Serotonin immunohistochemistry reveals immature EC cells.

Radioautographic studies of rat duodenal mucosa were carried out after single injections of 3H-thymidine, and the labelling indices of enterochromaffin (EC) cells identified by serotonin immunohistochemistry and by the diazo reaction were compared. The labelling index of EC cells by serotonin immunohistochemistry was 2.3% (mean percentage of 2 rats), while no labelled cells were found in diazo-positive EC cells. This finding indicates that serotonin immunohistochemistry reveals a less mature population of EC cells than conventional techniques such as the diazo reaction.

Differentiation of monoamine accumulating neuron systems in cultured chick retina: an immunohistochemical and fluorescence histochemical study.

Monoamine-containing neurons (dopamine and serotonin) have been reported to be localized among the cells in the inner nuclear layer (INL) of the chick retina. To examine the development of these neurons and whether they have an ability to synthesize serotonin (5-HT) or dopamine from exogenous precursors, we administered monoamines and their precursors in vitro as well as in situ and examined monoamine neuronal differentiation histochemically. In the chick retina, serotonin-containing neurons were found among amacrine cells. Two other types of serotonin-accumulating neurons were observed in the INL. One also had the ability to take up tryptophan and 5-hydroxytryptophan (5-HTP) and convert them to serotonin. In the monolayer culture from 61/2-day-old chick embryonic retinas, serotonin-containing neurons were observed on the 4th day of culture, increased in number until the 8th day, but had almost disappeared by the 15th day. After incubation of 8 day-cultures with 5-hydroxytryptophan, the number of serotonin-immunoreactive cells was at least twice that seen in untreated cultures. On the 20th day of culture, the same percentage of cells had the capability to take up serotonin, but the number of those which took up 5-hydroxytryptophan and decarboxylated it to serotonin had decreased markedly. Dopaminergic amacrine cells have also been found in the INL and shown to take up exogenously administered L-DOPA. In cultures, no dopamine-containing cells were observed with the present fluorescence histochemistry without prior incubation with DOPA.(ABSTRACT TRUNCATED AT 250 WORDS)

Siblings with carbamyl phosphate synthetase I deficiency.

This paper concerns with two autopsied cases of siblings who died from cerebral disturbances. In these patients hyperammonemia developed in the neonatal phase due to carbamyl phosphate synthetase I (CPS I) deficiency. The patient in Case 1 was admitted 2 days after birth because of oliguria and vomiting. Hyperammonemia developed and she died on the 43rd day. In Case 2 hyperammonemia developed from the 2nd day after birth and she expired on the 42nd day. In both cases the diagnosis of CPS I deficiency was established from autopsy findings of the liver.

Different response of left ventricular dimensions to nitroglycerin in patients with artificial pacemaker. A possible significance of reflex tachycardia for the reduction of LV cavity after nitroglycerin.

The effects of nitroglycerin (NTG) on cardiac function can be evaluated by echocardiography. In this paper, we discuss the effects of NTG on cardiac function in two groups of subjects, one with artificial pacemakers implanted with a fixed heart rate, and the other examined as the control group. Left ventricular dimension was obtained in the standard manner using the electronic sector scanning system of Toshiba SSH-11A. Left ventricular end-diastolic and end-systolic dimensions (Dd and Ds) were obtained in each case and the ejection fraction was calculated. After the sublingual administration of NTG (0.6 mg), the blood pressure decreased in both groups. The ventricular rate was fixed in the pacemaker group but increased significantly in the control group. Dd and Ds were found to decrease only in the control group. The ejection fraction showed an increase in the majority of the control group but remained unchanged in the pacemaker group. Heart rate change therefore may play a role in dimensional change and pumping function of heart after NTG.

[A nationwide survey on the use of organic solvents in Japan].

