Effects of irradiation on the ciliary activity of the eustachian tube and the middle ear mucosa.

Radiation-induced changes in the ciliary activity of the eustachian tube and middle ear mucosa were investigated in a laboratory model. Twenty-four guinea pigs were sacrificed, and a mucosa at the bony portion of each eustachian tube and from the middle ear proximal and distal to the tube were sampled. The mucosal samples were irradiated with 200 kV hard X-rays discretely at a dosage level of 0.5-30.0 Gy, and the induced changes in ciliary activity were expressed as a percent deviation from baseline levels before irradiation. The present study demonstrates that the cilia of the eustachian tube and middle ear mucosa show different reactions to irradiation according to their tympanic locations.

Topical immunology of nasal allergy and mucosal IgE antibodies.

In the present study, the in vitro tissue-radioallergosorbent test (t-RAST) was performed in two groups of patients: one with perennial attacks of sneezing, serous hypersecretion and nasal congestion, the other with nasal congestion only. The results obtained were compared with those obtained by a series of conventional allergy tests. We then found that t-RAST provided objective data comparable to those obtained with serum-RAST and that the t-RAST is a reliable means of quantitatively detecting specific IgE antibodies in the nasal mucosa. t-RAST is of special value to diagnosticians because it is able to discern unequivocally and easily those patients with localized nasal allergy.

Ciliary activity in patients with nasal allergies.

Nasal allergy is a disease ensuing from type-I allergic reactions in the nasal cavity. A correct understanding of this process requires further investigation of the non-allergic pathology present. In the allergic state, the function of the nasal cilia is considered to be involved in the invasion of allergens into the nasal mucosa, and may also be involved with the manifestation and progression of the disease produced. In the present study, we examined the ciliary activity of the nasal mucosa of 35 patients with nasal allergies by using the photo-electric method of Ohashi and Nakai to better understand the non-allergic pathologies present. The nasal cilia of four healthy volunteers served as controls. In addition, a multivalent analysis was made according to quantifying theory I in order to define the effects of various factors on ciliary activity in the nasal mucosa during the allergic state. The following results were found in our present study. Ciliary activity was observed in 32 of 35 patients with nasal allergy. The mean value of the ciliary activity in these patients was 498 +/- 195 beats/min, while that of the normal controls was 753 +/- 46 beats/min. The ciliary activity in the nasal mucosa in the allergic state declined as the morbid period was prolonged; this decline in ciliary activity was relatively great in cases of perennial allergies aggravated by season-specific allergens.

Induction of SOS responses in Escherichia coli by Panfuran-S, 3-di(hydroxymethyl)amino-6-(5-nitro-2-furylethenyl)-1,2,4-triazine.

The inducibility of SOS responses by Panfuran-S, which has been used as an antimicrobial medicine in Japan, was studied in Escherichia coli cells having different DNA-repair capacities for UV lesions. Panfuran-S induced mutations at high frequencies in uvrA and the wild-type strains, and significant killing effects of Panfuran-S were detected in DNA-repair-deficient strains, uvrA and recA. The effective prophage induction was detected in two kinds of lambda-lysogenized cells treated with Panfuran-S. The expression of the umuC+ gene was apparently induced in uvrA and the wild-type strains, but not induced in lexA and recA strains. In particular, high inducibility of the gene expression was detected in uvrA strain as compared with the wild-type strain. From these results, we conclude that Panfuran-S is a DNA-damaging agent and may induce the error-prone SOS responses.

Blood loss with use of the Shaw scalpel for the treatment of oral cancer.

The Shaw scalpel was used in seven radical operations for oral cancer. The amount of bleeding and postoperative exudate and the occurrence of postoperative complications were compared with that from 12 operations performed with the conventional steel scalpel. The blood loss during the radical neck dissection performed with the Shaw scalpel was 39% of the control value, and no blood transfusions were necessary. There was no increase in the amount of postoperative exudate in the Shaw scalpel group compared with the control group. Skin incision with the Shaw scalpel was accompanied by superficial wound dehiscences in three patients, which healed without any treatment.

The ciliary activity of the middle ear lining--functional and morphological observation.

