Cytotoxicity of 2-ethenyl-2,3-dihydrophthalazine-1,4-diones in murine and human tumor cultured cells.

2-Etheny1-2,3-dihydrophthalazine-1,4-diones were successfully synthesized and proved to be effective cytotoxic agents against the growth of suspended murine and human leukemias and lymphomas. Selected compounds were also active in human HeLa uterine carcinoma, suspended effusion breast MCF-7 and glioma HS683 screens. These agents suppressed P388 lymphocytic leukemia DNA synthesis after 60 min at 100 microM. Their target appeared to be the de novo synthesis pathway with significant inhibition of the activities of both regulatory enzymes of the pathway, i.e. PRPP-amide transferase and IMP dehydrogenase resulting in a reduction in the d[NTP] pool levels for DNA incorporation. The compounds did not affect de novo pyrimidine synthesis and its regulatory enzymes. Very minor reduction by the agents was noted for the nucleoside kinases and the DNA and RNA polymerase activities within 60 min. DNA was not a target of the agents in that there was no alkylation of the nucleotide bases, intercalation between base pairs or cross-linking of the DNA strands; however, the agents did cause P388 DNA strand scission after 24 h at 100 microM.

mRNA and protein expression of Fas associated death domain protein in apoptosic hepatocyte induced by tumor necrosis factor-alpha.

OBJECTIVE: To study the expression of protein and mRNA of Fas associated death domain protein (FADD) in the apoptosic hepatocyte induced by D-galactosamine (GalN) and TNF-alpha. METHODS: Fulminant hepatic failure (FHF) was induced by injection of GalN into sensitized BALB/c mice by TNF-alpha. Hepatocyte apoptosis was examined by electric microscopy, in site end labeling (ISEL) and DNA electrophoresis on 1.5% agarose gel. The expression of protein and mRNA of FADD were detected by immunohistochemistry and PT-PCR. RESULTS: GalN/TNF-alpha may induce hepatocyte apoptosis, necrosis and liver failure. The expression of protein and mRNA of FADD were positively correlative with the apoptosic hepatocytes. CONCLUSION: TNF-alpha induces hepatocyte apoptosis by up regulation expression of FADD protein.

Quantification of cellular acid sphingomyelinase and galactocerebroside beta-galactosidase activities by electrospray ionization mass spectrometry.

BACKGROUND: Diagnosis of Niemann-Pick (A and B) and Krabbe diseases is achieved by measurement of the lysosomal enzymes acid sphingomyelinase (ASM) and galactocerebroside beta-galactosidase (GCG), respectively. Conventional assays use radiolabeled or fluorescent substrates and do not allow simultaneous determination of two or more enzymes in the sample. METHODS: We developed a sensitive and specific method to assay ASM and GCG in skin fibroblast homogenates using biotinylated substrate conjugates. The products were purified by bioaffinity capture on streptavidin-agarose beads and, following release, were analyzed by electrospray ionization mass spectrometry. Quantification was achieved using stable-isotope-labeled internal standards that were chemically identical to the products of the enzymatic reactions. RESULTS: The method demonstrated excellent linearity of ASM and GCG enzymatic product formation with the amount of cellular protein and incubation time. The range of ASM activities in fibroblast lysates from six healthy patients was 39-70 nmol. mg(-1). h(-1) compared with 3.7-5.1 nmol. mg(-1). h(-1) in cell lysates from two patients affected with Niemann-Pick A disease. The GCG activities toward the corresponding substrate conjugate were 4.0-6.8 nmol. mg(-1). h(-1) in cell lysates from healthy patients compared with 0.1-0.2 nmol. mg(-1). h(-1) in cell lysates from two patients affected with Krabbe disease. The amounts of substrate conjugates needed per analysis were 15 nmol (14 microg) for both ASM and GCG. CONCLUSIONS: Electrospray mass spectrometry combined with the use of biotinylated substrate conjugates and bioaffinity purification represents a new approach for the diagnosis of lysosomal storage diseases as demonstrated for Niemann-Pick A and Krabbe diseases. No radioactive substrates are used, and the method uses a single instrumental platform to determine both ASM and GCG in one cell sample.

[Immune response induced by hepatitis B DNA vaccine pCI-S-IRES- ProT alpha].

