Glycophorin variants and contents of sialic acid and total sulfhydryl groups on erythrocyte membranes of residents in a malaria hyperendemic area.

OBJECTIVE: To conduct a screening survey of glycophorin (GP) variants and observe the content changes of sialic acid (SA) and total sulfhydryl (SH) groups on the erythrocyte membranes among residents in a tertian malaria hyperendemic area of Guizhou Province. METHODS: GP variants were detected in the erythrocyte hemolysates of 173 local residents at two villages of Libo County by SDS-PAGE on 10% to 15% gradients gel and Western immunoblotting. Their SA and total SH group contents were estimated in erythrocyte membranes by spectrophotometric methods. 114 healthy subjects in Changsha and 49 individuals at a neighbouring village of the above area showing low morbidity of malaria served as normal and endemic controls respectively. RESULTS: Three distinct types of GP variants were found among 19 propositi in this hyperendemic area. The incidence of GP variants was 7.9% (8/101) at Yaolu Village whose population was mainly composed of Yao ethnic group; while that of Buyi ethnic group at Maolan Village was higher (15.3%; 11/72). The erythrocyte membrane contents of SA in residents at both villages exhibited a very significant tendency of decline (P < 0.01), whereas those of total SH groups increased prominently in residents of Yaolu Village only (P < 0.05). CONCLUSIONS: The frequency of GP variants in this hyperendemic area does not depend upon the severity of malarial prevalence. The evident reduction of SA contents in the residents may be related to the breaking down of the SA residues on membrane GPs by the invasion of Plasmodium vivax.

Pharmacokinetics of intravenous arsenic trioxide in the treatment of acute promyelocytic leukemia.

OBJECTIVE: To study the pharmacokinetics and metabolism of arsenic trioixide (As2O3) and its main side effects. METHOD: As2O3 was administered intravenously at the dose of 10 mg per day for the treatment of 8 relapsed acute promyelocytic leukemia (APL) patients. The arsenic content was measured by Gas-phase chromatography. RESULTS: The plasma maximal concentration (Cpmax) was 0.94 +/- 0.37 mg/L (x +/- s), time to peak concentration (Tp) was 4 hours, plasma distribution half-time (t1/2 alpha) and elimination half-time (t1/2 beta) were 0.89 +/- 0.29 hours and 12.13 +/- 3.31 hours, respectively. Apparent distribution volume (Vc) was 3.83 +/- 0.45 L, system clearance (CLs) was 1.43 +/- 0.17 L/h, and area under curve (AUC) was 7.25 +/- 0.97 mg.h/L. The continuous administration of As2O3 did not alter its pharmacokinetic behaviors. During As2O3 treatment, 24-hour arsenic content in urine accounted for 1%-8% of the daily dose (10 mg). When arsenic accumulation in hair and nail increased continuously, the peak concentration could be five to seven-fold higher than that of pre-treatment. Importantly, arsenic contents in both urine and hair or nail declined gradually after drug withdrawal. No bone marrow suppression or severe organ-impairment was found. CONCLUSION: As2O3 is a relatively safe and effective remedy in the treatment of patients with relapsed APL, in spite of certain degree of arsenic accumulation in some tissues.

[Diverse effects of glucocorticoids on diverse leucocytes apoptosis].

