A bifunctional transposon mini-Tn5gfp-km which can be used to select for promoter fusions and report gene expression levels in Agrobacterium tumefaciens.

A mini-Tn5 transposon derivative, mini-Tn5gfp-km, has been constructed which contained a promoter-less artificial operon consisting of two open reading frames, green fluorescent protein (GFP) and neomycin phosphotransferase II (NptII). When this transposon was used to mutagenize Agrobacterium tumefaciens, all the mutants selected in the presence of kanamycin exhibited GFP expression, which could be conveniently monitored by a fluorometer. The transposon appeared to be bifunctional and could provide both selection and reporter functions. Even the mutants showing minimal levels of GFP expression were still resistant to kanamycin. This suggests that this transposon can be used to select for insertions downstream of both weak and strong promoters, as long as the insertions themselves are non-lethal. This system was used to identify A. tumefaciens genes that were upregulated in response to acidic pH. Screening only 20 colonies led to identification of two promoters that were specifically induced by low pH and one promoter that was specifically induced by acetosyringone in a minimal medium of pH 5.5.

Localization and possible functions of phospholipase D isozymes.

The activation of PLD is believed to play an important role in the regulation of cell function and cell fate by extracellular signal molecules. Multiple PLD activities have been characterized in mammalian cells and, more recently, several PLD genes have been cloned. Current evidence indicates that diverse PLD activities are localized in most, if not all, cellular organelles, where they are likely to subserve different functions in signal transduction, membrane vesicle trafficking and cytoskeletal dynamics.

Kinetic properties and metal content of the metallo-beta-lactamase CcrA harboring selective amino acid substitutions.

The crystal structure of the metallo-beta-lactamase CcrA3 indicates that the active site of this enzyme contains a binuclear zinc center. To aid in assessing the involvement of specific residues in beta-lactam hydrolysis and susceptibility to inhibitors, individual substitutions of selected amino acids were generated. Substitution of the zinc-ligating residue Cys181 with Ser (C181S) resulted in a significant reduction in hydrolytic activity; kcat values decreased 2-4 orders of magnitude for all substrates. Replacement of His99 with Asn (H99N) significantly reduced the hydrolytic activity for penicillin and imipenem. Replacement of Asp103 with Asn (D103N) showed reduced hydrolytic activity for cephaloridine and imipenem. Deletion of amino acids 46-51 dramatically reduced both the hydrolytic activity and affinity for all beta-lactams. The metal binding capacity of each mutant enzyme was examined using nondenaturing electrospray ionization mass spectrometry. Two zinc ions were observed for the wild-type enzyme and most of the mutant enzymes. However, for the H99N, C181S, and D103N enzymes, three different zinc content patterns were observed. These enzymes contained two zinc molecules, one zinc molecule, and a mixture of one or two zinc molecules/enzyme molecule, respectively. Two enzymes with substitutions of Cys104 or Cys104 and Cys155 were also composed of mixed enzyme populations.

The tryptophan biosynthesis gene cluster trpCDEGFBA from Pyrococcus kodakaraensis KOD1 is regulated at the transcriptional level and expressed as a single mRNA.

The entire gene cluster encoding enzymes involved in biosynthesis of L-tryptophan in the hyperthermophilic archaeon Pyrococcus kodakaraensis KOD1 has been cloned and sequenced. Seven ORFs, which encode indole-3-glycerol phosphate synthase (trpC), anthranilate phosphoribosyltransferase (trpD), the two subunits of anthranilate synthase (trpEG), phosphoribosyl anthranilate isomerase (trpF) and the two subunits of tryptophan synthase (trpAB), were identified. The gene order is trpCDEGFBA, covering a region of 6045 bp. In order to confirm the function of the gene products, we expressed the first gene, Pk-trpC, in Escherichia coli. The protein product was purified, and was found to show the expected indole-3-glycerol phosphate synthase activity, with a temperature optimum of 85 degrees C. We could clearly identify a single mRNA transcript by Northern analysis using probes in the central and 3'-regions of the gene cluster, indicating that the gene cluster is transcribed as an operon. A significant increase in trp mRNA level was observed in cells grown in medium depleted of L-tryptophan, compared to cells grown in medium supplemented with L-tryptophan, indicating that expression of the gene cluster is regulated at the transcriptional level.

