6-hydroxydopamine-induced lesions of nigrostriatal dopamine cells, intranigral senktide infusion increased dopamine turnover in the surviving dopamine neurons and reduced functional dopamine asymmetry (reflected by spontaneous rotations) over the 2-week period tested. Thus nigral neurokinin receptors can modulate normal dopamine cell activity and may provide a therapeutic target in the treatment of Parkinson's disease.

Results of pneumonectomy for cancer in patients with limited ventilatory function.

It is well established that patients with compromised pulmonary function have a greater incidence of morbidity and mortality following lung resection. The prognosis of 36 (9.7%) patients with poor respiratory function (forced expiratory volume in ls (FEV1) and FEV1/FVC (forced vital capacity) ratio were equal to or less than 50% of the predicted value) of a total of 369 patients who underwent pneumonectomy due to non-small cell lung carcinoma over 10 years were reviewed. All but three patients were male with a median age of 62.5 years. Right pneumonectomy was carried out in 12 (33%) and left in 24 (67%) patients. Median FEV1 and FEV1/FVC were 1.51 (46%) and 46.5% respectively. Three (8%) patients died within 30 days of surgery. The postoperative complication rate in patients with poor respiratory function was 44%. Nine (27%) of the hospital survivors died due to non-malignant causes (recurrent chest infection/respiratory failure) and 12 (36%) due to recurrent tumour. The cause of death in one patient was second primary lung tumour and it was unknown in three (9%) patients. Eight (24%) long-term surviving patients did not have severe respiratory symptoms; their FEV1 and FEV1/FVC were remeasured and revealed a median 1.05 l (38%) and 50%, respectively of the predicted value. Actuarial 5-year survival was 29%. Poor respiratory function is associated with postoperative complications and non-malignant deaths arising secondary to respiratory failure. The survival profile demonstrates that patients were successfully treated with pneumonectomy and suggests that surgery should not be withheld from those with limited lung function if detailed investigations predict adequate residual lung function.

Influence of heparin thromboprophylaxis on plasma leucocyte elastase levels following lobectomy for lung carcinoma.

Recent in vitro studies and animal investigation indicate that plasma leucocyte elastase (PLE) can dissolve pulmonary structural proteins, such as elastin, and produce lesions in the lung similar to that seen in adult respiratory distress syndrome (ARDS) and emphysema. In contrast, heparin strongly inhibits PLE and protects elastin from elastolysis. On the basis of these findings, PLE levels were monitored in 24 patients with non-small cell lung carcinoma (NSCLC) undergoing lobectomy. Ten patients from Killingbeck Hospital (Group 1) received 5000 IU subcutaneous (s.c.) heparin commenced 2 h prior to surgery and continued at 8 h intervals until the patient was fully ambulatory. Fourteen patients from Bradford Royal Infirmary (Group 2) received no heparin as standard policy. There was no significant difference in pre-operative PLE levels between groups. The post operative PLE levels in both groups increased significantly (P < 0.02) on the first post operation day (POD). However, PLE levels of Group 2 were 2.5 to 5.3 times higher than those of Group 1 at each postoperative interval (first, third, and seventh POD) respectively (0.002 < P < 0.02). There was no difference in blood loss between groups (P = 0.17). These results indicate that post operative PLE activity is elevated in NSCLC patients following lobectomy and s.c. heparin administration as thromboprophylaxis may inhibit PLE activity post operatively without increasing blood loss. Therefore, heparin may have a role to play in protecting lung tissue against the pulmonary lesions caused by proteolytic activity of PLE, and theoretically reduce post-operative complications, such as ARDS or emphysema.

Growth responses to angiotensin II in bovine adrenal glomerulosa cells.

The effects of angiotensin II (ANG II) on growth responses of primary cultures of bovine adrenal glomerulosa cells were studied to explore the mechanism(s) by which ANG II leads to hyperplasia and hypertrophy of the glomerulosa layer in sodium deficiency. ANG II did not increase [3H]thymidine incorporation during the first 5 days of culture, but mitogenic responses to ANG II became evident after longer periods of culture and were most prominent between 8 and 11 days after seeding. At this time, cell cycle analysis showed that ANG II increased the proportion of cells in the S phase and did not cause accumulation of cells in the G2 phase. Consistent with this finding, ANG II also stimulated proliferation of glomerulosa cells during treatment for 3 days in the presence of 1% serum. The mitogenic effect of ANG II was not inhibited by pretreatment with pertussis toxin and was mediated by AT1 receptors as indicated by its sensitivity to the subtype-selective antagonist DuP-753. Also, there was no emergence of AT2 receptors in glomerulosa cells during prolonged culture. These results indicate that intracellular mechanisms that mediate growth responses become more active during prolonged culture of glomerulosa cells. Thus, in addition to regulating the steroidogenic and secretory functions of the zona glomerulosa, ANG II exerts mitogenic actions that depend on the functional state of the glomerulosa cells.
