[Electron microscopic studies on pathogenesis of idiopathic pulmonary fibrosis].

OBJECTIVE: To observe ultrastructural changes and study the pathogenesis of idiopathic pulmonary fibrosis. METHODS: The clinical manifestations in 9 patients with idiopathic pulmonary fibrosis were selected. Transbronchial lung biopsy materials were obtained for light microscopic and electron microscopic examination. RESULTS: The major morphologic changes were as follows: histologically, alveolar inflammation and interstitial fibrosis were observed. electron microscopic findings were: 1. alveolar type I cells were degenerated, broken-down and desquamated, endothelial cells were swelled, with inter cellular tight junction shortened, alveolar type II cells hyperplastic, basement membrane thinned and deformed; 2. alveolar macrophages and interstitial macrophages were hyperplastic; 3. mast cells were infiltrated and degranulated; 4. electron-dense deposits were present at alveolar wall; 5. myofibroblasts, fibroblasts, collagen and basement membrane like material were hyperplastic. CONCLUSIONS: Electron microscopic observations show that the alveolar epithelial cells, endothelial cells and basement membrane are all injured, interstitial macrophages hyperplasia, mast cells infiltration and degranulation, electron-dense deposits at alveolar wall are present. These morphologic changes provide basis for formation of idiopathic pulmonary fibrosis.

[Detection of apoptosis exposed to 10-hydroxycamptothecin in T24 human urinary bladder cancer cells].

OBJECTIVE: To study whether 10-hydroxycamptothecin (HPT) can induce apoptosis in bladder cancer cell and establish methods for detecting apoptotic cells. METHODS: Human urinary bladder cancer cell line (T24) was exposed in vitro to different concentrations of 10-hydroxycamptothein for various lengths of time. Flow cytometry, Hochest 33258 and Hematoxylin staining were used to determine the induction of apoptosis after use of HPT. DNA gel analysis was also carried out to detect DNA fragmentation. RESULTS: Cell shrinage, nuclear fragmentation and condensed chromosomes showed that apoptosis can be induced by HPT within the concentration of 0.01 - 10 microg/ml. The flow cytometry analysis showed that the percentage of apoptotic cells were related to the concentration and the time of induction. T24 cell line exposed to HPT experienced internucleosomal DNA fragmentation by producing a typical ladder pattern on agarose gel electrophoresis. The detection of minimum exposure time for HPT-induced apoptosis in T24 cells showed that 3 hours of exposure to HPT were enough to trigger internucleosomal DNA fragmentation. Compared to Hochest 33258 staining, Hematoxylin staining was more easy, rapid and accurate to detect apoptosis. CONCLUSIONS: The induction of apoptosis exposed to HPT in T24 human urinary bladder cancer cells is a good model for further studying urinary bladder cancer. Hematoxylin staining is a useful method for detecting apoptosis.

[The expressions of Fas and Fas-L in human breast invasive ductal carcinomas and their surrounding tissues].

OBJECTIVE: To study the relationship of the expression of Fas and Fas-L with the pathogenesis and differentiation of breast invasive ductal carcinomas. METHODS: Both carcinoma tissues and their surrounding tissues from 50 patients with breast invasive ductal carcinomas were studied for the expression of Fas and Fas-L by immunohistochemical staining. RESULTS: The expression level of Fas in breast ductal carcinoma was significantly lower than that of tissues surrounding carcinomas, and the expression level of Fas-L in breast ductal carcinoma was significantly higher than that of tissues surrounding carcinomas. There was no significant difference in the expression of Fas and Fas-L in carcinomas with different differentiation grades. CONCLUSIONS: The expression of Fas is downregulated and the expression of Fas-L is upregulated in human breast invasive ductal carcinomas. Human breast invasive ductal carcinomas may enhance their development through downregulating the expression of Fas and upregulating the expression of Fas-L. The expression level of Fas and Fas-L has nothing to do with the differentiation of human breast invasive ductal carcinomas.

[A study of anterior segment structures in primary infantile glaucoma eyes by ultrasound biomicroscopy].

