Studies on the castability of Co-Cr alloy for cast plates. Part 2: Effect of form of sprue attachment to the wax pattern.

Co-Cr alloy is used more frequently than Ni-Cr alloy as a non-precious alloy for cast plates in Japan. However, since the melting point of Co-Cr alloy is very high, about 1300 degrees C, and since it oxidizes easily, a vacuum-pressure casting machine capable of melting this alloy in a reducing atmosphere has recently been developed. Using this vacuum-pressure casting machine, the authors studied the effects on the castability of Co-Cr alloy due to the form of sprue attachment to the wax pattern. The results clarified that in the vacuum-pressure casting method, the form of sprue attachment to the wax pattern has a significant effect (p less than 0.01) on the castability of Co-Cr alloy.

Electroporation-transformation system for coryneform bacteria by auxotrophic complementation.

We evaluated electroporation as an alternative system for genetic exchange for one of the coryneform bacteria, Brevibacterium flavum MJ233. The maximum number of transformants, 6 x 10(4) cells, was obtained when cells were cultured with Penicillin G (1 U/ml) and harvested at the middle-log phase. Electroporation was done using 12.5 kV/cm of pulse field strength, 1 x 10(10) cells, and 1 microgram of plasmid DNA. Other coryneform bacteria, Brevibacterium lactofermentum ATCC 13869, Corynebacterium glutamicum ATCC 31830, and B. stationis IFO 12144 were also transformed by electroporation. Electroporation has the advantage that intact cells can be used as host cells without the need for protoplast formation and regeneration. Moreover, minimal medium can be used, so auxotrophic complementation of the transformants is possible.

[A clinical study on cefdinir in pediatrics].

Clinical evaluations of cefdinir (CFDN, FK482) were carried out. The obtained results are summarized as follows. 1. Clinical responses to CFDN of 43 patients with pediatric infections were excellent in 23, good in 15, fair in 4 and poor in 1. The overall efficacy rate was 88%. 2. Bacteriologically, the eradication rate for 34 isolates presumed to be pathogens was evaluated and the eradication rate was 85.3%. 3. Side effects observed were diarrhea in 4 of 48 patients. The incidence was 8.3%. Abnormal laboratory findings were an elevation of GOT, an elevation of eosinophils and an increase in platelet counts. The results suggested that CFDN might be a very useful and safe drug for the treatment of pediatric infections.

Psammoma bodies in metastatic carcinoma to the thyroid.

In a review of the 427 cases of thyroid with malignant neoplasm that were surgically resected at the Cancer Institute Hospital during the 10-yr period from 1978 to 1987, five cases of metastatic carcinoma to the thyroid were found. Psammoma bodies were histologically observed in three of the five cases. Primary thyroid carcinoma was highly suspected in the three cases, because psammoma bodies were detected during preoperative examinations that included image diagnosis and aspiration biopsy cytology. Thus, the presence of psammoma bodies in any kind of thyroid tumor should be carefully evaluated, both clinically and histologically, to ensure that the possibility of metastatic origin is not overlooked.

[The relationship between responsiveness to splenectomy and clinical course after splenectomy in chronic idiopathic thrombocytopenic purpura (ITP)].

Fifty-four ITP patients were treated by the protocol proposed by Japanese Idiopathic Disease of Hematopoietic Organ Research Committee. Forty-eight patients were treated with corticosteroid (CS). Twenty-eight patients out of 30 patients who were refractory to CS were splenectomized. To study natural courses of splenectomized patients, 25 patients were splenectomized without elevation of platelet counts at the operation and were followed up their platelet counts and PAIgG for one month without treatment. Clinical courses after splenectomy were classified into four types by the platelet increment followed by splenectomy and the platelet counts at one month after splenectomy; type I (markedly over-shooted platelet peak and normalized platelet counts), type II (moderate platelet peak and 50 approximately 100 x 10(3)/microliters platelets), type III (minimal platelet peak and less than 50 x 10(3)/microliters platelets), and type IV (elevation of platelet counts at day 7 or later of splenectomy and normalized platelet counts). The results of analyses from 25 patients indicated that the numbers of type I, II, III and IV were 10, 5, 6 and 4, respectively. The clinical course of type IV suggests that platelet production may be impaired by anti-platelet antibodies.

