[The production and reactivity of the monoclonal antibody (MCA-97) to endometrial adenocarcinoma].

Spleen cells from BALB/c mouse immunized with the human endometrial adenocarcinoma cell line (ISHIKAWA) were fused with mouse myeloma cell line (NS-1) in the presence of polyethylene glycol (Mr 1000). One monoclonal antibody, MCA-97 (IgM subclass), which showed reactivity with the ISHIKAWA-cell line, was obtained by a limiting dilution technique. In a cellular enzyme-linked immunospecific assay, the MCA-97 antibody reacted with all of 7 adenocarcinoma cell lines tested. In immunoperoxidase testing of formalin-fixed paraffin-embedded sections, the MCA-97 antibody reacted with most endometrial adenocarcinomas and endometrioid adenocarcinomas of the ovary, but did not react with squamous cell carcinomas or ovarian serous adenocarcinomas. In addition, it reacted with the glandular epithelium of normal tissues, such as proliferative endometrium, fallopian tube, uterine cervix and gastrointestinal tract. The reversed passive hemagglutination method demonstrated the antigen defined by MCA-97 in the sera of 5 out of 11 patients with endometrial carcinomas, and 3 out of 4 of those with ovarian endometrioid adenocarcinomas. It was not demonstrable in sera of most normal female volunteers, or any patients with cervical squamous cell carcinomas or ovarian serous adenocarcinomas. Thus, MCA-97 is of potential clinical application in diagnostic serology and pathology.

Effects of a synthetic human atrial natriuretic polypeptide on regional blood flow in rats.

The effects of alpha-hANP on systemic hemodynamics and regional blood flow were examined in conscious WKY and SHR. An intravenous infusion of alpha-hANP (3 micrograms/kg per min) resulted in a rapid and marked fall of blood pressure and of total peripheral resistance but with no change in cardiac output in both strains. alpha-hANP decreased vascular resistance in most organs and there was a redistribution of renal blood flow. Thus, the acute hypotensive effect of alpha-hANP is probably related to a vasodilator action.

Experimental studies on myocardial glucose metabolism of rats with 18F-2-fluoro-2-deoxy-D-glucose.

The myocardial uptake of 18F-FDG was investigated under various conditions, and compared with brain and tumor uptake as a function of blood glucose level. The uptake by the heart of normal feeding rats was rapid and remained essentially unchanged up to 2 h after 18F-FDG injection, approximately 3%-4% dose/g tissue. On the other hand, the myocardial uptake of fasted rats was significantly lower than that of control rats throughout the course of the study, and it was about 0.3%-0.4% dose/g tissue. Myocardial uptake of 18F-FDG was relatively constant at glucose levels under about 120 mg/100 ml and increased steeply at higher blood glucose levels. In contrast, brain uptake decreased linearly with increasing levels of blood glucose, revealing a strong negative correlation between brain uptake of 18F-FDG and blood glucose levels. The tumor uptake pattern remained relatively unchanged, irrespective of blood glucose levels. It was revealed that the glucose demands of brain, heart, and tumor were entirely different. After a glucose load, the myocardial uptake of fasted rats increased only slightly from 0.4% to 0.6% dose/g tissue, in spite of transitional hyperglycemia. In contrast, insulin caused myocardial uptake to increase extraordinarily, although it caused a decrease in blood glucose levels.

Studies on 18F-labeled pyrimidines III. Biochemical investigation of 18F-labeled pyrimidines and comparison with 3H-deoxythymidine in tumor-bearing rats and mice.

Metabolic studies of 18F-labeled 5-fluoro-2'-deoxyuridine(FdUrd), 5-fluorouridine(FUrd) and 5-fluorouracil (FUra) were performed in tumor-bearing rats and mice. Also, the usefulness of 18F-FdUrd and 3H-deoxythymidine (dThd) for tumor detection was compared. In the tumor, 2 h after the injection of the 18F-pyrimidines, 3%-11% and 6%-14% of the 18F was present in the nuclear and microsomal fractions, respectively, and 17%-34% and 19%-24% of the 18F was incorporated into the acid-insoluble and nucleotide fractions, respectively. Of the three 18F-pyrimidines, 18F-FUrd demonstrated the highest incorporation rate, while 18F-FUra showed the lowest incorporation rate. The incorporation in the spleen, small intestine, and liver was less than that in the tumor. 3H-dThd and 18F-FdUrd were injected into the same mice. The 3H-dThd was accumulated in the spleen, small intestine, and tumor, and in these three tissues significant amounts of the 3H were incorporated into acid-insoluble materials. However, the clearance of 18F-FdUrd was slow in the tumor but rapid in the spleen and small intestine. In the autoradiograms of the tumor, 18F and 3H showed a slightly different distribution. Both distribution patterns were unchanged when the soluble materials were rinsed out with perchloric acid. For tumor detection, 18F-FdUrd gives the same information as radio-dThd, and further information can be obtained by positron-emission tomography.