OBJECTIVE: To investigate immune response in mice inoculated with DNA vaccine. METHODS: pCI-S and pCI-S-IRES- ProT alpha were constructed by gene technology, transferred into cell line NIH-3T3 mediated by lipofectamine. Mice were inoculated with these plasmids too. HBsAg and HBsAb were detected by ELISA, and transcriptions of these plasmids were detected by RT-PCR. RESULTS: pCI-S -IRES- ProT alpha, and pCI-S were effectively expressed in cultured cells and vaccinated animals. Humoral immune and specific T-cell proliferative responses were stronger in pCI-S-IRES group than pCI-S group. CONCLUSIONS: Plasmids coexpression of prothymosin alpha and hepatitis B surface antigen can improve immune responses significantly.

Nitric oxide induces murine thymocyte apoptosis by oxidative injury and a p53-dependent mechanism.

Previously, we showed that NO induces thymocyte apoptosis via a caspase-1-dependent mechanism [(1) ]. In the present study, we investigated the role of heme oxygenase, catalase, bax, and p53 in this process. The NO donor, S-nitroso-N-acetyl penicillamine (SNAP), induced DNA fragmentation in thymocytes in a time- and concentration-dependent way. SNAP (100 microM) induced 50--60% apoptosis; higher doses did not increase the rate of apoptosis significantly. SNAP decreased catalase and heme iron (Fe) levels without affecting superoxide dismutase, glutathione, or total Fe stores in thymocytes. SNAP significantly increased the expression of heme oxygenase 1 (HSP-32), p53, and bax but not bcl-2. Treatment with the heme oxygenase inhibitor, tin protoporphyrin IX inhibited SNAP-induced thymocyte apoptosis. Furthermore, thymocytes from p53 null mice were resistant to NO-induced apoptosis. Our data suggest that NO may induce its cytotoxic effects on thymocytes by modulating heme oxygenase and catalase activity as well as up-regulating pro-apoptotic proteins p53 and bax.

[Improvement of proliferation of intrasplenically transplanted fetal hepatocytes after immunoadsorption].

OBJECTIVE: To investigate whether removing the compartments of hemopoietic stem cells by immunoadsorption would improve the proliferation of fetal hepatocytes after transplantation. METHODS: The purification of fetal hepatocytes was performed by immunoadsorption, and the proliferative capability of fetal hepatocytes after intrasplenic transplantation was evaluated by image analysis and flowcytometry analysis. RESULTS: The yield of fetal hepatocytes after immnoadsorption was (2~3)+/-10(7) in total per fetal liver and the viability is over 85%. The square densities of fetal hepatocytes with image analysis at 3, 7, and 14 days after transplantation were (1603+/-337), (3788+/-605), (8129+/-1025) micrometer(2)/mm(2), and the adult hepatocytes were (1007+/-128), (2013+/-242), (3780+/- 772) mum(2)/mm(2) accordingly. The S phase rates of fetal hepatocytes by flowcytometry analysis were (14.25+/-4.11)%, (16.07+/-5.35)%, and (17.32+/-5.17)%, and the rates of adult hepatocytes were (5.66+/-1.02)%, (4.98+/- 0.74)%, and (6.83+/-1.37)% correspondingly. The S phase rates and the square densities of fetal hepatocytes were significantly higher than those of adult hepatocytes. CONCLUSIONS: Fetal hepatocytes are highly proliferative and potentially suitable as donor cells in hepatocytes transplantation after immunoadsorption.

[Effect of vascular endothelial diphosphate glucuronosyltransferase mRNA regulated by alcohol].

OBJECTIVE: To investigate the effect of vascular endothelial growth factor(VEGF) gene transfection on tissue vascularization in transplanted site and the proliferation of transplanted hepatocytes. METHODS: Samples were gained at ten days after the pcDNA3 VEGF121 gene transfected hepatocyte was transplanted into the spleen of SD rats. The changes of microvascular density(MVD) counts in the transplanted site and proliferating cell nuclear antigen(PCNA) index of the transplanted hepatocytes were examined. RESULTS: In vitro protein expression of VEGF gene was enough to induce proliferation of human umbilical venous endothelial cells. In vivo VEGF gene transfected hepatocytes could form mass colonization of transplanted hepatocytes and reconstituted liver tissue. Difference of MVD counts in the transplanted site on all groups were not significant. P>0.05; PCNA index of pcDN-A3 VEGF121 gene transfection group increased significantly compared with pcDNA3 control group and no transfection group, P<0.01. PCNA index were 13.13 +/- 2.75, 4.75 +/- 1.58, and 4.63 +/- 1.41, suggesting that in vivo many factors affect the tissue vascularization, and expression of VEGF gene can promote not only proliferation of transplanted hepatocyte but formation of reconstituted liver tissue. CONCLUSIONS: VEGF ex vivo gene transfection is an effective way to induce reconstitution of transplanted liver tissue.