Pseudomonas syringae pv tomato induces the expression of tomato EREBP-like genes pti4 and pti5 independent of ethylene, salicylate and jasmonate

The tomato genes Pti4 and Pti5 encode ethylene-responsive element binding protein-like transcription factors that bind to the GCC box, a conserved cis-element in many defense-related genes. The Pti proteins have previously been shown to interact with the tomato disease resistance protein Pto. Here we report that the expression of both Pti4 and Pti5 are induced by a virulent strain of Pseudomonas syringae pv tomato. The expression of Pti5 is further enhanced by the interaction of the Pto gene in tomato and the corresponding avrPto gene in the bacterium. The enhancement of Pti5 expression by Pto-avrPto interaction requires a functional Prf gene in the plant. Pti5 appears to be expressed specifically during biotic stresses, suggesting a specific role in plant defense. Pti4 and several EREBP-like genes are induced by ethylene, salicylate and wounding. However, the Pseudomonas bacterium induced a wild-type level of Pti4 and Pti5 transcripts in tomato plants carrying the nahG transgene, the Nr mutation, or the def1 mutation. In addition, the ethylene action inhibitor norbornadiene did not inhibit the induction of Pti4 and Pti5 either in the compatible or incompatible interactions. The results suggest that the Pseudomonas bacterium induces Pti4 and Pti5 expression through a pathway independent of salicylic acid, ethylene and jasmonic acid.

Overexpression of Pto activates defense responses and confers broad resistance.

The tomato disease resistance (R) gene Pto specifies race-specific resistance to the bacterial pathogen Pseudomonas syringae pv tomato carrying the avrPto gene. Pto encodes a serine/threonine protein kinase that is postulated to be activated by a physical interaction with the AvrPto protein. Here, we report that overexpression of Pto in tomato activates defense responses in the absence of the Pto-AvrPto interaction. Leaves of three transgenic tomato lines carrying the cauliflower mosaic virus 35S::Pto transgene exhibited microscopic cell death, salicylic acid accumulation, and increased expression of pathogenesis-related genes. Cell death in these plants was limited to palisade mesophyll cells and required light for induction. Mesophyll cells of 35S::Pto plants showed the accumulation of autofluorescent compounds, callose deposition, and lignification. When inoculated with P. s. tomato without avrPto, all three 35S::Pto lines displayed significant resistance and supported less bacterial growth than did nontransgenic lines. Similarly, the 35S::Pto lines also were more resistant to Xanthomonas campestris pv vesicatoria and Cladosporium fulvum. These results demonstrate that defense responses and general resistance can be activated by the overexpression of an R gene.

Intravenous angiotensinogen antisense in AAV-based vector decreases hypertension.

Angiotensinogen (AGT) has been linked to hypertension. Because there are no direct inhibitors of AGT, we have developed antisense (AS) inhibition of AGT mRNA delivered in an adeno-associated virus (AAV)-based plasmid vector. This plasmid, driven by the cytomegalovirus promoter, contains a green fluorescent protein reporter gene and AS cDNA for rat AGT. Transfection of the plasmid into rat hepatoma cells brought a strong expression of the transgenes and a significant reduction in the level of AGT. In the in vivo study, naked plasmid DNA was intravenously injected into adult spontaneously hypertensive rats at different doses (0.6, 1.5, and 3 mg/kg). Expression of AGT AS mRNA was present in liver and heart, and it lasted longer in the liver. All three doses produced a significant decrease in blood pressure (BP). BP decreased for 2, 4, and 6 days, respectively. The lowest dose decreased BP by 12 +/- 3.0 mmHg, whereas the higher doses decreased BP by up to 22.5 +/- 5.2 mmHg compared with the control rats injected with saline (P < 0.01). The injection of the plasmid with liposomes produced a more profound and longer reduction (8 days) in BP. Consistent changes in plasma AGT level were observed. Sense plasmid had no effect. No liver toxicity was observed after injection of AS plasmid with or without liposomes. Our results suggest that the systemic delivery of AS against AGT mRNA by AAV-based plasmid vector, especially with liposomes, may have potential for gene therapy of hypertension and that further studies with the plasmid packaged into a recombinant AAV vector for a longer-lasting AS effect are warranted.

