[The changes in quantities of elements around cementum-dentin junction in human natural tooth--electron probe microanalysis].

To study the changes of inorganic and organic substance around cementum-dentin junction in the tissues of human natural tooth, the method of electron probe microanalysis was used in this research project. Determinations of weight percentage (wt%) were made for Ca, P and S elements around cementum-dentin junction. The results showed that the changes in quantities of Ca, P and S elements around cementum-dentin junction were not significant(P > 0.1). These suggest that the quantities of inorganic and organic substance have not obviously varied with the anatomic structure of natural tooth.

[Construction and identification of recombinant fowlpox virus for expressing haemagglutinin-neuraminidase glycoprotein of Newcastle disease virus strain F48E8].

For the construction of transfer vector pFGHN1175-1, the gene encoding haemagglutinin-neuraminidase(HN) glycoprotein of newcastle disease virus(NDV) strain F48E8 was removed from plasmid pGEMHN, and inserted into the HindIII site of insertion vector pFG1175-1, downstream of P7.5 promotor. Chicken embryo fibroblast (CEF) cell cultures which had been infected with fowlpox virus (FPV) Chinese vaccine strain 282E4 for 3-4 hours were transfected with pFGHN1175-1 plasmid DNA by liposomal transfection. Recombinant FPV with blue plaques were selected and purified 3 times in CEF cell cultures overlaid agarose containing X-gal. PCR analysis and DNA dot-blotting hybridization assay indicated that the HN gene had inserted into the FPV genomic DNA. The expression of the NDV HN gene in recombinant FPV was confirmed by indirect immunofluorescence assay with specific monoclonal antibody.

[A quantitative measurement of retinal nerve fiber layer thickness by optical coherence tomography in normal Chinese people].

PURPOSE: To measure retinal nerve fiber layer(RNFL) thickness in normal Chinese people for the use of early diagnosis in glaucomatous patients. METHODS: Optical Coherence Tomography(OCT) was used in 111 eyes of 77 Subjects to create cylindrical cross sections of the RNFL around the optic nerve head with scan diameter of 3.46 mm. The differences in RNFL thickness were analyzed with age, sex, either eye. RESULTS: We found a decrease in RNFL thickness with aging especially in the superior quadrant(P < 0.05), but there weren't significant difference between right and left eyes, neither the sex(P > 0.05). The Mean RNFL thickness of different ages(mean +/- s) was as following: the superior quadrant 128.00 +/- 10.31-149.00 +/- 13.17, the inferior quadrant 132.95 +/- 9.54-142.33 +/- 10.60, the overall mean RNFL thickness 109.00 +/- 4.72-114.28 +/- 7.98 microns. CONCLUSION: The preliminary study showed that neither sex nor either eye were associated with significant differences in RNFL thickness. It is necessary to define the normative value by age group on a larger scale.

Hemodynamics of ophthalmic artery and central retinal artery and correlation with other factors in patients with primary open angle glaucoma.

OBJECTIVE: To investigate the hemodynamics of ophthalmic artery (OA) and central retinal artery (CRA) in patients with primary open angle glaucoma(POAG) and analyse the correlation between hemodynamics of OA and CRA and other factors. METHODS: The hemodynamics of OA and CRA in normal persons (102 eyes) and glaucomatous patients (102 eyes) were measured by Colour Doppler Image (CDI). The linear correlation analysis was made between OA and CRA about each of the following parameters: the peak systolic flow velocity (Vmax), the end diastolic velocity (Vmin). The multiple stepwise regression analysis was taken to investigate the correlation between each of the following hemodynamics of OA and CRA: Vmax and Vmin in 74 eyes with POAG and each of the following related factors in hemorrheology: whole blood apparent viscosity at low, medium and high shear rate, plasma viscosity and hematocrit. The multiple linear regression was applied to investigate the correlation between each of the following hemodynamics of OA and CRA: Vmax and Vmin in 69 eyes with POAG and each of the following blood vessel filling time: the arm-retinal artery (A-AT) and retinal artery-venous (A-VT) of the fundus fluorescein angiography(FFA). RESULTS: The Vmax, Vmin and time-averaged maximum velocity (Vmean) of OA and CRA in patients with POAG were lower than those in normal persons, but the resistance index (RI) of OA and CRA was higher than those in normal. There was a positive correlation between OA and CRA in Vmin, but a negative correlation between Vmin of OA and plasma viscosity. The whole blood apparent viscosity at low shear rate was clodely related to Vmax and Vmin of CRA. Also there was a negative correlation between Vmax of CRA and A-AT of the FFA. CONCLUSION: There was abnormal hemodynamics, which is influenced by blood viscosity, of OA and CRA in patients with POAG. Abnormal hemodynamics can affect the blood supply to optic disc and retina in POAG.

