[Study on nuclear DNA content and image cytometry in cervical cancer and precursor lesions].

OBJECTIVE: The aim of this research was to study the biological and clinical features of cervical cancer and precursor lesions. METHODS: Nuclear DNA was analyzed by image cytometry (ICM) in 125 embedded tissue 5 microns sections stained with Feulgen stain. Samples included normal cervical squamous epithelium (n = 11), cervical intraepithelial neoplasia I (CIN I) (n = 22), CINII (n = 17) and CINIII (n = 13), cervical neoplasm (n = 62). RESULTS: The mean DNA content, nuclear area increased progressively from normal cervical epithelium, CINI, CINII, CINIII to invasive squamous carcinoma. Statistical analysis revealed significant difference (P < 0.05, < 0.01). Histograms of cellular DNA ploidy distribution showed different pattern, where 9 components (9C) cells were observed in cervical cancer and CINIII. No differences in DNA mean ploidy (DMP), DNA index (DI) and nuclear area were observed between cervical neoplasm. CONCLUSIONS: It is increased progressively from normal cervical epithelium to cancer, in which CINIII is turning point.

[The CPBC types used for clinical choice and observation on their microstructure].

UNLABELLED: According to the primary properties of calcium phosphate bone cement (CPBCs), we have chosen certain types of them for use in cranioplasty and have observed the microstructure of their set bodies by means of SEM. Five kinds of CPBCs powder have been prepared, including: octacalcium phosphate precipitated hydroxyapatite (OCP-PHA) type, octacalcium phosphate hydroxyapatite (OCP-HA) type, octacalcium phosphate (OCP) type, calcium deficient hydroxyapatite (CDHA) type and hydroxyapatite (HA) type. The mixing liquids were deionized water and 0.25 M Na2HPO4/NaH2PO4 buffer solution. The setting time was measured by Gillmore method. The compressive strengths were measured using the TS-14 automatically measured instrument-II of single granule's compressive strength. The CPBC types used for clinical cranioplasty were defined according to Ginebra's Criteria of the CPBCs, and the ultrastructure of the set-bodies of the defined CPBC type was observed by SEM before and after its immersion in Ringer's Solution. RESULT: Two out of five CPBCs, OCP-PHA-CPBC and CDHA-CPBC were selected for clinical cranioplasty. Before and after they were immersed in Ringer's Sol., the SEM found their set-bodies to be structurally porous and to dissolve with the increase of immension time. These data indicate that OCP-PHA-CPBC and CDHA-CPBC are sufficient for the reconstruction of non-stress-bearing bone, and the porous structure of their set-bodies is advantageous to fibroplasty or fibrovasculization in their set bodies.

[Effect of hypertonic saline solution on the viscoelasticities of erythrocyte membrane in rats subjected to hemorrhagic shock].

We have studied the effect of hypertonic saline solution on the viscoelasticities of erythrocyte membrane in hemorrhage-shocked rats using micropippette aspiration technique. Wistar rats were randomly divided into three groups of 0.9% NaCl(NS), 7.5% NaCl (HS) and 5% NaCl-3.5% NaAc (HSA), respectively. The animals were bled to reach a mean arterial pressure of 5.3 kPa in 10 minutes and maintained in shock for 90 minutes. 4 ml/kg NS, HS and HSA was given intravenously and respectively in 5 minutes following hemorrhagic shock. The blood was collected to determine the viscoelasticities of erythrocyte membrane at baseline, shock and after treatment. The results showed that the elastic moduli and viscous coefficients of erythrocyte membrane were increased obviously following hemorrhagic shock. HS raised elastic moduli and reduced viscous coefficients significantly compared with NS after treatment. The elastic moduli and viscous coefficients of erythrocyte membrane were decreased remarkably in HSA group than in NS and HS group. These data suggested that HSA could improve the viscoelasticities of erythrocyte membrane significantly in rats subjected to hemorrhagic shock.

Clinical significance of examining IL-2R in peripheral blood lymphocytes of patients with gastric cancer.