OBJECTIVE: To investigate anterior segment structures in primary infantile glaucoma (PIG) eyes by ultrasound biomicroscopy (UBM) in vivo. METHODS: UBM high frequency transducer (50 mHz) was used to measure and observe the anterior segment of 38 PIG cases (58 eyes). RESULTS: No matter the severity of the disease or the age at onset, the most significant characteristics of the diseased eyes are relative positional changes between scleral spur and angle apex. In three forth of the eyes, the scleral spur lay in the lateral or posterior-lateral site of the angle apex and in one forth of the eyes the site of the tip of scleral spur was parallel with the iris root insertion. The thickness and length of ciliary processes in the eyes of PIG were greater than that of the normal infantile eyes. The ciliary processes were anteriorly displaced, pulled towards the lens, and part of them got into contact with the iris. CONCLUSIONS: The relative positional changes between scleral spur and angle apex in diseased eyes indicate that the poor development of scleral spur and the iris anterior insertion are the basic pathogenesis in PIG.

[Qualitative and quantitative studies on Chinese drug maidong].

OBJECTIVE: To explore a better approach to controlling the quality of Maidong. METHOD: The differences between Ophiopogon japonicus and the plants belonging to Liriope genus were identified by TLC. The total saponins in plants collected from different areas were determined by spectrophotometry, using HClO4 as the staining agent. RESULTS: The quantity of Maidong markedly varies with producing areas. CONCLUSION: This method is convenient and effective, and the MeOH extract of samples can be determined directly after reaction with HClO4 without preliminary treatment. The RSD was 0.75% (n = 5).

[The standardized techniques for preparing samples used in pathologic diagnostic transmission electron microscopy].

Transmission electron microscopy (TEM) has been used widespreadly in the field of diagnostic pathology. Based on the practical domestic condition and aimed at fully satisfying the needs of the pathologic diagnostic TEM, a set of standardized techniques were established for preparing different kinds of samples. The techniques included the preparation and storage of fixatives and embedding media, the requirement of sampling, the 5 h and 24 h embedding procedures for fresh specimens, the embedding procedure for suspension specimens and specimens collected from fine needle aspiration biopsy, as well as the procedures for preparing samples taken from paraffin embedded blocks or paraffin sections. In practical use, we found that the advantage of these techniques was that the ultrastructures of the tissues and cells were well preserved and they perfectly met the needs for diagnosis of tumors, kidney diseases, et al. The main points of the standardized techniques have been discussed in detail in this paper.

[DNA typing for HLA-A by polymerase chain reaction with sequence-specific primers].

The authors have established a method of DNA typing for HLA-A antigen in Chinese by using polymerase chain reaction with sequence-specific primers (PCR-SSP). HLA-A alleles were successfully typed in all clinical samples and 36 standard DNAs by PCR-SSP. A total of 635 A antigens were typed in 345 donor-recipients, including 55 homozygotes and 290 heterozygotes. Twenty alleles of HLA-A were accurately distinguished by DNA typing. No false positive or false negative typing results were obtained. Reproducibility was 100%. The overall time of DNA typing was 5 hours. The typing results were concordant with the results of UCLA Tissue Typing Lab. Comparative study of HLA-A typing was carried out using PCR-SSP and standard serology. The results showed that the discrepancy rate of serology was 9.0%, containing 13 individuals incorrectly interpreted by serology and 16 serological "blanks" turned out to be a definable alleles by PCR-SSP. In conclusion, DNA typing for HLA-A by PCR-SSP has proved to be a high-resolution, high-specificity, rapid and accurate technique, suitable for clinical application with a greater precision than serology.

[Study of the immunological tolerance induced by peripheral intravenous infusion of donor-specific antigens].

In the experiment, donor specific tolerance was induced by DST, DSSL and DSBM through peripheral intravenous transfusion in rat cervical cardiac transplantation model. The results showed there were significant differences in graft survival time between the blank control group and the other groups (P < 0.05). The mean graft survival time days of the DST, DSSL and DSBM groups were 28.9 +/- 13.7, 42.9 +/- 6.6 and 40.3 +/- 11.6 days, respectively. The effect of component infusion was obviously better than that of DST (P < 0.05), but no statistical difference was observed between DSSL and DSBM. CsA gave a noticeable synergistic effect to the induction of immunological tolerance. MLR and CD8+ T cell were remarkably reduced(P < 0.05) in the cases of component transfusion. This study suggested that components transfusion may be of clinical use for donor-specific tolerance inducing.

[Light and electron microscope observations of heart and vascular tissues in animals acutely infected with tubercle bacilli].

