Leukemic lymphosarcoma (LLS) with monoclonal IgM: idiotypic specificity on the cell surface and in the cytoplasm of lymphosarcoma cells.

A case of leukemic lymphosarcoma with a small amount of monoclonal IgM in the serum is described. Immunologic investigations showed that about 10% of the abnormal cells formed EAC rosettes, that most of these cells had IgMlambda on the cell surface, and that 40% of them contained IgMlambda in the cytoplasm. Monoclonal IgM was detected in the 19S fraction of the serum despite the lack of monoclonal change as determined by serum electrophoresis or immunoelectrophoresis. The common idiotypic specificity was found in the monoclonal serum IgM, surface Ig, and cytoplasmic Ig of lymphosarcoma cells using antiidiotypic antiserum prepared against purified serum IgM. After chemotherapy, cells with idiotypic determinants disappeared and the serum IgM changed from a monoclonal to a polyclonal distribution.

Mutation of an inosine-producing strain of Bacillus subtilis to DL-methionine sulfoxide resistance for guanosine production.

An inosine-producing strain of Bacillus subtilis was mutated to resistance against the antagonist of glutamine, DL-methionine sulfoxide. Among the mutants derived, guanosine producers were observed frequently. The best strain, 14119, produced 9.6 g of guanosine per liter at a weight yield of 12% from consumed sugar. Inosine production decreased concomitantly. When resistance was increased further by exposure to higher doses of DL-methionine sulfoxide, another strain, AG169, was obtained that did not excrete inosine but produced increased amounts of xanthosine. In these strains, the specific activity of 5'-nucleotidase was lower and that of inosine 5'-monophosphate (IMP) dehydrogenase was higher than the parent strain. It is speculated that the metabolic flow from IMP to xanthosine 5'-monophosphate proceeds more smoothly than that from IMP to inosine and yields more xanthosine and guanosine.

Effects of radiation after enucleation without implantation on orbital development of patients with retinoblastoma.

Orbital growth of the patients treated with enucleation and radiation in infancy for retinoblastoma was studied by roentgenographic measurements. The patients radiated following enucleation without implantation were found to be many in the present series, and the effects of radiation and the insertion of an implant to the development of the orbit in infancy were discussed. Judging from the results of statistical analysis of roentgenographic measurements of the orbit, growth retardation of the orbit radiated after enucleation was greater than that of the orbit only enucleated. Radiation therapy after enucleation for intraocular malignant tumor in infancy should be avoided as much as possible unless the extrascleral invasion is found at operation.

LH- and FSH-releasing activities of synthetic tetrapeptide and synthetic LH-RH in rats.

Changes in serum LH and FSH were measured following injection of synthetic tetrapeptide (Glu-Tyr-Arg-Trp-NH2) or synthetic LH-RH into the femoral vein of oophorectomized rats. A considerable secretion of LH occurred after injection of synthetic tetrapeptide although its LH-releasing activity was less prominent than that observed after injection of LH-RH. No appreciable FSH-releasing activity was noted following injection of the tetrapeptide. A significant rise of LH and an observable, but extremely modest, rise of FSH with an indistinct peak were noted after single injection of synthetic LH-RH.
