Subcellular localization of D-glucanases in Bacteroides oralis Ig4a.

Three D-glucan-hydrolysing enzymes from Bacteroides oralis Ig4a have been isolated. Two of them are dextranases which hydrolyse (1----6) but not (1----3) linked alpha-D-glucans; one (EC 3.2.1.11, 1,6-alpha-D-glucan 6-glucanohydrolase) is localized in the periplasm, and the other, which is an exo-enzyme (EC 3.2.1.70, 1,6-alpha-D-glucan glucohydrolase), in the cytoplasm. The third is a mutanase (EC 3.2.1.59, 1,3-(1,3;1,4)-alpha-D-glucan 3-glucanohydrolase) that hydrolyses (1----3) but not (1----6) linked alpha-D-glucans, and is present only in the cytoplasm.

Comparison of disappearance from blood and lymphatic delivery of human fibroblast interferon in rat by different administration routes.

Disappearance from the blood and lymphatic delivery of human fibroblast interferon (HuIFN-beta) in rats by the various administration routes were studied. Intravenous injection showed a rapid disappearance of HuIFN-beta from the blood circulation, however, the clearance rate was greatly decreased after 2 h and the half-lives were 15 min and 2.0 h. Intramuscular, subcutaneous and intraperitoneal injections maintained readily detectable levels in the blood for 12 h, and the half-lives were 2.3, 1.7 and 1.2 h, respectively. Lipid-surfactant mixed micelles greatly promoted the absorption of HuIFN-beta from the large intestine with extremely high selective lymphatic delivery, whereas intravenous and intramuscular injections showed no selective transfer into the lymphatics.

Indications for and limitations of coronary thrombolysis.

Experimental studies in dogs with coronary thrombi induced by copper wire confirmed the optimal method of intracoronary thrombolysis, and showed that a high-dose, brief intravenous infusion of urokinase can lead to recanalization. The thrombolytic effects of intracoronary thrombolysin at a rate of 50 IU/kg over 10 minutes are similar to the effects of intracoronary urokinase at a rate 500 IU/kg over 20 minutes. Overall reperfusion rates of 83-86% have been achieved. These results indicate that the thrombolytic effect of thrombolysin is 20 times stronger than that of urokinase. The effect of a brief intravenous infusion of urokinase was less than that of intracoronary urokinase. The reperfusion rate in the same experimental model was 40%. Later, a clinical trial of intracoronary urokinase was performed in 47 patients with acute myocardial infarction who were admitted within 12 hours of the onset of symptoms. In 25 of 33 (75.8%) patients with complete occlusion, selective or ostial infusion of urokinase 500 IU/kg over 20 minutes was successful. When given intravenously, recanalization was achieved in 11 of 15 (73%) patients with complete occlusion who were admitted within 6 hours. Both reperfusion rates were similar except for dosage and the duration of infusion.

Properties of binding of partially purified glucocorticoid receptor from rat liver with glucocorticoids of different biopotencies.

To elucidate the relationship between binding parameters and biopotencies of glucocorticoids, we partially purified the receptor from the liver cytosol of rats in a dexamethasone-bound and unactivated form by precipitation with protamine sulfate, gel filtration and DEAE-cellulose chromatography (approximately 100-fold) and examined the interaction of the preparation with 3 glucocorticoids of different biopotencies (dexamethasone; Dex, corticosterone; Cort and prednisolone; Pred). The partially purified receptor (PPR) was stable at -20 degrees C for at least 2 months in the presence of bovine serum albumin, glycerol, molybdate and dithiothreitol. Treatment of the PPR with p-hydroxymercuribenzoate liberated the ligands and the treated PPR reassociated 3H-glucocorticoids efficiently following the addition of dithiothreitol. The reassociated PPR was bound to the DNA-cellulose after a brief heating. Metabolic activity on ligands and inactivation of the binding sites in the PPR were insignificant under the conditions used. Kd's were approximately 0.9, approximately 3 and approximately 6 nM for Dex, Cort and Pred, respectively (at 0 degree C). Relative binding affinity of ligands to the PPR which was estimated by competitions was higher in the order of triamcinolone acetonide greater than Dex greater than Cort greater than Pred greater than progesterone greater than cortexolone. Association of Dex and Cort was relatively rapid and significantly accelerated by raising the incubation temperature, while the association of Pred was slower and effects of the temperature was moderate. The rate of dissociations was also varied with ligands. The rate of dissociation of Dex was the lowest among the 3 ligands and was elevated by raising the temperature. Because the effect of temperature was more pronounced in the dissociation than in the association, apparent Ka's decreased at higher temperature. Thermodynamic examinations of glucocorticoid binding in the PPR revealed that the binding reaction proceeds at a higher rate in the order of Dex greater than Cort greater than Pred. Because the relative biopotencies of these 3 glucocorticoids in vivo is higher in the order of Dex greater than Pred greater than Cort, from the results obtained in the present study, it appears that biopotency of glucocorticoids in vivo does not correlate with the affinity of the binding to the receptor estimated in vitro.

