Purification and physicochemical characterization of Schistosoma mansoni egg allergen recognized by mouse sera obtained at an acute stage of infection.

A major allergenic component recognized by mouse sera obtained at an acute stage of infection was purified from soluble egg antigen preparation (SEA) of Schistosoma mansoni by anion-exchange chromatography on DE52 and gel chromatography on Sephadex G-150. The purified allergen showed homogeneity by immunoelectrophoresis and by polyacrylamide gel electrophoresis. Its apparent molecular weight was 210,000 by gel chromatography on Sephadex G-200. This purified allergen could bind to Con A-Sepharose 4B, indicating its glycoprotein nature. After amino acid analysis, aspartic acid, glutamic acid, serine and theonine were found as the major amino acids. The allergenic activity was destroyed by heating at 100 degrees C for 60 min and by pronase or periodate treatment. By double diffusion in agar gel, this purified allergen gave a strong single band against acute (8 w) stage serum, which fused to the major band formed by crude SEA. On the other hand, it showed a very faint band against chronic (22 w) stage serum, which is apparently different from the main band formed between crude SEA and the chronic stage serum. When specific IgE or IgG antibody titers in the serum of human schistosomiasis mansoni cases were measured by ELISA using this purified allergen, the results showed good correlation with those obtained by using crude SEA. Thus, this purified allergen is not only a major allergen in the acute stage of murine schistosomiasis but also an allergen in human schistosomiasis mansoni.

A case of human B cell leukemia that implicates an autocrine mechanism in the abnormal growth of Leu 1 B cells.

Production of B cell growth factor (BCGF) from B-chronic lymphocytic leukemia (B-CLL) cells was demonstrated. Freshly isolated monoclonal B-CLL cells expressed surface mu, delta, B1, and Leu 1, but not Ba (an antigen expressed only on activated B cells). Upon stimulation with anti-IgM, they secreted BCGF, which could act on anti-IgM-stimulated autologous leukemic cells as well as anti-IgM-stimulated normal B cells. Cell lines established from these leukemic cells also constitutively secreted BCGF. The BCGF from B-CLL cells or established cell lines induced neither proliferation nor enhanced HLA-DR expression in resting B cells. These results show the presence of B cell-derived BCGF, which is distinct from BSF-1 and effective only on activated B cells. They also suggest that an autocrine mechanism may operate in the growth of B-CLL cells.

Detection of high molecular weight eosinophil chemotactic factor in murine schistosomiasis sera.

Eosinophil chemotactic activity in sera from mice undergoing an acute stage of schistosomiasis japonica and mansoni was examined. Eosinophilotactic activity in the serum was dependent on the dose and time of infection. Eosinophilotactic activity in sera from S. japonicum-infected mice was higher than that from S. mansoni-infected mice when they were compared at the comparable dose and time of infection. After gel chromatography on Sephadex G-200, eosinophilotactic activity in sera from mice infected with 30 cercariae of S. japonicum for 5 weeks was detected in the high molecular weight component. On the other hand, when sera from mice infected with 30 cercariae of S. japonicum for 8 weeks was chromatographed through Sephadex G-200 columns, eosinophilotactic activity was segregated into high (greater than 455,000) and low (less than 13,000) molecular weight components. High molecular weight ECF in sera from mice infected with 30 cercariae of S. japonicum for 8 weeks had high affinity to Con A, and was stable to heating or pronase digestion, but was sensitive to periodate oxidation, indicating its polysaccharide or glycoprotein nature. This high molecular weight ECF could be adsorbed by, and eluted from immunoaffinity beads coated with rabbit IgG anti-S. japonicum adult worm antibody. Thus, at least some part of circulating high molecular weight ECF would be derived from adult parasites.

Purification and characterization of a neutrophil chemotactic factor from Dirofilaria immitis.

A neutrophil chemotactic factor (NCF-Di) was purified from a crude extract of Dirofilaria immitis adult worm by a combination of anion-exchange chromatography on DE52 and gel filtration on Sephacryl S-200. NCF-Di showed a single protein band by both polyacrylamide gel electrophoresis (PAGE) and sodium dodecyl sulfate (SDS) PAGE. The molecular weight of NCF-Di was estimated to be 17,000 by gel filtration on Sephadex G-150, and 14,000 by SDS-PAGE. NCF-Di was an acidic protein with isoelectric point of 4.5. NCF-Di was absorbed neither to lentil lectin-Sepharose nor to concanavalin A-Sepharose. The chemotactic activity of NCF-Di was heat labile (56 C, 1 hr), but was resistant to periodate oxidation. These results suggest that NCF-Di is a simple peptide which has few or no sugar chains. These physicochemical properties of NCF-Di were compared to previously reported parasite-derived chemoattractants or purified allergen of D. immitis.

Identical expression of cell surface membrane antigens on two parent and eighteen cloned cell lines derived from two different neuroblastoma metastases of the same patient.

