[Color differences between light-cured composite resin made shade guides and manufactured shade guides].

The purpose of this study was to investigate color differences between shade guides and light-cured composite resins. Resin specimens were made with resins and opaque resins of Silux Plus and Silux (3 M Co.) in a shade-guide metal mold. The resin specimens and the shade guides therefore were identical in shape and size. The sample resin specimens were compared with the shade guides provided with Silux Plus and Silux light-cured composite resins. Color measuring was performed with the Murakami Color Research Laboratory Fast Spectrophotometer C M S -500 and the Flexible Sensor F S -1. Specimens were placed on a plastic disc fixed to the top of the flexible sensor, and identical areas of their incisal, central, and cervical parts were measured. Conclusions. 1) The delta E*ab values between all shade guides and resin specimens showed color differences detectable to the naked eye. 2) Color differences between shade guides and resin specimens were great for the incisal colors (I, X L) and the deep and dark colors (D G, D Y, Y B, D G O, D Y O, Y B O, D D Y O). Color differences were relatively small, however, for the general colors (U, Y, G, U O, Y O, G O). 3) The L* values of shade guides ranged more widely than those of resin specimens. For many colors, L* values of the shade guides were lower than those of the resin specimens. 4) The a* values of shade guides and resin specimens were much more limited. The a* values of all colors showed minus values. The magnitude of a* values of shade guides were lower than those of resin specimens. 5) The b* values of shade guides were higher for some colors and lower for some colors than those of resin specimens.

Effects of cleaning, polishing pretreatments and acid etching times on unground primary enamel.

The purpose of this study was to investigate the influence of cleaning and polishing pretreatments and etching times on unground primary enamel smooth surfaces with respect to the quality of etching patterns. Labial surfaces of 117 extracted or exfoliated caries free primary anterior teeth were used. Various cleaning and polishing methods were used before etching. Etching times with 40% phosphoric acid gel were 10, 30 and 60 seconds and all specimens were washed with an air water spray for 30 seconds. The following observations were obtained using the SEM. 1. There was not a significant difference between the appearance of prism structures by etching and the pretreatment methods in the 60 and 30 seconds etching. 2. The shorter the etching time, the higher was the rate of the cases which did not show any prism structure after etching.

Rapid methods for purification of human recombinant interleukin-5 (IL-5) using the anti-murine IL-5 antibody-coupled immunoaffinity column.

A large scale production of human recombinant IL-5 (hIL-5) was performed by way of recombinant DNA technology. In this study, we transfected Chinese hamster ovary cells with pdKCR-hIL-5gene-dhfr plasmid and selected a cell line, with the use of methotrexate, producing large amounts of hIL-5. The recombinant hIL-5 thus obtained induced IgM production of murine B cell leukemia BCL1, and its activity was inhibited by TB13 anti-mouse IL-5 monoclonal antibody. hIL-5 could be purified from the cell-free supernatants of the transfectants with high recoveries by using anti-mouse IL-5 antibody-coupled immunoaffinity column in combination with a gel permeation column chromatography. N terminal amino acid sequence analysis of purified hIL-5 revealed that a single amino-terminal amino acid (isoleucine) is detected and hIL-5 consists of 115 amino acid residues.

Direct projection from the dorsal hypothalamic area to the nucleus raphe pallidus: a study using anterograde transport with Phaseolus vulgaris leucoagglutinin in the rat.

A hypothalamic projection to the nucleus raphe pallidus of the medulla was examined using the anterograde tracing technique based on Phaseolus vulgaris leucoagglutinin (PHA-L) in the rat. After the iontophoretic application of PHA-L to the dorsal hypothalamic area, labeled fibers that finally ended in the nucleus raphe pallidus were observed descending through the most medial part of the ventral tegmental area and the nucleus reticularis tegmenti pointis to reach the medial aspect of the pyramid. Many varicose fibers forming a loose plexus were observed in the nucleus raphe pallidus, especially ventrally. The ventral surface of the pyramid and the most ventral region of the nucleus reticularis paragigantocellularis lateralis (PGCL) contained labeled varicose fibers. At the electron microscopic level, the labeled profiles in the nucleus raphe pallidus were small-sized unmyelinated axons and axon terminals. Labeled axon terminals containing spherical synaptic vesicles formed synapses on spine-like protrusions or small-sized dendritic shafts. These results strongly indicate that neurons in the dorsal hypothalamic area have a direct connection with neurons in the nucleus raphe pallidus and the ventral part of the PGCL. The possible involvement of this pathway in cardiovascular regulation was discussed.