[Primary lung cancer in young adults].

Despite a recent trend of rapid increase in the incidence of lung cancer in Japan, lung cancer has been detected rarely among young patients. The findings in 15 patients under 40 years of age who underwent surgery for primary lung cancer at Cancer Institute Hospital (C.I.H.) are presented. Fourteen were located in the region of the periphery. Ten of the 14 peripheral lesions were adenocarcinoma. The chances of cure or even long survival are very small. The case report presents a case with lung cancer in a 22-year-old woman who was the youngest in C.I.H. On histological examination, the excised tumor proved to be moderately differentiated adenocarcinoma.

[A case report of anesthesia for concomitant right pneumonectomy and coronary artery bypass operations].

A 61-year-old man with previous myocardial infarction, was diagnosed as having lung cancer. Coronary arteriogram revealed stenoses of left anterior descending artery. We did the concomitant pulmonary and cardiac operations. Anesthesia was induced with fentanyl 1.5 mg and pancuronium 6 mg. A 37 Fr double-lumen endotracheal tube was inserted. Then a pulmonary artery catheter was inserted. The patient was given nitroglycerin for prevention of myocardial ischemia. The tumor had invaded pulmonary artery and therefore right pneumonectomy was necessary. After resection of right lung, coronary artery bypass operation was performed. On weaning from cardio-pulmonary bypass, pulmonary artery pressure increased to 48/20mmHg. Therefore he required dopamine and dobutamine each 4 micrograms.kg-1.min-1 for weaning. But we experienced no serious complications such as hypoxemia or perioperative myocardial infarction.

[Study of action spectrum of papulovesicular light eruption (PVLE)].

Eight patients suffering from papulovesicular light eruption (PVLE) were phototested for reproduction of skin lesions with FL20SE and FL20BLB. Of the 8 patients, 50.0% developed typical lesions of PVLE at the test site by provocative phototesting with FL20SE (3 daily exposure of 2MED). In these patients, similar lesions were induced by the provocative test with FL20BLB for 2 consecutive days without filter, but no eruptions were induced when a sharp cut filter UV-35 was used. In conclusion, it is important to consider not only UVB but also the border spectrum between UVB and UVA (320-340 nm) as the action spectrum of PVLE.

Pulmonary sclerosing hemangioma of the lung. A type II pneumocytoma by immunohistochemical and immunoelectron microscopic studies.

Three cases of pulmonary sclerosing hemangioma were studied by immunohistochemical and immunoelectron microscopic methods using a panel of antibodies. Six cases of adenocarcinoma of the lung, three cases of normal mesothelium, and three cases of mesothelioma were used as controls. The cytoplasm of some of the sclerosing hemangioma tumor cells was positive for the anti-lung surfactant apoprotein monoclonal antibody (PE-10). These cells were the pale cells of the solid areas, the cells covering the papillary projections, and the cells lining the cleft-like spaces. These cells also were positive for conventional epithelial cell markers. Some cells also were positive for vimentin. Electron microscopic study showed that the predominant cell was a poorly differentiated pneumocyte. Immunoelectron microscopic study also demonstrated that PE-10 existed in the rough endoplasmic reticulum of some of the cells in the solid areas, in the same way as normal type II pneumocytes. We concluded that the sclerosing hemangioma is an epithelial tumor with differentiation towards type II pneumocytes.

Dopamine receptors in the stellate ganglion of the dog.