A total of 1,179 organic solvent products used as thinners, paint, degreasers, inks, adhesive and others, were collected nationwide from factories of various sizes and kinds in Japan, and analyzed by gas chromatography. Solvent components per sample, frequency of each solvent component, and contents of each component were analyzed and classified by use. Paints, inks, and adhesives which contained nonvolatile substances were analyzed by head space techniques. Seventy-seven percent of all samples, 93% of paints, 85% of thinners, 73% of adhesives, and 52% of degreasers contained mixed organic solvents. The average numbers of solvent components per sample were 4.14 (max. 13) in thinner, 3.29 (max. 7) in paint, 2.23 (max. 5) ink, 2.19 (max. 6) in adhesive, 1.71 (max. 6) in degreaser, when gasoline was excluded. Toluene, xylene, methylalcohol, isopropylalcohol, ethylacetate, methylethylketone, industrial gasoline were widely used in the above-mentioned products. Toluene was the most popular component in the solvent products, excluding degreasers. Many kinds of solvent components were used in thinner and paint. However, toluene, xylene and industrial gasoline were their main components and contents per sample were very high. Acetates, alcohols, ketones and glycols were frequently detected in solvent products, but their contents per sample were relatively low, because these solvent components were usually used as subordinate solvents. The chlorinated hydrocarbons such as trichloroethylene, tetrachloroethylene and 1,1,1-trichloroethane, and industrial gasoline were the most popular components in degreasers. 1,4-dioxane was detected in a relatively large number of the samples, but its content per sample was very low, because it is usually used as an additive to 1,1,1-trichloroethane. Toluene was the most popular component in ink solvents, too. In addition, isopropylacohol, methylalcohol, methylethylketone were frequently detected in inks. Toluene was also the most popular component in adhesives, and n-hexane was also frequently detected in them. In order to prevent poisoning by industrial solvents, special attention should be paid to toluene and xylene, which are frequently and largely used for wide purposes, trichloroethylene which is largely used as a degreaser and has a severe toxicity, n-hexane which is frequently used in adhesives and has a high volatility and severe neurotoxicity, and other popular and toxic solvents. The severely toxic solvents such as benzene, chloroform, carbon tetrachloride, 1,2-dichloroethane, 1,1,2,2-tetrachloroethane and carbon disulfide were detected in few samples.(ABSTRACT TRUNCATED AT 250 WORDS)

Serotonin-containing neurons in the rat and cat brain, especially in the hypothalamus, following monoamine oxidase inhibitor pretreatment: an immunohistochemical study using anti-serotonin antiserum.

The presence of serotonin-containing neurons in the hypothalamus of the rat and cat was studied by immunohistochemistry. In the rat, a group of serotonin-immunoreactive neurons was observed in the nucleus dorsomedialis hypothalami following nialamide pretreatment at a high dosage (over 300 mg/kg). In the cat, serotonin-immunoreactive neurons were sparsely distributed in the ventral part of the middle to the caudal lateral hypothalamic area after high dosage (500 mg/kg) nialamide pretreatment.

[Assessment of the exposure of pest control operators to organophosphorus pesticides. Organophosphorus pesticides in blood and alkyl phosphate metabolites in urine].

Pest control operators usually spray pesticides in small areas such as a kitchen in a restaurant and are exposed to various pesticides, especially those of the organophosphorus (OP) type. In order to evaluate their occupational exposure to OP pesticides during the work, OP pesticides in blood and alkyl phosphate metabolites in urine of these operators were analyzed and the relationship between pesticide exposure and analytical results were studied. OP pesticides in blood were analyzed by gas chromatography with flame photometric detector (FPD-GC) after separation of phospholipid in blood with silicagel column chromatography. OP pesticides were not detected in any blood samples (the limit of detection was 1 ng/ml). Dimethylphosphate (DMP) and dimethylthiophosphate (DMTP), being urinary metabolites of OP pesticides, were analyzed by FPD-GC after benzyl derivatization. This method eliminated interfering peaks in gas chromatograms. The ratio of two isomeric derivatives of DMTP was found to be constant. Both DMP and DMTP of the exposed group were significantly higher than those of the non-exposed group, DMP being higher than DMTP. The ratio of DMP to DMTP in the fenitrothion-dichlorvos-exposed group was significantly higher than that in the fenitrothion-exposed group. It was considered that the ratio might reflect a result of pesticide exposure. The urinary metabolites of OP pesticides tended to become lower with the lapse of time since the last exposure. However, small amounts were detected in a few samples even 5 days after the last exposure.