OBJECTIVE: To observe effects of glucocorticoids on the process of apoptosis in main inflammatory cells related to pulmonary inflammation. METHOD: Human peripheral blood neutrophil(PMN), eosinophil (EOS) and T lymphocyte(TLC) were cultured in vitro under glucocorticoids condition. In situ end-labeling technique of fragmented DNA was used to detect the dynamic fesatures of apoptosisin experimental collected cell. The peripheral PMN apoptosis in acute respiratory distress syndrome(ARDS) and EOS, TLC apoptosis in bronchial asthma were determined by the same method before and after treatment with glucocorticoids. RESULT: PMN, EOS and TLC showed signs of apoptosis spontaneously and no signs of surviving cells after 12 h/43 h, 15 h/65 h and 65 h/128 h of culture respectively. Dexamethasone accelerated EOS and TLC apoptosis, while inhibited PMN apoptosis. The PMN apoptosis in ARDS and EOS, TLC apoptosis in bronchial asthma were in various degreof inhibitory state. The above time of PMN, EOS and TLC apoptosis were 41.5 h/92.6 h,29.3 h/100 h,92.4 h/152.2 h respectively. The inhibitory state of PMN apoptosis was not influenced by glucocorticoids therapy, but the EOS and TLC apoptosis were relieved from their inhibitory state. CONCLUSION: Effects of glucocorticoids are different in diverse leucocyte apoptosis. This could have an differential significance in pulmonary inflammation.

[Determination of serotonin in rat brain by gas chromatography/mass spectrometry].

A GC/MS method for the determination of serotonin (5-hydroxytryptamine, 5-HT) has been developed. 5-HT in homogenized rat brain was acylated and extracted into EtOAc. The acylate was further derivatized with heptafluorbutyl anhydride and formed a spirocyclic pentafluoropropionyl derivative. Subsequent analysis of 5-HT derivative by gas chromatography-electron capture negative ion chemical ionization mass spectrometry was highly specific and sensitive. The assay is linear from 0.5 to 50 micrograms/L with a linear equation of Y = 0.134 8X - 0.07995, r = 0.999. The recovery was 98.2% +/- 3.8% (n = 10) and the limit of detection was 0.5 microgram/L. The reproducibility of 5-HT determination in rat brain was less than 10% (RSD) at both levels of 1.0 microgram/L and 50.0 micrograms/L. The method has been applied to analysis of 5-HT in the dose rat brain successfully.

[Effect of Jianpi Yiqi recipe on the heat shock proteins in rats with acute injury of gastric mucosa].

OBJECTIVE: To investigate the mechanism of Jianpi Yiqi Recipe (JPYQR) in inhibition of gastric mucosa injury. METHODS: A rat model of acute injury of gastric mucosa was established by binding and immersing in water. Ulcer index (UI) was measured, change of heat shock protein (HSP70) in plasma and gastric tissue was examined, and the gastric HSPs was studied immunohistochemically. RESULTS: The UI in rats pretreated with JPYQR or Lizhudele was lower than that of model group (P < 0.001), and UI of JPYQR group was lower than that of Lizhudele group (P < 0.01). On the other hand, the HSP70 level in the two pretreated groups were significantly higher than that of the model group (P < 0.05), and the plasma HSP70 level of JPYQR group was higher than that of Lizhudele group (P < 0.05). The difference of gastric HSP70 level between the two pretreated groups was insignificant (P > 0.05). Immunohistochemical study revealed a high level expression of HSP70 and HSP60 by glands and the level of the expression in model group was the highest, followed by those in Lizhudele group and JPYQR group. CONCLUSION: JPYQR could enhance the expression of HSPs in rats, and so as to prevent the gastric mucosa from injury. HSPs might involve in the defending mechanism of gastric mucosa.

[Changes of plasma endocrine hormone in pilots under Coriolis acceleration].

Plasma endocrine hormones were studied in both 24 motion sickness (orthostatic intolerance) and healthy pilots. Coriolis acceleration of 3.75, 5.00 and 6.25 pi 2 cm/s2 were given with intervals of 3-4 min AT-II, insulin, cortisol, Aldosterone and gastrin were determined by radioimmunoassay. It was found that aldosterone, AT-II, gastrin increased with increase of coriolis acceleration in all pilots. (P < 0.05), but cortisol and insulin only increased in healthy pilots (P < 0.05). It suggests excitation of the autonomic nervous system might be insufficient in orthostatic intolerant pilots and that determination of endocrine hormones may be useful in the evaluation of autonomic nervous activities.

The change of arteriovenous carbon dioxide and pH gradients during severe hemorrhagic shock and resuscitation.

