A case of a 77 year old woman with a heterotopic pancreas in the distal common bile duct is reported herein. The patient had no symptoms, but an ultrasound examination showed bile duct dilatation and subsequent endoscopic retrograde cholangiography demonstrated a spherical filling defect in the distal common bile duct. Under suspicion of a benign neoplasm in the common bile duct, resection of the common bile duct and hepaticojejunostomy using a Roux-en Y jejunal limb were successfully performed. Pathological examination revealed heterotopic pancreatic tissue in the distal common bile duct. This is only the ninth reported case of heterotopic pancreas occurring in the common bile duct or ampulla of Vater, and thus, a review of the literature is also given.
Two temperature-sensitive alleles of Drosophila melanogaster, Cha(ts1) and Cha(ts2), have been previously identified and are thought to be point mutations in the structural gene for the neurotransmitter biosynthetic enzyme choline acetyltransferase. In order to clarify the molecular nature of these alleles and characterize the presumed amino acid substitutions, we have used the polymerase chain reaction to amplify choline acetyltransferase messenger ribonucleic acid fragments from both mutant genotypes. Amplified mutant complementary deoxyribonucleic acid was cloned and used to construct chimeric complementary deoxyribonucleic acid clones containing approximately two-thirds of the wild-type sequence which would code for N-terminal amino acids and one-third of the mutant sequence coding for the C-terminal amino acids. After in vitro translation of complementary ribonucleic acid produced from the chimeric complementary deoxyribonucleic acid clones, choline acetyltransferase activity was determined and shown to be thermolabile. Sequence analyses of these clones showed that one amino acid substitution due to single base substitution is crucial in each chimeric choline acetyltransferase complementary deoxyribonucleic acid to generate a thermolabile choline acetyltransferase product. The point mutations of the structural gene for choline acetyltransferase are thus confirmed and shown to regulate the thermolability of the enzyme produced by Cha(ts1) and Cha(ts2).
A case of an unusual neoplasm of the buccal mucosa nearly identical to an ameloblastoma is reported. The tumor probably arose from the overlying squamous epithelium in the vicinity of the parotid duct.
As part of our search for active agents against systemic fungal infections, a new series of triazole compounds with a thiolane ring was synthesized. Their antifungal activities were investigated in vitro and in vivo. Some of these thiolanetriazoles showed promising activity, comparable to that of ketoconazole, against a mouse systemic Candida albicans infection, after oral or parenteral dosing.
Triazole compounds with an oxazolidine ring were designed and synthesized as a potential inhibitor of the fungal cytochrome P450 14 alpha-demethylase. In testing for antifungal activity against a mouse systemic Candida albicans infection, (4R,5R)-3-acyl-4-methyloxazolidine derivatives 4 exhibited remarkably high efficacy after oral or parenteral dosing. The potent activity of 4 is hypothesized to be a consequence of a structural similarity between 4 and lanosterol, a target molecule of the cytochrome P450 14 alpha-demethylase. Highly stereoselective synthesis of these oxazolidines is also described.

This clinico-pathological study was conducted to examine biopsies and surgical specimens for histopathological diagnosis from April 1983 to March 1988 in Meikai University (formerly Josai Dental University) Hospital. The following statistical results were obtained: 1) The total number of the specimens examined was 2705, 2026 from the dental clinic and 679 from the medical clinic. 2) Concerning age distribution, most specimens from the dental clinic were from patients in the 40's age bracket, whereas most from the medical clinic were from those under 10 years of age. With regard to sex distribution, the difference between males and females was not so much in the dental clinic specimens, but the number from the medical clinic was greater for males than for females. 3) Concerning classification of the tumors from the dental clinic, 181 cases were benign; and of them 41 were odontogenic and 140 were non-odontogenic tumors. Among the odontogenic tumors, ameloblastoma, cementoma, and odontoma were the most numerous. Among non-odontogenic tumors, fibroma was the most common, followed by pleomorphic adenoma. Malignant tumors comprised 77 cases. Of them squamous cell carcinoma was the most commonly found, with the grade-2 type being most numerous. Of the medical clinic tumors, nevus was the most common benign tumor observed; and adenocarcinoma, the most common malignant one. 4) Concerning the distribution of main lesions in the dental clinic specimens, most specimens were cysts or cystic lesions, followed by inflammatory lesions and tumors. Among medical clinic specimens, inflammatory lesions were the most common. 5) Regarding the dental clinic specimens, the most frequently observed case was the radicular cyst (301 samples) among the odontogenic cysts (514 samples); and among non-odontogenic cysts (290 samples), the most common was the mucous cyst (159 samples). 5) In terms of the distribution by location, tissue or organ, most tumors from the dental clinic were from the jaws, followed in descending frequency by those from gingiva and alveolar mucosa, maxillary sinus, lip, buccal mucosa, and tongue. Among the medical clinic tumors, the paranasal cavity and tonsils were the most frequent locations.