[Clinical effectiveness of KL 007 (oxybutynin hydrochloride) in urinary disorders].

The clinical effectiveness of KL 007 (oxybutynin hydrochloride) was studied on patients with urinary disturbance, mostly complaints of urinary frequency, who were suffering from nervous pollakisuria, irritable bladder or neurogenic bladder. Fifty two patients were administered KL 007 three times daily at the dose of 6 mg, 9 mg, or 12 mg per day for one or two weeks. An overall clinical effectiveness of KL 007 was seen in the cases of hypertonic bladder. As to the clinical effectiveness of subjective symptoms, KL 007 improved the symptoms of urinary frequency, urinary incontinence, discomfort upon micturition, urinary urgency and sense of residual urine. Transient side effects were observed in 23 cases (41.4%), and dry mouth was most observed. No severe side effects were noticed.

Plasma cell leukemia accompanied by polyclonal gammopathy and congenital tumor in the atrioventricular node.

A 32-year-old man with plasma cell leukemia and congenital tumor of the atrioventricular node is described. Although M protein could not be detected by immunoelectrophoresis, the examination of bone marrow atypical plasma cells with immunofluorescent and immunoperoxidase showed that most of these plasma cells produced IgG or IgA. Also some plasma cells possessed concomitantly both IgG and IgA. An electrocardiogram revealed complete atrioventricular block. The patient died of aspergillosis and bleeding in the lung. Postmortem examination revealed the infiltration of atypical plasma cells in the bone marrow, spleen, liver and lungs, and infiltration of mesothelial tumor cells in the atrioventricular node.

Scanning electron microscopical observation on the isolated mucosa of rat small intestine: with special reference to the intestinal crypt.

The small intestinal mucosa of rats (male, aged 8 weeks) was isolated by the Bjerknes and Cheng's method (1981): the anesthetized rats were perfused through the left ventricle with a solution of ethylenediaminetetraacetic acid (30 mmol/l) in calcium-magnesium-free Hanks' solution, and then the mucous epithelium was removed from a segment of the duodenum, jejunum and ileum by means of vibration. Three-dimensional images of crypts on the isolated mucosa were obtained by scanning electron microscopy. The length and number of the crypts between the villi were observed to decrease along the intestine from the duodenum to the ileum. The majority of intestinal crypts were simple non-branched tubular glands, however, budding and bifurcation of the crypt ends were often observed. It was relatively rare to find longitudinally split crypts. The shapes of the crypts seems to denote various stages of the proliferation of crypts.

Assignment of two Japanese xeroderma pigmentosum patients to complementation group D and their characteristics.

Japanese xeroderma pigmentosum sib patients XP58TO and XP59TO were assigned to complementation group D on the basis of cell hybridization studies. Ultraviolet and 4-nitroquinoline-1-oxide hypersensitivity and reduced unscheduled DNA synthesis of cells of these XP patients were also characteristic of authentic group-D cells. The patients have not yet developed either apparent neuromental abnormalities or skin cancers.

DNA repair and its possible involvement in the origin of multiple cancer.

Multiple skin cancers and other cancers in patients with xeroderma pigmentosum (XP) were investigated in relation to DNA repair defects in the cells of the patients. Multiple skin cancers of the same or different histopathological types were found in many patients and six patients had cancers, one each, in organs other than the skin. The frequency of basal cell carcinoma was higher than that of squamous cell carcinoma in XP patients, while the two types were reported to be approximately equal in frequency in all skin cancers in Japanese. Defect in the excision-resynthesis type of repair presumably enhances the error-prone type of repair of DNA damage in XP patients and may lead to the development of cancer. Although a similar DNA repair defect of damage caused by ionizing radiation has been suspected in ataxia telangiectasia (AT), induction of mutation by gamma-rays in AT cells was lower than that in normal cells at the same survival levels. The high incidence of malignancy in AT patients could be due to factors not associated with DNA repair.