The objective of this study was to observe the heart and vascular pathological lesions of some animals acutely infected with H37Rv tubercle bacilli. C57 BL/6N mice and rabbits were used for the establishment of the models. Light sections were stained with HE, Z-N, MCT and BCGS-P. Ultrathin sections were stained with uranyl and lead citrate and examined by electron microscope. The results showed that tubercle bacilli were present in heart and vascular tissues. Myocardial fibre, pericardium and vascular tissues had various degrees of lesions. The lesions of myocardial fibre and pericardium were more serious than that of the vascular tissues. These findings suggest that the toxin of the tubercle bacilli can damage the tissues of the infected animals, and the excessive reproduction can be also a factor detrimental to the infected tissues.

[A method of preparing the section of eye tissue for transmission electron microscopy].

A method of preparing the section of eye tissue for transmission electron microscopy (TEM) is recommended. The tissue was prefixed with a mixed solution of 4% paraformaldehyde and 2.5% glutaraldehyde and followed by softening in 3% EDTA solution for 20 minutes, then the tissue was fixed in 1% osmium tetroxide, dehydrated in series acetone, infiltrated in Epox 812 for a longer, and embeded. Ultrathin sections were cut with glass knives or diamond knife, stained with uranyl acetate and lead citrate and examined with H600-IV. The advantage of this method is that the ultrastructures of tissues are well preserved without any damage and distortion. The main points of the procedure of preparation have been discussed.

[Studies on detection of Plasmodium falciparum and Plasmodium vivax in blood samples by multiplex polymerase chain reaction].

AIM: To establish a sensitive and specific PCR-based method to detect Plasmodium falciparum and P. vivax in blood samples in a single amplification reaction. METHODS: Malaria parasite DNA in blood was amplified by the multiplex polymerase chain reaction using two sets of primers derived from the P. f. moderately-repetitive DNA sequence and COIII gene of P.v. RESULTS: A 206-bp product for P. f. and a 370-bp product for P.v. were amplified by multiplex PCR, being able to detect parasitemia level as low as 5 x 10(-7) for P. f. and 1.02 x 10(-6) for P. v. and having no cross-reaction with human leucocyte DNA. A total of 783 blood samples on the filter paper collected from patients attending to malaria clinics in malaria endemic areas were detected. The positive rate of multiplex PCR was 85.8%, the misdiagnosis rate was 0, and the under-diagnosis rate was 0.1%, while these three rates of microscopic examination were 84.9%, 3.1% and 1.0%, respectively. The concordance between the two methods was 95.8%. CONCLUSION: The multiplex PCR method made the malaria detection more sensitive and specific than the microscopic examination and should be suitable for the diagnosis of malaria in mixed endemic areas, large-scale epidemiological studies, follow-up of drug treatment and donor blood screening.

[Changes in chloroquine resistance of Plasmodium falciparum in Hainan province].

AIM: To observe the variation in resistance of Plasmodium falciparum after the cessation of chloroquine medication. METHODS: WHO standard in vitro microtest and in vivo test were used. RESULTS: In vitro test showed that the rate of chloroquine-resistant P. falciparum dropped from 97.9% in 1981 to 26.7% in 1997 (P < 0.001). The mean concentration of chloroquine for complete inhibition of schizont formation declined from 10.46 +/- 7.14 pmol/ul blood in 1981 to 1.63 +/- 1.47 pmol/microliter blood in 1997(P < 0.001). In vivo test showed that the rate of chloroquine-resistant P. falciparum decreased from 84.2% in 1981 to 18.4% in 1997(P < 0.001). The proportion of R III cases to the total resistant cases dropped from 53.1% in 1981 to 14.3% in 1997. CONCLUSION: A tendency of progressive decline of resistance of the parasite was revealed after the cessation of chloroquine medication.

[Remote effects of local injection of botulinum toxin type A].

