Partial amino acid sequence of human plasma retinol-binding protein. Isolation and alignment of the five cyanogen bromide fragments and the amino acid sequences of four of the fragments.

Studies are reported on the primary structure of human retinol-binding protein (RBP), the specific plasma transport protein for vitamin A. The protein consists of a single polypeptide chain of 186-187 amino acids. RBP was cleaved by cyanogen bromide into five fragments, CB-I (27 residues), CB-11 (25 residues), CB-III (20 residues), CB-IV (15 residues), and CB-V (99-100 residues). The cyanogen bromide fragments were isolated, their compositions were determined, and they were aligned after studies that included the tryptic digestion of maleylated, reduced, and carboxymethylated RBP and subsequent enzymatic digestion of some of the resulting tryptic peptides. The amino acid sequences of four of the five cyanogen bromide fragments were determined, and the sequence of almost two-thirds of the NH2-terminal portion of the RBP molecule was determined as: H2N-GLU-Arg-Asp-Cys-Arg-Val-Ser-ser-Phe-Arg-Val-Lys-Glu-Asn-Phe-Asp-Lys-Ala-Arg-Phe-Ser-Gly-Thr-Trp-Tyr-Ala-Met-Ala-Lys-Lys-Asp-Pro-Glu-Gly-Leu-Phe-Leu-Gln-Asp-Asx-Ile-Val-Ala-Glu-Phe-Ser-Val-Asx-Glx-Gly-Thr-Met-Ser-Ala-Thr-Ala-Gly-Lys-Arg-Val-Arg-Leu-Leu-Asn-Asn-Trp-Asp-Val-Cys-Ala-Asp-Met-Val-Gly-thr-Phe-Thr-Asp-Thr-Glu-Asp-Pro-Ala-Lys-Phe-Lys-Met-Lys-Tyr-Trp-Gly-Val-Ala-Ser-Phe-Leu-Gln-Lys-Gyl-Asn-Asp-Asx-His-Trp-Ile-Val-Asp-Thr-Asx-Thr-Tyr-Tyr-Ala-Val-Glu-Tyr-Cys-Ser-Arg---.

Precocious induction of pepsinogen in the stomach of suckling mice by hormones.

Effects of hormones on pepsinogen activity in mouse stomach were investigated by enzyme assay and electron microscopy. Administration of hydrocortisone alone to mice on days 5--10 increased the enzyme activity in the stomach to as much as 4.5-fold that of untreated mice and the increase was dose dependent. Thyroxine also evoked precocious differentiation of the stomach. The effects of thyroxine and hydrocortisone were additive. Injections of insulin had little effect when given alone, or in combination with other hormones. Injection of hydrocortisone alone or plus thyroxine also caused morphological differentiation of the chief cells in the stomach mucosa. Administration of thyroxine to mice on days 15--20 induced as much enzyme activity as that induced by hydrocortisone, but neither of these hormones had any effect when injected after day 23. These results suggest that besides hydrocortisone, thyroxine is also involved in differentiation of the stomach in mice for the first 20 days after birth and that the normal increase of pepsinogen activity in the stomach of mice during the late suckling period is brought about by serum glucocorticoids, possibly with thyroxine.

Electron microscopic study on phagocytosis of staphylococci by mouse peritoneal macrophages.

Macrophages from the mouse peritoneal cavity were examined by electron microscopy at various time intervals up to 24 h after intraperitoneal administration of each of three strains of staphylococci different in virulence for mice: high-virulent, low-virulent, and avirulent strains. After engulfment, avirulent bacteria were highly liable to intracellular digestion, resulting in almost complete degradation within 24 h after injection, whereas high-virulent bacteria were more resistant to digestion, some showing figures suggestive of a dividing process; the gross configuration of most of the ingested bacteria was relatively well preserved over the 24-h period. Time-dependent morphological changes of low-virulent bacteria were intermediate. Among the most distinct cytoplasmic responses to the ingested bacteria was the formation of phagocytic vacuoles around them, the type of which was dependent on the staphylococcal strain infected; ingestion of avirulent bacteria led to formation of vacuoles in which the bacteria were surrounded by a halo of amorphous material of moderate density, which may be the lysosomal content. In contrast, larger vacuoles developed after ingestion of high-virulent bacteria and contained only a small quantity of such amorphous materials. Both types of phagocytic vacuoles were seen around the low-virulent bacteria ingested. Some degenerative changes were found in the macrophages ingesting high- or low-virulent bacteria, but were not in those ingesting avirulent bacteria. Thus, resistance to intracellular degradation, as well as cytotoxicity toward phagocytes of staphylococcal strains, can be correlated with their virulence.

Hepatic lymphatics as opacified by percutaneous intrahepatic injection of contrast medium. Analysis of hepatic lymphograms in 125 cases.

Hepatic lymph vessels were opacified in 125 patients with various hepatobiliary diseases during percutaneous transhepatic cholangiography or portography. Contrast medium deposited in the parenchyma flowed relatively quickly in winding channels, usually toward the hepatic hilus. No correlation was demonstrated between the locus of the intraparenchymal deposit and the direction of the flow. Contrast medium also opacified the lymphatics near the hepatic hilus and abdominal aorta. Neither jaundice nor liver disorder were prerequisite for the visualization of the hepatic lymphatics. The intrahepatic injection of contrast medium provides a safe and useful method for the study of hepatic lymph and its pathways.

False-positive liver scan in a patient with hepatic amyloidosis: case report.

A case of secondary hepatic amyloidosis exhibiting a large liver and multiple defects on the radiocolloid scintigraph is presented. Biopsy and angiographic studies indicated that the areas of reduced colloid uptake represented heavy amyloid deposition, and the area of the left lobe with contrasting high activity most probably represented compensatory hypertrophy.

Hepatic vascular anomalies in nonparasitic cysts of the liver.

Vascular abnormalities demonstrated by angiography in 3 patients with nonparasitic liver cyst are described. In two cases of polycystic liver, dilated and mutually anastomosing hepatic vein branches were seen around a cyst as opacified by hepatic venography, and in one case of solitary liver cyst, anastomoses between large portal branches and a right hepatic vein was demonstrated by celiac angiography. These vascular abnormalities most likely represent developmental malformation along with aberrant bile duct formation expressed in the form of cystic liver. To exclude the possibility of these changes being secondary to expansion of the cyst, an angiographic study of excised polycystic liver was carried out, and it was found that cyst enlargement produced displacement and thinning but no anastomosis of the intrahepatic blood vessels.

Demonstration of growing casts of hepatocellular carcinoma in the portal vein by celiac angiography: The thread and streaks sign.

In 9 of 112 patients with hepatocellular carcinoma (HCC) examined by selective celiac and hepatic arteriography, an unusual finding, the "thread and streaks" sign, was observed. It began to appear shortly after the start of injection and was most clearly seen at 3.0-3.5 seconds. Histopathologic and radiographic studies of excised livers have demonstrated that these thin streaks correspond to the blood spaces and vessels which run longitudinally in and around a tumor cast growing in a large branch and trunk of the portal vein.
