[Management of abnormal blood vessels of donor kidney in renal transplantation (report of 128 cases)].

The experience on management of abnormal blood vessels in 128 cases of donor kidney during the tailoring operation was reported. The various techniques used for different types of abnormal arteries and veins, and the critical points which should be paid attention to have been discussed. It was concluded that the multiple renal arteries should be treated in a single renal artery and anastomosed with internal iliac artery or/and external iliac artery. The appropriate management given to abnormal renal blood vessels during the tailoring operation may shorten the warm ishemia time, ensure the renal blood supply, reduce the renal vasular complication, and promote the recovery of renal function.

[Study on the effects of hyaluronic acid-streptomycin perfusion through the round window on the function and morphology in guinea pig inner ears].

To investigate the effects of the hyaluronic acid-streptomycin (HA-SM) perfusion through round window on the function and morphology of the inner ear in guinea pig, membrous labyrinth mapping, temporal bone section after celloidin embedding, transmission electron microscopy, electrocochlegraphy (ECochG) and electronystagmography (ENG) were examined. The nystagmus duration induced by caloric test was obviously reduced in comparison with that of the preoperation (P< 0.01), while the action potential(AP) by ECochG was not obviously changed. The sensory cells of estibular organs were severely damaged, while the morphology of corti's organs were significantly damaged after HA-SM perfusion. The results suggest that the HA-SM perfusion through the round window may selectively destroy the vestibular function, whereas the auditory function is not obviously damaged.

[Human cytomegalovirus infection and congenital malformation].

OBJECTIVE: To study the relationship between intrauterine cytomegalovirus (HCMV) infection and congenital malformation, and to determine the distribution of tissues infected. METHODS: Autopsy samples of 41 infants with congenital malformation and 19 infants with normal appearances were studied. Using polymerase chain reaction (PCR) technique the paraffin embedded specimens of main organs were examined for HCMV infection. In-situ hybridization (ISH) was performed in some of the PCR positive tissues in order to define the distribution of HCMV DNA. RESULTS: 19 of the 41 infants (46.34%) with congenital defects were HCMV positive, while 1 in 19 (5.26%) were positive in the control group, and there was significant difference between the 2 groups (P < 0.05), 20.46% (35/171) of the fetal organ samples were HCMV DNA positive in the malformation group, but only 1 out of 78 samples (1.28%%) was positive in the pulmonary tissue of the control group. More malformations of the digestive system were presented in HCMV infected babies but no statistical significant difference when compared with other systems. Brain tissue had the highest HCMV infection rates (41.37%, 12/29), which was significantly higher than other organs. By ISH technique HCMV DNA was found only in 6 out of 17 PCR positive samples, and they were located at neurons, neurogliocytes, epithelium and interstitial cells of the kidney, and epithelial cells of pulmonary alveolar. CONCLUSION: There are strong correlation between HCMV infection and congenital malformation, and brain is more susceptible to HCMV. By combining PCR and ISH, both sensitivity and distribution of HCMV could be obtained.

[Patterns of cisplatin induced apoptosis in ovarian cancer cell line COC1].

OBJECTIVE: To elucidate the patterns of chemotherapeutic drugs induced apoptosis and its role in cancer chemotherapy. METHODS: Apoptosis in cultured ovarian cancer cell line COC1 induced by ciplatin was investigated in vitro by applying cytohistochemistry stain, florescence stain and electron microscopy scanning combining with DNA gel electrophoresis and flow cytometry analysis. RESULTS: After exposure to cisplatin, COC1 cells manifested typical apoptotic morphological features. Apoptosis persisted throughout 30 hours following addition of cisplatin and augmentated gradually. The rate of apoptosis was enhanced within tested dose range in a dose dependent pattern. At the point of 12 hour cisplatin exposure, the necrosis rate was only 12.4% while the apoptotic rate accounted for 30.8% already. CONCLUSIONS: Chemotherapeutic drugs induced apoptosis is one of the primary mechanism of anti-cancer chemotherapy.

[An immunohistochemical study of the proliferating activity of ameloblastic fibroma and ameloblastic fibrosarcoma].

Six cases of ameloblastic fibroma (AF) and three cases of ameloblastic fibrosarcoma (AFS) were immunohistochemically investigated with a proliferating cell nuclear antigen (PCNA) monoclone antibody. Data on the PCNA labeling index (LI) of both epithelial and mesenchymal components were analyzed. The results showed that the PCNA LI of mesenchymal component of AFS (40.8%) was significantly higher than that of AF (3.2%) (P < 0.01) and that of the epithelial component of AFS (5.3%) was significantly lower than that of the mesenchymal one of AFS (P < 0.01). The results suggested that PCNA LI be a useful marker for the differentiation diagnosis of AF and AFS.

[Antisense to cyclin D1 inhibits the proliferation of glomerular mesangial cells].