OBJECTIVE: To investigate clinically useful markers for determining the severity of hemorrhagic shock and adequacy of resuscitation. METHODS: Prospective study was undertaken in 12 dogs, using an established model for hemorrhagic shock. The anesthetized dogs were bled to a mean arterial pressure of 40 mmHg which was maintained for 3 hours. Then each animal was resuscitated with heperinized whole blood followed by intravenous infusion of dobutamine at a rate of 5 micrograms.kg-1.min-1 for 10 minutes. Arterial and mixed venous blood gases, arterial lactate concentrations and hemodynamic parameters were measured throughout the study. RESULTS: A difference in the PCO2 and pH values between arterial and mixed venous blood was observed. Arterial-venous PCO2 and pH difference increased significantly after sustained shock. The arteriovenous carbon dioxide and pH gradients recovered more rapidly than arterial lactate levels after successful resuscitation with blood and dobutamine. CONCLUSION: Arterial blood gases fail to reflect the acid-base status of tissues during hemorrhagic shock. The differences in PCO2 and pH values between arterial and mixed venous blood could be used as clinical indicators for assessing the severity of shock and efficacy of resuscitation.

[Treatment of arteriovenous malformation by linear accelerator radiosurgery].

OBJECTIVE: To summarize the effectiveness of radiosurgery for cerebral arteriovenous malformations (AVMs). METHOD: We treated with linear accelerator radiosurgery 17 patients harboring AVMs in the brain. 15 AVMs were located supratentorially and others infratentorially. All the patients were treated with single fraction. The doses at AVM margin were 15 to 30 Gy. RESULT: AVMs disappeared in 13 of the 17 patients, reduced in 3 and unchanged in only 1 at 2 year follow up. Vasogenic edema observed in 2 patients, their neurologic deficits were all improved with steroid. CONCLUSION: Linear accelerator radiosurgery is effective in dealing with cerebral AVMs. It provides with a new means of treatment for AVMs which can't be effectively treated with microsurgery and embolization.

[Immunohistochemical investigations of congenital pseudarthrosis of the tibia].

OBJECTIVE: To investigate the etiology and pathology of congenital pseudarthrosis. METHOD: Sixty-three specimens were taken from 21 CPT patients. The antibodies, which were used for immunohistochemical studies, were produced against (1) NF-200 or S-100 protein; (2) type I or III collagen; (3) PCNA. The control specimens were taken from neurofibromatosis and traumatic pseudarthrosis. SABC staining was performed. RESULT: Low positive expression of NF-200 (3.2%) and S-100 (4.8%) was noted in the CPT lesion, whereas high expression of NF-200 (85.2%) and S-100 (88.9%) in the lesion of neurofibromatosis. The soft tissues in CPT had a higher ratio of type III to I collagens in comparison with traumatic pseudarthrosis. Positive expression of PCNA (95.2%) was significantly higher in the soft tissues than that (2.5%) in traumatic pseudarthrosis. CONCLUSION: CPT is a disease of non-neuro origin. Its occurrence has no direct relation with neurofibromatosis. CPT has its characteristic pathology in the periosteum rather than in the bone. The pathological fracture or non-union of CPT is caused by fibromatosis, which has a strong activity of cellular proliferation and corrosion. CPT has its pathological property similar to a tumor.

[Determination of azathiopurine and 6-mercaptopurine in serum by reversed-phase high performance liquid chromatography].

A method for simultaneous determination of azathiopurine(AZP) and its metabolite 6-mercaptopurine (6-MP) concentration in serum has been developed. Reversed-phase high performance liquid chromatography with gradient elution and dual wavelength detection was employed. After protein precipitation with acetonitrile, the serum sample was evaporated to dryness with a gentle N2 stream at 37 degrees C and 100 microL of the mobile phase were added to dissolve the residue. After centrifugation, 20 microL of the supernatant was injected directly into the HPLC. The detection wavelength was set at 278 nm (AZP) and 325 nm (6-MP). The internal standard was metronidazole. The minimum detectable mass concentration for both AZP and 6-MP in serum was 5 mg/L. The average recoveries were (100.6 +/- 4.2)% for AZP and (102.4 +/- 4.5)% for 6-MP.

