The effect of bu shen sheng xue fang on beta-thalassemia at gene level.

Isoelectrofocusing electrophoresis, PCR-SSCP analysis with DNA direct sequencing and RT-PCR were used to analyze the constituents of hemoglobin, determine the pattern of globin gene mutation, and detect its mRNA transcription extent. The results indicated that Bu Shen Sheng Xue Fang (BSSXF) significantly increased the contents of hemoglobin (Hb) and fetal hemoglobin (HbF), and the chain ratio in patients with beta-thalassemia. The drug is markedly effective in heterozygous, but not effective in homozygous or genetic compound patients. It was suggested that the beta-globin gene defects were compensated by elevating the gamma/beta + gamma ratio, promoting the transcription and expression of gamma-globin gene in synthesizing HbF.

[A prospective study on etiologic bacteria in 200 patients with pneumonia].

The etiologic agents in 200 patients with pneumonia were studied by the bacterial culture of sputums obtained from the protected single catheter brush or quantitative expectoration at one morning or three-morning expectoration. Two hundred patients were divided into 3 groups. Group 1 was Nosocomial pneumonia (NP patients). Group 2-1 and Group 2-2 were community acquired pneumonia (CAP patients). All cases in Group 1 and Group 2-2 suffered from significant underlying diseases while Group 2-1 did not. Gram-negative bacilli(GNB) were isolated from the specimens in Group 1 (87%) and Group 2-2 (75%), respectively. Pseudomonas (30.8%) and klebsiella (20.5%) were the predominant bacteria (in Group 1 and pseudomonas bacteria) in Group 1 and pseudomonas (27.3%), acinetobacter (23%) and kledsiella (18%) were the major etiologic agents in Group 2-2. The commonest pathogens in Group 2-1 were gram-positive cocci (75%), in which streptococcus (38%) and staphylococcus aureus (25%) were the dominant agents. Compared with Group 2, Group 1 suffered from more mixed bacteria and the agents presented severer drug-resistant. The prognosis was worse in Group 2-2 than in Group 2-1. The results showed that the GNB pneumonia was more common in the cases who had underlying disease, no matter whether the pneumonia was NP or CAP. These patients had more trouble on their antibiotic therapy. Thus it is important that doctors should use vigorous antibiotics timely while treating these patients' underying diseases.

[A new fluorescent scanning technique in quantitative determination of isoenzymes of creatine kinase].

To increase the sensitivity and specificity and decrease the cost on determining the isoenzymes of creatine kinase (CK), including CK-MM, CK-MB, and CK-BB, we used the electrophoresis bath with different buffers to isolate the isoenzymes and used the fluorescent scanner to determine the content of each isoenzyme. We found that the concordance appeared in sensitivity and specificity in comparison with the MOPS electrophoretic system. There was a positive correlation between our method and MOPS, and the cost was much less than that of the MOPS.

[Scavenging effect of total flavonoids of lycium barbarum L on active oxygen radicals and inhibitory effects on heat output from L1210 cells].

The scavenging effects of total flavonoids of Lycium barbarum L. (TFL) were studied by using ESR-spin trapping technique and the inhibitory effects on heat output of both polymorphonuclear leukocyte(PMN) respiration burst and L1210 cells were measured by using microcalorimetric technique. TFL (0-217 mg/L) could scavenge O2-. in xanthine/xanthine oxidase (Xan/XO)system, with scavenging rate of 0-51%. TFL(7.5-200 mg/L)could scavenge OH. produced in Fenton reaction and the scavenging rate is between 20% to 72%. Those effects were concentration-dependent. Furthermore, TFL(0.56 g/L)could completely inhibit the heat output from PMA-stimulated PMN and TFL(1.0-5.0 g/L)could inhibit the heat output from L1210 cells.

[Effect of ischemia/reperfusion in cardiac surgery with cardiopulmonary bypass on nitric oxide levels of peripheral blood in patients].

To examine the effect of ischemia/reperfusion in cardiac surgery with cardiopulmonary bypass (CPB) on nitric oxide (NO) levels of peripheral blood, venous levels of NO2- and NO3- were measured at multiple time-points before, during, and after CPB by means of high-pressure liquid chromatograph to reflect NO concentration indirectly in 20 patients. The results showed there was a linear regression between the decreased trend of NO release at the period of peri-CPB is related to the endothelial cell activation caused by ischemia/reperfusion at the same time. Therefore it is necessary to explore the regulation and action of NO release during systemic inflammatory response procedure in CPB.

[Sjögren's syndrome and malignant lymphoma].

To diagnose and study the early stage of Sjögren's syndrome associated with malignant lymphoma, the authors used the PCR technique of B malignant lymphoma gene for lacrimal gland tissues in four patients with Sjögren's syndrome and in five patients suffering from B malignant lymphoma. The results showed that all cases of Sjögren's syndrome were negative and all cases of B malignant lymphoma were positive. These suggest that in general the patients with primary or secondary Sjögren's syndrome have no malignant lymphoma genes, but those who have chronic fever and enlarged superficial lymph nodes should under go this technique for early diagnosis and treatment of Sjögren's syndrome associated with malignant lymphoma.

