Two cases of a renal epithelial tumour resembling immature nephron.

Two cases of renal epithelial tumours are reported in females aged 46 and 66 years respectively. In spite of the large size of the tumours, neither invasive growth nor metastasis was observed. Histologically, the tumours were composed of immature epithelial cells forming tubules with abortive glomeruloid structures. Electron microscopy of tumour cells revealed poorly developed polarity and intracytoplasmic organelles. They showed similar immunohistochemical reactions to those of developing nephrons, particularly to those of the S-shaped body. The nuclear DNA content of the tumour cells was almost euploid. We conclude that the lesions were epithelial tumours of the kidney histologically mimicking developing renal parenchyma.

[Visual evoked potential and relative afferent pupillary defect in patients with unilateral optic neuritis].

The relationships between relative afferent pupillary defect (RAPD) and visual evoked potential (VEP), visual acuity or visual field in 14 patients with idiopathic unilateral optic neuritis, were investigated during the period of recovery from an acute episode. Grading of RAPD into four steps mainly based upon an escape phenomenon in a swinging flashlight test, was used to determine the degree of RAPD. The degree of RAPD significantly correlated with a reduction in VEP amplitude, visual acuity and loss of central visual field, while it did not correlate with delay in VEP latency. These results suggest that RAPD is mainly related to the integrity of central vision, and that grading of RAPD, as well as a comparison of VEP amplitude between the affected and contralateral eyes, is useful for an objective evaluation of visual functions of the affected eyes in patients with unilateral optic neuritis.

Family history of cancer in a female cohort population in Japan.

This study was aimed at examining the effect of familial predisposition on cancer incidence in a cohort population. Family history (FH) of cancer and reproductive history were obtained by a simple questionnaire from about 40,000 participants in a cervical cancer screening program visiting screening clinics in 1977. Number and age structure of family members were not known. About 40% of subjects reported positive FH which was not medically verified. Comparing the observed and expected numbers of pair-wise combination of various cancers observed among the family members, we found more intense cluster of cancers of the same than different sites. This population was followed until 1987 by using the population-based cancer registry and 638 cancer cases were detected. By matching residence and birth year, the same number of nonaffected controls was chosen from the original cohorts. Positive effect of FH was detected for large bowel and breast cancers, but not for stomach and cervical cancers. Older age at marriage and lower parity were noted for breast cancer cases.

Establishment of an epithelioid malignant schwannoma cell line (YST-1).

A novel cell line, YST-1, was established from an epithelioid malignant schwannoma (EMS) that occurred in the upper arm of an 8-year-old girl. YST-1 cells were polygonal and stellate in shape, contained abundant free ribosomes, mitochondria, lysosomes and rough-surfaced endoplasmic reticulum, and grew stably with a population doubling time of 40 h. Immunohistochemically, vimentin, S100 protein and S100 protein beta subunit were positive in the cytoplasm. The xeno-transplanted tumor in nude mice was composed of cells with an epithelioid arrangement similar to the original tumor. The borders of the tumor cells were connected intimately without desmosomal junctions, and there were abundant organelles in the cytoplasm. YST-1 cells were considered to be of value for studying the nature and histogenesis of EMS.

A study of recording accuracy in linear measurements displayed by sirognathograph.

The recording accuracy of Sirognathograph (Siemens, West Germany) was conducted by means of an experimental apparatus. Diagrams of mandibular border movements in three dimensional directions were performed. The results showed the distortion was not recognized within 5 mm of primary condition in a subject, but slightly between 5 mm and 10 mm. In a distance of more than 15 mm, a considerable distortion was increased until at the maximum opening position.

[Immunobiological activities of Veillonella parvula isolated from infected root canals].

The primary etiological agent of all periapical lesions has long been considered to be bacterial. Furthermore, bacteria from infected root canals are potential antigens capable of initiating immunological reactions in periapical tissues. The purpose of this study, therefore, was clarify the immunological potentials of Veillonella parvula (V. parvula), which was frequently isolated from root canals with periapical lesion. Immunobiological activities of V. parvula sonic extracts were investigated on the enhancement of monocytes migration, induction of interleukin-1 (IL-1) production, mitogenicity and polyclonal activation of B cell. Following results were obtained: 1. Both LPS and protein of V. parvula sonic extracts strongly enhanced the activity of human peripheral monocytes migration. 2. Induction of IL-1 on C3H/HeN mice macrophage by V. parvula sonic extracts were stronger than that of S. typhimurium LPS as positive control. 3. It was found that mitogenicity of LPS from V. parvula on splenocytes was stronger than that of the protein, however mitogenicity on thymocytes was not shown in both preparation. 4. The polyclonal activation of B cell on splenocytes of BALB/c nu/nu mice by V. parvula was induced by the protein and LPS. 5. These findings indicate that both the protein and LPS from V. parvula have a regulation of immunobiological responses against macrophages and lymphocytes.

[Studies on the pressor mechanism of 19-hydroxyandrostenedione].

In the present study, the pressor activity of 19-hydroxyandrostenedione (19-OH-AD) (which is a kind of androgen) and the presence or absence of the possibility of its role as an amplifier of mineralocorticoid were investigated. 34 Wistar rats of 8 weeks of age were used as the experimental subjects. The rats were divided into 3 groups, the control group (n = 12), 19-OH-AD group (n = 12) and DOCA group (n = 10). The control group was given 0.2 ml of sesame oil, the 19-OH-AD group was given 10 mg of 19-OH-AD dissolved in 0.2 ml of sesame oil, and the DOCA group was given 10 mg of DOCA dissolved in 0.2 ml of sesame oil. This was subcutaneously administered three times a week for 4 weeks, and body weight and blood pressure were observed. At the termination of the experiment, blood was sampled by decapitation, and the serum sodium concentration, erythrocyte sodium content, plasma renin activity (PRA), plasma atrial natriuretic hormone (alpha-rANP), and contents of DOC, corticosterone and aldosterone in adrenal gland were determined. The following results were obtained. The blood pressure at the termination of the experiment was higher than that at the initiation of the experiment by 9.9 +/- 1.9 (S.E.), 31.3 +/- 1.7, and 46.2 +/- 2.0 mmHg in the control, 19-OH-AD and DOCA groups, respectively, where significant elevations were observed in both the 19-OH-AD and DOCA groups as compared with the control group. As to the serum sodium concentration, only the DOCA group showed a significant elevation (p less than 0.001) as compared with the other two groups. However, the erythrocyte sodium contents of both the 19-OH-AD and DOCA groups increased significantly as compared with the control group. Moreover, the increase of the DOCA group was significantly higher (p less than 0.001) than that of the 19-OH-AD, and a significant positive correlation between the elevation of blood pressure and the erythrocyte sodium content was observed (p less than 0.001). As to the PRA value, only the DOCA group showed a significant lowering as compared with the other two groups, whereas alpha-rANP value of the DOCA group showed a significant elevation as compared with the other two groups. As to the contents of steroid hormones in the adrenal gland, all of the steroid hormones were significantly lower in the DOCA group, while those of the 19-OH-AD group were unchanged as were those of the control group.(ABSTRACT TRUNCATED AT 400 WORDS)

Changes of T-kininogen levels in plasma and liver during development of rats.

T-kininogen levels of plasma and liver were measured in rats during development by RIA. We found that higher levels of T-kininogen in plasma and liver of neonates and of the mother rats at around term. The plasma level of mature male rats was as low as 1/5 to 1/2 of that of mature female rats. T-kininogen levels in male and female rats increased after treatment with estradiol, indicating that sex hormones may regulate the physiological level of T-kininogen in rats.