Effects of fenoldopam, a selective DA1 dopamine receptor agonist, and dipropyl dopamine and propylphenethyl dopamine, preferential DA2 dopamine receptor agonists, on ganglion transmission were studied in pentobarbital-anesthetized, open-chest dogs. Tachycardia induced by electrical stimulation (supramaximal voltage, 0.5 ms duration, 1-2 Hz) of the preganglionic cardio-accelerator nerves was monitored as a measure of ganglionic transmission. Drugs were injected into the costocervical artery (i.a.) close to the arterial supply of the ganglion. Doses required to produce 30-40% inhibition of ganglionic transmission by the i.a. route were 2-8 micrograms for dipropyl dopamine, 4-16 micrograms for propylphenethyl dopamine, and 100 micrograms for fenoldopam. At these doses none of the agonists affected tachycardia induced by electrical stimulation of the postganglionic nerve. Domperidone (5 micrograms/kg i.v.), a selective DA2 dopamine receptor antagonist, markedly antagonized the effects of dipropyl dopamine and propylphenethyl dopamine, but had only minor (and statistically insignificant) effects on the inhibitory effect of fenoldopam. SCH 23390 (5 micrograms/kg i.v.), a selective and potent DA1 antagonist, failed to modify the effects of any of the agonists. In a separate series, infusion of fenoldopam, 20 micrograms/kg per min i.v. for 5-7 min, facilitated postganglionic nerve stimulation and blocked the inhibitory effect of UK 14,304, an alpha 2-adrenoceptor agonist, on the postganglionic nerve. These results confirm and support the presence of DA2 but do not support the presence of the prototypal DA1 dopamine receptor in the mammalian ganglia. Furthermore, the alpha 2-adrenoceptor blocking property of fenoldopam points to the complication of using i.v. administration for studying its ganglionic actions while monitoring the target tissue effects in response to preganglionic nerve stimulation.

Reduced levels of UV-induced unscheduled DNA synthesis in epidermal keratinocytes of patients with xeroderma pigmentosum and correlation with development of skin neoplasms.

Primary epidermal keratinocytes obtained from 25 patients with xeroderma pigmentosum (XP) (nine with XP-A, one with XP-C, two with XP-D, five with XP-E, and eight with XP-variant) exhibited less UV-induced unscheduled DNA synthesis (UDS) than did those from 34 normal subjects. Levels of UDS depended greatly on the type of XP; i.e., 3-17% of the control in XP-A, 14% in XP-C, 33-53% in XP-D, 38-77% in XP-E and 58-98% in XP-variant. The extent of UDS in epidermal keratinocytes was almost the same as that in dermal fibroblasts in XP-C, D, and E, but in three out of eight of the XP-variant the level of UDS in epidermal keratinocytes was significantly lower than that in normal subjects. Clinically, three out of nine XP-A patients developed skin neoplasms before 20 years of age. Both patients with XP-D developed skin neoplasms around 40 years of age. In the five XP-E patients, two developed multiple basal cell epithelioma on sun-exposed areas during the forth decade, and one of them also developed squamous cell carcinoma at the age of 50. Four out of the eight patients with the XP-variant developed various skin neoplasms during their 20s and 30s. These results suggest that a defect in UV-induced UDS in epidermal keratinocytes of XP patients is responsible for skin carcinogenesis and the extent to which this defect occurs tends to relate to the age of onset of skin neoplasms.

Effects of cholecystokinin and carbamylcholine on Paneth cell secretion in mice: a comparison with pancreatic acinar cells.

To confirm whether the Paneth cells of mice (ICR, male, 10-12 weeks old) have the same secretory response to hormonal and cholinergic stimulation as do pancreatic acinar cells, ultrastructural changes of Paneth cells and pancreatic acinar cells 1 hr after administration of various doses of cholecystokinin (octapeptide, CCK-8) and carbamylcholine were morphometrically assessed. After maximal (1.5 micrograms/kg intraperitoneally [i.p.]) and supramaximal (15 micrograms/kg, i.p.) stimulation by CCK-8, pancreatic acinar cells showed, respectively, degranulation or disturbance of secretion (e.g., an increase in lysosome-like bodies, aggregation of zymogen granules). The Paneth cells, however, were almost unchanged in the parameters examined. After carbamylcholine injection (1,000 micrograms/kg, subcutaneously [s.c.]), both pancreatic acinar cells and Paneth cells showed degranulation. Paneth cells sometimes developed large vacuoles, probably formed after massive exocytosis; such vacuoles were not observed in pancreatic acinar cells. It is suggested that Paneth cells and pancreatic acinar cells have different secretory responses. Paneth cell secretion, which possibly plays a role in controlling the intestinal bacterial milieu, may be stimulated by cholinergic rather than hormonal mechanisms.

Characterization of human residual catalase of an acatalasemic patient by isoelectric focusing and sodium dodecyl sulfate-polyacrylamide gel electrophoresis followed by electrophoretic blotting and immunodetection.