Two neuroblastoma cell lines established from tumor tissue taken from one individual are described. The first of these was established from a bone marrow aspirate (RT-BM) and the other from a right axillary lymph node (RT-LN) of a 1-yr 2-mo-old patient with Stage IV disease. The original lines were cloned in soft agar to yield six clones of the bone marrow-derived line (RT-BM 1-6) and 12 of the lymph node line (RT-LN 1-12). Chromosomal analysis of the original lines and clones showed they all have either identical or very similar karyotypes, with a deletion of chromosome 1p. Transmission electron microscopy indicates all contain neurosecretory (dense core) granules and neurotubules. In addition catecholamine metabolites of dopamine and noradrenaline have been identified. Different growth characteristics of the lymph node and bone marrow lines have been identified. RT-LN lines grow in a single cell layer with neurite processes, whereas bone marrow-derived lines form focal aggregates with neurite processes. In addition the colony-plating efficiency of the lymph node-derived lines is higher than those derived from bone marrow. Comparison of the cell surface antigen profile of the original tumor tissue, parent lines, and clones demonstrates they all bind seven of a panel of nine monoclonal antibodies. The expression of these antigens has remained stable in vitro for 25 passages undertaken over a 2-yr period. The definition of antigens that are expressed on the membranes of neuroblastoma cells in a stable form can aid in the differential diagnosis of neuroblastoma from other "small round cell tumors of childhood" and hopefully contribute to a greater understanding of the biology of this highly malignant tumor.

[Autosomal translocation and associated male infertility].

Four balanced autosomal reciprocal translocations were found through mitotic chromosome analysis among 72 subfertile males, 27 with azoospermia and 45 with sperm counts below 40 X 10(6)/ml. They were 46, XY, t(3; 20; 21) with azoospermia, 46, XY, t(14; 21) with sperm counts below 1 X 10(6)/ml, 46, XY, t(1; 19) lqh+ with azoospermia and 46, XY, t(3; 16) with sperm counts 27 X 10(6)/ml. Histological, cytogenetic and hormonal analysis were performed. Testicular biopsies from the first 3 carriers revealed complete spermatogenic arrest at the spermatocyte stage and meiotic studies of the same biopsies showed severe reduction in numbers of cells in 2nd meiotic division. In spite of severely defective spermatogenesis, serum gonadotropins of the carriers were within normal range, except for LH of the 4th case. Other chromosomal aberrations observed were 5 Klinefelter's syndrome, 2 autosomal minor variants (46, XY, 15p+ and 46, XY, 14s+) and 1 small Y.

Partial purification and characterization of eosinophil chemotactic factors from soluble extract of Fasciola species.

Eosinophil chemotactic factor (ECF) was partially purified from common liver flukes (Japanese strain of Fasciola sp) by a combination of anion-exchange chromatography on DE52 and gel filtration on Sephacryl S-200. The molecular weight of ECF was estimated to be approximately 27,000 by high-pressure liquid chromatography. The ECF was heat labile (56 and 100 C for 30 minutes) and was not bound with lentil-lectin Sepharose. The results of isoelectric focusing showed that ECF comprises at least 2 components; one was a major ECF with an isoelectric point of 3.1, and the other, a minor ECF with an isoelectric point of 3.8 to 4.5. These results indicate that ECF of common liver flukes are acidic proteins.

Effect of intraluminal bile on the feedback regulatory mechanism of pancreatic enzyme secretion in conscious rats.

The effect of intraluminal bile on the well-known feedback regulatory mechanism of exocrine pancreatic secretion exerted by intraluminal trypsin was investigated in conscious rats with pancreatic, biliary and duodenal fistulae. The stimulated pancreatic enzyme secretion caused by diversion of bile-pancreatic juice from the intestine was apparently suppressed by intraduodenal reintroduction of pancreatic juice or bile-pancreatic juice, while it was slightly suppressed by intraduodenal reintroduction of bile. Although additional reintroduction of bile did not alter the already suppressed pancreatic enzyme secretion by the presence of pancreatic juice in the intestine, diversion of bile stimulated the suppressed pancreatic enzyme secretion by intraluminal bile-pancreatic juice. Infusion of sodium taurocholate into the duodenum with diversion of bile-pancreatic juice effectively inhibited pancreatic enzyme secretion. The inhibitory effect seemed to be dependent on the concentration of taurocholate infused into the duodenum. The results suggest that bile and bile acid have an important role in the feedback regulatory mechanism of pancreatic enzyme secretion, at least partly directly inhibiting the secretion.

Schistosoma japonicum: identification and characterization of neutrophil chemotactic factors from egg antigen.

