Industrial Application of Artificially Induced Diploid Strains of Torulaspora delbrueckii.

Diploid strains of Torulaspora delbrueckii were tested for industrial application. Because the cell volume of the diploid strain was three times as large as that of the parental haploid strain, collection and subsequent dehydration to make compressed yeast cakes were greatly improved with the diploid YL3. The time required for dehydration of the diploid strain was shortened to 1/2.5 that of the parent strain under conventional conditions. Moreover, for the diploid cells frequent filter changes for dehydration were not required, which was the case with parental cells. Fermentation activity and tolerance to freeze-thawing in dough were succesfully inherited by the diploid strains. The diploid YL3 showed nearly the same activity as the diploid F31 in bread making. However, the endurance period of yeast cakes when stored at 30 degrees C without softening to lead to liquefaction was much longer in YL3 (199 h) than in F31 (132 h). This superiority was ascribed to the fact that YL3 was induced through direct diploidization and had no genetic defect on chromosomes because the wild-type strain was employed as the parent, whereas F31 was obtained through protoplast fusion from two auxotrophic mutants and carried at least two mutagenized genes that were masked by heterolallelism.

Cell surface antigen of encapsulated Staphylococcus epidermidis ATCC 31432.

The cell surface antigen (CSA) from encapsulated Staphylococcus epidermidis ATCC 31432 was isolated and fractionated by DEAE-Sephadex ion-exchange chromatography. It yielded a single precipitin line against rabbit antiserum and was composed of galactose, glucosamine, and two (so far) unidentified carbohydrates linked to a protein backbone. Glycerol could not be detected. In an experimental mouse infection, CSA showed some protective activities, enhancing the stimulation of granulocytes as well as the immunoglobulin M response. Apparently, CSA is different from teichoic acid.

Necrobiosis-lipoidica-like skin manifestation in lymphomatoid granulomatosis (Liebow).

A 58-year-old man developed 3 indurative erythematous lesions like necrobiosis lipoidica on the right lower leg. He had had similar cutaneous lesions 1.5 years previously. These had been surgically excised, and a histologic diagnosis of necrobiosis lipoidica was made at another hospital. He was diagnosed as having lymphomatoid granulomatosis by lung biopsy in our hospital. Nasal involvement was confirmed in later examinations and the skin lesions were also considered to be the cutaneous manifestation of lymphomatoid granulomatosis. This outlines that cutaneous manifestations may allow early diagnosis of lymphomatoid granulomatosis.

Molecular cloning of cDNA of S100 alpha subunit mRNA.

The primary structure of the bovine S-100 alpha mRNA on the basis of molecular cloning and sequence analysis of the cDNA are described. The sequence is composed of 532 bp which include the 282 bp of the complete coding region, 89 bp at the 5'-noncoding region, 161 bp at the 3'-noncoding region, polyadenylation signal, ATTAAA and poly(A) tail. Northern blot analysis shows that the size of S-100 alpha mRNA is about 700-800 bases long and a single mRNA occurs in bovine brain. Bovine brain contains both S100 alpha and beta subunits and their mRNAs. In contrast, the rat brain contains only S100 beta subunit and its mRNA.

Three distinct activities possibly involved in mRNA splicing are found in a nuclear fraction lacking U1 and U2 RNA.

A nuclear extract from HeLa cells was fractionated by DEAE-Sepharose chromatography, and the fractions were assayed for the binding activity for a small RNA transcript carrying a splice junction or branch point sequence. The binding activity for the RNA carrying a 5' splice junction was localized in the small nuclear ribonucleoprotein (snRNP) fraction together with binding activity for the RNA carrying a 3' splice junction or branch point sequence. However, stronger binding activities for the 3' splice junction RNA and for the branch point RNA were discovered in the flow-through fraction where no small nuclear RNA were detected. When the flow-through fraction was added to purified U1 ribonucleoprotein, the binding activity for the 5' splice junction RNA was markedly enhanced. We propose that the factors responsible for the three types of activities found in the flow-through fraction play a role in the splicing of mRNA precursor.

A case of virus-associated haemophagocytic syndrome with hypoplastic anaemia.

A 4-year-old girl with virus-associated haemophagocytic syndrome is described, suffering from fever, jaundice, rectal bleeding and a loss of consciousness. Severe pancytopenia was a prominent finding. The bone marrow biopsy showed hypocellularity, and bone marrow aspiration revealed proliferation of histiocytes containing many erythrocytes and thrombocytes. The patient recovered within several weeks of supportive therapy. Viral studies demonstrated a recent infection with Epstein-Barr virus.

[Neonatal subdural hematoma secondary to tentorial tearing--clinical analysis on 48 survived cases].

In order to evaluate the treatments of subdural hematoma in neonates and to observe their prognosis, we carried out clinical analyses on 48 survived cases in the past three years from Jan. 1979 to Dec. 1981. Based on CT observations, hematomas were grouped into four types according to their locations as follows: Type I, localized around the posterior interhemispheric fissure in 25 cases (52%); Type II, extending from the posterior interhemispheric fissure to the hemispheric convexity in five cases (10%); type III, extending from incisura to the posterior fossa in 15 cases (31%); and type IV, having subdural hematoma accompanied by intracerebral hemorrhage in three cases (7%). Intracranial pressure was measured via the anterior fontanelle in 13 cases. Ten patients who had extensive hemorrhage were found to exceed above 200 mmH2O in the pressure. Ages of the patients studied were 0 to 7 days old. There were 36 mature infants (75%) and 12 cases immature (25%). The maternal history of delivery was primiparous in 27 cases (56%). The fetal presentations were 38 cephalic (79%) and 10 suction deliveries (21%). The fundus oculi was examined on 32 patients. Retinal hemorrhage was noted in 12 cases. Operations were performed on 13 patients in total; one case in type I, four cases in type II, five cases in type III and three cases in type IV. Functional prognoses were found to be as follows; Type I; normal, 15 cases, abnormal, four and undetermined, six: type II; normal, four and abnormal, one: type III; normal, 13, abnormal, one, and undetermined, one: and type IV; normal, two and abnormal, one case.
