[Effects of 3,4,5-trimethoxy-N-(3-piperidyl) benzamide (KU-54) on the incorporation (excretion) of 14C-glucosamine in the gastric mucosa and the liver of rats].

Effects of KU-54 on the biosynthesis of glycoprotein in the gastric mucosa and the liver, as measured by the rate of incorporation of 14C-glucosamine, were investigated in rats under various conditions after a single administration of 14C-glucosamine of (9.88 microCi/animal, ip). 14C-glucosamine was incorporated with relative ease in the acid-insoluble fraction of the gastric mucosa. KU-54 at 100 mg/kg was orally administered twice daily for 5 days in rats (though it was given once on the 5th day) before injection of 14C-glucosamine. The rate of 14C-glucosamine incorporation into the acid-insoluble fraction of the gastric mucosa was significantly increased by KU-54, but that of the hepatic tissue was not increased. In addition, hydrocortisone (20 mg/kg) also produced a drop of incorporation of 14C-glucosamine in the gastric mucosa, but oral KU-54 at 100 mg/kg twice daily for 5 days (though it was given once on the 5th day) significantly inhibited the drop of incorporation of 14C-glucosamine in the gastric mucosa, but that in the hepatic tissue was not inhibited. Therefore, the effects of KU-54 were greater in the gastric mucosa than in the hepatic tissue. On the 5th day of the ulcer produced by acetic acid in rats, the specific radioactivity in the mucosa of the margin of the ulcer increased significantly compared with that in the normal (non-ulcerative) mucosa, but this phenomenon was not affected by KU-54.(ABSTRACT TRUNCATED AT 250 WORDS)

Cryoglobulin removal with cryofiltration in the treatment of cryoglobulinemia.

The technique of cryofiltration has been applied in the treatment of cryoglobulinemia. Good clinical improvement has been demonstrated accompanied by decreases of cryoglobulin levels. Three treatments in a week produced transient improvement of clinical symptoms and reduction of cryoglobulin levels. Follow-up treatments performed at adequate frequency and volume processed maintained relief of symptoms and serologic improvement. The cryofilter demonstrates good selectivity for cryoglobulin removal in cryogel, confirmed by electrophoretic analysis of both the cryogel filtrate from the cryofilter used clinically. Plasma cryofiltration appears to be a safe alternative to plasma exchange, requiring no replacement fluids.

[A case of two-chambered left ventricle with echoes of an abnormal floating mass].

A case of a two-chambered left ventricle with an abnormal floating mass was reported. A 13-year-old boy was admitted to our hospital because of a grade III/VI transsystolic apical murmur. Chest radiography showed mild cardiomegaly. The electrocardiogram showed QS patterns in leads V1-V3, and left ventricular high voltage with wide QRS complexes. A two-dimensional echocardiogram revealed two chambers divided by the septum in the left ventricle, and a mass floating in the apical chamber. A levogram showed the double chambered left ventricle incompatible with the findings of echocardiography. The walls of the accessory chamber and the septum were hypokinetic, but not dyskinetic. Coronary arteriograms were unremarkable. At surgery, tangled trabeculae carneae and chordae tendineae were observed on the apical surface of the septum. The accessory chamber communicated freely with the main chamber through an orifice with a diameter of 1.5 cm. No other congenital malformations were found. Microscopic examination of the accessory chamber wall showed the thickened endocardium and hypertrophic myocardial fibers, and there was no area of fibrosis.

[Effects of 3,4,5-trimethoxy-N-(3-piperidyl) benzamide (KU-54) on respiration of the gastric mucosa and liver in rats].

Effects of 3,4,5-trimethoxy-N-(3-piperidyl) benzamide (KU-54), an antiulcer drug, on the tissue respiration of the gastric mucosa and the liver were studied in rats. Oral KU-54 at 100 mg/kg twice daily for 5 days (though it was given only once on the 5th day) caused an increase in oxygen consumption of the gastric mucosa in rats, but did not affect that of the liver. Thus the principal active site of KU-54 on tissue respiration was found to be the gastric mucosa. Oral KU-54 at 100 mg/kg once daily for 11 days significantly accelerated the oxygen consumption of marginal gastric mucosa of acetic acid ulcer in rats. The effect of oral gefarnate at 200 mg/kg was about half that of KU-54 at 100 mg/kg, but it was not significant. In addition, oral KU-54 at 100 mg/kg twice daily for 5 days (though it was given only once on the 5th day) significantly inhibited the decrease of oxygen consumption of the gastric mucosa in hemorrhagic shocked rats. The effect of oral gefarnate at 100 mg/kg was not like that at KU-54 at 100 mg/kg in conscious rats. When KU-54 was added in the incubation medium with small gastric mucosal fragments of rats, the increase of oxygen consumption of the gastric mucosa did not occur. Oral KU-54 at 100 mg/kg significantly accelerated a glycogen consumptive stimulation of the gastric mucosa of the corpus in ischemic rats, but the respiration of the antral mucosa was not accelerated under anaerobic incubating conditions. Oral gefarnate at 200 mg/kg accelerated an anaerobic glycolysis of the gastric antral mucosa in rats.

A role of valency of concanavalin A and its chemically modified derivatives in lymphocyte activation. Monovalent monomeric concanavalin A derivative can stimulate lymphocyte blastoid transformation.

A concanavalin A (ConA) derivative of a monovalent monomeric nature, including a monomeric molecular weight at pH 7.4, significantly induces lymphocyte blastoid transformation. The derivative was recently obtained by Tanaka, I., Abe, Y., Hamada, T., Yonemitsu, O., and Ishii, S. ((1981) J. Biochem. (Tokyo) 89, 1643-1646) by a novel procedure of photochemically induced alkylation of tryptophan residues of native tetravalent ConA using a high pressure mercury lamp in the presence of chloroacetamide followed by two steps of column chromatography. This monovalent monomeric ConA (Mm-ConA) was demonstrated to be almost equally potent in producing the maximal response of lymphocytes when compared with native tetrameric ConA, although Mm-ConA was required at about 70 times as high as the concentration of native ConA on a weight basis to attain the maximal response of lymphocyte activation. The binding potency of the former to lymphocytes was about two-thirds as potent as that of the latter. Mm-ConA failed to agglutinate sheep erythrocytes at concentrations 1800-fold higher than native tetravalent ConA, but showed a weak but definite agglutinating activity against guinea pig erythrocytes at a relatively high concentration (approximately 80 micrograms/ml). Cell cluster formation was observed in lymphocyte cultures for 24 to 48 h with Mm-ConA where DNA replication in stimulated lymphocytes was observable. No significant difference was observed between sizes of cell clusters formed in the presence of Mm-ConA and of native tetravalent ConA at this phase of lymphocyte activation. The present results suggest that the multivalency of ConA with respect to sugar-binding sites may not be a stringent requirement for lymphocyte activation, and that another binding site for cell membrane, which has been suggested to exist in the ConA protomer and to be hydrophobic (membranophilic), may play a subsidiary but important role in triggering lymphocyte blastoid transformation as well as hemagglutination with Mm-ConA. The biological significance in lymphocyte activation of lectin valency with respect to sugar-binding sites has been discussed, comparing the effects of various ConA derivatives of different valencies, including a monovalent monomer (Mm-ConA), divalent dimers (beta-ConA, sulfomethylamino-ConA, and succinylated ConA), and tetravalent tetramers (alpha-ConA), on the cell surface.