OBJECTIVE: To understand the clinical significance of membrane receptors of interleukin-2 (CD25) in peripheral blood lymphocytes (PBL) of patients with gastric cancer. METHODS: CD25 percentages in PBL from 110 patients with gastric cancer and 100 normal individuals were examined by flow cytometry. Results were analyzed clinically and pathologically. RESULTS: Mean CD25% in PBL from normal individuals and patients with gastric cancer were 17.24 +/- 5.33 and 22.47 +/- 6.82, respectively, and the difference was significant (P < 0.01). Abnormally elevated CD25% in PBL from patients with gastric cancer were correlated with tumor site, macropathology type, tumor size, differentiation degree and clinical stage. CONCLUSION: Measuring CD25% in PBL from gastric cancer patients before surgery may provide a reference for diagnosis, immune treatment and judging prognosis.

[Significance of the CT scan in renal tuberculosis].

OBJECTIVE: To elucidate the features and clinical significance of CT scan in the diagnosis of renal tuberculosis. METHODS: The CT findings and the clinical characteristics of 42 cases with renal TB were retrospectively analyzed. RESULTS: Unilateral renal TB was found in 34 cases, bilateral in 8 cases. The typical CT features were characterized by one or more cysts surrounding the calyx with thin adjacent cortex (34/50), thickened wall of ureters and calyx, hydronephrosis (22/50) and kidney calcification (8/50). Atypical features presented with single or multiple low density nodes in the renal parenchyma (6/50). 30 patients receiving contrast enhanced CT scan showed that intensified Hu (by 20 approximately 120 HU) in the affected kidney. Multiple lymph nodes calcification in abdomen was found in 2 patients, calcification and low density nodes in spleen and liver in 4 patients, vertebral damage and paravertebral abscess in 6 patients and post-abdominal abscess in 1 patient. CONCLUSION: CT scan examination is not only useful in the diagnosis of renal tuberculosis, but also of significance in assessing the severity of the disease, renal function and the involvement of other organs in the abdomen.

PDA support for outpatient clinical clerkships: mobile computing for medical education.

This project provides educational support for students enrolled in a family practice clerkship by supplying PDA-based clinical references and tools to collect information about the patients they see and the seminars they attend. Each student is supplied with a Handspring Visor Deluxe to use during the clerkship. Supplied software includes a clinical reference (Five Minute Clinical Consult) and Lexidrugs drug reference and a medical calculator. The data collection software consists of a patient log for recording simple demographic and diagnostic information about each patient seen during the clerkship, a seminar evaluation module for recording student feedback about each didactic presentation during the course, and an evaluation form where they students supply their reactions to the use of the Visor. Despite encountering a number of problems, the devices appear to be beneficial tools. The applications provide useful references for students who do not have access to the resources of the University in their family medicine clerkship. They also provide an improved, though not perfect, means to capture data regarding patient encounters and course evaluations. However, the challenge remains to better integrate the PDA with the student s workflow in the clinic.

[Epidemiological study on the influence of pig-derived Ascaris to the transmission of human ascariasis].

OBJECTIVE: An experimental epidemiological study was designed to investigate possible relations of pig-derived Ascaris to the transmission of human ascariasis which might be caused by cross-infection between the pig and humans. METHODS: In Xinjian County, Jiangxi province, two adjacent villages, Laozhi and Panzhi, with similar baseline of ascariasis in pig and human populations were selected as the study sites with a one-year longitudinal epidemiological follow-up study. People in the two villages received mass chemotherapy. Pigs received mass chemotherapy only in Panzhi village and was twice repeated with two months interval, while pigs in Laozhi village were not treated at all. In the following year after treatment on humans, ascariasis among villagers was studied cross-sectionally five times with Kato-katz technique. RESULTS: No significant difference of the re-infection patterns of human ascariasis was found between the two villages in terms of prevalence and intensity during the following year in all the five surveys (chi(2) < 0.658, F < 1.658 and P > 0.1). CONCLUSION: The results suggested that pig-derived Ascaris were mainly transmitted among pigs but had no important role on the transmission of human ascariasis. The results were in good accordance with the findings through a recent molecular genetic research on the local human-and pig-derived Ascaris worms. Possible confounding factors were discussed.

[Mycobacterial spindle cell pseudotumor of lymph nodes].

