Transcriptional regulation of c-Fes in myeloid leukemia cells.

The c-Fes proto-oncogene encodes a myeloid-specific protein-tyrosine kinase that is expressed preferentially in differentiated myeloid cells, but not in early myeloblast progenitor cells. To examine the basis for the phenotypic expression of c-Fes, the transcription initiation sites of the human c-Fes gene were mapped in myeloid leukemia cells and regulatory elements in the genomic c-Fes sequence were characterized. Two major transcription initiation sites were found in the myeloid leukemia cell line THP-1 which delineated exon 1 to be 72-83 bp. When the activity of the CAT reporter gene under the control of the c-Fes promoter region, untranslated exon 1 and intron 1 was measured in TF-1, K562 and MCF-7 cells, only TF-1 cells exhibited chloramphenicol acetyltransferase activity. In contrast, all cell lines supported reporter gene activity when intron 1 was deleted. Deletion analyses revealed a negative regulatory region in intron 1, which was localized by Southwestern analysis and DNA footprinting to a 14 bp region. This negative regulatory region suppressed reporter CAT activity in K562 and TF-1 cells when inserted downstream to the SV40 early promoter. These results suggest that the tissue-specific expression of c-Fes may result, in part, from the negative regulation of transcription in myeloid and nonmyeloid cells.

Effects of oxidized low density lipoprotein on endothelin secretion by cultured endothelial cells and macrophages.

The aim of this study was to determine how oxidized LDL affects endothelin secretion by endothelial cells, monocytes and macrophages. It was found that different degrees of oxidation of LDL had different effects on endothelin production. Extensively oxidized LDL inhibited endothelin secretion from cultured endothelial cells. It also attenuated endothelin secretion from phorbol ester-activated macrophages. The inhibitory effect on endothelin secretion required a substantial degree of LDL oxidation as reflected by an increase in absorbance at 234 nm (conjugated diene) of 0.7 AU with 125 nM LDL and a two- to three-fold increase in migration distance on electrophoresis. Oxidized LDL inhibited thymidine incorporation in porcine aortic endothelial cells, hence in these cells cytotoxicity may account for at least part of the inhibition of endothelin secretion. Acetyl LDL slightly increased basal endothelin release by endothelial cells, but native LDL or mildly oxidized LDL had no significant effect. Overall, the present findings argue against a stimulatory effect of oxidized LDL on endothelin release as contributing to increased vasoreactivity in atherosclerosis. In fact, the apparent inhibition of endothelin release by extensively oxidized LDL might tend to attenuate vasoreactivity near atherosclerotic lesions.

Characterization of a CO-responsive transcriptional activator from Rhodospirillum rubrum.

In Rhodospirillum rubrum, CO induces the expression of at least two transcripts that encode an enzyme system for CO oxidation. This regulon is positively regulated by CooA, which is a member of the cAMP receptor protein family of transcriptional regulators. The transcriptional start site of one of the transcripts (cooFSCTJ) has been identified by primer extension. The ability of CooA to bind to this promoter in vitro was characterized with DNase I footprinting experiments using extracts of a CooA-overproducing strain. CooA- and CO-dependent protection was observed for a region with 2-fold symmetry (5'-TGTCA-N6-CGACA) that is highly similar to the consensus core motifs recognized by cAMP receptor protein/FNR family. In vivo analysis in a heterologous background indicates that CooA is sufficient for CO-dependent expression, implicating it as the likely CO sensor.

Distribution of glutamate receptor subunit GluR1 and GABA in human cerebral neocortex: a double immunolabelling and electron microscopic study.

Specimens of human cerebral neocortex were obtained during neurosurgical operations and studied by immunocytochemistry and electron microscopy, using antibodies to the glutamate receptor subunit GluR1 and gamma-aminobutyric acid (GABA). Many GluR1-positive pyramidal neurons and fewer GluR1-positive non-pyramidal neurons were present in the cortex. Non-pyramidal neurons were more heavily labelled for GluR1 than pyramidal neurons. Most GABAergic neurons were labelled for GluR1. The white matter was unstained, except for occasional labelled neurons. This pattern of GluR1 immunostaining is similar to that in rat cerebral cortex, but is different from that in the hippocampus and amygdala, where large numbers of pyramidal or projection neurons, but few non-pyramidal or GABAergic neurons, were labelled for GluR1.

The preliminary research on the relationship between TNF- and egg-induced granuloma and hepatic fibrosis of schistosomiasis japonica.