Difference in distribution of central terminals between visceral and somatic unmyelinated (C) primary afferent fibers.

1. In the guinea pig, the central projections of somatic and visceral C-afferent fibers were compared by tracing arborizations labeled through injection of Phaseolus vulgaris leucoagglutinin (PHA-L) intracellularly into single neurons of the 13th thoracic dorsal root ganglia (DRG). 2. Two of 27 somatic C-afferent neurons that responded to electrical stimulation of the 13th thoracic (subcostal) nerve (conduction velocity: 0.69 +/- 0.14 m/s, mean +/- SD) were well enough marked to allow delineation of their central processes. In both cases, the entering axon ran rostrally, giving off branches that converged on a single terminal field located in the substantia gelatinosa (lamina II) with some extension in lamina I. The terminal field in each case extended approximately 400 microns rostrocaudally and 100 microns mediolaterally. 3. Intracellular recordings were obtained from 31 afferent units that responded to electrical stimulation of the celiac ganglion. Units with onset latencies of greater than 15 ms were classified as having visceral C-afferent fibers because the shortest course from the celiac ganglion stimulation electrodes to the DRG was greater than 7 mm (i.e., a conduction velocity of less than 0.5 m/s). 4. Seven visceral C-afferent fibers were labeled well enough to follow their central trajectories. Each had a main ascending and a descending central branch. Each main branch in turn issued several collaterals that terminated in the superficial dorsal horn (laminae I and II), laminae IV, V, and X, and occasionally in the dorsal and lateral funiculi. A few collaterals reached the contralateral laminae V and X.(ABSTRACT TRUNCATED AT 250 WORDS)

[Chromatic study of composite resins. Report 1. The influence of the color measuring instruments and the background colors on the chromatic values].

The purpose of this study was to investigate the influence of the structural difference of the color measuring instruments and the background colors on the chromatical values of the composite resins. Composite resin disks which were 10 mm in diameter and 1 mm and 2 mm in thickness, and made by the light-cured composite resin Silux Plus (3M Co.) were chromatically measured. The colors of the composite resin used in this study were universal, yellow, dark-yellow and light. The color measuring instruments used in this study were the Murakami Color Research Laboratory Fast Spectrophotometer CMS-500 used with the Flexible Sensor FS-1, FS-2 and FS-3, and the Murakami Color Research Laboratory Color Difference Meter CD-270 used with the Improved Light Guide CD-270. The diameters of the head which receives the light from the object were: FS-1 (2 mm), FS-2 (3 mm), FS-3 (5 mm) and Improved Light Guide CD-270 (2 mm). The chromatical values of the resin specimens were measured without back color, backed by a white plate and backed by a black plate. The chromatical values of the composite resin disks were measured with the different color measuring instruments under the different background colors. The XYZ, L*a*b* and xy values were compared and the following results were obtained. 1) The chromatical XYZ, L*a*b* and xy values of the composite resins showed different value patterns among the color measuring instruments. 2) Different influences of the background colors on the chromatical values of the composite resins were obtained among the different colors and different thicknesses of the resins in the cases of some color measuring instruments were used. 3) The most precise color measuring instrument for chromatical measuring of the composite resins among the instruments used in this study was the Fast Spectrophotometer CMS-500 used with the FS-3. 4) Prior to the comparison of the chromatical values measured by the different color measuring instruments, the chromatical values obtained by measurement with the same specimens should be compared and the structural differences, the precision and the sensitivities of the instruments should be understood.

[Effect of acid etching on the primary enamel smooth surfaces. (3) A study of the etching times].

The purpose of this study was to investigate the influence of etching times for the effect of acid etching on the primary enamel smooth surfaces. Fifty labial surfaces of the extracted or exfoliated caries-free primary anterior teeth were used. The cleaning and polishing pretreatment used in this study was polishing with a brush cone (Howe-Neos-Dental Co.) with water at a low speed (600-6000 r.p.m.). The etchant used in this study was 40% phospholic acid gel and the etching times were 10, 30, 60, 90 and 120 seconds. All specimens were washed with an air water spray for 30 seconds after etching. The following observations were obtained using the SEM. 1. The shorter the etching time, the higher was the rate of the cases which did not show any prism structure. 2. The numbers of the cases which did not show any prism structure were: 4 (40%) out of the 10 in 10 seconds etching, 2 (20%) out of the 10 in 30 seconds etching and 1 (10%) out of the 10 in 60 seconds etching. All cases showed prism structure in 90 seconds and 120 seconds etching. 3. The most common etching patterns in all etching times were poorly structured etching patterns. 4. The longer the etching time, the higher was the rate of the cases which showed peripheral etching patterns. 5. Tooth to tooth differences in the etching effects and patterns were observed under the same etching times. Various etching effects and patterns on the same enamel surface were observed. 6. Among the etching times used in this study, 90 seconds was an adequate time for etching on the unground primary enamel smooth surfaces polished with a brush cone.

