Human DNA polymerase alpha. Compensation for heat-labile mouse DNA polymerase alpha and its gene localization on the X chromosome.

The chromosomal location of human DNA polymerase alpha gene was determined by studies on somatic cell hybrids between a temperature-sensitive mutant cell line of mouse FM3A cells and normal human lymphocytes or a line of human diploid fibroblasts derived from a patient with the fragile X syndrome. A temperature-sensitive mutant, FT20-M6, a 6-thioguanine-resistant derivative of tsFT20, has heat-labile DNA polymerase alpha. Interspecific cell hybrids between FT20-M6 and human cells grew at the non-permissive temperature, indicating that some human chromosomes can compensate for the temperature-sensitive defect of tsFT20 in mouse-human cell hybrids. Three of these hybrid clones were examined further, and were shown to contain heat-stable DNA polymerase alpha that was neutralized with human DNA polymerase alpha-specific monoclonal antibody. Subcloning and segregation tests of these hybrid clones showed a positive correlation between the expression of human DNA polymerase alpha and the presence of the human X chromosome. Two subclones, however, did not conform to this relationship: they grew at the nonpermissive temperature but not in hypoxanthine/amethopterin/thymidine medium. Detailed examination of the human chromosomes in these subclones revealed that these clones had only one human chromosome, an X chromosome with a terminal deletion of the long arm including the locus of the gene for hypoxanthine phosphoribosyltransferase (EC 2.4.2.8). From these data, the functional DNA polymerase alpha gene was located on the human X chromosome.

Primary isolation of cytopathic bovine rotaviruses on fetal rhesus monkey kidney cells.

Primary isolation of bovine rotaviruses was successfully performed on rolling cultures of MA104 cells following trypsin treatment of fecal samples and cells. Fifty-one fecal samples were obtained from 22 herds affected with naturally-occurring acute diarrhea in calves during a period of over two years. Rotavirus particles were demonstrated in only 10 fecal samples by electron microscopy. Fourteen cytopathic bovine rotaviruses were isolated from positive samples and could be serially cultivated on MA104 cells. The presence of virus was identified by specific immunofluorescence in infected cells. These data indicated that approximately 30% of the herds affected with acute diarrhea in their calves were associated with rotavirus infection.

Effect of protein quality on dietary induction of hepatic ornithine decarboxylase.

Dietary induction of ornithine decarboxylase (ODC) in rat liver depended on the quality of protein in the diet. Zein did not induce ODC unless it was supplemented with the deficient amino acids, tryptophan and lysine. Similar phenomena were observed with gelatin (tryptophan and methionine) and hemoglobin (isoleucine). However, ODC was found to be significantly induced by an amino acid mixture simulating zein. The difference between amino acid diet and protein diet effects could not be explained by digestibility of zein. Unphysiologically rapid influx of amino acids appeared to induce ODC by a mechanism different from that by which dietary protein induced ODC. After ingestion of zein, the concentration of tryptophan and lysine decreased markedly in plasma and liver. However, it was confirmed that their supply from intracellular protein degradation continued even after feeding. In contrast to ODC, tyrosine aminotransferase was induced by zein as well as by casein, indicating that the requirement of tryptophan and lysine as precursor amino acids for enzyme synthesis was satisfied by reutilization of the amino acids liberated by intracellular protein degradation. Therefore, it was concluded that good quality protein was required for ODC induction mainly as a signal but not as a source of precursor amino acids.

Serotypes of bovine rotaviruses distinguished by serum neutralization.

Fourteen cytopathic bovine rotaviruses isolated from naturally affected calves were characterized serologically by serum neutralization. Antisera were prepared in guinea pigs against the three isolates and the Lincoln strain of bovine rotavirus. These four rotaviruses were segregated into at least two distinct serotypes (designated types 1 and 2) by serum neutralization. The remaining 11 isolates were also placed into two groups according to neutralization specificity by using the antisera against types 1 and 2 bovine rotavirus. When colostrum-deprived calves were experimentally inoculated with type 1 or 2 bovine rotavirus, respectively, the calves produced neutralizing antibodies against the inoculated serotype but not against the other serotype. Calf sera obtained from six herds with naturally occurring rotavirus infections possessed neutralizing antibodies against type 1 or 2 or both. Thus, distinct serotypes of bovine rotavirus were demonstrated by serum neutralization. The possible role of serotypes in the epidemiology of rotavirus infection among bovine species is discussed.

Family trees representing the finitely proliferative nature of cultured rat liver cells.

Family trees of rat liver epithelial-like cells in primary culture were obtained by cinemicrographic analysis. With few exceptions the proliferative profiles reconstituted from these family trees showed limited proliferation. This limitation was not due to post-confluence inhibition of proliferation or to nutritional deprivation during time lapse cinemicrography, but to the finite potential of clonal proliferation. In these family trees a cell entered mitosis, incomplete division, death, or a long interphase with no detectable termination. These family trees successfully related the life phases of the individual cells that compose the population to the limited nature of proliferative potential as observed in clonal cell populations.

Experimental studies on vertical infection of mice with Japanese encephalitis virus. IV. Effect of virus strain on placental and fetal infection.

An experiment was carried out to examine the effect of an inoculated strain of Japanese encephalitis virus on the establishment of experimental vertical infection of mice with this virus. In it, closed-colony mice of the CFW strain were inoculated intravenously with seven strains of the virus at 7 days of pregnancy. After that, an attempt was made to recover the virus from placenta and fetus, so that the infection rate of each strain might be determined. As a result, the infection rate was high for both placenta and fetus in the case of the AS-6 and Sagara strains both of which had undergone three passages in the mouse brain. The placental infection rate was high and the fetal infection rate relatively low in the case of the JaGAr01 and Fuji strains which had undergone 7 and 150 passages, respectively, in the mouse brain. The infection rate was very low for both placenta and fetus in the case of the Nakayama-Yakken strain which had undergone more than 100 passages in the mouse brain. There was no difference in the severity of viremia after inoculation between the AS-6 and Fuji strains. Both placental and fetal infection rates were low in the case of the JaTH160 strain which had undergone passages in mice by intraperitoneal inoculation and which presented a strong peripheral infectivity and induced a severe viremia after inoculation. Neither placental nor fetal infection occurred in the case of the S- strain used as live virus vaccine. These results indicated that placental and fetal infection rates varied from one virus strain to another.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of serous cyst of the retroperitoneal space].

There have been very few reports on retroperitoneal serous cysts and only 23 cases have been reported in Japan. We report a case of a 47-year-old woman who presented with the complaint of dull pain in her lower abdomen. After careful examination, she was diagnosed to have retroperitoneal serous cyst.