The amount and distribution of tumor necrosis factor-alpha (TNF-alpha) in liver at various stages of Schistosomiasis japonicum-infected mice were immunohistochemically determined by SABC method. The results showed that the amount of TNF-alpha in liver began to increase at the 8th-12th week after infection, and reached a peak (9 mice reached 2+ grade or higher) 16 weeks after infection, which was higher than the levels 8 weeks after infection (P = 0.001) and was mainly scattered in and around egg granuloma. At the chronic stage of the infection, the amount of TNF-alpha in liver decrease not increase with an increased secretion of collagen in liver and development of hepatic fibrosis. On the contrary, FN, LN, type I and III collagen in liver began to rise 8-12 weeks after infection, and were linearly scattered around egg granuloma, reaching a peak (more than 70% of infected mice reached 3+(-)4+ grade) at the 20th and 24th week and they became wide, thick and retiform, and deposited around and in egg granuloma. After administration of recombinant TNF-alpha to the infected mice, the amount of FN and LN, the size and the inflammatory response of granuloma in liver showed no significant changes as compared with the untreated group at the same stage. However, the amount of type I collagen in liver increased, among which the amount of type I collagen was significantly higher than that in the untreated group at the same stage (P = 0.01), while similar changes were not observed when recombinant TNF-alpha was administrated to the healthy mice. It is suggested that TNF may play a role in stimulation of fibroblast in liver to proliferate and secrete collagen, but may need to cooperate with some factors.

Body size accounts for most differences in bone density between Asian and Caucasian women. The EPIC (Early Postmenopausal Interventional Cohort) Study Group.

We compared bone mineral density (BMD) of the whole body (and subregions: arm, leg, and pelvis), hip, spine, lateral spine, wrist, and forearm among Caucasian and Asian women at four geographic centers (Honolulu, HI; Nottingham, UK; Portland, OR; Copenhagen, Denmark). Data were derived from the baseline examination of 1367 Caucasian and 162 Asian women enrolled in the 1609-subject Early Postmenopausal Interventional Cohort (EPIC) study. After adjusting for age, study site, years postmenopause, and years of estrogen use, BMD was approximately 4-6% lower (P < 0.05) among Asian women at most skeletal sites, but there was no significant difference for wrist or forearm BMD. Adding height, lean body mass, fat mass, and/or quadriceps muscle strength to the regression models reduced the racial differences at most skeletal sites; after these additional adjustments, Asian women had significantly lower BMD only for the lateral spine (-4.4%; P < 0.005), arm (-2.20%; P < 0.05) and leg (-1.65%; P < 0.05), whereas the wrist was significantly greater (4.64%; P < 0.005) for Asian women. Further research is needed to determine why racial differences in BMD persist at certain skeletal sites, but not others, after adjusting for body size.

Exogenous nucleotides alter the proliferation, differentiation and apoptosis of human small intestinal epithelium.

The turnover of intestinal epithelial cells is a finely regulated process extending from undifferentiated crypt stem cells to terminally differentiated villus cells. The final phase of this maturation process is apoptosis and extrusion. Recent studies have shown that programmed cell death (PCD) occurs not only in senescent cells and in rapidly developing tissues but also in response to cellular stress preventing damaged cells from entering uncontrolled proliferation without repair. This study examined the role of exogenous nucleotides on cell proliferation, differentiation and apoptosis in organ cultures of human fetal small intestine. The addition of adenosine monophosphate (AMP), a putative stress reactant, to the culture media resulted in the suppression of crypt cell proliferation followed by the restoration of differentiation and the induction of apoptosis across a broad range of villus epithelium when compared with controls without added nucleotide. In contrast, the addition to cytidine monophosphate (CMP) to the culture media did not increase apoptosis, despite the nucleotide being taken up by the fetal small intestine in a similar dose- and time-dependent manner to AMP. Furthermore, Bax mRNA, an apoptosis-inducer gene, was increased with addition of AMP, suggesting that the induction of apoptosis may be channeled through the p53 pathway. These results suggest that a specific exogenous nucleotide, AMP, may have an important role in controlling the dynamic balance of cellular turnover in the developing human small intestine.

Changes in average blood pressure and incidence of high blood pressure 1983-1984 to 1987-1988 in four population cohorts in the People's Republic of China. The PRC-USA Cardiovascular and Cardiopulmonary Epidemiology Research Group.