[Resin adhesion to the ground enamel. Influence of the ground depths of the enamel and etching times (1)].

The purpose of this study was to investigate the influence of the ground depths of the enamel and etching times on the adhesion of the resin on the ground enamel. Seventy-five extracted and frozen bovine mandibular young permanent incisors were used. The etchant used in this study was 40% phospholic acid gel and the etching times were 0, 10, 20, 30 and 60 seconds. All of the specimens were washed with an air water spray for 30 seconds after etching. The bonding agent and composite resin used in this study were Photo Bond and Photo Clearfil A (Kuraray Co.). Shear bond strengths to the outer enamel layers and inner enamel layers of the same teeth were measured. After the shear bond strength test, all the test surfaces of the enamel and resin specimens were observed using the SEM. The following conclusions were obtained. 1) The etching times which showed the highest bond strength were 20 seconds on the outer enamel layers (bond strength: 35.25 +/- 6.60 MPa) and 30 seconds on the inner enamel layers (bond strength: 40.15 +/- 6.59 MPa). 2) When the enamel was etched with all of the etching times, the bond strengths were significantly higher than those of the enamel without etching on the outer and inner enamel layers. 3) In the etched groups, significant differences between the bond strengths were not obtained among the different etching times on the outer and inner enamel layers. 4) In the comparison of the bond strengths between the outer enamel layers and the inner enamel layers, the bond strengths of the inner enamel layer were higher than those of the outer enamel layer with 0, 10, 30 and 60 seconds of etching times. However, a significant difference between the bond strengths was obtained only in the cases with 30 seconds of etching time. 5) In the etched groups, such a tendency toward a higher bond strength was observed in cases which showed poor enamel prism structures and poor resin tags, and resin widely adhered on the enamel surface. 6) Thirty seconds is a sufficient time for etching on the ground young permanent enamel using Photo Bond and Photo Clearfil A.

[The state of the recall visit at the pediatric dentistry clinic of Nagasaki University Dental Hospital].

The purpose of this study is to discover the actual condition and the state of the recall visit of the patients who appeared at the Pediatric Dentistry Clinic of Nagasaki University Dental Hospital, and to compare the results of 1987 and 1985. Concerning the making of appointments for the recall visit, we sent cards to patients in 1987 though, in 1985 we did not send cards to the patients so the patients themselves had to make a telephone call to arrange an appointment. The results were as follows: 1) The rate of the patients who were making their first time recall visit increased by means of sending cards to patients. 2) Of the total number of 493 patients who appeared at our clinic in 1985, 127 (25.8%) did not finish their dental treatment and stopped visiting our clinic for treatment. 3) The rate of the patients who finished their dental treatment and were making the first time recall visit was 69.9% in 1987. 4) The average ages of the patients were 6 years 5 months in 1987 and 5 years 6 months in 1985. The lower the age of patients, the higher the rate of the recall visits, while the rate of pre-school children was higher than that of school children. 5) The rate of the patients who finished their dental treatment and made recall visits was higher in cases in which the chief complaints of the first visit were caries prevention (86.2%) and caries treatment (71.3%) than the cases of tooth extraction (50%), tooth shape anomalies (50%) and tooth pain (50.5%). 6) There was no significant difference in the number of caries decayed teeth at the first oral examination between the group of patients making their first recall visit and the group of patients rejecting a recall visit. 7) In the case of the patients to whom space retainers and orthodontic appliances were applied, the rate of making recall visits decreased more in 1987 than that of 1985. 8) There was no difference in the rate of those making their first time recall visit between the two categories of diseased children and nondiseased children.

Evaluation of various cleaning and polishing methods on primary enamel.

The purpose of this study was to investigate the effects of cleaning and polishing methods on primary enamel surfaces. Fifty-nine extracted or exfoliated caries free primary anterior human teeth were used. The cleaning and polishing methods used in this study were as follows: application of 10% NaOCl, polishing with a brush cone or a polishing brush at a low speed (600-6000 r.p.m.), polishing with a brush cone and various polishing pastes, cleaning with an air polisher Prophy-Jet or Quick-Jet with NaHCO3 powder and water. The following observations were obtained using the SEM: (1) Air polishing systems were useful for cleaning the enamel surface, (2) It was likely that materials such as organic films will remain on the enamel surface polished with a brush cone or polishing brush alone, and (3) The effect of 10% NaOCl application for 30 seconds was the lowest.