Electroretinographic oscillatory potentials in different aged rats: an analysis in the domains of time and frequency.

OBJECTIVES: Electroretinographic Oscillatory Potentials (Ops) are considered to be the optimal parameter to study the retinal circulation disturbances the present we compared parameters of OPs between different aged rats under dark adaptation. METHODS: The scotopic ERG were performed in anaesthetized rats aged 3, 12, 24 months. According a window of collective power spectrum field of the OPs was isolated by using the FFT and IFFT of the spectrum analysis program. The parameters in the time and frequency domains were calculated. RESULTS: Results showed that the amplitudes of wavelets and the summed amplitudes of OPs declined with the increase of age. A reduction of the total power and dominated power of OPs was also found after the FFT. But implicit time of each OP and dominant frequency did not change obviously. CONCLUSIONS: The present study indicated that change of OPs of rat might be related to senile degeneration of the retina and circulative deficiency in retina in elder rats.

[Spectroscopic characteristics of Ce3+ doped Ca3Al2M3O12 (M=Si, Ge) garnets].

The spectral properties of Ce3+ doped Ca3Al2M3O12 (M = Si, Ge) garnets have been reported under excitation of UV light. The emission spectra of Ce3+ ion in garnets consist of a broad band assingning to the 5d-4f transition of Ce3+ ion. The emission intensity Ce3+ ion in Ga3Al2Ce3O12 shows very weak, but in Ga3Al2SiO12 shows strong. The emission intensity of Ce3+ ion is greatly enhanced by adding Gd3+ or flux into Ca3Al2Si3O12: Ce garnet. At 295 and 77K, the fluorescence decay of Ce3+ ions in Ca3Al2Si3O12 garnet have been measured and the fluorescence lifetimes of Ce3+ ions are 46 and 49ns, respectively.

[Spectral properties and energy transfer of Ce3+ and Eu2+ ions in Ca8Mg(SiO4)4Cl2].

Ce3+ and Eu2+ codoped calcium magnesium chlorosilicate Ca8Mg(SiO4)4Cl2 (CMSC) was synthesized for the first time. The diffuse reflection spectra, excitation and emission spectra of CMSC : Ce3+, Eu2+ were measured and studied. Due to the good overlap between the Ce3+ emission bands and Eu2+ excitation bands, the energy transfer between Ce3+ and Eu2+ ions is expected to be very efficient and is confirmed by the emission spectra of CMSC : Ce3+, Eu2+ system. The fluoresence lifetime of Ce3+ in CMSC : 0.006Ce3+ and CMSC : 0.006Ce3+, 0.03Eu2+ was determined to be 30.1 and 17.2ns,respectively. By the calculation, the probability of energy transfer from Ce3+ to Eu2+ (PSA) is 2.49 x 10(7)/s and energy transfer efficiency (eta(SA)) is 42.9%.

Human vaginal mucosal immune system: characterization and function.

OBJECTIVE: The mucosal immune system has been recognized as the first line of defense against foreign antigens. The limited information available on the mucosal immunity of the lower reproductive organs have restricted our ability to fight infections, especially, the sexually transmitted disease. The aim of this study was to characterize in-vitro the human vaginal lamina propria lymphocytes (VLPL), their cell surface phenotypes, and cellular function. METHODS: VLPL were isolated from human vaginal mucosa by enzymatic techniques. Cell surface characteristics were investigated by immunohistochemistry and flow cytometric analysis. Cellular immune function was evaluated by 3H-thymidine incorporation studies and ornithine decarboxylase (ODC) activity. RESULTS: Immunohistochemistry and flow cytometric analysis showed that the CD4/CD8 ratio of the human vaginal mucosa is reversed compared to the gut lamina propria lymphocytes (0.55 +/- 0.17). PHA and ConA mitogens enhanced VLPL thymidine incorporation, while PWM did not have any significant effect. Very high level of ODC activity was observed in VLPL after PHA stimulation. CONCLUSIONS: The human VLPL can be isolated, characterized, and respond to a mitogenic stimulus. We postulate that further analysis of the vaginal immune system will enhance our understanding of local defence mechanisms which will help in the development of new therapeutic modalities against vaginal infections.