OBJECTIVE: To assess comparative changes in systolic and diastolic blood pressure and incidence of high blood pressure over 4 years, and factors related to these changes, in northern and southern, urban and rural adults in the People's Republic of China. SETTING: This is a prospective study of populations surveyed by standardized methods in 1983-1984 and 1987-1988 in north China, Beijing area steel mill workers and farmers, and south China, Guangzhou area shipyard workers and farmers. SUBJECTS: The subjects were 8805 men and women aged 35-54 years at baseline (1983-1984), of whom 7338 were non-hypertensive (systolic blood pressure < 140, diastolic blood pressure < 90 mmHg, not being administered antihypertensive drugs). MAIN OUTCOME MEASURES: The main outcome measures were 4-year changes in systolic and diastolic blood pressures and the 4-year incidence of high blood pressure in persons not hypertensive at baseline. RESULTS: At baseline, the average systolic and diastolic blood pressures were higher for Beijing than for Guangzhou cohorts. At resurvey the average systolic blood pressure had increased for seven of eight sex-city-setting cohorts (all but Guangzhou urban men). The Beijing urban cohorts had the greatest increases (men 6.2 mmHg, women 4.9 mmHg, slopes of 1.6 and 1.2 mmHg/year). The incidence of high blood pressure was higher for Beijing than for Guangzhou cohorts in all four comparisons (men and women, urban and rural). Beijing men, urban and rural, had the highest incidence rates (about 21%). In multiple logistic analyses by sex, variables significantly related to 4-year high blood pressure incidence were city, baseline systolic blood pressure and body mass index, change in weight, and, for men, baseline alcohol use and heart rate. CONCLUSION: Modifiable lifestyle traits, previously shown to be related to blood pressure and high blood pressure in cross-sectional studies, also relate to the incidence of hypertension.

Monoclonal antibodies for detection of the H7 antigen of Escherichia coli.

Two murine monoclonal antibodies (MAbs) (2B7 and 46E9-9) reactive with the H7 flagellar antigen of Escherichia coli were produced and characterized. A total of 217 E. coli strains (48 O157:H7, 4 O157:NM, 23 O157:non-H7, 22 H7:non-O157, and 120 non-O157:nonH7), 17 Salmonella serovars, and 29 other gram-negative bacteria were used to evaluate the reactivities of the two MAbs by indirect enzyme-linked immunosorbent assay (ELISA). Both MAbs reacted strongly with all E. coli strains possessing the H7 antigen and with H23- and H24-positive E. coli strains. Indirect ELISA MAb specificity was confirmed by inhibition ELISA and by Western blotting (immunoblotting), using partially purified flagellins from E. coli O157:H7 and other E. coli strains. On a Western blot, MAb 46E9-9 was more reactive against H7 flagellin of E. coli O157:H7 than against H7 flagellin of E. coli O1:K1:H7. Competition ELISA suggested that MAbs 2B7 and 46E9-9 reacted with closely related H7 epitopes. When the ELISA reactivities of the MAbs and two commercially available polyclonal anti-H7 antisera were compared, both polyclonal antisera and MAbs reacted strongly with E. coli H7 bacteria. However, the polyclonal antisera cross-reacted strongly both with non-H7 E. coli and with many non-E. coli bacteria. The polyclonal antisera also reacted strongly with H23 and H24 E. coli isolates. The data suggest the need to define serotype-specific epitopes among H7, H23, and H24 E. coli flagella. The anti-H7 MAbs described in this report have the potential to serve as high-quality diagnostic reagents, used either alone or in combination with O157-specific MAbs, to identify or detect E. coli O157:H7 in food products or in human and veterinary clinical specimens.

Mapping of the H7-serospecific domain of Escherichia coli flagellin.

The amino acid sequences responsible for H7 and H23 flagellum serology have been identified by using a genetic approach. The H7-specific domain was located between amino acids 352 and 374 of the H7 flagellin. The sequencing data also demonstrated that the difference between the H7 and H23 flagellins in this region results from a single substitution at amino acid 366 (Ser-->Thr). The common epitopes for H7 and H23 were located between amino acids 284 and 366.

Characterization of the region encoding the CO-induced hydrogenase of Rhodospirillum rubrum.