OBJECTIVE: To study the clinicopathological of mycobacterial spindle cell pseudotumor in lymph nodes. METHODS: Two cases of spindle cell pseudotumor in the lymph nodes of infants were examined using light microscopy. The sections were stained with HE, Ziehl-Neelsen and studied immunohistochemically. RESULTS: Both infants were 1 year of age and had a history of Bacilli Calmette Guerin (BCG) vaccination before discovery of their lymph node lesions. Clinically, the patients presented fever and enlarged lymph nodes. Histopathologically, the lesions showed proliferation of spindle cells arranged in bundles, storiform patterns with abundant capillaries and lymphocyte, plasma cell and neutrophil infiltration forming a spindle cell pseudotumor. Most of the spindle cells were phagocytic cells which contained large amounts of mycobactrium as observed by Ziehl-Neelsen staining. Immunohistochemical staining showed strongly positive result for Mac387 and lysosome, but negative for S-100 in spindle cells and histocytes. CONCLUSIONS: Mycobacterial spindle cell pseudotumor of lymph nodes may occur not only in immunodeficient patients (such as AIDS) but also in infants after BCG vaccination. The lesions may mimic spindle cell tumor in lymph nodes, therefore it is very important to recognize these lesions in order to treat the patient properly.

[Primary cutaneous CD30-positive anaplastic large cell lymphoma].

OBJECTIVE: To analyse the clinical manifestations, pathological and immunohistochemical features of 9 cases of primary cutaneous CD30-positive anaplastic large cell lymphoma (ALCL) in order to improve the criteria for its early and differential diagnosis. METHODS: Histopathological method was used to study the morphological characteristics. Immunohistochemical stainings (SP or ABC method) for leukocyte common antigen (LCA), CD20, CD30, CD45RO, CD68, epithelium membrane antigen (EMA), cytokeratin (CK) and HMB45 were applied to the routine paraffin sections of all the 9 cases. RESULTS: The 9 cases of primary cutaneous CD30-positive ALCL consisting of 6 males and 3 females, aged 31 to 84 years (mean 58.2 years). All patients presented with subcutaneous masses or papular eruptions on lower trunk and extremities. Histopathologically, the lesions were composed of numerous large round, oval or pleomorphic cells with abundant cytoplasm, amphophilic or basophilic, and large nucleus with distinct nucleolus. Mitosis was often observed. Multinucleated giant cells and Reed-Sternberg cells may be seen. The neoplastic cells displayed positive antigen markers for CD30 in 9 cases, 6 positive for CD45RO and 3 cases negative for both CD45RO and CD20. Two patients died of metastasis, 2 patients experienced recurrence and 5 patients still well during follow-up. CONCLUSIONS: Primary cutaneous CD30-positive ALCL is a morphologically distinctive neoplasm with a comparatively favorable prognosis. This tumor can be differentiated from other malignant tumors on the basis of histopathologic features and positive CD30 which is of significance.

[Application of triple-color flow cytometry for minimal residual disease detection in acute B-lymphoblastic leukemia].

OBJECTIVE: To set up a sensitive triple-color flow cytometric assay (FCM) for detection of residual leukemia cells. METHODS: Application of triple-color flow cytometry to the patients for detecting CD(10)(+)CD(22)(+)CD(45)(- or dim) leukemic cells in acute B lymphoblastic leukemia (B-ALL). RESULTS: Leukemic cells in B-ALL patients' bone marrow (BM) consistently showed the CD(10)(+)CD(22)(+)CD(45)(- or dim) phenotype. FCM analysis could detect leukemic cells at the level of 1 per 10(4) cells. The CD(10)(+)CD(22)(+)CD(45)(- or dim) cell population was absent in normal BM, regenerating BM, and BM from leukemia patients in continuous complete remission. This assay could identify residual leukemic cells in 50 days after starting induction chemotherapy. Furthermore, a gradual increase of leukemic cell population could be monitored by this assay before morphological confirmation of BM relapse. CONCLUSION: Triple-color FCM was a sensitive method for detecting BM CD(10)(+)CD(22)(+)CD(45)(minus sign or dim) leukemic cells and could be an additional useful tool to monitor MRD in B-ALL patients.

[A review on nasopharyngeal carcinoma in ancient Chinese literature].

