Effect of cobra venom factor on the response of Wistar-Furth rats to the Gross-virus-induced (C58NT)D lymphoma.

Wistar-Furth rats were injected with the anticomplementary cobra venom factor at the time of challenge with the Gross-virus-induced (C58NT)D lymphoma. Complement depletion and/or activation of the complement system in these animals led to the following findings: Multiplication of the tumor cells in vivo was accelerated and mortality rates were enhanced. In addition, serum levels of the immunoglobulins which mediate antibody-dependent cellular cytotoxicity were depressed.

Studies on drug-induced allergic intrahepatic cholestasis--hepatic cholestasis induced in dogs by lymphocyte culture supernatant.

When peripheral lymphocytes from patients with drug-induced allergic intrahepatic cholestasis were stimulated with a specific drug in vitro in the presence of a liver microsome fraction or soluble liver specific antigen fraction, lympholine production was seen in many cases. By the injection of culture supernatant of stimulated lymphocytes into the mesentery vein of dogs, cholestasis was induced in the liver, chiefly in the central zones of lobules. However, no cholestasis could be observed in dogs administered the supernatants of lymphocyte cultures prepared from normal individuals in the presence of drugs. Moreover, only slight swelling of the hepatocytes was observed in the liver when normal lymphocytes were stimulated with PHA-P and culture supernatant was injected into the mesentery vein of dogs. These results suggest that sensitized lymphocytes may produce a factor (or factors) by stimulation with a specific drug-carrier and this factor (or factors) causes cholestasis in the liver.

Immune receptors (IgG-Fc and complement receptors) in normal human organs.

The presence of immune receptors (IgG-Fc and complement receptors) was examined in normal human tissues from various organs. Sheep erythrocytes sensitized with rabbit IgG antibody (IgG-EA) or with rabbit IgM antibody and human complement (IgM-EAC) were used for the detection of IgG-Fc receptors and complement receptors, respectively. IgG-Fc receptors were detected on sinuses of the lymph node, splenic red pulps, hepatic lobules, renal glomeruli, alveolar wall of the lung, intestinal villi, superifical layer of the synovium, and subcutaneous tissue. The presence of complement receptors was demonstrated in the follicles and the sinuses of lymph nodes, white pulp of the spleen, renal glomeruli, alveolar wall of the lung, and lamina propria of the intestine. The specific binding of IgG-EA was consistently inhibited by heat-aggregated human IgG or by a high concentration of native human IgG. The detection of immune receptors in these various tissues might be helpful for understanding why the immune complexes are often detected when immunologically mediated disease processes involve these tissues.

Studies on the subpopulation and function of peripheral lymphocytes, and lymphocyte reactivity to colonic mucosal antigen and bacterial antigen in patients with ulcerative colitis and Crohn's disease.

Immunological studies were carried out in fifteen cases of ulcerative colitis, three cases of tuberculous colitis, four cases of Crohn's disease and one case of Behcet's disease diagnosed by X-ray, endoscopy or biopsy mainly from the standpoint of cellular immunity. In patients with ulcerative colitis the T-cell population and PHA responsiveness of peripheral lymphocytes were both depressed more than in the control group. At different stages of the disease, the deviation of the values of PHA responsiveness of lymphocytes showed a rather wide range in the active stage and there was also immunological instability. However, when the disease entered remission, the immunological conditions settled down and the pathological condition appeared stable. In contrast to the nonspecific immunological conditions mentioned above, characteristically, there were many examples of a positive lymphocyte response to colonic mucosal and bacterial antigens. In all cases of Crohn's disease the lesions are in the small intestine and there was a reduced response to PHA lymphocytic stimulation which was more pronounced than that in patients with ulcerative colitis. PHA response was also high in tuberculous colitis cases.

Eruptions. Possible involvement of carrier contained in cutaneous tissues.

In some types of drug eruptions, a positive blastoid transformation is frequently seen in peripheral lymphocytes when they are stimulated in vitro with the causative drug in the presence of autologous serum. In these cases, a soluble fraction prepared from guinea pig skin can replace the autologous serum in inducing the lymphocyte fransformation, whereas either the mitochondrial or microsomal fraction of guinea pig skin instead of the autologous serum is less effective. Moreover, in fixed drug eruption, the addition of soluble skin material fractionated by gel filtration with Sephadex G-100 (FrD1) is as effective as the autologous sera in inducing lymphocyte transformation. These results suggest that the soluble fraction of skin is related to the pathogenesis of fixed and exanthematic drug eruptions as a carrier substance of drugs.

Studies on an inhibitory factor to phytohemagglutinin-induced lymphocyte transformation found in the serum of patients with various liver diseases.

When normal lymphocytes were stimulated with PHA in the presence of serum taken from patients with various liver diseases, lower lymphocyte transformation was seen in many cases. The occurrence of a serum inhibitory factor in liver diseases tends to increase with the progress of the disease from acute hepatitis to cirrhosis of the liver. No such inhibitory factor was detected in two asymptomatic HBs-antigen carriers. When the inhibitory factor was fractionated by DE-52 column chromatography, an active component was obtained and shown to have mobilities from alpha2-globulin to beta-globulin in cellulose acetate membrane electrophoresis. However, two active fractions were separated by DEAE-cellulose column chromatography with stepwise increases of the concentration of the acetate buffer and their mobilities were consistent with those of alpha1-globulin and alpha2-globulin, respectively. In 11 cases of acute hepatitis which were followed for at least 6 months, three out of four inhibitory factor positive cases developed into the chronic form and the other became a protracted case. On the other hand, six out of seven inhibitory factor negative cases completely recovered and the remaining case followed a protracted course.
