[Visual disorders due to organic solvent poisoning].

Visual ability has increasingly become very important in recent years in industry which has become highly automated with the rapid introduction of computers. On the other hand, visual disorders due to occupational activity pose a serious occupational health problem. Many cases of visual disorders due to organic solvent poisoning have been reported, but there are few systematic studies on them. Many kinds and large volumes of organic solvents are being used in industry to which many workers are exposed. Therefore, it is considered very important for occupational health to review the literature on visual disorders due to organic solvent poisoning. Visual disorders due to carbon disulfide, n-hexane, methanol, toluene, trichloroethylene, butanols, methylethyl ketone (MEK), styrene and mixed or unidentified solvents were reviewed together with corneal ulcer, lenticular changes, visual field changes, retinal changes, visual acuity impairment, acquired dyschromatopsia and other visual disorders due to these solvents. Visual disorders are often accompanied with impairment of the central and/or peripheral nervous system and can serve as early indicators of their impairment due to organic solvent poisoning. Especially, acquired dyschromatopsia can be an indicator for the early diagnosis of CNS impairment due to organic solvent poisoning.

[Automated HPLC method for determining zinc protoporphyrin IX and protoporphyrin IX in erythrocytes of workers exposed to lead].

We developed an automated HPLC method for determining erythrocyte protoporphyrin. After a single extraction with N,N-dimethylformamide (DMF), zinc protoporphyrin IX (ZP) and protoporphyrin IX (PP) were separated by the column packed with a new type of reversed phase silica (capcell type) within 5 min. The column life was markedly extended to facilitate sample injection for about 1,000 times. The concentrations of ZP or TP (total protoporphyrin = 0.9 ZP + PP) determined by the present method correlated well with those determined by the conventional methods, i.e. by hematofluorometer (HF) or acid extraction method (FEP). In 150 workers exposed to lead, the ratio of 0.9 ZP/TP varied between 0.43 and 0.99 with the ratio being less than 0.6 in only 5 workers. The correlation coefficient between Pb-B and ZP was significantly higher than that between Pb-B and PP. Both anticoagulants, heparin and EDTA, could be used in the present method as well as in HF and FEP methods. Reference values for TP by HPLC were between 31.2 and 120.9 micrograms/dl RBC. Samples stored at 37 degrees C for 3 days were also used in the present method and the same results were obtained as in the samples stored at 4 degrees C.

[Reproduction studies of carboplatin (I)--Intravenous administration to rats prior to and in the early stages of pregnancy].

Carboplatin, an oncostatic drug, was administered intravenously to male Crj: CD (Sprague-Dawley) rats for 63 days and to female rats of the same strain for 14 days prior to mating at dose levels of 1, 2 and 4 mg/kg/day. These animals were then mated under the consecutive administration of this drug and the females confirmed to be copulated were further dosed from day 0 through 7 of gestation. The summarized results obtained are as follows: 1. Carboplatin 2 mg/kg and higher suppressed body weight gains accompanied by the decreases in food and water consumption in male rats. Further, body weight gains were suppressed in female rats followed by the decreases in food consumption at the same dose levels. 2. Though there were no differences between dosed animals and controls regarding the organ weights, the incidence of necrosis of the tails around the injection site was increased in male rats at 4 mg/kg. 3. Carboplatin failed to affect the reproductive ability of both sexes. 4. As for fetuses, the mortality was elevated at 2 mg/kg and higher and the number of live fetuses reduced at 4 mg/kg, but the influences on prenatal development were not apparently observed for live fetuses even at the highest dose level. Based on these results, the no-effect dose level of carboplatin under the present experimental condition was estimated to be 1 mg/kg/day against parent rats of both sexes and their offspring.

[Reproduction studies of carboplatin (II)--Intravenous administration to rats during the period of fetal organogenesis].

Carboplatin, an oncostatic drug, was administered intravenously to pregnant Crj: CD (Sprague-Dawley) rats from day 7 through 17 of gestation at dose levels of 1, 2 and 4 mg/kg/day. The summarized results obtained are as follows: 1. Carboplatin 4 mg/kg suppressed the maternal body weight gains from day 13 through 20 of gestation. 2. Uterine weights were reduced in F0 dams at term at carboplatin 4 mg/kg. 3. Carboplatin 4 mg/kg brought the inhibition of fetal growth accompanied by the lowered values in fetal weights, crown-rump distances and tail lengths. Furthermore, the elevated incidences of unossified 5th and 6th sternum , as well as retarded ossification of sacrococcygeal vertebrae were also noted in this dose level. 4. The birth rate was reduced in neonates (F1) at carboplatin 4 mg/kg. 5. Body weight gains in male F1 rats were suppressed at carboplatin 4 mg/kg from 4 to 8 weeks of age. 6. Carboplatin 4 mg/kg decreased the brain weights on an absolute basis in female F1 rats, but failed to affect their postnatal differentiations, early behavioral developments, learning ability, motor activity or emotional development. 7. Reproductive ability in F1 rats of both sexes were not affected by carboplatin. 8. Influences on prenatal development were not apparently observed for F2 fetuses derived from F1 rats whose dams had ever received carboplatin during the organogenetic period. Based on these results, the no-effect dose level of carboplatin under the present experimental condition was estimated to be 2 mg/kg/day against dams and their offspring.