Based on the clinical characteristics of nasopharyngeal carcinoma (NPC), a receptive standard was set up to screen related information with NPC from ancient literature of traditional Chinese medicine, while a rejection standard was also set up to prevent associated data in these literature with diseases such as lymphatic tuberculosis and lymphoma in the neck etc. Furthermore, a review was made on the etiology, pathogenesis and procedures used in the prevention and treatment of NPC recorded in ancient literature, according to the evaluation as above. The data showed that several hypotheses were present in ancient literature about etiology and pathogenesis of NPC, including healthy qi deficiency, cold in channels, or liver qi stagnation, pyrogenous toxin, phlegm coagulation and blood stasis etc. Therefore, prevention was emphasized and the main therapeutic and preventive methods for NPC were regulating emotions, nourishing qi and harmonizing ying (nutrient).

[Clinical study of detenia cecal-ascending colon continent cutaneous urinary reservoir].

OBJECTIVE: To construct a good continent urinary diversion which is easy to be performed and has a low incidence of complications. METHODS: 26 cases of bladder cancer were given radical cystectomy before the cecal-ascending colon was excluded and 15-20 cm of the cecal-ascending colon was isolated. The colon teniae were then incised at the interval of 0.5-1.0 cm to construct the detenia cecal-ascending colon continent urinary diversion open to the umbilicus. RESULTS: All the patients were followed up for 21.1 +/- 10.1 months. Reliable continence was achieved in all with a low incidence of complications. The capacity of the reservoirs reached 350-600 ml 6 month after operation. Self catherizations were carried out every 3 to 6 hours, Urodynamic data showed a mean maximum filling pressure of 58.7 +/- 24.5 cmH2O, and a mean maximum urethral (efferent) closure pressure of 104.3 +/- 33.8 cmH2O. CONCLUSION: Detenia cecal-ascending colon continent urinary diversion is an ideal method.

[Effect of para-aminobenzoic acid on the adhesion of Streptococcus mutans].

OBJECTIVE: To investigate the effects of Para-aminobenzoic acid (PABA) on the adhesion of Streptococcus mutans to saliva-coated hydroxyapatite (S-HA). METHODS: The adhesion model in vitro established by Gibbons was adopted in this study to quantify adsorptive cells through [3H] thymidine labelling. RESULTS: PABA solution over a range of concentration from 10(-9) to 10(-3) g/L could inhibit the attachment of S. mutans to S-HA effectively, and the inhibition declined when the concentration of PABA solution was 10(-3) g/L. CONCLUSION: PABA can inhibit the adherence of Streptococcus mutans to S-HA effectively.

[Clindamycin soluble suppository-production and its efficacy in treating bacterial vaginosis].

OBJECTIVE: To make the soluble suppository of clindamycin which is an effective drug for treatment of Bacterial vaginosis and to treat patients in clinical trials. METHODS: Outpatients who had vaginal infections from Sept. 1998 to Jan. 1999 were investigated. The diagnoses of bacterial vaginosis were made according to Amsel's criteria and 80 patients were included in this study. They were divided into two groups to be treated respectively by metronidanole suppository and clindamycin suppository which were made by our hospital pharmacy. RESULTS: The cure rate of metronidanole was 87.5% and that of clindamycin 90.3%; No significant difference was noted between the two groups(P > 0.05). The new clindamycin suppository was not irritant and it was easy to dissolve in vagina. There was no discontinued treatment because of side-effect. CONCLUSION: The clindamycin suppository does not irritate the mucosa of vagina, it is an efficacious remedy for bacterial vaginosis.

[A study on tongue cancer cell labeling using bromodeoxyuridine (BrdU) incorporation in vitro].