In the photosynthetic bacterium Rhodospirillum rubrum, the presence of carbon monoxide (CO) induces expression of several proteins. These include carbon monoxide dehydrogenase (CODH) and a CO-tolerant hydrogenase. Together these enzymes catalyze the following conversion: CO + H2O --> CO2 + H2. This system enables R. rubrum to grow in the dark on CO as the sole energy source. Expression of this system has been shown previously to be regulated at the transcriptional level by CO. We have now identified the remainder of the CO-regulated genes encoded in a contiguous region of the R. rubrum genome. These genes, cooMKLXU, apparently encode proteins related to the function of the CO-induced hydrogenase. As seen before with the gene for the large subunit of the CO-induced hydrogenase (cooH), most of the proteins predicted by these additional genes show significant sequence similarity to subunits of Escherichia coli hydrogenase 3. In addition, all of the newly identified coo gene products show similarity to subunits of NADH-quinone oxidoreductase (energy-conserving NADH dehydrogenase I) from various eukaryotic and prokaryotic organisms. We have found that dicyclohexylcarbodiimide, an inhibitor of mitochondrial NADH dehydrogenase I (also called complex I), inhibits the CO-induced hydrogenase as well. We also show that expression of the cooMKLXUH operon is regulated by CO and the transcriptional activator CooA in a manner similar to that of the cooFSCTJ operon that encodes the subunits of CODH and related proteins.

Widespread activation of barrel cortex by small numbers of neonatally spared whiskers.

Activity-dependent plasticity in rodent whisker barrel cortex was examined by means of high-resolution 2-deoxyglucose (2-DG) with immunohistochemical double labeling. Hamsters with all but one, two, or four follicles ablated on postnatal day 7 received 2-DG injections as adults. Autoradiograms of follicle-ablated animals showed heavy activation of the entire barrel field during normal behavior, despite the missing whiskers. The intensity of 2-DG labeling was significantly reduced if the whiskers spared after follicle ablation were trimmed prior to the 2-DG injection, demonstrating that the widespread activation was driven by the spared whiskers. This widespread metabolic activation of the adult barrel field after neonatal follicle ablation was in sharp contrast to the somatotopically appropriate 2-DG labeling in barrel fields of normal adults subject to acute trimming of most whiskers, but was similar to that seen in normal adult animals with all whiskers intact. The results demonstrate large-scale plasticity of barrel circuitry following neonatal sensory deprivation, and provide a powerful functional anatomical setting to investigate underlying mechanisms.

Effects of high-voltage galvanic stimulation of ST36 and ST37 acupuncture points on peripheral blood flow and skin temperature.

In traditional Chinese medicine, ST36 and ST37 acupuncture points are commonly recommended to enhance peripheral blood circulation and digestive functions; however, there is a paucity of empirical data in support of the above recommendations. This study was designed to evaluate the effects of transcutaneous electrical stimulation of ST36 and ST37 acupuncture points on peripheral haemodynamic functions. Eleven healthy subjects (five men, six women; mean age 28 years), voluntarily participated in the study. A 2 (pulse frequency) x 3(time-frame) experimental design was employed. The subjects' ST36 and ST37 acupuncture points were stimulated, at sensory threshold intensity, with the high-voltage galvanic (HVG) current using two (5 Hz and 120 Hz) different pulse frequencies. Skin temperature and haemodynamic functions (cutaneous blood flow, microvascular volume and erythrocyte velocity), between the first and second metatarsals of the subjects' dominant foot were monitored every 2 min, for 10 min at rest, during 20 min HVG stimulation treatment and 10 min post-stimulation. The results of the repeated-measures analysis of variance revealed no significant F-ratio for pulse frequency and time-frame main effects for any of the dependent variables. Based on our findings we conclude that percutaneously applied HVG stimulation of ST36 and ST37 acupuncture points does not increase peripheral haemodynamic functions in asymptomatic subjects.

[Limbal epithelial autograft transplantation for treatment of unilateral fibrous vascularized cornea caused by chemical burns].

OBJECTIVE: To provide healthy epithelial source for damaged cornea to prevent the invasion of conjunctival tissue for treatment of corneal vascularized opacity after chemical turns. METHODS: Limbal epithelial autograft transplantation was performed on 17 cases (17 eyes) with vascularized corneal opacity caused by chemical burns. RESULTS: Of 17 cases, the epithelial healing was stable in 15 cases, and there was corneal graft transparency, improvement of visual acuity and without neovascularization in 13 cases. CONCLUSION: Limbal epithelial autograft Limbal epithelial autograft transplantation applied for reconstruction of palisades of Vogt to provide new stem cell source for injured limbus is a reasonable therapeutic method for treatment of vascularized corneal opacity caused by chemical burns.