[In vitro testing of the intravenacaval respiratory assist device (I)--Pressure drop characteristics and oxygen transfer performance].

The intravenacaval respiratory assist device(IVOX) of three different sizes developed by the present authors were evaluated experimentally in vitro. The tests using in turn two different test chamber materials(rigid and elastic) showed that a pressure drop occured. The pressure drop was higher in rigid test chamber than that in the elastic one, and the pressure drop difference between these two kinds of chamber increased with the diameter of the bundle of IVOX. Oxygen transfer rate, with water as the medium in the tests, was augmented with increasing water flow rate and gas flow rate, and with decreasing inlet oxygen concentration in water. It was concluded that, to avoid bubbles produced by IVOX, the gas flow rate should be controlled with fixed outlet water pressure, and vacancy be used in the effluent gas system.

[FTIR microscope study on the change of membrane protein and lipids of erythrocyte damaged by free radicals].

The FTIR Microscope and Computer Aided Analysis were used to study not only the change of secondary structure of erythrocyte membrane protein attacked by the free radicals, but also the effect of oxygen centered free radicals on the membrane lipids in this paper. The results indicate that the content of alpha-helix in the secondary structure of protein clearly changed because of the attack of the free radicals. Damaged by free radicals in 30 minutes, the protein structure could not be restored after 3 hours. The number of the P=O (vp=0), C=O and C=C in the lipids of the membrane was changed. These changes were caused by the peroxidation of the oxygen centered free radicals.

Antineoplastic and cytotoxic activity of beta-alkylamino-(para-substituted)propiophenone and beta-alkylamino-(6-methyl)-naphthone derivatives in murine and human tissue culture cells.

The beta-alkylamino(para-substituted)propiophenone and beta-alkylamino-(6-methyl)naphthone derivatives were evaluated for their antineoplastic effects in vitro in CF1 mice at 8 mg/kg/day intraperitoneal for 9 days. A number of these agents showed over 70% inhibition of Ehrlich ascites carcinoma growth. In in vitro cytotoxicity assays, these agents significantly inhibited the growth of a number of cancer cell lines from both human and murine origins. Some agents showed more activity than several standard anticancer drugs against certain cell lines. Three analogs, beta-(4-methyl)piperidino-(para-methyl)propiophenone (4), beta-piperidino-(para-ethoxy)propiophenone (13), and beta-hexamethyleneimino-(para-ethoxy)-propiophenone (14), were selected for mode of action studies and all showed significant inhibition of DNA and RNA syntheses at 100 microM after 60 min incubation. However, the most significant site of action was the inhibition of the activity of dihydrofolate reductase.

Reversal by desferrioxamine of tau protein aggregates following two days of treatment in aluminum-induced neurofibrillary degeneration in rabbit: implications for clinical trials in Alzheimer's disease.

A clinical trial in patients with Alzheimer's disease has indicated that frequent intramuscular (i.m.) treatment with desferrioxamine (DFO) slows progression of the disease. Confirmatory trials have not been carried out, partly because of the rigors of twice daily intramuscular injections over a period of 2 years, even though the initial report gave promising results. The aim of the present study was to determine an optimal DFO treatment protocol in an animal model exhibiting Alzheimer's-like intraneuronal protein aggregates, previously shown to be partially reversed by such treatment. New Zealand white rabbits were injected intracisternally with either aluminum (Al) maltolate or with saline on day 0. Intramuscular injections of DFO were given to selected rabbits for 2 days prior to sacrifice on days 4, 6 or 8. Bielschowsky's silver impregnation demonstrated widespread neurofibrillary degeneration (NFD) in neuronal cell bodies and neurites of brain and spinal cord from Al-treated rabbits. Monoclonal antibodies Tau-2, AT8, PHF-1 and Alz-50, all of which characteristically stain neurofibrillary tangles associated with Alzheimer's disease, strongly labeled the Al-induced NFD. The number of positive neurons and staining intensities were much less in rabbits treated with Al and subsequently with DFO, than in animals only given Al. Control rabbit receiving intracisternal saline were negative for NFD. The results of quantitative immunohistochemistry using image analysis confirmed that immunostaining densities with all tau mAbs were higher in Al-treated than in Al-DFO-treated or in saline-treated controls. Furthermore, it appears that hyperphosphorylation of tau does not make this protein resistant to degradation once Al has been removed by DFO treatment. The effectiveness of only two days of DFO treatment in reversing Al-induced neurofibrillary degeneration suggests that further clinical trials of DFO for treatment of Alzheimer's disease should be attempted using much less frequent administration of DFO than in the initial study (Crapper McLachlan et al., 1991).