[NIES-90 microcystin producing algae strain genome library construction and one isolated gene analysis].

OBJECTIVE: To construct a plasmid microcystis genome library for microcystin (MC) pertaining gene family screening or microcystis strain specific gene screening. METHODS: Extracting genome DNA from MC producing strain NIES-90, then digesting DNA by HindIII and ligating to CIPase treated pUC18/HindIII fragment which later transformed to JM109. After checking the library by Xgal/Antibiotics plate, clones isolation, a few clones were sequenced and specific primers synthesized; finally, PCR was used to testify clone's NIES-90 strain specificity. RESULTS: Successfully constructed a NIES-90 genome plasmid library with following functions: a) cloning efficiency more than 95% by comparing the white/blue clones ratio in Xgal Plate; b) 200-700 bp average insert length with 3 x 10(5) independent clones by independent clones analysis; c) 10 times more coverage of original NIES-90 genome; d) strong strain specificity by cross PCR analysis of some clone's sequence in two different microcystis strains compared with FACHB-469. CONCLUSION: We successfully constructed a NIES-90 MC producing microcystis strain's genome plasmid library which could be used for screening MC pertaining gene family because of high strain specificity of some clones.

[Landscape structure and greenland spatial pattern in workshop district of Wuhan Iron and Steel Company].

The landscape structure and greenland spatial pattern in the workshop district of Wuhan Iron and Steel Company were investigated and analyzed on the basis of landscape ecological theory. The results showed that the main parts of the landscape structure were the artificial landscape elements such as building and concrete covering patches, highway and railway bed corridors, and industrial reservations. All of these artificial landscape elements accounted for 74.37% of patch area and 57.70% of patch dominance. In the landscape, the building and concrete covering land had the maximum percentage of patch area (56.43%) and the maximum dominance (35.59%). The special purpose greenlands had the highest fragmentation index (5.0591 ind.hm-2) and the wide-ranging distribution. The corridor density (13.78 km.km-2) was high, and the highway had more effects on landscape than the railway. All the results reflected not only the essential landscape features of industrial region, but also the rational trend of landscape structure in the workshop district of Wuhan Iron and Steel Company. In the greenland, the road greenbelts had the maximum patch number, area percentage (66.07%), and dominance (75.31%), the shelter forests being mainly composed of tree species had high patch average area (8454.45 m2) and environmental benefit, and the ornamental greenlands had abundant plant species and varied scenery. The greenland system was initially formed by four afforestation districts, using shelter forests and ornamental greenlands in large patch as a center connected by road greenbelts. It should be attached importance to the relatively high dominance of wild grasslands, the low area percentage of shelter forests and the imperfect road greenbelts to develop the uncovered lands and to improve the quality of the greenland system.

[Experimental study on resistance to cisplatin and taxol in human ovarian cancer SKOV3ip1 multicellular aggregates].

OBJECTIVE: To investigate the chemosensitivity of ovarian cancer SKOV3ip1 multicellular aggregates to cisplatin and taxol and to explore the possible mechanisms accounting for the effect. METHODS: Liquid overlay system was employed to obtain multicellular aggregates (MCA). We detected the resistance with trypan blue exclusion testing, clonogenic assay, cell cycle profiles and apoptosis with flow cytometry. RESULTS: MCA cells showed higher cell viability than monolayer cells (P = 0.045 and P = 0.003, respectively). After 40 mumol/L cisplatin exposure for 12 hours, no clone (> or = 50 cells) was formed. After 10 mumol/L taxol exposure for 12 hours, the clone formation showed significant difference in 100-cell group between multicellular aggregates and monolayer cells (P < 0.05). MCA cells in G0 + G1 phase was significantly increased (P = 0.003). After cisplatin exposure, the apoptosis rate of MCA cells were not significantly changed (P = 0.100), and so were cell cycle profiles. Taxol exposure brought about significantly decreased apoptosis rate in MCA cells (P = 0.012). Abrogation of G2 + M arrest was also showed in MCA cells (P = 0.002). CONCLUSIONS: SKOV3ip1 MCA cells show varying degree but increased resistance to cisplatin and taxol, in particular the latter. Cisplatin has nearly equal cytotoxicity to monolayer and MCA cells. Cell cycle redistribution, abrogation of G2 + M arrest and multicellular-mediated inhibition of apoptosis can partially account for the resistance.