A 51-year-old woman as admitted to our department because of multiple palpable tumors that were noted on the left breast, the perineum, and the neck, shoulders, arms, and legs. The breast tumor showed ulceration on the surface. The perineal tumor was 30 x 15 cm in size and had group slowly for 17 years. A chest X-ray revealed a bilateral coin lesion. She was diagnosed as having a left breast cancer with a skin lung metastasis and a perineal benign tumor. A simple mastectomy and a resection of the perineal tumor were performed. A postoperative examination revealed an adenocarcinoma of the breast and a fibroneurinoma of the perineum with a metastasis from the breast cancer.
We examined the anticoagulant effects of phosphotungstate (PTA). PTA inhibited factors IIa and Xa activity only in the presence of antithrombin III (ATIII). The inhibitory effect of PTA on IIa was much smaller than that on Xa. When PTA was added to heparin (Hep), the inhibitory effect of Hep on IIa was reduced. As a result of charge interaction, PTA showed an inhibitory effect on anion transport activity of the erythrocyte, a potent salting-in effect and a metachromatic reaction on toluidine blue (TolB). Silicotungstate (STA) showed almost the same results as PTA. These effects of PTA and STA were not replaced by their constituents, tungstate, silicate and phosphate, but may arise from their high anionic valency.
Mandibular condylar cartilage from normal and low phosphate-vitamin D deficient rats were investigated by lectin histochemistry utilizing wheat germ agglutinin (WGA). We obtained the following results: 1) The cartilage from low phosphate-vitamin D deficient animals consisted predominantly of hypertrophic chondrocytes with abundant extracellular matrix. 2) The territorial matrix of the hypertrophic cell zone of normal cartilage was intensely labelled with WGA, whereas the binding affinity of this region in low phosphate-vitamin D deficient animals was significantly diminished. 3) Compared to normal cartilage, WGA binding sites were demonstrated in the inter-territorial matrix of low phosphate-vitamin D deficient animals to a moderate degree. 4) Abolishment of the territorial WGA binding affinity in low phosphate-vitamin D deficient animals was presumed to indicate the accumulation of chondroitin sulfate which is believed to be a strong chelator of calcium ion.
The serum and bile tegafur (FT), 5-fluorouracil (5-FU) and uracil levels after administration of UFT were assessed in 13 cases of malignant biliary tumor accompanied by biliary obstruction in comparison with FT alone. The serum and bile FT and 5-FU levels showed almost the same transition pattern in both groups, reaching to the plateau in 1-2 weeks and revealing cumulative effect by continual administration. Correlation was obtained between serum and bile levels except for 5-FU level in UFT group (p less than 0.05). Correlation between 5-FU and uracil was obtained in the serum in both groups (p less than 0.05), but no effect of uracil was observed. In bile, correlation was seen only in UFT group (p less than 0.05), and the effect of uracil was observed in bile 5-FU level.

Kinetic studies of [14C]HCO3- and [35S]SO4(2-) transport show the existence of a common transport system for the two anions on the plasma membranes of porcine epididymal sperm. Km value for bicarbonate (8.3mM) is within the range of physiological concentration of bicarbonate. The anion transport activity is dependent on the temperature but independent of co-existence of Na+. Its pH dependency and sensitivity to the several anion transport inhibitors are similar to those observed in erythrocyte. Anion transport activity decreases during sperm maturation in epididymis.

We have identified and localized two incompatibility determinants (IncA and IncB) within a 1.3 kb segment of ColE2 sufficient for autonomous replication. The IncA determinant is localized in a region shorter than 250 bp and expresses incompatibility against both ColE2 and ColE3. The region which determines sensitivity to the IncA determinant seems to overlap with the region specifying the IncA determinant. The expression of the trans-acting factor(s) specifically required for replication of ColE2 interferes with expression of the IncA determinant against ColE2 but not against ColE3. The IncA determinant might be at least partly responsible for the copy number control of the plasmid. The IncB determinant is localized in a 50 bp region (origin) which is sufficient for initiation of replication in the presence of the trans-acting factor(s). The IncB determinant is specific for ColE2 and seems to be due to titration of the trans-acting essential replication factor(s) by binding.
