Localisation of ionotropic glutamate receptor subunits, and gamma-aminobutyric acid (GABA) in the monkey amygdala--a double immunolabelling and electron microscopic study.

The distribution of ionotropic glutamate receptor subunits GluR1 and NMDAR1, and the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) was studied by immunocytochemistry, in the monkey amygdala. The basal and lateral nuclei contained a higher density of GluR1-positive neurons than the corticomedial and central groups of nuclei, and the accessory basal nucleus. A higher density of NMDAR1 immunopositive cell bodies was also present in the lateral nucleus, compared to the other nuclei of the amygdala. Large multipolar or fusiform projection neurons, and not small local circuit neurons were GluR1 or NMDAR1-positive, and less than 0.5% of the GluR1-positive cells were double labelled for GABA. In contrast, almost all GluR1-positive neurons were also labelled for NMDAR1 in double labelled sections and vice versa. Electron microscopy also showed that GluR1- and NMDAR1-positive cells had distinctive ultrastructural features, compared with GABAergic cells, ant that there was very rare colocalisation, between GABA, and GluR1 or NMDAR1-positive cell bodies or dendrites. It is likely that not all projection neurons were GluR1 or NMDAR1-positive, however, since GluR1 or NMDAR1-positive neurons were only 2-3 times as common as GABAergic cells, whereas it has been estimated that projection neurons outnumber GABAergic local circuit neurons by 4 to 1 (McDONALD and AUGUSTINE, 1993; PITKANEN and AMARAL, 1994).

[The effect of gamma-interferon on hepatic fibrosis in schistosomiasis japonica].

OBJECTIVE: To study the effect of gamma-IFN on hepatic fibrosis in schistosomiasis japonica. METHODS: The amount and distribution of gamma-IFN and extracellular matrix, including fibronectin (FN), laminin (LN), I and II collagen which were used to judge the suppressing effect of gamma-IFN on hepatic fibrosis as markers of dynamics of extracellular matrix after administration of recombinant gamma-IFN, in liver obtained at different stages from S. japonicum-infected mice were determined by immunohistochemical streptavidin biotin-peroxidase complex method. RESULTS: The amount of gamma-IFN in liver reached a peak (3 mice respectively reached 2, 3 and 4 grade) at the 16th week after infection, which was higher than the levels of control groups and infected-mice at the 8-12th week (chi 2 = 15.39, 7.50, P < 0.01), and the majority of gamma-IFN was distributed round egg granuloma. FN and LN, and I and II collagen increased to 1 grade, individual 2 grade, at the 8th week after infection, which were higher than control (chi 2 = 12.44-15.39, P < 0.01), and were lineally distributed round egg granuloma. With chronicity and decrease of gamma-IFN, however, FN, LN and I, II collagen gradually increased, and reached a peak (over 70% infected mice reached 3-4 grade) respectively at the 20th week and 24th week, became wide, thick and retiform and deposited round and in egg granuloma. After administration of gamma-IFN, only 3 infected mice had 2 grade of FN at the 20th week, two mice had respectively 3 grade of type I and II collagen at the 24th week, and none of them reached 4 grade, which were significantly less than those in the untreated group at the same stage (chi 2 = 10.77, 5.05, 7.20, P < 0.01-0.05). CONCLUSION: The results indicate that gamma-IFN may play an important role in opposing the inflammatory response of egg granuloma, decreasing secretion and deposition of extracellular matrix in liver and suppressing hepatic fibrosis.

[The use of hyoid osteomuscular flap in extended partial laryngectomy].