OBJECTIVE: The aim of this study is to evaluate of Bromodeoxyuridine (BrdU) label in the proliferation of the tongue cancer. METHODS: The authors used 80 micrograms/ml BrdU or iododeoxyuridine (IdU) to label three cases of fresh tissue of tongue cancer and four bottles of Tca8113 cells in logarithmic growth period in vitro. They were fixed and embedded in the routine procedure, and then in serial sections. These specimens were labeled with BrdU polyclonal antibody (1:200), and PCNA monoclonal antibody (1:100), stained using immunohistochemical ABC method. The authors observed the shapes and sizes of those positive cells, counted their label indexes (LI) of BrdU- or IdU-positive cells and PCNA- positive cells respectively. Four bottles of Tca8113 cells were cultured for 1-4 days. One bottle of the cells was every day used in the experimental study about comparing the cells labeled BrdU with those of the S phase of FCM in same cultural time. RESULTS: The positive cells generally owned a larger round nucleus, sometimes in company with sister cells. There was no difference of positive cells in these two kinds of labeling methods between the incorporation of BrdU or IdU and the expression of proliferating cell nuclear antigen (P > 0.05). The cells of S phase with FCM method were much more than that method with BrdU incorporation (P < 0.001). CONCLUSION: The method of BrdU labeling could be used in tumor cellular proliferation studies.

[Effects of para-aminobenzoic acid (PABA) on growth of Streptococcus mutans].

OBJECTIVE: The aim of this study is to examine effects of para-aminobenzoic acid (PABA) on the growth of Streptococcus mutans (S. mutans). METHODS: Different concentrations of PABA (10(-10)-10(-3) g/L) were separately transferred to modified Carlsson medium. S. mutans (ATCC 25175) grew in modified Carlsson medium with different concentrations of PABA. All cultures were incubated at 37 degrees C anaerobically in an atmosphere of 80% of nitrogen (v/v), 10% of hydrogen (v/v) and 10% of carbon dioxide (v/v) for 48 hours. The absorbance values of S. mutans were measured by using a spectrometer (UV-1601). The colony forming units (CFU) were obtained by growing S. mutans in media with different concentrations of PABA (10(-10)-10(-3) g/L). RESULTS: Different concentrations of PABA had different stimulating effects on the growth of S. mutans (P < 0.05). But this kind of stimulating effects declined when the concentration of PABA was 10(-3) g/L. CONCLUSION: This experiment indicates PABA has stimulating effects on the growth of S. mutans, and PABA can promote growth of S. mutans.

[Effects of para-aminobenzoic acid (PABA) on growth of Lactobacillus acidophilus].

OBJECTIVE: The aim of this study is to examine effects of para-aminobenzoic acid (PABA) on the growth of Lactobacillus acidophilus (L. acidophilus). METHODS: Different concentrations of PABA (10(-10)-10(-3) g/L) were separately transferred to the modified Carlsson medium. L. acidophilus (ATCC4356) grew in these Carlsson media. All cultures were incubated at 37 degrees C anaerobically in atmosphere of 80% of nitrogen, 10% of hydrogen, and 10% of carbon dioxide for 48 hours. Absorbance values (lambda = 540 nm) of bacterial suspensions were measured using a spectrometer (UV-1601). Colony forming units (CFU) were obtained by growing L. acidophilus in Carlsson media with different concentration of PABA (10(-10)-10(-3) g/L). RESULTS: Different concentrations of PABA (10(-10)-10(-4) g/L) had different stimulating effects on the growth of L. acidophilus (P < 0.05). But stimulating effects declined, when PABA concentration was 10(-5) g/L, and when the concentration of PABA reached 10(-3) g/L, the stimulating effect disappeared. CONCLUSION: This study indicates PABA stimulates the growth of L. acidophilus, and PABA can promote growth of L. acidophilus.

[An experimental study on demineralization of several beverages on bovine enamel].

OBJECTIVE: The aim of this study is to investigate the demineralization effects of several beverages on bovine enamel. METHODS: The concentration of calcium and phosphate in these beverages were measured by using EL 312e Micro-Plate after the bovine enamel was exposed to beverages for a period of 7 days. RESULTS: The data indicated that almost all of these beverages could cause demineralization of bovine enamel, except mineral water, and the concentration of calcium and phosphate in these beverages changed after the bovine enamel was exposed to beverages. There were great significant differences in demineralization abilities among different beverages (P < 0.01). The concentration of calcium and phosphate in beverages also changed with time (P < 0.01). CONCLUSION: All these beverages, except mineral water have erosive effects on bovine enamel. Demineralization degree varied with kinds of beverages. The fruit juice has the highest erosive effect on the enamel, and the calcium milk has the lowest erosive effect on the bovine enamel, when comparing with other beverages.