[Electrochemical therapy and implanted ports treatment for unresectable carcinoma of body and tail of pancreas].

OBJECTIVE: To examine the efficacy and safety of electrochemical therapy (ECT) in treatment of patients with unresectable carcinoma of the body and tail of pancreas. METHODS: A total of 18 patients (14 men and 4 women; mean age 57.7) with unresectable carcinoma of the body and tail of pancreas were treated by ECT between May 1989 and June 1996 in our hospital (group A). Another 16 patients (12 men and 4 women; mean age 60.3) were treated by ECT plus intraperitoneal chemotherapy between July 1996 and June 1999 (group B). In group B, ECT was used during the operation. Meanwhile, 300 mg calcium leucovorin (CF), 500 mg 5-FU and 200 mg carboplatin were injected weekly into the abdominal cavity through the implanted ports. Pain was determined by using a 10-point visual analog pain scale (VAS) before and after the treatment, life quality periodically assessed according to the 5-step scale system, and tumor size measured using CT or MRI. RESULTS: All the patients were followed up for 5-30 months (mean = 10 months), showing that there were no operative deaths and such complications such as pancreatic fistula and bleeding in both groups. The tumor size in both groups was decreased to some degree. Ascites in 4 of 6 cases in group B were well controlled, whereas there was no changes in 6 cases of ascites in group A. In group A, the VAS basal score was reduced from 7.4 +/- 1.7 before to 2.66 +/- 1.05 after the treatment and the peak VAS from 9.3 +/- 0.9 to 3.71 +/- 0.96 (P < 0.05). In group B, they were decreased from 7.5 +/- 1.5 to 2.17 +/- 1.23 and 9.2 +/- 0.9 to 3.4 +/- 1.13, respectively (P < 0.01). Significant reduction of VAS score persisted for over 3 months after ECT. The satisfaction rate in group B was 37.5%, 62.5% and 25% in 7 days, 1 month and 3 months, respectively. CONCLUSIONS: ECT is a safe and effective procedure for treatment of unresectable carcinoma of the body and tail of pancreas and in relieving its pain and improving the life quality of patients. Supplementary injection of chemical drugs through drug delivery pump is helpful for control of ascites.

[Hypercoagulation aggravates renal dysfunction in patient with diabetic nephropathy].

OBJECTIVE: To investigate the relationship of hypercoagulation and renal dysfunction in patients with diabetic nephropathy(DN). METHODS: Forty six diabetes type II patients with nephrotic syndrome were divided into 3 groups according to 24 hr creatinine clearance (Ccr) as high, middle and low groups. Fourteen of the 46 patients received warfarin therapy. The parameters of coagulation and fibrolysis as well as renal function were examined. RESULTS: Patients with moderate and major renal dysfunction (M group and L group) displayed higher activities of coagulation than those with mild renal dysfunction (H group) did. Warfarin could obviously improve the status of coagulation and fibrolysis in patients with DN and delay the progress of renal dysfunction. CONCLUSION: Hypercoa-gulation was one of the causes of renal dysfunction in patients with DN.

[Gene dispersal risk of transgenic plants].

Gene flow is a key problem and top topic in risk assessment and management of transgenic plants. Five major study areas of gene dispersal in transgenic plants were reviewed in this paper, which include gene flow from transgenic plant to its wild species, characteristics of gene flow based on pollen, methods of doing experiment and risk assessment for gene flow, safety standards of risk assessment of transgenic plants, and long-term ecological impact of gene flow in the future. Some areas needed to be studied further were also proposed.

Hepatic artery angiography and embolization for hemobilia after hepatobiliary surgery.

OBJECTIVE: To evaluate the effectiveness of hepatic angiography and embolization in the diagnosis and treatment of hemobilia after hepatobiliary surgery. METHODS: Nine patients had upper gastrointestinal bleeding 7 days to 3 months after surgery. They underwent emergency hepatic artery angiography and were treated by embolization using Gelfoam particles only (8 patients) and Gelfoam particles plus microcoils (1 patient). RESULTS: Hepatic artery angiography revealed hepatic artery pseudoaneurysms in 3 patients, diffuse hemorrhage of the hepatic artery branches in 3, right hepatic artery-bile duct fistulas in 2, and hepatic artery-small intestine fistula in 1. Hemobilia was controlled with embolization in 7 patients, of whom 1 had recurrent bleeding 1 day after treatment. During the follow-up, 3 patients died of multiple organ dysfunction syndrome. Two patients whose hemorrhage could not be controlled due to technical reasons died several days later. CONCLUSION: When hemobilia after hepatobiliary surgery is suspected, patients should receive hepatic angiography as a first diagnostic procedure and be treated with minimally invasive procedure of selective embolization of the involved artery as soon as possible.