[CAG trinucleotide mutation detection in patients with hereditary spinocerebellar ataxia].

OBJECTIVE: To assess the frequency of the SCA1, SCA2,SCA3/MJD, SCA6, SCA7 and DRPLA CAG trinucleotide repeat expansions(CAG)n among individuals diagnosed with hereditary spinocerebellar ataxia(SCA) from Chinese families. METHODS: The SCA1, SCA2, SCA3/MJD, SCA6, SCA7 and DRPLA(CAG)n mutations were detected by polymerase chain reaction (PCR), denaturing polyacrylamide gel electrophoresis and silver staining technique in 167 patients with autosomal dominant SCA from 85 Chinese families and 37 sporadic SCA patients. RESULTS: Among 85 families, four families(4.70%) had seven SCA1 patients with the CAG repeat expanded to 53 to 62 repeats, five (5.88%) had twelve SCA2 patients with the CAG repeat expanded to 43 to 47 repeats, and 41 (48.23%) had 83 SCA3/MJD patients with the CAG repeat expanded to 68 to 83 repeats. Analysis of the mutation in these families showed a strong negative correlation between the size of the expanded CAG repeat and the age of disease onset. None of the SCA patients were positive for SCA6, SCA7, or DRPLA. Nor was any of the sporadic SCA patients positive for the CAG repeat expansion in the SCA1, SCA2, SCA3/MJD, SCA6, SCA7, or DRPLA gene. CONCLUSION: The frequency of SCA3/MJD is substantially higher than that of SCA1 and SCA2 in the autosomal dominant SCA from Chinese families. Chinese SCA3/MJD patients are non-Portuguese. Clinical expressions of the various SCAs overlap one another and hence can make the phenotype-based diagnostic classification inaccurate in many instances. It is important for SCA clinical studies to make an SCA gene diagnosis and genotype analysis.

[Study on the reversal of cancer multidrug resistance by Chinese medicine Fw13-te41 in nude mice].

We have reported that three reversal agents were sifted out from 32 Chinese galenicals through a series of cell culture tests. Among them, Fw13-te41 has the best effect of reversal cancer multidrug resistance (MDR) in vitro. In this study, the reversal action of Fw13-te41 in vivo was studied on the animal model of nude mice with human leukemia k562/ADR. Twenty SPF BALB/c-nu/nu nude mice with xenograft tumor were randomly divided into the control group (n = 6), VCR group [intraperitoneal (i.p.) VCR 250 micrograms/week, n = 5], VCR + Fw13-te41 group (i.p VCR 250 micrograms/week + Fw13-te41 0.2 ml/day, equivalent to crude drug 10 g/kg, n = 5), and Fw13-te41 group (i.p Fw13-te41 0.2 ml/day, equivalent to crude drug 10 g/kg, n = 4). After 18 days, the rate of tumor inhibition (RTI) of VCR group was 19.79%, but the RTI of VCR + Fw13-te41 group was as high as 86.95% (P < 0.05). There results demonstrate that the Chinese medicine Fw13-te41 has an evident reversal action of malignancy MDR in vitro and in vivo.

Carpal tunnel syndrome: a retrospective analysis of 262 cases and a one to one matched case-control study of 61 women pairs in relationship between manual housework and carpal tunnel syndrome.

OBJECTIVES: To get a more comprehensive recognition about carpal tunnel syndrome (CTS), especially the manual housework as its risk factor, and to facilitate early diagnosis and proper treatment. METHODS: 262 CTS patients (396 CTS hands) were analyzed retrospectively. A 1:1 matched case-control study of 61 woman pairs in relationship between manual housework and the occurrence of CTS was carried out. RESULTS: In 262 CTS patients, 84% were female, and the dominant hand was more frequently affected; repetitive and forceful movement of the hand and wrist might be associated with CTS. Typical clinical manifestations include pain and paresthesia in the median nerve territory, but 75.3% of our 396 CTS hands had all five digits involved. Conduction abnormalities appeared selectively in the median nerve distal to the wrist. In the case-control study, for manual washing, rolling or kneading dough, and knitting woolen sweater, the odds ratios (OR) of CTS between high and low intensities were 3.86 (95% confidence interval 1.79-8.33, chi 2 = 11.76, P < 0.01), 6.25 (95% confidence interval 2.50-15.63, chi 2 = 15.21, P < 0.01) and 1.13 (95% confidence interval 0.57-2.22, chi 2 = 0.125, P > 0.05) respectively; and those between long and short duration (i.e. the number of years engaging in these manual housework) were 2.33 (95% confidence interval 0.63-8.64, chi 2 = 1.6, P > 0.05), 1.88 (95% confidence interval 0.81-4.38, chi 2 = 2.13, P > 0.05) and 1 (chi 2 = 0, P > 0.05). CONCLUSIONS: The diagnosis of CTS requires confirmation of illness history, symptoms and signs with objective electrodiagnostic tests. Manual washing and rolling or kneading dough might be associated with the onset of CTS.