Effects of chronic LiCl and RbCl on muricide induced by midbrain raphe lesions in rats.

Midbrain raphe lesions in rats (raphe rats) induce aggressive behavior including muricide. A single administration of LiCl (Li) 100 mg/kg to raphe rats produced only 25% of muricide inhibition. However, the inhibitory effect of muricide in raphe rats significantly increased from the 5th day following repeated administration of Li. Chronic Li also inhibited muricide in olfactory bulbectomized (OB) rats. The inhibition of muricide lasted until the next day to some extent. In this point, the effect of Li on muricide is similar to that of antidepressants, but not of neuroleptics. On the contrary, RbCl (Rb) showed a tendency to induce muricide. The single re-administration of Li 100 mg/kg on the withdrawal on the 7th day after repeated administration for 14 days showed a significant inhibition of muricide in raphe rats, unlike that in OB rats. Li also showed a partial prophylactic effect on muricide when Li 100 mg/kg was administered for 1 week before raphe lesions. These results suggest that raphe rats may serve not only as an experimental model of depression, but also as that of manic illness.

Matrix glycosaminoglycan in urinary stones.

At first, urinary stones were classified according to their inorganic components (apatite, struvite, calcium oxalate monohydrate, calcium oxalate dihydrate and uric acid). Then, matrix glycosaminoglycan was extracted from the stones in each group and was analyzed by 2-dimensional electrophoresis. There were differences in the glycosaminoglycan content of matrices among different groups of urinary stones. The principal matrix glycosaminoglycan content consisted of hyaluronic acid in apatite and struvite stones, heparan sulfate in calcium oxalate monohydrate and uric acid stones, and hyaluronic acid and heparan sulfate in calcium oxalate dihydrate stones. We conclude that hyaluronic acid and/or heparan sulfate has an important role in urinary stone formation.

Effect of adenosine deaminase inhibitors on myocardial reactive hyperaemia following brief coronary occlusions.

Effect of two agents of adenosine deaminase inhibitor, 8-azaguanine and adenine, on myocardial reactive hyperaemia was tested in the anaesthetised open-chest dog. Reactive hyperaemic flow response of the circumflex coronary artery was observed following 5, 10, 15, 20 and 30 s coronary occlusions before, during and after infusion of 8-azaguanine and adenine, which are known as adenosine deaminase inhibitors. Intracoronary infusion of 8-azaguanine and adenine caused the minimum increase in the baseline coronary flow. Both the nucleic acids shifted the dose response curve of adenosine to the left. 8-azaguanine enhanced volume response of flow at all occlusion intervals tested. The infused dose of adenine also intensified volume response of flow after 5, 15, 20 and 30 s occlusions. Fifteen minutes after termination of the nucleic acid infusions, the reactive hyperaemia returned towards control levels. The results suggest that 8-azaguanine and adenine enhance myocardial reactive hyperaemia possibly by inhibiting adenosine deaminase to degradate myocardial interstitial adenosine to inosine.

Immunohistochemical study of amylase in common epithelial tumors of the ovary.

Cellular localization of amylase in various ovarian tumors was studied by the immunoperoxidase method using an antibody to human pancreatic amylase. Amylase was present in eight of 34 serous carcinomas and eight of 27 endometrioid carcinomas. However, only in one poorly differentiated serous carcinoma and two well-differentiated endometrioid carcinomas were a large number of amylase-reactive cells found. Five benign and three borderline serous tumors contained no amylase. Also, amylase was not detected in any of 34 mucinous tumors or five malignant clear cell tumors. The results obtained suggest that amylase will be a useful tumor marker, when present, for follow-up of endometrioid and serous carcinomas of the ovary.

Intracoronary adenosine enhances myocardial reactive hyperemia after brief coronary occlusion.

Adenosine is a prime candidate for the role of mediator between myocardial metabolic state and coronary blood flow. However, there are few reports concerning the direct effects of exogenously added adenosine on coronary autoregulation. The present investigation in the open-chest dog studied the effects of a threshold dose of intracoronary adenosine infusion on reactive hyperemia following brief coronary occlusions. The infused dose did not increase nonocclusive flow by greater than 10%. Adenosine enhanced total hyperemic flow at all occlusions tested (5, 10, 15, 20, and 30 s). Aminophylline pretreatment reduced reactive hyperemia below the control level even in the presence of an intracoronary infusion of adenosine. Adenosine injected into the left atrium and intracoronarily infused papaverine did not affect hyperemic response to 5- and 15-s coronary occlusions. The results suggest that a minimum dose of exogenously added adenosine enhances myocardial reactive hyperemia, possibly by potentiating the effects of endogenous adenosine released during ischemia.