Carboplatin, an oncostatic drug, was administered intravenously to pregnant Crj: CD (Sprague-Dawley) rats from day 17 of gestation through day 21 of postpartum at dose levels of 1, 2 and 4 mg/kg/day. The summarized results obtained are as follows: 1. Maternal body weight gains were suppressed during the former part of the lactation period at carboplatin 2 mg/kg and higher. 2. Thymic weights were decreased and lung weights were increased in dams (F0) at carboplatin 2 mg/kg and higher. Further, ovarian weights were reduced in dams (F0) at carboplatin 4 mg/kg. 3. Carboplatin failed to affect the parturition of F0 dams. 4. Carboplatin did not affect the viability of newborns (F1), and postnatal differentiations, early behavioral developments, learning ability, motor activity or emotional development in F1 animals. 5. Carboplatin 4 mg/kg brought a suppression of pituitary weights after mating in F1 male rats and decreases of adrenal and genital organ weights at weaning in F1 female rats, but failed to affect their reproductive ability. 6. Influences on prenatal development were not apparently observed for F2 fetuses derived from F1 rats whose dams had ever received carboplatin during the perinatal and lactation periods. Based on these results, the no-effect dose level of carboplatin under the present experimental condition was estimated to be 1 mg/kg/day against dams and 2 mg/kg/day against their offspring.

New antibiotics SF2315A and B produced by an Excellospora sp. II. The structural elucidation.

The structures of antibiotics SF2315A and B were determined to be [4aR-(4a beta,12b beta)] or [4aS-(4a alpha,12b alpha)]-4a,5,6,12b-tetrahydro-4a,8-dihydroxy-3- methylbenz[a]anthracen-1,7,12(4H)-trione and [1S-(1 beta,4a beta,6a beta,12 beta,12a beta,12b beta)] or [1 R-(1 alpha,4a alpha,6a alpha,12 alpha,12a alpha,12b alpha)]- 1,4,4a,5,6,6a,12a,12b-octahydro-1,4a,8,12-tetrahydroxy-3-methyl-6a ,12a- epoxybenz[a]anthracen-7(12H)-one, respectively from their spectroscopic analyses and single crystal X-ray diffraction analyses.

[A study of the cardiotocograms recorded by a separated 1MHz ultrasound transducer].

Fetal heart rate records were obtained in 12 antepartum and 13 intrapartum cases, with an ultrasonic Doppler transducer with five separated crystals and a conventional transducer. The loss of the FHR record due to increased noise in 30 minutes before delivery ranged from 1.1 to 23.2% (6.4 +/- 6.5%) in the separate type transducer, and it was 0.6 to 29.9% (10.5 +/- 7.6%) in the conventional one. During the 5 minutes before delivery, it was 3.3 to 58.0% (20.2 +/- 18.5%) in the separate type, and 3.3 to 70.0% (31.6 +/- 19.5%) in the conventional one. Clearer FHR records were obtained with the use of the separate type than the conventional one (p less than 0.05). Noise was mostly recorded in the 5 minutes before delivery. In the NST, the loss of the FHR record due to maternal posture change decreased when the new transducer (p less than 0.05) was used. The loss was 0 to 56.7% (14.9 +/- 18.2%) with the new type, and 2.0 to 100% (42.9 +/- 36.4%) with the conventional one for the maternal posture which showed the worst record. By using the separate type transducer, maternal posture need not to be limited in the NST, and the somatic and mental discomfort is less than when the conventional one is used.

Lysophosphatidylcholine----phosphatidylcholine converting activity of Penicillium notatum phospholipase B.

Phospholipase B from a fungus Penicillium notatum, which was previously shown to possess phospholipase B activity as well as lysophospholipase activity, was found to have another activity to convert lysophospholipid to phospholipid, to an extent comparable to its phospholipase B activity. The enzyme did not incorporate free fatty acid into phospholipid in the presence of CoA and ATP. The results shown here coincide with data reported on yeast phospholipase B and may imply the functional and structural kinship between two related enzymes.