Non-invasive assessment of myocardial risk and infarct area in canine model of myocardial reperfusion by intravenous contrast echocardiography.

OBJECTIVE: To evaluate the newly developed perfluoropropene-filled echo-contrast agent (FCT-188) in non-invasive assessment of risk areas (RA) and infarct areas (IA) with intravenous myocardial contrast echocardiography (MCE) in canine model of ischemia followed by reperfusion. METHODS: Eight chest-opened Beagle dogs with a 90-minute ischemia followed by a 240-minute reperfusion were studied. MCE was performed after a bolus injection of FCT-188 (0.025 ml/kg, i.v.) into a superficial vein of the forelimb at baseline, 20 minutes after occlusion, and 4 h after reperfusion to non-invasively assess the left ventricular myocardium area (LVMA), myocardial ischemic risk area (RA), and infarct area (IA) in a short-axis view of left ventricle. The accuracy of detecting myocardial perfusion with intravenous MCE was further assessed by in vitro myocardial staining of the matched cross-sections. Both RA and IA were expressed as percent of LVMA. RESULTS: LVMA, RA, IA, and IA/RA ratio were accurately assessed by MCE (LVMA: 6.60 cm2 +/- 0.76 cm2; RA: 35.7% +/- 6.68%; IA: 21.0% +/- 13.2%; IA/RA: 60.3% +/- 31.4%; n = 7) as compared with those of the matched cross-section (LVMA: 6.81 cm2 +/- 0.73 cm2, P = 0.062; RA: 35.3% +/- 9.9%, P = 0.84; IA: 25.10% +/- 14.5%, P = 0.07; IA/RA: 68.0% +/- 22.2%, P = 0.28, respectively). There was a significant correlation of MCE-assessed IA/RA ratio and its corresponding pathologically determined finding in vitro (Y = 1.21 x -21.6, r = 0.73, P = 0.015). No significant changes of electrocardiogram (ECG), mean artery pressures (MAP), pulmonary artery pressures (PAP), and pulmonary artery wedge pressures (PAWP) were found between pre- and post-intravenous injection of FCT-188 at each time point. CONCLUSION: These indicate that FCT-188 can be used to assess risk areas and infarct areas accurately and non-invasively with intravenous MCE in the canine model of a 90-minute ischemia followed by a 240-minute reperfusion and might have potential significance for non-invasive assessment of myocardial reperfusion clinically.

[Immune activation genes expression in CD28-costimulated lymphocytes and its application as a diagnostic tool in kidney transplantation].

OBJECTIVE: To investigate the detection of immune activation genes expression as an indicator for the diagnosis of immunological rejection in renal transplantation. METHODS: In 61 renal allograft recipients, the immune activation genes expression of CD28-costimulated peripheral blood mononuclear cells (PBMC) was studied by RT-PCR. RESULTS: The mRNA expression positive rate of IL-10, CD40L, Fas, Perforin, IL-2, IFN-gamma, FasL, bcl-xL, Granzyme B (GB), TGF-beta 1 and IL-4 was 91.8%, 70.5%, 88.5%, 96.7%, 41%, 54.1%, 60.7%, 67.2%, 67.2%, 54.1% and 32.8% respectively. IL-2, IFN-gamma, FasL and Granzyme B were screened out as indicators for the diagnosis of allograft rejection. Any three or more of IL-2, IFN-gamma, FasL, and Granzyme B genes expression could be used as a diagnostic tool for rejection with much favorable sensitivity (93%) and specificity (75%). CONCLUSION: The detection of IL-2, IFN-gamma, FasL and Granzyme B genes expression is beneficial for the evaluation of immunological state in renal allograft recipients. This non-invasive method can be used for clinical diagnosis of allograft rejection.