Ultrastructural features of functionally identified primary afferent neurons with C (unmyelinated) fibers of the guinea pig: classification of dorsal root ganglion cell type with reference to sensory modality.

Intracellular labeling of neurons permitted a direct correlation of neuronal profiles with sensory modality of cutaneous receptors. In the guinea pig, 47 neurons in the dorsal root ganglion (displaying C-fiber conduction velocities) were labeled with horseradish peroxidase (HRP) by iontophoresis after determining the sensory modality. Receptive field were explored with systematic "natural" stimuli. Cell areas of all C-fiber units were measured by tracing the cellular contour in light microscopy. The mean cellular diameter calculated from cell areas was 21.8 microns in the second cervical ganglion of the guinea pig. Mean cell diameter for high-threshold mechanoreceptors was 20.9 microns, 24.6 microns for polymodal nociceptors, and 25.7 microns for mechanical-cold nociceptors. Electron microscopic observations showed that all labeled neurons of C-fiber units had profiles of small, dark type-B neurons. Neurons representative of each sensory modality exhibited different cell features, each belonging to a distinct subtype of small B neurons. High-threshold mechanoreceptor and mechanical-cold nociceptor displayed a peripheral lamellar arrangement of cisternae of endoplasmic reticulum (ER), corresponding to the B1 subtype. Polymodal nociceptor units were characteristically of the B2 subtype, in which stacks of long and short cisternae of ER were distributed randomly throughout the cytoplasm, and the arrangement of Golgi bodies varied among these cells. Cooling receptors displayed poorly developed, flattened cisternae of ER and numerous vesicles, typical of the B3 subtype. These results imply that all C-fiber cells belong to the small B-type cell category and that the ultrastructural features of the neuron in the dorsal root ganglion may reflect the sensory modality of the receptive field.

[Results of the UFT sensitivity test using human urogenital cancers transplanted subcutaneously and under the subrenal capsule of nude mice].

Experimental chemotherapy with UFT was performed against human urogenital cancers, transplanted subcutaneously and under the subrenal capsule (SRC) of nude mice. The tumor take with subcutaneous assay was confirmed in 21 cell lines of human neoplasms, which consisted of 9 renal cell carcinomas, 5 renal pelvic carcinomas, 1 ureteral carcinoma, 3 carcinomas of the urinary bladder, 2 prostatic carcinomas and 1 testicular tumor. On the other hand, the tumor take with subrenal capsule assay was confirmed in 17 of the neoplasm, which consisted of 7 renal cell carcinomas, 4 renal pelvic carcinomas, 4 carcinomas of urinary bladder, 1 prostatic carcinoma and 1 testicular tumor. UFT was administered, starting on Day 14 for subcutaneous transplantation and starting on Day 0 for SRC assay. The dose of UFT was the same as clinical dose for both subcutaneous transplantation and subrenal capsule assays. Treatment with UFT was performed for 6 weeks in subcutaneous transplantation assay and for 4 weeks in subrenal capsule assay. Effect of UFT was evaluated by the inhibition rate (IR) calculated by the mean tumor weights of both treated and untreated groups. Responses to the treatment in subcutaneous transplantation and subrenal capsule assays were judged as effective when the IR were higher than 58% and 20%, respectively. Responses rates of UFT in subcutaneous transplantation and subrenal capsule assays were 43% and 82%, respectively.

[Results of drug sensitivity test of UFT, doxorubicin and cisplatin using human urogenital cancers transplanted subcutaneously into nude mice].

Experimental chemotherapy with UFT, doxorubicin and cisplatin was performed against human urogenital cancers transplanted subcutaneously into nude mice. The tumor take was confirmed in 33 cell lines of human neoplasms which consisted of 10 renal cell carcinomas, 5 renal pelvic carcinomas, 3 ureteral carcinomas, 8 carcinomas of the urinary bladder, 3 prostatic carcinomas and 4 testicular tumors. These three antitumor agents were administered starting at day 14 after subcutaneous transplantation. These agents were administered following doses as used clinical practice for 6 weeks. Effect was evaluated by the inhibition rate calculated by the mean tumor weights of both treated and untreated groups. These results were as follows: Synergic action by combination therapy was observed 8 strains (6%); reduced effect was found 37 strains (28%) and immutable group was detected 87 strains (66%). Our results suggest that there is a necessity to perform drug sensitivity test for combination therapy as well as sensitivity test for single drug.