The linear-quadratic model and most other common radiobiological models result in similar predictions of time-dose relationships.

One of the fundamental tools in radiation biology is a formalism describing time-dose relationships. For example, there is a need for reliable predictions of radiotherapeutic isoeffect doses when the temporal exposure pattern is changed. The most commonly used tool is now the linear-quadratic (LQ) formalism, which describes fractionation and dose-protraction effects through a particular functional form, the generalized Lea-Catcheside time factor, G. We investigate the relationship of the LQ formalism to those describing other commonly discussed radiobiological models in terms of their predicted time-dose relationships. We show that a broad range of radiobiological models are described by formalisms in which a perturbation calculation produces the standard LQ relationship for dose fractionation/protraction, including the same generalized time factor, G. This approximate equivalence holds not only for the formalisms describing binary misrepair models, which are conceptually similar to LQ, but also for formalisms describing models embodying a very different explanation for time-dose effects, namely saturation of repair capacity. In terms of applications to radiotherapy, we show that a typical saturable repair formalism predicts practically the same dependences for protraction effects as does the LQ formalism, at clinically relevant doses per fraction. For low-dose-rate exposure, the same equivalence between predictions holds for early-responding end points such as tumor control, but less so for late-responding end points. Overall, use of the LQ formalism to predict dose-time relationships is a notably robust procedure, depending less than previously thought on knowledge of detailed biophysical mechanisms, since various conceptually different biophysical models lead, in a reasonable approximation, to the LQ relationship including the standard form of the generalized time factor, G.

Effects of isovolumetric hemodilution treatment and superoxide dismutase on ischemic reperfused hearts in rabbit.

UNLABELLED: The isovolumetric hemodilution treatment reduces ischemic injury and anti-radical interventions may reduce reperfusion injury. However, the effects of the treatment with both interventions have never been investigated. In the present study, anesthetized rabbits underwent 45 min of coronary artery ligation, which was followed by 180 min of reperfusion. Ten animals in each group received: (i) normal saline only; (ii) isovolumetric hemodilution with Dextran 40 during ischemia; (iii) superoxide dismutase (SOD) 1 min before and during reperfusion; and (iv) combined treatment with hemodilution and SOD. RESULTS: (1) compared with those of the control group, left ventricular peak pressure (LVP) and +/- dp/dt-max improved significantly in the animals which received hemodilution or SOD alone, and left ventricular systolic function of the animals with the combination therapy improved further; (2) infarct size averaged 54.4 +/- 2.6% of the risk region in controls and was significantly smaller in the rabbits that were treated with hemodilution (32.8 +/- 1.8%) (p < 0.05), with SOD (42.1 +/- 3.2%) (p < 0.05) and with the combination therapy (23.4 +/- 1.5%) (p < 0.01). Therefore, (1) in groups where either hemodilution during ischemia or removal of oxygen radicals during reperfusion was done left ventricular systolic function and salvaged ischemic reperfused myocardium improved significantly; (2) in this acute model, SOD improved left ventricular systolic and diastolic functions and reduced infarct size further when given to the animals that had been treated with hemodilution.