[HLA-amino acid residue matching standard and immunogenic response].

OBJECTIVE: To provide a standard for HLA-amino acid residue matching (Res M) for Chinese population and to evaluate its immunogenic response and sensitization to allograft. METHODS: Based on the setting up of DNA typing technique and the analysis of the distribution of HLA antigen frequencies, a new matching policy of Res M of 17 residues (10 amino acid residues for class I and 7 DR supertypic groups for class II) was presented and prospectively applied to 163 first-cadaver kidney transplants. Comparing with the 6-antigen matching program, the immunogenic response and sensitization of Res M to allograft were assayed using the mixed lymphocyte culture (MLC), cytokine in culture liquid, immune competent cells in the peripheral blood and specific anti-HLA IgG antibodies with PRA-STAT technique. RESULTS: The rate of Res-matched transplants increased significantly from 1.2% by Ag M to 39.9%. Immunogenic response and sensitization to allograft of recipients with Res-matched, containing index of stimulation (SI) of MLC, gamma-IFN representing Th1 cell, IL-10 representing Th2 cell, CD4+, CD8+, CD28+ T cell and the ratio of CD4/CD8, as well as the level of sHLA-IgG representing sensitization to allograft, were similar to those of Ag-matched transplants. Those parameters showed no difference between Res-matched and Ag-matched recipients, but showed significant difference in Res-mismatched or Ag-mismatched transplants. CONCLUSION: The New Res M can increase the rate of Res-matched transplants by a big margin, minimize the sensitization and immunogenic response to allograft, and be suitable to the clinical application to organ transplantation in Chinese Han population.

[A comparison of serological and high resolution DNA methods for HLA-B40 cross-reactive groups typing].

OBJECTIVE: To compare the high-resolution DNA typing method with serological method for HLA-B40 cross-reactive groups (CREG) typing in Chinese population. METHODS: A total of 199 consecutive samples were entered into the study, including 177 identified serologically as B40 CREG positive, 12 ambiguous and 10 negative controls. Double-blind typing for HLA-B40 CREG was carried out using PCR-SSP high-resolution DNA typing and serological one-step monoclonal antibody technique, respectively. RESULTS: In 3 of the samples serological typing could not be performed owing to poor cell viability, while in 1 PCR-SSP typing could not be done owing to poor DNA. Among samples that were successfully typed by both methods, all alleles of HLA-B40 CREG could be accurately distinguished by PCR-SSP. It was proved that PCR-SSP was a high-sensitivity and high-specificity technique. However, serology showed 11.2% misassignments. The overall time for serological typing was 2 hours, and for DNA typing, 5 hours. The percentages of HLA-B40 CREG alleles in Chinese population were 67.4% for B60, 20.8% for B61, 9.6% for B4801 and 2.2% for B4005. CONCLUSION: High resolution PCR-SSP typing for HLA-B40 CREG could be used in routine clinical practice with a greater precision than serology.

[Preliminary study on the antitumor immuno-protective mechanism of beta-elemene].

OBJECTIVE: To study the antitumor effects and their molecular mechanism of beta-elemene, which is one of the effective monomers of Oleum Curcuma aromatica, a Chinese anticancer medicament. METHODS: The expression of membrane protein HSP70 on H22 cells treated with beta-elemene or mitomycin C(MMC) was analysed by immunofluorescence and flow cytometric techniques and the antitumor effects of the treated H22 cells was examined in C57BL/6 mice. RESULTS: The expression rate or intensity of membrane protein HSP70 of H22 cells treated with beta-elemene, and those treated with MMC was both significantly higher than that of the untreated H22 cells (P < 0.001). Increase in HSP70 expression was most marked when H22 cells were treated with beta-elemene and MMC in combination, with an expression rate of 95.1%. Heat shock treatment of the H22 cells combined with other stress-inducing factors could further increase the HSP70 expression intensity. Specific antitumor immunity could be elicited in C57BL/6 mice immunized with beta-elemene-treated H22 cells, and the effect could be partially blocked by anti-HSP70 mAb. In mice immunized with beta-elemene-treated H22 cells, tumor developed in 3 of 11 mice, while in the immunized mice given anti-HSP70 mAb, 5 out of 10 mice developed tumor, and all of the control mice developed tumor. The average tumor weight (g) on day 28 after challenge with untreated H22 cells was 0.17 +/- 0.33, 0.66 +/- 0.77 and 1.11 +/- 0.58 in the three groups of mice, respectively. CONCLUSION: beta-elemene increases tumor cell immunogenicity by inducing, at least in part, elevated expression of heat shock protein 70 on tumor cell surface.