The surgical defect after conventional partial laryngectomy practically needs no special technique to repair. On conservation surgery for late (T3 and T4) cases a large part of the larynx is to be resected so that the margin of surgical clearance is ample and, at the same time, an appropriate method of reconstruction is mandatory to restore the essential functions of the larynx. For this purpose we developed a technique of osteomuscular flap to repair the defect left behind after a three-fourths laryngectomy (for supraglottic case) or an extended vertical partial laryngectomy (for glottic case). In both cases the arytenoid cartilage or the cricoarytenoid joint was resected to ensure the safety margin of resection. The mylohyoid muscle was severed at the upper border of the body of the hyoid bone. A small piece of the hyoid bone (about 5 mm) and its attached strap muscles were used. The muscles were made a 90 degree bent toward the laryngeal cavity. The hyoid bone was used to substitute the removed arytenoid and the muscles to bridge over the laryngeal lumen. The postericoid mucosa was advanced to cover the transplanted hyoid bone. Then the cut edge of the thyroid cartilage was lifted up and sutured with the base of the tongue to close the wound. 108 cases were treated within a period of 12 years (1979-1990). The three year survival rates for stage II and stage IV of supraglottic type (66 cases) were 78.6% and 66.7% respectively, and for stage II of glottic type (42 cases), 79.3%. The decannulation rate was 75%-85%, and 80% of the patients enjoyed a socially acceptable voice. All cases resumed normal mouth-food-taking.

[The effect of interferon in combination with ribavirin on the plus and minus strands of hepatitis C virus RNA in patients with hepatitis].

The effect of interferon in combination with ribavirin on the plus and minus strands of hepatitis C virus (HCV) RNA in patients with chronic hepatitis C (CHC) was studied by means of nested RT-PCR. The results showed that in those who respond to the combination of antiviral therapy, their increased serum ALT levels decreased to normal range, but more than half (55.56%) of these patients relapsed 24 weeks after cessation of the antiviral therapy. The positive rate of the plus strand HCV RNA in serum (92.31%) decreased significantly to 38.46% (P < 0.005) and that of the minus strands HCV RNA in peripheral blood mononuclear cells (PBMC, 76.92%) to 38.46% (P < 0.05) at the end of the treatment, but little effect on the plus strand HCV RNA in PBMC in these patients was found. Relapse occurred in patients whose plus and minus strands of HCV RNA in PBMC remained positive during treatment. These data indicated that absence of HCV RNA in serum do not mean complete clearance of HCV and can not predict a sustained therapeutic response. For evaluating the antiviral effect and prognosis it is essential to measure the plus and minus strands of HCV RNA in serum and PBMC simultaneously. The results of the combined antiviral therapy were similar to that of single interferon treatment; it seems that rib-a-virin do not enhance the antiviral effect of interferon.

[The effect of phospholipid liposomes on atherosclerosis and serum lipid in rabbits].

In order to study the effect of oral phospholipid liposomes on regression of atherosclerosis process, serum total cholesterol triglyceride, lipoprotein cholesterol and atherosclerotic lesion on the aorta wall in different groups of 20 New Zealand rabbits were observed. Results showed that phospholipid liposomes could obviously increase high density lipoprorein cholesterol concentration and decrease serum total cholesterol triglyceride and very low density lipoprorein cholesterol concentration. Phospholipid liposomes could also reduce the size of atherosclerotic lesion on the aortic wall. In comparision with the results observed in phospholipid group, there was significant difference (P < 0.05, P < 0.01 or P < 0.001). It suggested that oral phospholipid liposomes could effectively regress atherosclerotic lesion on the arterial wall and regulate serum lipid to reduce cholesterol deposit on the arterial wall in experimental rabbits. It might provide some experimental basis for human being to prevent and treat atherosclerosis.

[Value of measuring resistance of airway for diagnosis of asthma].

Resistance of airway (Raw), sGaw and Maximal expiratory flowvolume curve (MEFV) were measured in 35 normal subject, 43 patients with remission asthma and 100 patients with remission chronic bronchitis (some complicated by obstructive emphysema). The measured values showed that in simple asthma Raw was significantly higher than that in normal subject and in patients with chronic obstructive bronchitis (COB), but MEFV was in normal range. In chronic bronchitis the value of Raw was higher than that in normal subject, but lower than that in asthma (0.05 > P > 0.02); however the FEV1, MMEF, V75 in the curve of MEFV were significantly decreased at the stage of COB, with the most remarkable decrease in MMEF. The results suggest that Raw is a sentitive test for diagnosis of simple asthma, in addition to MEFV measurement; it may be helpful for differential diagnosis of simple asthma and COB and may has potential value for clinical practice.