Isolation and cultivation of porcine hepatocytes for extracorporeal artificial liver support system.

OBJECTIVE: To develop procedures for the successful harvesting of large quantities of viable and functional pig liver cells from abattoir organs. METHODS: The procedure included partial liver lobe retrograde perfusion and mechanical/enzymatic digestion of the liver tissue, followed by separation of the hepatocytes, based on size and density, from contaminating cell types. RESULTS: Digestion of the partial liver lobe resulted in an average yield of 1.39 x 10(9) cells (9.9 x 10(8) cells/g liver) with an average viability of 92.5%. The yield and viability of cells were improved by dispase/collagenase resultant digestion. The emergence of blebby cells was blocked by supplying oxygen to the cell isolation buffers. Isolated hepatocytes seeded onto polystyrene surfaces remained viable and functional at a level comparable to that of rat hepatocytes, although their function decreased over time. CONCLUSIONS: Adult pig hepatocytes can be harvested with high yields and retain viability and differentiated function using this method. Abattoir pig livers can be an excellent source of hepatocytes for use as the biological component of artificial liver assist devices.

Activity identification of anti-caspase-3 mRNA hammerhead ribozyme in both cell-free condition and BRL-3A cells.

OBJECTIVE: To study the transcription effects and cleavage activities of rat caspase-3 specific hammerhead ribozyme (Rz107) in both cell-free conditions and BRL-3A cells. METHODS: Rat caspase-3 gene fragment was cloned into the pGEM-T EASY vector under the T7 promoter control. The 32P-labeled caspase-3 transcript was the target-RNA. Rz107 genes designed against caspase-3 mRNA were cloned into vector p1.5 between 5'-cis-Rz and 3'-cis-Rz. 32P-labeled ribozyme transcripts were incubated with target-RNAs at different conditions and autoradiographed after denaturing gel-electrophoresis. Rz107 was electroporated into BRL-3A cells and the Rz107 expression was analyzed by RT-PCR. RESULTS: In cell-free conditions, Rz107 was active at 37 degrees C. The optimal temperature was 50 degrees C. The Km and Kcat were 14.13 nmol/L and 2.31.min-1 respectively. Intracellular cleavage efficiency of Rz107 was 37%, as analyzed by RT-PCR. This indicated that the design of Rz107 was correct, and Rz107 had the activity of common enzymes. CONCLUSIONS: Rz107 in cell-free conditions possessed perfect specific catalytic cleavage activity, and it can also cleave the target RNA successfully in cells. The results illustrate the feasibility of ribozyme therapy as a potential alternative approach for treating liver disease caused by apoptosis.

[Significance of lung perfusion scanning with technetium labeled macroaggregated albumin and pulmonary function assay for diagnosis of early hepatopulmonary syndrome].

OBJECTIVE: To evaluate the values of pulmonary function assay and dynamic pulmonary perfusion imaging with technetium labeled macroaggregated albumin ((99m)TcMAA) in the early diagnosis of hepatopulmonary syndrome (HPS). METHODS: The pulmonary function assay and (99m)TcMAA scans were performed in 28 patients with HPS, 30 cirrhotic patients (CP) without HPS, and 21 healthy controls (HC). RESULTS: In the patients with HPS, PaO(2), SaO(2) and diffusion capacity for carbon monoxide of lungs (DLco) was significantly lower than that in CP (P<0.01) and HC (P<0.01), and alveolar-arterial gradient [P((A-a))O(2)] was significantly increased (P<0.001). Results from (99m)TcMAA scans showed that the radionuclides were distributed over the spleen, kidney, liver and brain, and the ratios of arterivenous shunt were significantly higher than that in CP (P<0.001) and NC (P<0.001). In cirrhotic patients, DLco significantly decreased (P<0.05), P((A-a))O(2) and shunt ratios increased (P<0.01 and 0.001). CONCLUSIONS: Pulmonary function assay and dynamic pulmonary perfusion imaging with (99m)TcMAA are sensitive methods for diagnosis of the early HPS.