OBJECTIVE: To assess the severity and temporal profile of remote effects of botulinum toxin type A, BTX-A (Botox from Allergan Inc, USA and CBTX-A made by Lanzhou Biological Products Institute, China) injected locally on neuromuscular junction. METHODS: Patients had enrolled in a prospective study for their movement disorders, 18 cases with Botox and 22 cases with CBTX-A were studied. Single fiber electromyography (SFEMG) in the extensor digitorum communis muscle or tibialis anterior muscle was performed before and 2-3 weeks, 5-8 weeks, 4-5 months after injection of Botox or CBTX-A. RESULTS: It was measured totally 119 times. Significant increase of jitter was demonstrated 2-3 weeks after injections in both groups and MCD was in direct proportion to dose of injections. Fiber density value increased at the same time or later and lasted until 4-5 months after injections. CONCLUSIONS: Subclinical effects on neuromuscular transmission of remote uninjected muscles after injections of Botox and CBTX-A are demonstrated, which indicates that the toxin spread distantly from the site of injection. SFEMG is valuable for studying the pathogenesis of the remote effects of BTX-A and for avoiding reinjection too frequently.

[Effects of extract from Pegasus laternarius on the weight of immune organs and the antistress in mice].

Extract from Pegasus laternarius Cuvier (PLE) could increase the splenic and thymic weight in immunity-suppressed mice induced by hydrocortisone or cyclophosphamide and could increase the thymic weight in senile mice. PLE could prolonged the survival time of anoxic mice under normal pressure or chemically anoxic mice poisoned by potassium cyanide. It could also prolong the survival time of mice swimming in ice-water and enhance the tolerance of mice without food to eat and water to drink.

[Extraction and purification of acidic mucopolysaccharide from Holothuria atra].

Mucopolyaccharide with molecular weight of 10253 Da was extracted and purified from fresh Holothuria atra Jaeger by means of enzymic and alkaline hydrolysis, potassium acetate and ethanol fractional precipitation. It was tested to be purified ingredient with agarose electrophoresis. The percentage content of galactosamine, glucuronic acid, fucose and sulfate in the mucopolysaccharide was 16.12%, 17.88%, 11.66% and 23.52% respectively.

[A new absorbance ratio-derivative method and its application to simultaneous determination of two components in a mixture].

In this paper, the principle of a new absorbance ratio-derivative method was described. The experimental methods of derivation with subtraction technique on ordinary spectrophotometer and simultaneous determination of phenol and resorcin were studied. The result is satisfying. The recovery of resorcin and phenol is in the range of 99.4%-104.2% (n = 25, RSD = 1.1%) and 97.1%-102.8% (n = 25, RSD = 1.6%) respectively.

Different systems in human brain are involved in presemantic discrimination of pictures as revealed by event-related potentials.

It is uncertain if the several brain areas in response to presemantic picture processing are functionally homogeneous. Subjects were asked to discriminate visual stimulus pairs and event-related potentials (ERPs) were recorded from the scalp. A line-drawing picture (S1) was followed by another stimulus (S2) 600 ms later. S2 was the same as S1 in condition 1, a reversed picture of S1 in condition 2, the corresponding Chinese word in condition 3 and the syllabic alphabet of S1 in condition 4. Amplitudes of ERPs over right posterior scalp with time window from 102-180 ms after the onset of S2 in condition 1 and 2 were significantly larger than condition 3 and 4. The N2 component on the right anterolateral scalp with time window from 182-240 ms was significantly larger in condition 1 than other conditions, whereas the N2 on the left anterolateral scalp with time window from 262-350 ms in condition 2, 3 and 4 was significantly larger than condition 1. It is suggested that pictures and words are processed in different systems at the presemantic stage: repetitive and physically deviant stimuli evoke different responses in different brain regions.

Analysis of CAG/CTG repeat size in Chinese subjects with schizophrenia and bipolar affective disorder using the repeat expansion detection method.

BACKGROUND: Family studies of schizophrenia and bipolar affective disorder provide evidence for genetic anticipation, which (in common with a number of mendelian disorders), may be caused by triplet repeat expansion. This hypothesis is strengthened by evidence from repeat expansion detection (RED) analysis revealing association between the psychoses and long CAG/CTG trinucleotide repeats. METHODS: We performed RED on Han Chinese subjects with schizophrenia (82), bipolar affective disorder (43), and normal controls (61), using a CTG10 oligonucleotide. RESULTS: Comparison between cases and controls revealed no significant association between long repeats and affected status. We also found no detectable association with age at onset and repeat length in either bipolar affective disorder or schizophrenia. Overall, the size distribution of CAG/CTG repeats in Chinese subjects was not significantly different from those reported previously for Caucasian subjects. CONCLUSIONS: These findings indicate that CAG/CTG repeat expansion is not likely to be a major etiological factor for psychosis in Chinese populations.