Since the middle ear lining is an extension and a modification of the respiratory mucosa, it is conceivable that it has a mucociliary system and it plays an important role in clearance of the tympanic cavity. It was already noted in morphological studies that the middle ear lining had ciliated cells. To our knowledge, however, ciliary activity has never been observed directly. In the present paper, the ciliary activity of the middle ear mucosa was investigated directly and quantitatively by the photo-electric method of Ohashi and Nakai. In this work, special reference was made to the frequency of ciliary beating at various sites within the tympanic cavity. Ciliary activity was found to exist in the Eustachian tube and middle ear same as in other respiratory epithelium and this activity by ciliated cells was more active as their location became more distal to the Eustachian tube. Although the present results are preliminary, the techniques used in this study will provide a new avenue of the investigation of pathological middle ear lining.

The ciliary activity of the middle ear lining in some pathological states.

The ciliary activity in some pathological middle ear lining was discussed with special reference to the morphology of the epithelial cells in this paper. The changes in ciliary activity due to in vitro Staphylococcus aureus injection did not practically show any differences between the sites. On the other hand, the time-course changes in ciliary activity after in vivo S. aureus injection were found to differ between the sites. Ciliary activity was rather accelerated at the proximal site, while it was decelerated at the distal site. Few changes but for sporadic small compound cilia were observed at the proximal site, and some inflammatory changes were noted at the epithelium of the distal site. SO2 exposure affected the middle ear lining of rabbits. Fourteen days after 4 week's exposure to SO2, the middle ear lining displayed declined ciliary activity and many signs suggestive of secretory asthenia. In addition, the middle ear lining of rabbits given about 600 mg carbocysteine, both during and 2 weeks after the exposure displayed excellent ciliary activity and normal morphology of the epithelial structure.

Lacrimal gland autoimmunity in New Zealand mice.

We studied the spleen cell-mediated as well as antibody-mediated cytotoxicity to lacrimal gland acinar cells in New Zealand mice, an excellent animal model of Sjögren's syndrome. We found no difference in spleen cell-mediated cytotoxicity between young (3 months old) and old (6 to 12 months old) animals. With age, however, New Zealand mice developed an increasing level of serum autoantibodies to the lacrimal gland acinar cells. Such antibodies may play a role in the tissue damage of the lacrimal gland in these mice.

Tape attachment therapy for correction of congenital malformations of the auricle: clinical and experimental studies.

After the auricular cartilage of 5-week-old rabbits was bent and fixed by suture, the cartilage increased in thickness in a short period of time. The thickness was maximal 6 weeks later and slightly greater than that of the control cartilage 12 weeks later. At this stage, the auricle was fixed in curved form. There was no difference in the morphology of cartilage cells between the bent group and the control. Tape attachment therapy is very simple. We propose that surgical treatment should be designed after a few months of tape attachment therapy. In the present work, the effect of taping was graded good in 4 cases of Stahl ear, 1 case of atypical cryptotia, and 2 other cases. In 2 cases of cup ear the result was fair, but this deformity with marked auricular aplasia was not curable by this conservative treatment.

Dietary fat and immune function. I. Antibody responses, lymphocyte and accessory cell function in (NZB x NZW)F1 mice.

The influence of dietary fat on autoimmunity in lupus-prone (NZB x NZW)F1 mice has been demonstrated. In defining further the effects of dietary lipid on the immune system of this strain, female weanling mice were placed on four diets differing in quantity and type of fat. Their immunologic response was then studied by a variety of tests at 4 and 7 mo of age. Few differences were seen among the four groups at 4 mo of age. At 7 mo of age, however, the mice receiving diets high in saturated and unsaturated fats had a reduced mitogenic response to T cell mitogens and an enhanced response to the B cell mitogen LPS. Immunoglobulin levels and delayed hypersensitivity responses did not show any consistent differences among the diet groups. At 7 mo, however, mice receiving diets high in unsaturated fat demonstrated hyperresponsiveness to injected sheep red blood cells as measured by the hemolytic plaque technique. In addition, peritoneal leukocytes from the same diet group exhibited an increased response to bromelain-treated autologous erythrocytes which was decreased after treatment with anti-Thy-1 antiserum and complement. Phagocytosis by peritoneal macrophages was significantly decreased in the animals fed high-fat diets, particular high saturated fat. Similarly, natural killer cell activity was markedly reduced in the mice with a high intake of saturated lipid, a finding which correlated with the in vitro production of interferon. These results indicate that diets high in fat influence immune responses and thus can affect the onset and severity of autoimmune disease. A low-fat diet can reduce the development of disease by maintaining normal immune responses. The data also suggest that unsaturated fat may influence T helper cell activity and therefore antibody production, whereas saturated fats may affect cellular immune responses which are dependent on membrane contact.