OBJECTIVES: To investigate the role of Cyclin D1 in the proliferation of glomerular mesangial cells. METHODS: We first detected the expression of Cyclin D1 protein in mesangial cells by using immunohistochemical method, set up a retroviral vector mediated genetransfer system expressing Cyclin D1 antisense RNA with recombinating techniques, and assay the proliferating situation of genetransferred mesangial cells by MTT method and flow cytometry respectively. RESULTS: Cyclin D1 protein was expressed in the nuclei of cultured mesangial cells in vitro. The exgoenous gene was transducted into mesangial cells mediated by retrovirus and transcripted and expressed antisense RNA in high effiency. The response of antisense Cyclin D1 genetransferred mesangial cells to Endothelin-1 (OD570)(18 hr 0.105 +/- 0.009 vs 6 hr 0.096 +/- 0.013) was not significant (P > 0.05), but normal cells (18 hr 0.259 +/- 0.080 vs 6 hr 0.100 +/- 0.007) and only vector transferred cells (18 hr 0.278 +/- 0.020 vs 6 hr 0.097 +/- 0.008) were significant (P < 0.01). The progression of cell cycle was prevented from G1 to S phase by antisense Cyclin D1, and the percentage of cells of S phase in antisense Cyclin D1 genetransferred cells (12.70 +/- 3.39) was significantly reduced (P < 0.01) as compared with normal cells (29.98 +/- 3.26). CONCLUSIONS: Cyclin D1 is a key regulatory protein of G1 progressing of cell cycle of mesangial cells, and the cellular proliferation stimulated by Endothelin-1 can be suppressed prominently in antisense Cyclin D1 gene transducted mesangial cells.

[Diagnostic evaluation of magnetic resonance urography in urinary tract dilatation and obstruction].

OBJECTIVE: To evaluate the diagnostic value of magnetic resonance urography (MRU). METHODS: Two hundred and one patients were examined, 92 patients with urinary tract dilatation using a modified, heavily T2-weighted fast spin-echo pulse sequence and fat-suppression pulse aid. Postprocedure processing was performed with a maximum-intensity-projection (MIP) algorithm. RESULTS: In the 92 patients with urinary tract dilatation, 86 had obstructive urinary tract dilatation 6 non-obstructive urinary tract dilatation. In the 86 patients, 14 had ureteral carcinoma, 19 calculus, and 13 benign stricture, 23 congenital ureteral stricture/or with anomalies, 7 ureteral obstructive caused by extrinsic pelvis disease, and 10 bladder carcinoma involved ureter. In the 6 patients, 5 had nervous bladder. MR urography provided the high-resolution image of the urinary tract and determined the presence of obstruction. CONCLUSION: MR urography is a reliable noninvasive method for depicting the urinary tract.

[Sudden deafness in the elderly].

To study the clinical features of the sudden sensorineural hearing loss (SSHL) in the elderly, forty-five cases with SSHL, in the age more than 60 years, and fifty-seven young cases with SSHL as controls were investigated. The results showed that: 1. there was a higher incidence of the cardiovascular disease in the elderly group than in the young group (P < 0.05), 2. prognosis of hearing in elderly patients was worse than that in the controls (P < 0.05).

[The determinations of tumor necrosis factor and interleukin 6 in serum of patients with sudden sensorineural hearing loss].

Serum level of tumor necrosis factor(TNF alpha) and interleukin 6(IL-6) were determined in 32 patients with sudden sensorineural hearing loss (SSHL), 15 patients with progressive sensorineural hearing loss(PSHL) and 20 normal controls by using enzyme-linked immunosorbent assay. The results showed that plasma TNF alpha and IL-6 levels in patients with SSHL were higher than those in PSHL group and in normal control group (P < 0.05). The study suggested that the TNF alpha and IL-6 played an important role in the pathogenesis of SSHL.

Glycophorin variants and contents of sialic acid and total sulfhydryl groups on erythrocyte membranes of residents in a malaria hyperendemic area.

OBJECTIVE: To conduct a screening survey of glycophorin (GP) variants and observe the content changes of sialic acid (SA) and total sulfhydryl (SH) groups on the erythrocyte membranes among residents in a tertian malaria hyperendemic area of Guizhou Province. METHODS: GP variants were detected in the erythrocyte hemolysates of 173 local residents at two villages of Libo County by SDS-PAGE on 10% to 15% gradients gel and Western immunoblotting. Their SA and total SH group contents were estimated in erythrocyte membranes by spectrophotometric methods. 114 healthy subjects in Changsha and 49 individuals at a neighbouring village of the above area showing low morbidity of malaria served as normal and endemic controls respectively. RESULTS: Three distinct types of GP variants were found among 19 propositi in this hyperendemic area. The incidence of GP variants was 7.9% (8/101) at Yaolu Village whose population was mainly composed of Yao ethnic group; while that of Buyi ethnic group at Maolan Village was higher (15.3%; 11/72). The erythrocyte membrane contents of SA in residents at both villages exhibited a very significant tendency of decline (P < 0.01), whereas those of total SH groups increased prominently in residents of Yaolu Village only (P < 0.05). CONCLUSIONS: The frequency of GP variants in this hyperendemic area does not depend upon the severity of malarial prevalence. The evident reduction of SA contents in the residents may be related to the breaking down of the SA residues on membrane GPs by the invasion of Plasmodium vivax.