Expression of apoptosis regulatory genes by retinal pericytes after rapid glucose reduction.

PURPOSE: Retinal capillary pericytes underwent apoptosis in vitro after they had been exposed long-term to high levels of glucose followed by a rapid glucose reduction. The present work was designed to study the expression of bcl-2 family members and apoptosis regulatory genes and to determine the status of oxidative stress induced by high concentrations of glucose in this in vitro apoptosis model. METHODS: Pericytes were grown in normal or high glucose concentrations (5, 20, 30, and 40 mM) for 10 days and then exposed to a rapid reduction of glucose to 0.5 mM or 5 mM. Pericyte cell death was evaluated by determining the loss of cell viability and the fragmentation of DNA using agarose gel electrophoresis. In parallel, the quantitative reverse transcription-polymerase chain reaction technique was used to determine the expression of bcl-2, bax, p53, and glutathione peroxidase (GSH-Px) genes. The intracellular level of glutathione (GSH) and the DNA fragmentation were determined simultaneously for pericytes treated with or without exogenous GSH monoethylester. Retinal capillary endothelial cells, experiencing the same glucose variation, were studied as a comparison. RESULTS: For pericytes, downregulation of bcl-2 was observed as early as 24 hours after rapid glucose reduction, whereas DNA fragmentation was not detectable at that time. After 72 hours, a decreased protein ratio of Bcl-2 to Bax was concomitant to evident loss of pericyte viability. During the period of high glucose and the following glucose reduction, p53 expression essentially was unchanged. Decreased levels of GSH induced by high concentrations of glucose (>30 mM) became further depleted when the glucose levels were rapidly reduced. Addition of GSH monoethylester to the medium restored the level of GSH in pericytes and prevented pericyte apoptosis induced by glucose variation. Moreover, the mRNA levels of GSH-Px were significantly elevated. By contrast, with the same glucose reduction endothelial cells did not undergo apoptosis. Their mRNA levels of bcl-2, bax, and GSH-Px essentially were unchanged. CONCLUSIONS: High levels of glucose and rapid reduction of glucose modulate the expression of bcl-2 family genes in retinal pericytes. Upregulation of GSH-Px and depletion of GSH indicate a reparative process of accelerated elimination of reactive oxygen species following rapid glucose reduction. These findings indicate that the aggravated oxidative stress and the weakened antioxidant defense induced by the combined effects of high levels of glucose and subsequent rapid glucose reduction cause pericyte apoptosis. Prevention of DNA fragmentation of pericytes by exogenous GSH further supports this notion. Because endothelial cells did not show similar pathologic changes, this proposed mechanism seems to be specific to pericytes.

Educational attainment and transitions in functional status among older Taiwanese.

Despite considerable research examining the influence of socioeconomic status on health, few studies have considered this relationship as it pertains to older adults in non-Western societies. We attempt to ascertain the influence of education on changes in physical functioning in a rapidly developing country. Data come from the 1989 Survey of Health and Living Status of the Elderly in Taiwan and a follow-up interview in 1993 (N = 4,049, age = 60+). Individuals are conceptualized to be in a state of functional independence or functional limitation at the time of origin, based on their ability to perform three physical functioning tasks. The outcome at the follow-up interview is categorized as functionally independent, limited, or dead, allowing for six probabilities, one from each state of origin to each outcome. These are calculated using a multinomial logit model, controlling for other factors often thought to be associated with health transitions. High levels of educational attainment result in a decreased incidence of functional limitation for those originating in a state of independence. Contrary to expectations, however, education has little influence on those who originate functionally limited. Thus, higher education plays a substantial role in primary prevention of morbidity, delaying the onset of disability, but other factors are more important once limitations begin. We speculate on the reasons behind these findings, including that the results may be culturally dependent.