[Detailed deletion mapping of chromosome 9p21-22 in nasopharyngeal carcinoma].

OBJECTIVE: To further refine the extent of deletion on chromosome 9p21-22 in nasopharyngeal carcinoma(NPC) and provide evidence for discovering new tumor suppressor genes. METHODS: Loss of heterozygosity (LOH) on chromosome 9p21-22 was analyzed in 25 paired blood and tumor samples by using 11 high-density microsatellite polymorphic markers. RESULTS: Of the 25 cases, 17(68.0%) showed LOH at one or more loci. Higher frequencies of LOH were found at four loci: D9S161(35.0%), D9S1678(31.6%), D9S263 (33.3%) and D9S1853(33.3%). In 6 cases, contiguous stretch of allelic loss was found. CONCLUSION: The minimal common region of deletion might be defined between D9S161 and D9S1853 (approximately 2.7 cM in length) at 9p21.1, suggesting that inactivation of one or more tumor suppressor genes located in this region may be an important step in NPC.

[Study on early application of Chinese medicinal herbs after total gastrectomy].

OBJECTIVE: To evaluate the effect of Chinese medicinal herbs on digestive function, nutritional status, immune function and complication rate in patients after total gastrectomy. METHODS: Thirty-nine patients of gastrectomy were divided into two groups and were intubated into small intestine for administering postoperatively. Twenty-one of them belonged to the treated group and were dripped with Chinese medicinal herbs intraluminally during the early postoperative stage, while 18 cases of the control group were dripped with 0.9% sodium chloride. RESULTS: The digestive function of the treated group recovered rapidly, and the nutritional status and immune function improved markedly as compared with those of the control group (P < 0.001-0.05). CONCLUSION: Early application of Chinese medicinal herbs after total gastrectomy could promote the digestive function to recover to normal, the nutritional status and immune function to improve. The occurrence of complication was reduced.

[The diagnostic significance of antineutrophil cytoplasmic antibodies in ulcerative colitis].

OBJECTIVE: To inquire into the diagnostic significance of antineutrophil cytoplasmic antibodies (ANCA) in ulcerative colitis (UC). METHODS: Serum ANCA from 58 UC patients, 43 non-UC patients and 58 healthy donors were detected with indirect immunofluorescence (IIF), enzyme linked immunosorbent assay (ELISA)and Western blot. RESULTS: The sensitivity and specificity of the presence of ANCA in the diagnosis of UC were 37.93% and 100% respectively. In the group of UC, patients with mild, moderate and severe clinical manifestations had positive ANCA rates of 17.65%, 41.67% and 52.94% respectively. The occurrence rates of mucosal vasculitis and grade III-V mucosal inflammation were 78.95% and 78.95% respectively in ANCA-positive group, but 37.04% and 44.44% in ANCA-negative group. The levels of the binding of the five kinds of ANCA antigens i.e. myeloperoxidase (MPO), bactericidal/permeability increasing protein (BPI), lactoferrin (LF), cathepsin G (CG) and proteinase-3 (PR-3) with the serum of UC patients were 13.99%, 13.79%, 10.34% 10.34% and 8.62% respectively. Specific protein strips were observed in 48.28% of the UC patients with Western blot technique, 47,000 polypeptide being the most common and making up 22.41% of them. CONCLUSION: The detection of ANCA was an auxiliary method for the diagnosis of UC. At present, UC-associated target antigens were not yet clarified, 47,000 polypeptide may be one of them.