[Sudden sensorineural hearing loss and jugular bulb diverticulum].

The relationship between high placed jugular bulb (diverticulum) and inner ear disorder is not well known. Three of 19 patients with sudden sensorineural hearing loss (SSHL) treated in 1995 had right side jugular bulb diverticulum revealed by CT scan and MRA. One of the 3 SSHL cases complicated with delayed endolymphatic hydrops. The exact mechanism of causation of inner ear symptom is not clear, but may partly be due to pressure effects with the jugular fossa encroaching on inner ear structure such as the cochlear aqueduct and vestibular aqueduct, and due to turbular flow in the diverticulum striking the inner ear. It is needed to further study the influence of the diverticulum on the inner ear.

Androgen receptor in primary hepatocellular carcinoma and its clinical significance.

OBJECTIVE: To assess retrospectively the clinical value of androgen receptor (AR) levels in primary hepatocellular carcinoma (HCC) as a prognostic factor of the disease. METHODS: Fresh HCC tissue and the surrounding liver tissue were obtained surgically from 32 patients with HCC, and preserved in liquid nitrogen. The levels of AR in all specimens were determined by radio-ligand binding assay. RESULTS: The median level of AR was 42.8 fmol/mg protein in the cancerous tissue and 48.3 fmol/mg protein in the surrounding non-cancerous liver tissue. The overall survival rate of the patients with AR < 30 fmol/mg protein in either HCC or the non-cancerous liver was significantly higher than that of the patients with AR > or = 30 fmol/mg protein (P < 0.05 and P < 0.01, respectively). The relative risk on prognosis was 3.27 (P < 0.01) for AR level in HCC and 6.06 (P < 0.001) for AR level in the non-cancerous tissue. The main prognostic factors except the tumor size were not different between the group with higher AR level and that with lower AR level in HCC. The AR level in HCC had a positive correlation with the tumor size (r = 0.44, P < 0.05). CONCLUSIONS: AR can be detected in HCC and the AR status might be a prognostic parameter that provides additional predictive information on the survival. Different AR status might define a real difference of biological characteristics between HCCs.

[The effect of antioxidant sodium ferulate on human lymphocytes apoptosis induced by H2O2].

OBJECTIVE: Sodium ferulate is a potent antioxidant purified from ferula assafoetida L. The effect of sodium ferulate on apoptosis of human lymphocytes induced by H2O2 in vitro was studied. METHODS: Apoptosis was detected by light and electron microscopic, DNA electrophoresis and Flow cytometric cell cycle analysis. mitochondrial function was detected by MTT method. BCL-2 and Bax expression was detected by confocal laser scanning microscope. RESULTS: The morphological and biochemical changes characteristic of apoptosis were partially restored after treatment with sodium ferulate, Immunofluorescence assay showed a decrease in BCL-2 and an increase in Bax oncogene proteins in lymphocytes treated with H2O2 and an improvement of cell apoptosis was found after sodium ferulate treatment. CONCLUSION: Oxidative stress can induce apoptosis in lymphocytes, and this induction can be partly prevented by sodium ferulate.

[The powder X-ray diffraction analysis with Fourier spectrum of Chinese material medica Cortex Cinnamomi].

OBJECTIVE: To develop a method for the identification of Chinese medicinal materials in diffraction space. METHOD: Using powder X-ray diffraction analysis to obtain the Fourier spectrum of 10 samples of Cortex Cinnamomi. RESULTS: The analysis shows that each of the 10 samples has its own Fourier pattern topology and characteristic peaks which are connected with its chemical composition. CONCLUSION: Cortex Cinnamomi can be identified successfully according to its topology and characteristic peaks.

[HPLC determination of two flavonoids in pollen Typhae (puhuang) and its different processed products].

OBJECTIVE: To study the changes of chemical constituents in Pollen Typhae(Puhuang) after processing. METHOD: The contents of two flavonoids, typhaneoside(I) and isorhamnetin-3-O-neohesperidoside(II), in Pollen Typhae (Puhuang) and its different processed products were determined by HPLC. RESULT: Compared with the crude drug, the contents of I and II decreased by 40.22% and 49.87% respectively in the stir-fried product. I and II were not detected in the carbonized product. CONCLUSION: The decrease of contents of I and II after processing shows that the chemical constituents in Pollen Typhae (Puhuang) have changed after processing.