[A study on the course of corneal epithelial healing in diabetic rabbits].

OBJECTIVE: In order to approach the course of corneal epithelial healing of diabetes mellitus. METHODS: The experimental rabbit diabetes models were produced by the injection of alloxan. The corneal epithelium of each rabbit was removed by freezing with a 6 mm diameter brass probe cooled in liquid nitrogen. The rate of corneal epithelial healing following freezing in the normal rabbit was compared to that in the diabetic one. Contrasted with the normal rabbits, the ultrastructural changes of the epithelium with and without freezing were also observed in the diabetic rabbits. RESULTS: It is shown that there is no statistical significant difference in the overall rate of epithelial healing between the normal and diabetic rabbit. In contrast with the normal rabbit, the epithelial basement membrane of the diabetic rabbit without freezing revealed markedly thickened, and the mitochondria and glycogen particles in the epithelial cell increased. In the diabetic rabbit with freezing, the epithelial basement membrane appeared absent or discontinuous. After complete healing, the corneal epithelium was edematous in diabetic rabbit. CONCLUSION: The abnormal basemenet membrane and edematous epithelial cell were responsible for the diabetic corneal epithelial poor healing and recurrent erosion.

[Comparison of various types of imidazole derivatives for treatment of filamentous fungal keratitis].

OBJECTIVE: 73 eyes of 73 patients with filamentous fungal keratitis were treated by various types of imidazole derivatives. METHODS: The mean patient age was 36 (range, 15-62) years. The areas of corneal ulcer were 2-4 mm x 2-5 mm. The hypopyon was 1-3 mm in height. The patients were randomly divided into 4 groups: I 23 eyes were treated by 1% miconazole suspension. II 15 eyes by imidazole mixed oil (i.e. econazole and miconazole in castor oil). III 19 eyes by dusting mixed imidazole powder onto focal corneal lesions. IV 16 eyes by focal corneal lesion resection combined with dusting mixed imidazole powder. RESULTS: The cure rates were 21.74%, 66.67%, 73.68% and 93.75%, and the average days of treatment were 65, 68, 35 and 19, respectively, in the 4 groups. The differences in the cure rates and in the means of treatment durations between each of the former three groups and the 4th group are remarkable (P <0.01). CONCLUSION: Focal corneal lesion resection combined with dusting mixed imidazole powder is relatively effective in treatment of filamentous fungal keratitis.

[Reasonability of extracting procedures for huiyangfanben decoction].

The reasonability of extracting procedures for traditional Chinese prescriptions was studied with Huiyangfanben Decoction as sample. The comparative results of the ingredients in extractions prepared by different procedures revealed. For the prescriptions with complicated ingredients, more attention should be paid to the ingredients and their characteristics when designing the extracting procedurs, and in extraction the drugs should be classified according to the interaction of the ingredients. More extracting solvents could be used according to the characteristics of the active ingredients.

[Preparation of hepatic targeting antivirus agent NGA-ACV and its targeting property].

Neoglycoalbumin (NGA), a special ligend of asialoglycoprotein receptor on the hepatocyte, was linked via a butanediacyl bridge to acyclovir to form a conjugate NGA-ACV. By using DTA (Differential thermoanalysis) and HPLC analysis, ACV was shown to be connected with NGA by covalent bonds and stable in blood. The radio-biodistribution of 131I-NGA-ACV with high drug density in vivo was carried out in mice. The maximum absorption of 131I-NGA-ACV in liver was 81.7 +/- 10.4% at 5 min. The radioimage of 131I-NGA-ACV with high or low drug density in rabbit showed no significant difference in liver targeting property. The competitive connection tests indicated that 131I-NGA-ACV was concentrated in liver through receptor mediated mechanism. A tentative test of antihepatitis B of NGA-ACV and ACV in vitro showed that the effective dose of the former was significantly lower than that of the latter.