Two neutrophil chemotactic factors were identified in soluble egg antigen preparations of Schistosoma japonicum. The higher-molecular-weight neutrophil chemotactic factor was not separable from eosinophil chemotactic factor by means of gel filtration, anion-exchange chromatography, isoelectric focusing, or affinity chromatography; this neutrophil chemotactic factor is apparently identical to the higher-molecular-weight eosinophil chemotactic factor which we purified previously from the soluble egg antigen. The chemotactic activity of the eosinophil chemotactic factor for neutrophils was stable to periodate oxidation but was notably affected by heating or Pronase digestion, suggesting that the determinant for neutrophil chemotaxis exists on the peptide moiety of the eosinophil chemotactic factor. The lower-molecular-weight neutrophil chemotactic factor was separable from the higher-molecular-weight eosinophil chemotactic factor by gel filtration or anion-exchange chromatography. This neutrophil chemotactic factor was rather hydrophobic and heat-stable, but was sensitive to Pronase or carboxypeptidase A digestion. These results suggest that the receptors on the surfaces of neutrophils and eosinophils for those chemoattractants would be different from each other. We suppose that neutrophil chemotactic factors and eosinophil chemotactic factors from the eggs are responsible for neutrophil and eosinophil accumulation around the eggs in schistosomiasis japonica.

Airflow, volume, and durational characteristics of oral reading by the hearing-impaired.

This study examined airflow, volume, and durational characteristics of speech produced by hearing-impaired subjects using a face-mask-pneumotachometer-pressure-transducer system. Results or oral reading of a standard passage indicated that, in comparison to the normal-hearing subjects, the speech respiration of the hearing-impaired was characterized by high air consumption manifested as high air expenditure per syllable, high average expiratory airflow rates, and high peak-airflow rates; frequent inspirations; inspirations at linguistically inappropriate places; short duration of expiration; and large individual differences. These characteristics were especially prominent among the more severely hearing-impaired individuals. Correlational analysis revealed that more frequent inspirations were associated with poorer overall speech proficiency while high air consumption was closely related to a breathy voice quality.

Isolation of Schistosoma japonicum egg-derived neutrophil stimulating factor: its role on eosinophil chemotactic factor release from neutrophils.

Neutrophil stimulating factor (NSF), which can stimulate polymorphonuclear neutrophils (PMNs) to release eosinophil chemotactic factor (ECF), was isolated from Schistosoma japonicum soluble egg extract (SEA). The release of ECF from PMNs began as early as 5 min after stimulation and reached a peak at 40 min, and was dependent on the concentration of SEA. After Sephadex G150 gel filtration, toluene extraction, Sephadex G25 gel filtration and Dowex 1 X 8 anion exchange chromatography, NSF was identified as a hydrophilic and negatively charged component with a molecular weight of about 1,000 daltons. It was heat-stable at 100 degrees C for 60 min. NSF was easily separable from SEA-derived neutrophil chemotactic factor or from SEA-derived ECF. PMNs are suggested to be one of the sources of ECF in the eosinophil accumulation of granulomatous lesions around the deposited eggs in schistosomiasis japonica.

In vitro and in vivo induction of neutrophil and eosinophil chemotactic responses by Schistosoma japonicum cercaria.

High neutrophil and eosinophil chemotactic activities of soluble extract of Schistosoma japonicum cercariae (SjCe-ext) were detected in vitro by using blind-well chemotaxis chambers and Millipore filters in a dose-dependent manner. Low doses (total of 0.25 and 2.5 micrograms) of SjCe-ext induced significant neutrophilic infiltrations in normal and S. japonicum-infected (8 week) guinea pig tissues beginning at 2 hr after the intradermal injection. Eosinophilic responses were observed in the infected animals especially in early phase (2-8 hr post-injection of SjCe-ext). SjCe-ext elicited strong immediate skin reactions (at 30 min) in the infected guinea pigs. Possible roles of these chemotactic activities and hypersensitivity reaction in S. japonicum infection are discussed.

Neutrophilic nodules in the intestinal walls of Japanese monkeys associated with the neutrophil chemotactic activity of larval extracts and secretions of Oesophagostomum aculeatum.

High neutrophil chemotactic activity was detected in the culture medium from Oesophagostomum aculeatum larvae in vitro using blind-well chambers with Millipore filters, and guinea pig leucocytes as indicator cells. Neutrophil chemotactic activity was also detected in the extract from larval worms in a dose dependent fashion. This activity was detected in the low molecular weight fractions adjacent to a sodium chloride marker by gel filtration on Sephadex G200. These results were further confirmed with monkey neutrophils. The possible role of this activity in the formation of granulomatous lesions rich in neutrophils found in O aculeatum infections in the Japanese monkey is discussed.

[Corticosteroid as an anti-emetic agent in cancer chemotherapy with cis-diammine-dichloroplatinum (II)].

The anti-emetic effect of corticosteroid during cancer chemotherapy with cis-diamminedichloroplatinum (II) (CDDP) is analyzed retrospectively. From November 1981 to April 1984, 23 of 50 patients receiving CDDP were given corticosteroid as an anti-emetic. Corticosteroids, methylprednisolone (80-500 mg/day) or hydrocortisone (200-480 mg/day), were administered intravenously for 1 to 3 doses. Including 4 patients who had no symptoms, 60% of patients were free from nausea and vomiting. The incidence of gastrointestinal symptoms was significantly lower than that in patients who were not given corticosteroid. Corticosteroid reduced gastrointestinal symptoms in 2 of 3 patients who had prior CDDP treatment. There were no remarkable adverse reactions except acne in 2 patients. A further controlled study is needed to determine the appropriate dose and modality of administration of corticosteroid.