[Simultaneous assay of Ca(2+)-ATPase and Na+, K(+)-ATPase activities of osteoblast rat by malachite green colorimetic method].

The aim of this study was to find a method to measure simultaneously the activities of Ca(2+)-ATPase and Na+, K(+)-ATPase of rat osteoblast membrane. In this test, Ca(2+)-ATPase and Na+, K(+)-ATPase released inorganic phosphate (Pi) from ATP, and Pi was determined by colorimetric method using malachite green dye to show ATPase activities. Ca2+ concentration and osteoblast culture generation were important factors affecting ATPase activities. Ca2+ could elevate membrane ATPase activity. When Ca2+ concentration reached 40 mumol/L, ATPase activities were improved. Ca(2+)-ATPase activity decreased with the increase of osteoblast culture generation. Ca(2+)-ATPase activity was lower than Na+, K(+)-ATPase activity in osteoblast membrane. This method is simple, sensitive and reliable.

[Study of magnesium and calcium levels of plasma and within erythrocyte before and after magnesium sulfate treatment in patients with pregnancy induced hypertension].

OBJECTIVE: To study the changes of calcium and magnesium levels in patients with pregnancy induced hypertension (PIH) and the therapeutic effect of magnesium sulfate (MgSO4). METHODS: The calcium and magnesium levels in plasma and erythrocytes of 36 patients with PIH and 14 healthy gravidi in their late pregnancy were determined and the changes after 4 hours infusion of MgSO4 as well. RESULT: Plasma calcium decreased significantly in moderate and severe PIH patients, but calcium increased and magnesium decreased in their erythrocytes. After MgSO4 infusion for PIH, magnesium levels in both plasma and erythrocytes increased, while calcium increased in plasma and decreased in erythrocyte to normal levels. CONCLUSION: Magnesium deficiency is the basis of calcium and magnesium disturbances in PIH, and MgSO4 treatment may improve the situation effectively.

[Analysis of 196 cases of trial of labor with borderline oligohydramnios assessed by ultrasound].

OBJECTIVE: To determine the effect of borderline oligohydramnios on perinatal outcome. METHOD: Patients admitted for delivery had an amniotic fluid index (AFI) assessment. Color Doppler was used to examine the systolic/diastolic ratio of the blood flow velocity in fetal umbilical arteries. Normal amniotic fluid volume was defined as an AFI of 8.1-18 cm (n = 200), and borderline oligohydramnios as 5.1-8.0 cm (n = 196). The two groups were compared on perinatal outcomes as all of the patients had trial of labor. RESULTS: There was no difference in the incidence of fetal distress and neonatal mortality between the two groups. The incidence of emergency section in borderline oligohydramnios group was significantly higher. CONCLUSION: The patients with borderline oligohydramnios may have a trial of labor under proper intensive care.

[Effects of retionoic acid and GM-CSF on apoptosis of HL-60 cell induced by mild heat].

The effects of several factors on the apoptosis of HL-60 cells, a human acute myeloid leukemia cell line, were investigated. The apoptosis model was established with mild heat. Typical changes of cell morphology and characteristic DNA ladder pattern were found. The effect of apoptosis induced by mild heat was enhanced, when HL-60 cells were pretreated with the retinoic acid. By contraries, that of apoptosis was displayed with the slight inhibition while HL-60 cells were pretreated with rhGM-CSF. We also observed the alterations of mRNA expressions of c-myc by the dot hybridization. The results indicate that both the mild heat and the combination of mild heat and retinoic acid obviously reduce the mRNA expression of c-myc compared to the control group.

[Protective effect of selenium on human erythrocyte rheology].

OBJECTIVE: To assess the protective effect of selenium on RBC rheology during myocardial ischemia/reperfusion period in patients with VSD or ASD who underwent cardiopulmonary bypass. METHODS: The blood samples were taken from the coronary sinus, and ascorbyl free radical (A.), peroxidants (MDA), antiperoxidative activities (plasma SOD, erythrocyte GSH-Px), the membrane shear elastic module of RBC (mu), molecular rotational correlation times of lipids and proteins of RBC membrane (tau l, tau p) were measured and compared between control and selenium groups. RESULTS: After myocardial reperfusion only for one minute, in both groups A. was significantly increased (The increased percentage in the control group +68%, in the selenium group +66%), but the selenium group had lower MDA and GSH-Px activity. On contrary to the control group with high A. level, A. of the selenium group was dropped about half since reperfusion for 5 minutes and recovered to the pre-ischemia level. After myocardial reperfusion, the control group had specific changes in membrane mu values as well as evidently prologed tau l and tau p. Simultaneously, except tau p at 5 minutes of reperfusion was obviously prolonged, no changes in the membrane mu and tau l were observed in the selenium group during the cardiac ischemia/reperfusion periol. CONCLUSION: Selenium can at least partly protect RBC rheology from free radical damage during the myocardial ischemia/reperfusion period.