The mouse obese gene. Genomic organization, promoter activity, and activation by CCAAT/enhancer-binding protein alpha.

The obese gene product, leptin, regulates adiposity. Mice homozygous for a nonfunctional obese gene become massively obese and develop diabetes mellitus due to overeating and increased metabolic efficiency. The cDNA sequence of obese was recently reported (Zhang, Y., Proenca, R., Maffei, M., Barone, M., Leopold, L., and Friedman, J. L. (1994) Nature 372, 425-432; Correction: (1995 Nature 374, 479). We have determined the genomic organization of the 5' end of the mouse obese gene. The coding sequence is in exons 2 and 3. A single TATA-containing promoter was found upstream of exon 1. A minority (probably approximately 5%) of the obese mRNA contained an extra, untranslated exon between exons 1 and 2. Transcription of the obese gene was detected only in adipose cells. A 762-base pair obese gene promoter driving a luciferase gene yielded abundant activity in transiently transfected rat adipose cells in primary culture. The obese promoter was inactive in erythroid K562 cells. Deletion of bases from -762 downstream to -161 did not affect promoter activity in transfected adipose cells. The -161 minimal promoter contained consensus Sp1 and CCAAT/enhancer-binding protein (C/EBP) motifs. Cotransfection with C/EBP alpha (a transcription factor important in adipose cell differentiation) caused 23-fold activation. These data suggest that the obese promoter is a natural target of C/EBP alpha.

Alloantigens introduced into the anterior chamber of the eye induce systemic suppression of delayed hypersensitivity to third-party alloantigens through "linked recognition".

It has been recognized for over a century that the anterior chamber of the eye is endowed with unique immunological properties that permit the long-term survival of histoincompatible grafts that would otherwise be rejected at extraocular sites. Immune privilege in the anterior chamber of the eye has been attributed, at least in part, to the active down regulation of systemic delayed-type hypersensitivity (DTH) that is induced when antigens are introduced into this ocular compartment. The antigen-specific suppression of systemic DTH that is induced by anterior chamber priming has been termed anterior chamber-associated immune deviation (ACAID) and has been demonstrated with a variety of antigens. The present report summarizes a systematic evaluation of various categories of histocompatibility antigens for their capacity to induce ACAID. The results indicate that ACAID can be induced against a wide variety of alloantigens ranging from a single minor histocompatibility antigen (H-Y) to mismatches involving the entire major histocompatibility complex plus multiple minor histocompatibility loci. Further investigation revealed that it was possible to induce suppression of DTH to third-party alloantigens providing the alloantigens introduced into the anterior chamber were coexpressed with the third-party alloantigens on cells used for extraocular immunizations.

Type I, II and III inositol 1,4,5-trisphosphate receptor co-immunoprecipitation as evidence for the existence of heterotetrameric receptor complexes.

The recently described co-expression of type I, II and III inositol 1,4,5-trisphosphate (InsP3) receptors in the same cell type has raised the issue of whether these proteins exist as homotetramers or heterotetramers. To address this question, InsP3 receptors were immunoprecipitated with specific antibodies and then probed for co-immunoprecipitating proteins. This revealed that type I, II and III InsP3 receptors co-immunoprecipitate and thus, exist in heteromeric complexes. This situation was maintained when the relative abundance of InsP3 receptors was altered radically during cell differentiation. Thus, heterotetrameric InsP3 receptors are likely to contribute towards signaling in cells expressing more than one receptor type.

ApoE immunoreactivity and microglial cells in Alzheimer's disease brain.

The spatial relationship of apolipoprotein E (apoE)-like immunoreactivity (IR) to amyloid beta-peptide (A beta), astrocytes and microglial cells in the brain of Alzheimer's disease was studied by double immunolabelling. Diffuse apoE-like IR was seen in A beta diffuse deposits, and markedly increased in the core of classic senile plaques. Microglial cells, sometimes immunoreactive for apoE, were frequent in areas of apoE-like IR, where they often grouped into clusters in the core of apoE-labelled senile plaques. Although astrocytic processes were seen within these senile plaques, the cell bodies were always at a distance from the core. None of these astrocytes expressed apoE-like IR. Microglial cells, some of them immunoreactive for apoE, were seen in the center of apoE-labelled senile plaques. These data suggest that microglial cells play a more significant role than astrocytes in apoE deposition in senile plaques of Alzheimer disease.

