Establishment of a human glioma cell line bearing a homogeneously staining chromosomal region and releasing alpha- and beta-type transforming growth factors.

A human glioma cell line (YKG1), which was positively identified for glial fibrillary acidic (GFA) and S-100 proteins, was established from a surgical specimen of a patient with glioblastoma. Chromosome analysis of the cells revealed a homogeneously staining region (HSR) on a marker chromosome. The assay for transforming growth factors (TGFs) in the conditioned medium of the cell line revealed that it contained high levels of alpha- and beta-type TGFs, which might regulate the growth of glioblastoma and influence on the peritumoral tissues.

Establishment and characterization of an immature epithelial cell line (ENU-T-1) derived from a rat nephroblastoma.

A new cell line designated ENU-T-1 has been established from a xenotransplanted experimental rat nephroblastoma. The cultured cells are spindle-shaped or polygonal and are arranged in a wavy fashion morphologically similar to cultured embryonal renal epithelial cells. The cells exhibit a number of epithelial characteristics. Enzyme histochemistry gives positive reactions for gamma-glutamyltranspeptidase and alkaline phosphatase, both of which are present in renal tubular epithelial cells. Immunofluorescence studies show positive reactions for vimentin and cytokeratin. When inoculated into athymic nude mice, the cultured cells form tumors composed of sheets of epithelial cells with focal tubular formation. This cell line may be of value in studying differentiation of nephroblastoma, and possibly normal nephrogenesis.

[A case of thoracic kidney].

An asymptomatic 37-year-old woman visited our outpatient clinic for evaluation of an abnormal shadow of the right thorax detected by a mass roentgenographic survey. P-A chest roentgenogram showed a round homogeneous mass with a convex superior margin in the posterior portion of the right thorax. The CT with contrast enhancement and intravenous pyelography demonstrated the ectopic high location of the right kidney. Renal arteriography showed the right renal artery originated from the abdominal aorta at a normal level. Renal function tests showed no abnormality.

Detection of tetrodotoxin by thin-layer chromatography/fast atom bombardment mass spectrometry.

A new method for detection of tetrodotoxin (TTX) by thin-layer chromatography/fast atom bombardment (FAB) mass spectrometry was developed. TTX and/or related substances were separated by TLC on LHP-K high-performance precoated plates, with a solvent system of pyridine:ethyl acetate:acetic acid:water (15:5:3:4). The plates were subjected to positive FAB mass spectrometry, under scanning within a mass range from m/z 100 to 500. TTX was identified by selected ion-monitored chromatograms at m/z 320 (M + H)+ and 302 (M + H - H2O)+, along with full scan positive ion FAB mass spectrometry. The limit of detection for TTX was about 0.1 micrograms. TTX was also detected by cellulose acetate membrane electrophoresis/FAB mass spectrometry.

Developmental and sexual differences of T-kininogen levels in rat plasma and liver.

T-kininogen levels in plasma and liver microsomes were measured by radioimmunoassay in female and male rats of various ages. High levels of T-kininogen were found in plasma and liver of 1-day to 1-week old male and female rats and their mothers. The levels in newborns gradually decreased along with their development. In mature male rats the levels were as low as 1/5-1/2 of those in mature female rats. Treatment with estradiol increased the plasma and the liver levels of T-kininogen significantly in both sexes, but testosterone decreased the level in female rats and had no effect in male rats. These results suggest that sex hormones may regulate the physiological level of T-kininogen in rats.

A case of renal angiomyolipomas associated with multiple and various hamartomatous microlesions.

A case of 50-year-old woman who demonstrated large intrarenal and externally protruding angiomyolipomas (AMLomas), unusual intraglomerular lesions and multiple hamartomatous microlesions of various types within the kidney is reported. This type of intraglomerular lesion has been briefly described only twice previously in patients with tuberous sclerosis (TS), and is unique in its location and histological appearance. The lesions are mainly composed of proliferating mesangial cells and are hamartomatous in nature. Although no stigmata of TS other than renal AMLomas are apparent in this case, the presence of many renal hamartomatous lesions suggests the possibility of a forme fruste of TS.

[Transthoracic approaches to the lesions of thoracic cord and thoracic vertebrae].

Experiences of transthoracic approaches to the thoracic cord lesions were reported. Since 1983, we have performed six transthoracic approaches to the thoracic lesions; one thoracic OPLL, one dumbbell-shaped neurinoma, two thoracic soft disc, one epidural metastatic tumor to thoracic vertebrae. From the viewpoint of surgical anatomy, the thoracic vertebrae show a physiological kyphosis and the subarachnoid space of the ventral site is narrower than that of the dorsal site. Due to such anatomical characteristics, the thoracic laminectomy for decompression is not so effective as in the cervical or lumbar region and a relatively small mass lesion can bring a paraplegic state. The lesion of the ventral site of the thoracic cord has been regarded as no man's land because of poor results of posterior approaches. Instead of posterior approaches, anterior or anterolateral approaches with transthoracic route have been adopted. In the present paper, we used transthoracic anterolateral approaches for four patients and anterior sternum-splitting approach for two patients. The operative procedures of the approaches were described in detail. By these approaches, we could treat four patients with favourable results but the result of thoracic OPLL was poor. The cause of this poor result seemed to depend upon the intraoperative compression of the thoracic cord. For the troublesome complication, we described the postoperative cerebrospinal fluid leakage into thoracic cavity with respiratory disturbance. Several devices to prevent such troublesome complication were discussed.

Induction of suppressor T cells by intravenous administration of monoclonal anti-I-A antibody.

Intravenously administered monoclonal anti-I-A antibodies successfully induced suppressor T (Ts) cells specific for alloantigen-specific delayed-type hypersensitivity (DTH) responses in mice. These Ts cells exerted their effects in the effector phase and had no H-2 restrictions. Phenotypic analysis revealed L3T4 antigens on their cell surface but failed to reveal Lyt-2 antigen. Ts cell activity was abrogated by a 30-min incubation with the anti-I-A antibodies used for Ts cell induction. Incubation in the anti-I-A-antibody-coated plate also abrogated the Ts cell activity. Since anti-I-A antibody is idiotypic for the I-A antigen, it is suggested that these Ts cells might express antiidiotypic receptors for I-A antigens. These findings are considered to be consistent with previous observations of hapten-specific systems, in which antiidiotypic Ts cells are inducible by idiotypic antibodies.