[Effects of erythropoietin or nitric oxide synthesis inhibitor on hyperdynamic circulatory state in cirrhotic rats].

OBJECTIVE: To observe the effects of erythropoietin or nitric oxide synthesis (NOS) inhibitor NG-nitro-L-arginine methyl ester (L-NAME) on hyperdynamic circulatory state in rats with cirrhosis. METHODS: Cirrhotic rat model was made. Cirrhotic rats were treated with NOS inhibitor L-NAME (0.5 mg.kg-1.d-1) by gavage or with erythropoietin (100 U/kg) injected subcutaneously for two weeks. The hemodynamic parameters in cirrhotic rats treated with L-NAME or with erythropoietin were determined by using 57Co-labled microsphere technique. Serum nitric oxide (NO) levels were also measured by using a fluorometric assay. RESULTS: Hyperdynamic circulatory state was observed in all rats with cirrhosis. Serum NO levels in cirrhotic rats were significantly higher than that in normal controls. Hyperdynamic circulation status in cirrhotic rats treated with erythropoietin or with L-NAME was markedly attenuated. As compared with untreated-cirrhotic rats, serum NO concentration in erythropoietin-treated and L-NAME-treated cirrhotic rats were significantly lower. CONCLUSION: L-NAME treatment could reverse the hyperdynamic circulatory state in cirrhotic rats which might be ameliorated by inactivation of overproduced NO by increasing hemoglobin with erythropoietin.

[Effects of hypertonic solution on the membrane potential of isolated outer hair cell in the guinea pig cochlear].

To learn effects of hypertonic solution on the membrane potential and cell length of isolated outer hair cell in the guinea pig cochlear, we determined the membrane potential and cell length during hypertonic activation with ACAS 570 and the membrane potential sensitive probe DiBAC4(3). The results showed that the hypertonic activation depolarized the outer hair cell and made it elongate. We think that the outer hair cell has activity, the depolarization and elongation of the outer hair cell caused by hypertonic solution are cyto-electrophysiological basis of hearing loss and recruitment in Meniere's disease.

Gene expression of receptor for advanced glycosylation end products and its modulation by aminoguanidine in diabetic kidney tissue.

OBJECTIVE: To explore the relationship between receptor for advanced glycosylation end products (RAGE) gene expression in the kidneys of diabetic rats and the role of aminoguanidine on it. METHODS: Among four groups of rats, 2 groups received streptozotocin (STZ) to induce diabetics (DM) and were followed by either aminoguanidine (AG) treatment for 12 weeks or no therapy. Two groups of normal rats in which one received AG and the other given water served as control (Con). Reverse transcription-polymerase chain reaction (RT-PCR) was used to measure the amount of receptor of advanced glycosylation and products (RAGE) messenger RNA (mRNA) expression. RESULTS: After 4 weeks of diabetes inducement, RAGE mRNA levels showed a continuous increase up to 12 weeks both in diabetic renal cortex and medulla compared with mRNA levels at the end of 2 weeks. The RAGE mRNA levels in renal cortex and medulla of control rats did not change significantly with age. AG as the prototype inhibitor of advanced glycosylation end products (AGEs) formation, when administered to diabetic rats for 4 weeks, had no visible effect on the alterations of renal RAGE mRNA in diabetes. By continuous administration up to 8 weeks, those alterations of RAGE mRNA both in diabetic renal cortex and medulla were all ameliorated [P < 0.05, (diabetes without AG) versus (diabetes with AG)]. In addition, diabetic animals had a progressive increase in urinary protein excretion compared to control rats from week 4 onwards. Similarly, after 8 weeks diabetic rats had significantly higher glycated Hb (GHb) (DM without AG vs Con: 7.71 +/- 0.22% vs 2.95 +/- 0.52% P < 0.001). AG treatment for 8 weeks decreased GHb by 24.78% and retarded the rate of rise in albuminuria in diabetic rats as compared with that in untreated diabetic rats (DM with AG vs DM without AG: 22.04 +/- 0.91 micrograms/24 h vs 70.25 +/- 13.05 micrograms/24 h, P < 0.001). CONCLUSIONS: Gene expression of RAGE in renal cortex and medulla was altered in diabetic rats and excessive gene expression of RAGE in kidney tissue may enhance the interactions between advanced glycosylation end products (AGEs) and its receptor (RAGE) which would contribute to the development of diabetic nephropathy. Cumulative AGEs play an important role in increasing RAGE gene expression in vivo. The decrease of AGEs level by AG therapy ameliorate the abnormal regulation of RAGE gene expression.