Inhibition of the growth of various human and mouse tumor cells by 1,15-bis(ethylamino)-4,8,12-triazapentadecane.

Effects of 1,15-bis(ethylamino)-4,8,12-triazapentadecane (BE3333), the least toxic bis(ethyl)pentaamine, on the growth of tumor cells were studied in in vitro systems and with tumor xenografts in mice. BE3333 suppressed ornithine decarboxylase and S-adenosylmethionine decarboxylase, induced spermidine/spermine N1-acetyltransferase, and thus decreased the amount of polyamines. BE3333 accumulated in cells at a concentration 3-5-fold that of spermine in control cells through the polyamine transport system. The accumulated BE3333 inhibited protein synthesis, especially mitochondrial protein synthesis, and decreased the amount of ATP. The inhibition of protein synthesis was correlated with the subsequent inhibition of cell growth. BE3333 showed inhibitory effects in in vitro systems against the growth of mouse FM3A mammary carcinoma cells, human SW480 and SW620 colon tumor cells, Lu-65A and A549 lung tumor cells, MCF-7 breast tumor cells, and MALME-3M and A375 melanoma cells at a range of 0.5-10 microM. Intravenous (30 mg/kg) or i.p. (50 mg/kg) daily injections of BE3333 for 5 or 7 days greatly suppressed the growth of human colon tumor SW620 xenotransplanted into nude mice. Similar antitumor activity was obtained with continuous infusion of BE3333 into the peritoneal cavity (80 mg/kg), but not with p.o. administration (200 mg/kg). BE3333 also showed inhibitory effects against the growth of lung tumors (Lu-65, Lx-1, Lc-1, and Lu-61), stomach tumors (Sc-6 and St-15), and melanoma (SEKI) xenotransplanted into nude mice. The results indicate that BE3333 is effective against both rapid- and slow-growing tumors, with reasonable short-term host toxicity.

A quantitative model of post-laminectomy scar formation. Effects of a nonsteroidal anti-inflammatory drug.

STUDY DESIGN: A quantitative model of peridural post-laminectomy fibrosis in rats was designed in this study. Concurrently, the effects of a nonsteroidal anti-inflammatory drug (ketoprofen) were evaluated. OBJECTIVES: To quantify the amount of fibrotic tissue, the extent of adhesion to the dura mater, and the cell nature and density for drug trials requiring large series of animals. SUMMARY OF BACKGROUND DATA: Most of the previous analyses of experimental post-laminectomy fibrosis were qualitative in nature. Only one quantitative analysis was reported in the rabbit, and the rat has seldom been used. The effects of nonsteroidal anti-inflammatory drugs on post-laminectomy scar formation have never been evaluated. METHODS: L5 laminectomies were performed in 32 rats. The treated group (16 rats) received a systemic injection of ketoprofen (5 mg/kg, once daily from the day before the operation to the seventh postoperative day), the other 16 rats constituted a control group. The post-laminectomy scar formation was evaluated on postoperative days 8, 15, 30, and 90 using a semiautomatic image analysis system. RESULTS: The course of post-laminectomy scar formation was similar to that described in larger animals. There was a good correlation among the measurements of fibrous tissue area obtained by 2 independent observers. The mean amount of peridural scar tissue was significantly smaller in the treated group than in the control group. The extent of adherence to the dura mater and the density of fibroblasts and fibrocytes was not different between the two groups. The density of inflammatory cells was significantly less in the treated group than in the control group only at day 8. CONCLUSION: Quantitative evaluation of post-laminectomy fibrosis can easily be performed in rats and is reproducible. This model could allow trials with drugs administered locally or systemically as preventive treatment of post-laminectomy scar formation. Nonsteroidal anti-inflammatory drugs could decrease the amount of fibrotic tissue.