Isoelectric points and subunit sizes of catalases in human blood and human cultured skin fibroblasts from acatalasemic and normal subjects were analyzed by isoelectric focusing in agarose gel and by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, respectively, followed by electroblotting to polyvinylidene difluoride membranes for immunodetection. The results indicated that the isoelectric point of residual catalase in the C fraction prepared from acatalasemic erythrocytes was identical with that of catalase prepared from normal erythrocytes. The residual catalase in homogenates of acatalasemic cultured skin fibroblasts also reacted with anticatalase rabbit serum and had an isoelectric point identical with that of normal catalase. Subunit sizes of normal and acatalasemic catalases in the C fractions of erythrocytes were also found to be identical on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, followed by electroblotting and immunoenzymatic amplification. The results indicated no substantial difference in molecular size and charge of catalase proteins between normal and acatalasemic erythrocytes and fibroblasts.

Influence of conventionalization on cecal wall structure of germ-free Wistar rats: quantitative light and qualitative electron microscopic observations.

The structural changes of the cecal wall in germ-free rats were observed at regular intervals after the inoculation of fecal microflora from conventional rats. Quantitative light microscopy showed that most of the elements in the cecal wall increased at 12 or 24 h and reached peak values at 4 days after inoculation. On the 7th day, they decreased approximately to the values for conventional rats. The crypts were bent or widely open till 24 h but were not after the 4th day. Hyperplasia of the crypt epithelial cells including mucous-type cells was observed following microbial inoculation. Electron microscopy revealed that most of the epithelial cells lining the mucosa were typical columnar cells. Desquamation of the epithelial cells and contraction of the muscle fibers were often seen on 4th day. The mucous-type cells were divided into two types, goblet and non-goblet mucous-type cells. Reduction of cecal volume after microbial inoculation may be mainly caused by muscle contraction in the early period and hyperplasia and desquamation of the epithelial cells may suggest their role as the first and non-specific defense line prior to operation of the specific immune system.

Tyrosine-hydroxylase-immunoreactive nerve fibers in the separated capsule of the rat adrenal gland.

The present study applied the separated adrenal capsules of rats for wholemount immunocytochemistry and used tyrosine hydroxylase (TH) antibody as a marker for catecholamines. TH-immunoreactive nerve bundles without varicosities and fibers with varicosities were seen to run along or to encircle blood vessels entering the adrenal capsule from the outside, and then to run along a network of blood vessels in the intracapsular region. Also, the TH-immunoreactive nerve bundles and fibers were found to run along blood vessels in the subcapsular region. Some TH-immunoreactive nerve fibers and bundles with varicosities, unassociated with the blood vessels, were seen in the subcapsular region. In this region, TH-immunoreactive nerve fibers with varicosities were often seen to be closely associated with the cortical cells. Some TH-immunoreactive nerve fibers without varicosities were visible within the splanchnic nerve in the subcapsular region. The present study suggests that numerous catecholaminergic nerve fibers are associated with blood vessels forming a network in the superficial region of the rat adrenal gland.

Studies on the monoamine oxidase (MAO) inhibitory potency of TL-1, isolated from a fungus, Talaromyces luteus.

The compound tentatively named TL-1 was isolated from Talaromyces luteus as a metabolite having monoamine oxidase (MAO) inhibitory potency. TL-1 showed mixed-type inhibition of MAO in mouse liver when kynuramine was used as a substrate, and the IC50 was 6.6 microM. The inhibition constants (Ki) for MAO-A and -B in mouse liver were 39.9 and 7.85 microM, respectively. On the other hand, the Ki values for MAO-A and -B in mouse brain were 74.0 and 0.71 microM, respectively. Despite the marked structural resemblance between TL-1 and 7-episclerotiorin, the latter compound had little inhibitory effect on MAO.

Preventive effect of taurine against acute paraquat intoxication in beagles.

The continuous infusion of taurine markedly protected against paraquat (PQ)-induced oliguria in beagles. Pharmacokinetic studies revealed that taurine infusion increased the blood concentration of PQ and reduced the net content in the kidney and, to a lesser extent, in the lung. The excretion of PQ into the urine was unaltered. The infusion of glycine did not have such effects.