Stimulation of bull seminal RNase by various basic proteins.

The activity of purified bull seminal RNase was markedly stimulated by various basic proteins. At the half concentration of substrate RNA, basic proteins such as histones, high-mobility group chromosomal proteins and cytochrome c stimulated the enzyme activity 4-6 fold. Other non-basic proteins such as bovine serum albumin and human gamma-globulin were far less effective. In addition to enzyme-stimulating activity, basic proteins showed a marked enzyme-stabilizing activity, indicating the presence of a strong interaction between the enzyme and basic proteins.

Suppression of secondary herpes simplex uveitis by cyclosporine.

The authors studied the effect of an immunosuppressive agent, cyclosporine (CyA), on experimental secondary herpes simplex (HS) uveitis. Secondary HS uveitis was induced in a rabbit eye that had recovered from primary HS uveitis by challenging it with an intravitreal injection of herpes simplex virus (HSV) antigen. Daily intramuscular injections of CyA (25 mg/kg body weight) for 7 days prior to the intravitreal challenge with HSV antigen significantly suppressed the induction of secondary HS uveitis, but daily injections of CyA after the challenge with HSV antigen was ineffective. Intravitreal injections of CyA (5 mg) 7 days and 3 days prior to the HSV challenge were less effective, but the combined treatment with seven daily intramuscular CyA and two intravitreal CyA injections prior to the HSV challenge was most effective in the prevention of the uveitis. The daily intramuscular treatment with CyA resulted in a marked reduction of cell-mediated immunity while leaving the level of circulating HSV specific antibody high. No reactivation of latent HSV was detected in trigeminal and superior cervical ganglia of CyA-treated rabbits.

Effects of azelastine on allergen- and exercise-induced asthma.

The effects of the new anti-allergic drug, azelastine, on allergen- and exercise-induced asthma were studied. In six allergen inhalation tests for five asymptomatic asthmatic patients, the maximum percentage fall in FEV1.0 immediately after inhalation of allergen extract was 37.2 +/- 6.4 per cent (mean +/- SEM). As compared with a placebo, the maximum percentage fall in FEV1.0 with azelastine after inhalation of allergen extract in the same manner as with the placebo was 17.3 +/- 6.9 per cent. The difference was statistically significant (p less than 0.05). The percentage fall in FEV1.0 with placebo and azelastine in late asthmatic response (n = 4) was 36.0 +/- 5.3 per cent and 10.0 +/- 5.2 per cent, respectively. The difference was also statistically significant (p less than 0.01). An exercise test was carried out on seven asymptomatic asthmatic patients using an inclined treadmill. The maximum percentage fall in FEV1.0 without drugs, with diphenhydramine and azelastine was 38.9 +/- 5.0 per cent, 20.1 +/- 3.8 per cent and 11.3 +/- 3.1 per cent, respectively. Significant differences were found among each group (p less than 0.05). Azelastine was regarded as having sufficient potency to inhibit exercise-induced asthma; however, placebo effects cannot be ruled out with regard to the effects of diphenhydramine. These results suggests that chemical mediator release is involved not only in allergen-induced asthma but also in exercise-induced asthma, suggesting the clinical utility of azelastine.

The presence of cytotoxic autoantibody to lacrimal gland cells in NZB/W mice.

New Zealand Black and White F1 hybrid mice (NZB/W mice) spontaneously develop an autoimmune disease which provides us with a suitable animal model for Sjögren's syndrome. With increasing age, these mice develop foci of mononuclear cell infiltration in the lacrimal and salivary glands, which closely resemble the lesions seen in patients with Sjögren's syndrome. We studied the cell-mediated and antibody-mediated immune responses of NZB/W mice to lacrimal gland cells. Lacrimal gland acinar cells were isolated from 2-month-old NZB/W or BALB/c mice for the target of 51Cr-release assay. There was no statistically significant difference in the spleen cell-mediated cytotoxicity to lacrimal gland cells among NZB/W mice of different ages (2, 5, and 8 months old). With increasing age, on the other hand, we found a statistically significant increase in the titers of autoantibodies to lacrimal gland cells in NZB/W mice, while aged BALB/c mice did not develop such antibodies. Fractionation of pooled positive sera by gel filtration revealed that this cytotoxic activity was mostly recovered in the IgM fraction. The tissue absorption study showed that these antibodies cross-reacted with salivary gland and kidney.
